      • 水深變化 및 反射率을 考廬한 港內波高分布에 관한 硏究

        A numerical model is presented for the prediction of wave height distribution in a harbor due to the linear wave theory. The method is based upon the solution of a time-dependent analysis including wave refraction, diffraction, and arbitrary reflection coefficient by the finite difference approach. The present numerical model has been validated in several stages. The result obtained from wave shoaling effect has been compared with hydraulic experimental result presented by Ito and Tanimoto. The portion of the diffracted wave heights in a harbor has also been compared with the experimental works proposed by Nisimura et. al.. Good agreement was obtained between the present model and previously obtained results. Therefore it is concluded that the model can be applied to predict the wave height distribution in a harbor with varying depth and arbitrary reflection coefficients. Finally, a numerical example of this application for the Hwa-Sun harbor is shown.

        The numerical model is presented for the prediction of wave fields in a harbor with partially reflecting boundaries. The water depth is assumed to be constant, and the solution was obtained by the boundary element approach. The present numerical study is based on the linear diffraction theory with regular waves. The results of the present model have been compared with the results of previous physical modeling. Good agreement was obtained in all cases. Based on these comparisons, the present numerical model is and accurate and efficient tool to predict the wave fields in a harbor with partially reflection boundaries in constant water depth.

        열공형과 비열공형 피질하 혈관성 치매에서 위험인자의 차이에 관한 비교 연구

        Backgrounds and Objectives: Vascular dementia is a group of dementing disoders arising from various stroke syndrome. Among these. subcortical ischemic vascular dementia (SIVD) is regarded as a relatively distinct clinical entity. However, MRI patterns of SIVD are not homogenous. In some patients, lacunes are dominant, and in others, subcortical white matter changes are. This study was designed to compare risk factor profiles between SIVD with and without multiple lacunes. Methods: We divided 47 subjects (22 males, mean age. 68 years) recruited from VADAPET (Multicenter Trial For Evaluation Of The Changes In the PET Images Of Subcortical Vascular Dementia Patient) study into two groups one with more than 5 lacunes in deep gray matter (lacune group) and the other with 5 or less(non-lacune group) Clinical characteristics and laboratory findings of two groups were compared. Results: Nineteen of 47 patients (40%) belonged to the lacune group. The lacune and non-lacune groups d d not differ in the following variables: age, hypertension, diabetes mellitus, hyperlipidemia heart disease, history of stroke or TIA, history of trauma or major surgery, family history of hypertension stroke, or dementia, age at diagnosis of dementia, body mass index, white blood cell count, ESR, CRP, fibrinogen, hemoglobin A1C, total cholesterol. LDL cholesterol creatinine, proteinuria, glucosuria, and microhematuria. However, male sex, smoking alcohol. hemoglobin, and HDL cholesterol were possibly associated more with lacune group SIVD than with non-lacune group (p<0 1) Multivariate analyses revealed that smoking, hemoglobin, and HDL cholesterol were independent predictors of SIVD with multiple lacunes Conclusion: Our study suggests that SIVD with multiple lacunes may be significantly different in smoking habits hemoglobin, and HDL cholesterol from SIVD without multiple lacunes.

      • Characterization of the SCF ubiquitin ligase 'proteome'

        SCF ubiquitin ligases are comprised of Skp1, Cdc53(human Cull or Cullin), Hrt1(human Roc1 or Rbx1) and one member of a large family of substrate receptors(F-box proteins, or FBPs). To investigate how the subunits of SCF are integrate into protein networks in vivo, we employed sequential rounds of epitope-tagging, affinity purification, and mass spectrometry. Sixteen Skp1- and Cdc53-associated proteins were identified in budding yeast, including all components of SCF, nine FBPs, Yjr033(Rav1), and Ydr202(Rav2). Reiterative purification revealed that Rav1 and Rav2 form a novel protein complex with Skp1 named RAVE(Regulator of (H+)-ATPase of the Vacuolar and Endosomal membranes), which associates with the V1 domain of the vacuolar membrane (H+)-ATPase(V-ATPase). V-ATPase are conserved throughout eukaryotes, and have been implicated in tumor metastasis and multidrug resistance. Genetic and mechanistic dissection revealed that RAVE promotes the glucose-triggered assembly of V-ATPase holoenzyme. Systematic genome-wide two-hybrid screen yield 17 Skp1- and Cdc53-interactors, only 3 of which overlap with those that we found. Thus, reiterative rounds of purification/mass spectrometry provide a distinctive view of the web of interactions that link proteins into a comprehensive cellular network. To fully exploit the potential power of mass spectrometry for deciphering networks of interacting proteins, we have developed an epitope tag system('HTM' for His8-TEV2-Myc9) that enables a C-terminally tagged protein to be purified in consecutive affinity steps. The target protein is bound to anti-myc affinity resin, eluted with Tobacco Etch Virus protease(which cleaves at the TEV recognition sites in the tag), and recaptured on NTA resin. Using Skp1 as a test-bed, we have shown(in collaboration with John Yates at Scripps) that the MudPIT(multidimensional protein identification technology) approach developed in the Yates laboratory can be used to rapidly identify multiple interacting partners of Skp1 in a single LC/MS run. We tagged 25 proteins, which involved in cell cycle or transcription regulation, with the HTM epitope, and used MudPIT to identify the interacting partners. Iterative application of this approach on a large scale has the potential to reveal the web of protein interactions that underlie cell function and regulation.

      • Macrobenthic Communities on the Tidal Flats around Yongjong and Yongyu Islands, Inchon, Korea

        Four tidal flats, covering two dissipative type beaches and two other tidal mud flats, around Yonaiong-Yongyu Islands, Inchon, Korea were studied in May 1991 to analyze the spatial distribution patterns of benthic communities on macrotidal flats. From the statistical test on spatial patterns of species number, density, and biomass, significant trends were found in species number and biomass. Although quantitative analysis was not performed, the interpretation showed that the variations were ruled out by environmental gradients such as sediment grain size and tidal elevation. The eight communities revealed in this study are as follows: Moerella-Mactra(Group 1), llyoplax-Glycera -Magelona japonica-Magelona sp.-Pe riserrula ( Group 2), Amphiura-Nephtys californiensis-Bullacta-Eohaustorius (Group 3), Leonnates-Heteromastus-Pretankyra-Nephtys polybranchia (Group4), Nephtys chemulpoensis-Macrophthalmus (Group 5), Ceratonereis-Scopimera (Group 6), Haustorioides-Urothoe (Group 7) and Cycladicama-Armandia-Minuspio community (Group 8). Some of these communities were classified into substratum-specific (Group 1 to 3) and tidal elevation-specific communities (Group 4). It was difficult to identify the predominant governing agent in Groups 5,6,7 and 8 because of their presence at extreme types of sediment and at uppermost elevations. Mixed effects seemed to act upon the latter four communities (Group 5 to 8). Based on the number of samples, 60% of the target samples were influenced by substratum properties, 20% by tidal elevation effect and 20% by mixed effect of the two factors. From this, it was concluded that substratum properties serve as the most important factors on soft bottom inhabitants in the area studied.

        산삼 배양액 급여가 육계의 육질에 미치는 영향 연구

        본 연구에서는 산삼의 조직 배양 후 폐기되는 산삼 배양액을 육계에 음수를 통하여 다양한 농도로 급여한 후 계육의 일반 성분, 물리화학적인 성상 및 이화학적 성상 변화를 분석하여 산삼 배양액의 효능과 첨가제로서의 가능성을 규명하였다. 산삼 배양액을 급여한 육계 가슴육의 수분 함량, 조단백질과 조지방의 함량에 유의적인 차이는 보이지 않았지만 조회분의 함량은 산삼 배양액 32 mL/L 급여군이 다른 처리군에 비하여 유의적으로 증가하는 것을 볼 수 있었다. 또한, 산삼 배양액 급여군에서 육즙 손실, 조리 감량, 전단력과 저장성은 감소되는 경향을 보이나 유의적인 차이는 보이지 않았지만 산삼 배양액 급여군의 가슴육 pH는 유의성 있게 감소하였다. 이러한 결과는 산삼 배양액의 급여에 의해 육제품에서 미생물의 번식을 억제하고 보존이나 내구성을 강화시키거나, 육계의 골격 형성과 육질 내 삼투압이나 항상성 유지에 도움을 줄 것으로 사료된다. TCM-KWG (tissue culture medium waste after harvest of Korean wild ginseng) (panax ginseng) is left over of tissue culture medium used to grow Korean wild ginseng (KWG). The present study was conducted to investigate the effect of TCM-KWG on meat quality and the possible of application as additives in broiler chickens. A day old broiler chickens randomized in 6 groups (n=60/groups) were administered orally with 2, 4, 8, 16, 32 mL/L TCM-KWG through drinking water with one untreated control group. After administration for five weeks, we analyzed chemical composition and meat quality. Crude ash increased approximately 20% in TCM-KWG treatment groups as compared to control group whereas the concentration of moisture, crude protein and crude fat did not show any significant difference. Crude ash is essential to enhance skeleton formation and physiological function. TCM-KWG treatment gradually decreased the pH value of breast meat whereas it did not change the purge loss and cooking loss. The pH value of meat is important for preservation of meat for longer periods and high durability strength. These results suggest that TCM-KWG treatment may improve the quality of meat and can be apply as food additives in chickens.

        Extrapyramidal Signs and Cognitive Subdomains in Alzheimer Disease

        <P>Objective: Extrapyramidal signs (EPS), commonly observed in Alzheimer disease (AD), predict cognitive impairment and functional decline. This study investigated the association between EPS and five cognitive subdomains in a large number of participants with AD. Design: Cross-sectional analyses of the nationwide Clinical Research of Dementia of South Korea (CREDOS) study, 2005-2012. Setting: Multicenter clinical settings. Participants: 1,737 participants with AD drawn from the CREDOS study. Measurements: The EPS group was defined by the presence of at least one EPS based on neurologic examination. We assessed five cognitive subdomains: attention, language, visuospatial function, memory, and frontal/executive function using the Seoul Neuropsychological Screening Battery-Dementia version. The associations of EPS with each cognitive subdomain were analyzed with a multiple linear regression model after controlling for confounding factors: sex, age, years of education, severity of dementia (Clinical Dementia Rating Sum of Boxes), and white matter hyperintensities. Results: 164 AD participants (9.4%) had EPS. AD participants with EPS showed lower performance compared with those without EPS in two cognitive subdomains: attention and visuospatial function. The language, memory, and frontal/executive subdomains did not differ between the EPS-positive and the EPS-negative groups. In addition, we found a significant moderating relationship between EPS and deep white matter hyperintensities on visuospatial function score. Conclusions: EPS in AD are associated with severe cognitive impairment in attention and visuospatial function. Careful screening for EPS in patients with AD may assist in prediction of cognitive profile.</P>
