        Early Phase of UVB - induced GM - CSF Upregulation in Epithelial Cell Line is not Totally Dependent on IL - 1α

        Backgrounds : It was demonstrated that ultraviolet(UV) B light induces the release of IL-la in cultured human epithelial cell line and augmentation of GM-CSF production by UVB is reported to be mediated by IL-1α in the murine keratinocyte cell line Pam 212. Objective : The purpose of this study was to evaluate the effects of UVB on kinetic profile of IL-1 and GM-CSF mRNA expression and to see whether synthesis of GM-CSF by UVB can be completely inhibited by blocking IL-1α mediated pathway. Method : We used a competitive RT-PCR for measuring cytokine gene expression in epithelial cell line after UV radiation. Results : The IL-1α mRNA increased as early as 1h after UV irradiation, and then decreased at 3h after the irradiation. Thereafter, the response of IL-1α mRNA was upregulated with a second peak at 6h after the UV irradiation. However, mRNA for GM-CSF increased at 1h after UV light exposure and anti-IL-1α antibodies could only partially inhibit UV-augmented GMCSF production. Conclusion : UVB induced GM-CSF production seemed to be mainly mediated by UVB induced IL-1α but these results suggest that UVB may also induce GM-CSF production through an IL-1α independent pathway.

        Pollen metabarcoding reveals the floral resources and niche overlap of honeybees in forest areas

        We investigated the floral resources of the honeybee (Apis mellifera)bypollenDNA metabarcoding as the first step in honeybee conservation. We attempted to identify as many floral resources and as wide resource availability as possible in areas and compared the richness of floral resources between forests and national parks. We found that both forest and national parks can sufficiently support honeybee floral resources; however, national parks were more stable habitats with more complex vegetation structure than those of forests. Also, honeybee’s preferences for floral resources were determined using niche breadth, niche overlap, and network analysis among the floral resources in forest areas. Quercus and Actinidia were the most frequently detected taxa of pollen resources in both forests and national parks by pollen metabarcoding. Even though we anticipated that more various pollen resource taxa would be found in national parks than forests, no differences in the number pollen resource taxa were detected between them. According to the niche overlap analysis result, the honeybee preferred the pollen resources in both forests and national parks, and the co-occurrence pattern analysis of pollen resource taxa in both forests and national parks showed a simple network. Altogether, our findings suggested that DNA metabarcoding of pollen collected by the honeybee can provide useful information on the flowering plants in a given area. As the use of floral pollen resources varies spatially and temporally, periodic monitoring of pollen resources can identify which pollen is required to maintain the health of honeybee colonies.

        To evaluate antibiotic-induced endotoxin release(AIER) and its correlation with some cytokines, we measured endotoxin level and tumor necrosis factor alpha(TNF-α) and interleukin6(IL-6) production in mononuclear cells in vitro after exposure of Pseudomonas aeruginosa to antibiotics belonging to different class with two extreme concentrations. The tested concetration of antibiotics were set up according to peak serum level. The low concetration of ceftazidirne and low concentration of imiperiem increased AIER, but high concentration of ceftazideme, high concentration of ciprofloxacin, high concentration of cefoperazone/sulbactam, high concentration of amikacin, and high concentration of meropenem reduced AIER.Interestingly, combined treatment of these antibiotics markedly reduced AIER, But the major cyotkines, TNF-α and IL-6 were not affect by type and concettration of antibiotics, combined treatment of antibiotics, and level of endotoxin released by antiboitics. In this study, we observed AIER was different according to type of antibiotics, concentration of antibiotics, and combination of antibiotics, But AIER had poor correlation with TNF-α and IL-6 in Pseudomonas aeruginosa. It suggests that cytokine release is not solely dependent to endotoxin, but more complex cascade is needed. More invesfigations, such as endotoxin induced cytokine mRNA expression, relationship with penicillin-binding proteins and endotoxin-neutralizing effect of antibiotic itself, must be performed.

        Original Articles : Anti-inflammatory Effects of Ethanolic Extracts from Codium fragile on LPS-Stimulated RAW 264.7 Macrophages via Nuclear Factor kappaB Inactivation

        Bacterial lipopolysaccharide (LPS) induces expression of pro-inflammatory cytokines and enzymes producing nitric oxide (NO) and prostaglandins (PGs) in immune cells. This process is mediated by the activation of nuclear factor kappaB (NF-κB). In this study, we investigated the anti-inflammatory characteristics of Codium fragile ethanolic extract (CFE) mediated by the regulation of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) using LPS-stimulated murine macrophage RAW 264.7 cells. CFE significantly inhibited LPS-induced NO and PGE2 production in a dose-dependent manner and suppressed the expression of iNOS and COX-2 proteins in LPS-stimulated RAW 264.7 cells with no cytotoxicity. Pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-6, and tumor necrosis factor-α, were significantly reduced by treatment of CFE in LPS-stimulated RAW 264.7 cells. CFE inhibited the promoter activity of (NF)-κB in LPS-stimulated macrophages. Treatment with CFE suppressed translocation of the NF-κB p65 subunit by preventing proteolytic degradation of inhibitor of κB-α. These results indicate that the CFE-mediated inhibition of NO and PGE2 production in LPS-stimulated RAW 264.7 cells is mediated through the NF-κB-dependent transcriptional downregulation of iNOS and COX-2, suggesting the potential of CFE as a nutraceutical with anti-inflammatory activity.

        Potential of Lilium lancifolium in Alleviating Pain and Inflammation in a Rat Model of Monosodium Iodoacetate-Induced Osteoarthritis

        Lilium lancifolium (LL) is widely cultivated in East Asia and used to attenuate airway diseases. Our current study aimed to investigate the therapeutic effect of LL on pain level and inflammatory response in a rat model of monosodium iodoacetate (MIA)-induced osteoarthritis (OA). We first examined the effect of LL on inflammatory cytokines and inflammatory mediators in IL-1β-treated HTB-94 cells. The LL extract was found to significantly inhibit the levels of Interleukin-6 (IL-6), Interleukin-8 (IL-8), and prostaglandin-E2 (PGE-2) in Interleukin-1 β (IL-1β)-stimulated HTB-94 cells in a dose-dependent manner. Moreover, chronic oral administration of LL effectively restored the weight-bearing distribution in the rat model of MIA-induced OA. In addition, administration of LL inhibited inflammatory cytokines and inflammatory mediators, such as C-reactive protein (CRP), IL-6, leukotriene B4 (LTB-4), PGE-2, and matrix metalloproteinase-9 (MMP-9). Our findings collectively suggested LL as one of the potential therapeutic agents for OA, owing to its properties of reducing pain and inflammatory responses.

        Effects of <i>Ecklonia cava</i> ethanolic extracts on airway hyperresponsiveness and inflammation in a murine asthma model: Role of suppressor of cytokine signaling

        <P><B>Abstract</B></P><P><I>Ecklonia cava</I> (EC) is a brown alga that evidences radical scavenging activity, bactericidal activity, tyrosinase inhibitory activity, and protease inhibitory activity. However, its anti-allergic effects remain poorly understood. In the current study, we attempted to determine whether pretreatment with EC induces a significant inhibition of asthmatic reactions in a mouse asthma model. Mice sensitized and challenged with ovalbumin (OVA) evidenced typical asthmatic reactions, as follows: an increase in the number of eosinophils in bronchoalveolar lavage fluid; a marked influx of inflammatory cells into the lung around blood vessels and airways, and airway luminal narrowing; the development of airway hyperresponsiveness; the detection of tumor necrosis factor-alpha (TNF-α) and Th2 cytokines, including IL-4 and IL-5 in the bronchoalveolar lavage (BAL) fluid; and the detection of allergen-specific immunoglobulin E (IgE) in the serum. However, the administration of EC extract prior to the final airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. We also demonstrated that EC extracts treatment resulted in significant reductions on matrix metalloproteinase-9 (MMP-9) and Suppressor of cytokine signaling-3 (SOCS-3) expression and a reduction in the increased eosinophil peroxidase (EPO) activity. The treatment of animals with EC extracts resulted in a significant reduction in the concentrations of the Th2 cytokine (IL-4 and IL-5) in the airways, without any concomitant increase in the concentration of Th1 cytokines. These findings indicate that EC extracts may prove useful as an adjuvant therapy for allergic airway reactions via the inhibition of the Th2 response. Accordingly, this study may provide evidence that EC extract performs a critical function in the amelioration of the pathogenetic process of asthma in mice.</P>

        레토르트 파우치 추어탕과 우렁 된장국의 세균상 조사

        레토르트 파우치 추어탕과 우렁 된장군의 세균상을 조사한 결과 레토르트 살균 전 추어탕과 우렁 된장군 제품에 대한 각 배지별 생균수는 추어탕은 TSA 3.80×10 exp (6), Endo agar 2.27×10 exp (2), SS agar 5.20×10 exp (2)이었고, 우렁 된장국은 TSA 4.03×10 exp (6), Endo agar 8.91×10 exp (2), SS agar 2.57×10 exp (2)이었으며, SDA와 TCBS 배지에서는 두 제품 모두 어떤 균도 검출되지 않았다. 비 살균 추어탕과 우렁 된장군 제품에 대한 각 배지별 분리균을 동정한 결과, 총 16종이 분리되었으며 추어탕의 경우 TSA 배지에서는 B. pantothenticus가 우점종으로 나타났고 다음은 S. dysenteriae, C. sporogenes 순이었으며, C. perfringens, E. ictaluri, E. gergoviae, E. coli, X. nematophilus, K. subsp. rhinoscleromatis 등도 일부 분리되었다. 우렁된장국에서는 B. marinus가 우점종이었고 다음은 S. aureus, S. saccharolytics 순이었으며 P. tetradicus, S. adorifera, E. ictaluri, E. gergoviae, E. coli 등도 분리되었다. Endo agar에서는 두 제품 모두 비슷한 경향으로 장내세균인 E. gergoviae와 E. coli가 동정되었고 추어탕 일부에서 K. subsp. rhinoscleromatis 등도 분리되었다. SS 배지에서는 추어탕에서 S. ficaria와 P. prevotii가, 우렁된장국에서는 S. ficaria와 P. tetradius가 각각 분리되었다. 시판중 부패된 레토르트 파우치 추어탕과 우렁 된장국에서 분리된 균을 동정한 결과, 총 13종이 분리되었으며 추어탕에서는 B. pantothenticus, S. dysenteriae, C. sporogenes, P. prevotii, E. gergoviae, S. ficaria, K. subsp. rhinoscleromatis균이, 우렁 된장국에서는 B. marinus, S. aureus, S. saccharolytics, P. tetradius, E. ictaluri, S. ficaria, S. adorifera 균이 각각 분리·동정되었다. The present study is concerned with the bacteriological examination of retort pouched loach soup and soybean paste soup containing mud snail. It was found that before sterilization, viable cell counts (c.f. u/ml) in their media were for the loach soup 3.80×10 exp (6) on TSA, 2.27×10 exp (2) on Endo agar, 5.20×10 exp (2) on SS agar and for the soybean paste soup, 4.03×10 exp (6) on TSA, 8.91×10 exp (2) on Endo agar, 2.57×10 exp (2) on SS agar. With the culture media, SDA and TCBS, no microorgarmsms were isolated from the both soups. Sixteen species of microorgarnisms were identified for the unsterilized soups. In the loach soup on TSA, B. pantothenticus was the dominant species, followed by S. dysenteriae, C. sporogenes and some others were also identified, such as C. perfringens, E. ictaluri, E. gergoviae, E. coli, X. nematophilus, K. subsp, rhinoscleromatis. In the soybean soup on TSA, B. marinus was the dominant species, followed by S. aureus, s. saccharolytics and P. tetradius, S. adorifera, E. ictaluri, E. gergoviae, E. coli were also identified. On Endo agar, the two soups showed a similar bacteriological pattern, in which entrobacterium such as E. gergoviae and E. coli were identified. Particularly K. subsp. rhinoscleromatis for the loach soup. On SS agar, S. ficaria and P. prevotii in the loach soup, S. ficaria and P. tetradius in the soybean paste soup were indentigied respectively. Bacteriological examination was also carried out for the spoiled retort pouched soup in the market, in which thirteen microorgarnisms were isolated and its pattern almost similar to that before sterilization. They were B. pantothenticus, S. dysenteriae, C. sporogenes, P. gergoviae, E. ictaluri, S. ficaria, K. subsp. rhinoscleromatis in the loach soup and B. marinus, S. aureus, S. saccharolytics, P. tetradius, E. ictaluri, S. ficaria, S. adorifera in the soybean soup.

        Maximal voluntary muscular Peak Torque, Deficit, %Flexor/Extensor, Total Work, Endurance Ratio of the quadriceps and hamstrings during isokinetic exercise was studied in order to obtain the isokinetic normative data of the left & right knee joint. The results obtained were as follows : 1. Peak torque of left knee joint flexor were shown significantly(p<.028), while I Group had increased trend in a few. Right knee joint flexor were shown significantly(p<.000), while I Group had increased trend in a few. And Peak torque of left knee joint extensor were not shown significantly(p<.176), while I Group had increased trend in a few. Right knee joint Extensor were not shown significantly(p<.583), while between Group had trend in a equivalent. 2. Deficit ratio of flexor were not shown significantly(p<.165), while ⅡGroup had increased trend in a few. Extensor were not shown significantly(p<.172), while between Group had trend in a equivalent. 3. %Flexor/Extensor of left knee joint were not shown significantly(p<.457), while ⅡGroup had increased trend in a few. And %Flexor/Extensor of right knee joint were not shown significantly(p<.443), while ⅠGroup had increased trend in a few. 4. Total work of left knee joint flexor were shown significantly(p<.017), while I Group had increased trend in a few. Right knee joint flexor were shown significantly(p<.001). while ⅠGroup had increased trend in a few. And Total work of left knee joint extensor were not shown significantly(p<.459), while ⅠGroup had increased trend in a few. Right knee joint extensor were not shown significantly(p<.710) , while between Group had trend in a equivalent. 5. Endurance ratio of left knee joint flexor were shown significantly(p<.040). while ⅠGroup had increased trend in a few. Right knee joint flexor were not shown significantly(p<.636), while ⅡGroup had increased trend in a few.
