Glioblastoma multiforme is the most lethal malignant brain tumor. Despite many intensive studies, the prognosis of glioblastoma multiforme is currently very poor, with a median overall survival duration of 14 months and 2-year survival rates of less than 10%. Although viral infections have been emphasized as potential cofactors, their influences on pathways that support glioblastoma progression are not known. Some previous studies indicated that human Kaposi’s sarcoma-associated herpesvirus (KSHV) was detected in healthy brains, and its microRNA was also detected in glioblastoma patients’ plasma. However, a direct link between KSHV infection and glioblastoma is currently not known. In this study, we infected glioblastoma cells and glioma stem-like cells (GSCs) with KSHV to establish an in vitro cell model for KSHV-infected glioblastoma cells and glioma stem-like cells in order to identify virologic outcomes that overlap with markers of aggressive disease. Latently KSHV-infected glioblastoma cells and GSCs were successfully established. Additionally, using these cell models, we found that KSHV infection modulates the proliferation of glioma stem-like cells.
Retinal Artery Occlusion and the Risk of Stroke Development : Twelve-Year Nationwide Cohort Study

<P>Background and Purpose-Our aim was to evaluate the risk of subsequent stroke development after retinal artery occlusion (RAO). Methods-National registry data were collected from the Korean National Health Insurance Service, comprised 1 025 340 random subjects. Patients diagnosed with RAO in 2002 and 2003 were excluded. The RAO group was composed of patients with an initial diagnosis of either central or other RAO between January 2004 and December 2013 (n=401). The comparison group was composed of randomly selected patients (5 per RAO patient; n=2003) who were matched to the RAO group according to sociodemographic factors and year of RAO diagnosis. Each sampled patient was tracked until 2013. Cox proportional hazard regression was used. Results-Stroke occurred in 15.0% of the RAO group and in 8.0% of the comparison group (P < 0.001). RAO was associated with an increased risk of stroke occurrence (hazard ratio, 1.78; 95% confidence interval, 1.32-2.41). The magnitude of the RAO effect for stroke was larger among younger adults aged < 65 years (hazard ratio, 3.11) than older adults aged >= 65 years (hazard ratio, 1.26). However, the risk of subsequent stroke was significantly increased in older adults aged >= 65 years at the 4-year follow-up (hazard ratio, 1.58; 95% confidence interval, 1.01-2.48). Conclusions-RAO was significantly associated with subsequent stroke after adjusting for comorbidities and sociodemographic factors. These findings are limited by uncontrolled confounding factors and need to be replicated by other observational studies.</P>
The Structural Design of ITER Tokamak Sub-assembly Tools

ITER (International Thermonuclear Experimental Reactor) tokamak assembly mainly comprises sub-assembly and in-pit assembly. The tools for sub-assembly procedure consist of the tools for upending, sector lifting, vacuum vessel support and bracing, and sector sub-assembly, among which the design of three tools, excluding sector lifting, is described herein. The basic structures for these tools have been developed. The structural stabilities have been studied by using ANSYS code, against the seismic loads of SL-2 level for the two candidate ITER sites, Cadarache in France and Rokkasho in Japan, respectively1. Dynamic analyses using FRS (Floor Response Spectrum) have been performed. The upending and sub-assembly tool turned out safe against the seismic loads, while vacuum vessel support and bracing one gave large toroidal bending of 68 mm and the resultant high stress of 856 MPa at the lower region of vacuum vessel. Design improvement for this tool is on-going.?

The tools for in-pit assembly procedure consist of the tools for Toroidal Field (TF) magnet gravity support assembly, TF magnet inner support, TF coil bracing, and TF coil, vacuum vessel, and thermal shield assembly, among which the design of TF magnet inner support and TF coil bracing tools with sector lifting tool, one of the sub-assembly tool, is described herein. The basic structures for these tools have been developed. The structural stabilities have been studied by using ANSYS code, against the seismic loads of SL-2 level for the two candidate ITER sites, Cadarache in France and Rokkasho in Japan, respectively1. Dynamic analyses using FRS (Floor Response Spectrum) have been performed for the TF magnet inner support and TF coil bracing tools, while an equivalent static analysis was done for the sector lifting tool. The sector lifting tool turned out safe against the seismic loads. The dynamic analyses for the TF support and bracing tool, however, showed a relatively large displacement of 30 mm, which can lead to contact between components. Design improvement for this tool is on-going.

Nogo receptor (NgR) has been shown to inhibit the migration and invasion of human glioma cells. However, little isknown regarding the regulatory mechanisms of NgR in glioblastoma multiforme (GBM). In this study, we propose anovel mechanism that regulates the maturation process of NgR through an interaction with vimentin. The inhibition ofTGFβ1 activity by LY2109761 attenuated the migration/invasion of GBM cells by upregulating cell-surface NgR. Conversely, the treatment of GBM cells with TGFβ1 suppressed NgR maturation. We showed that NgR and vimentininteract, which could be a possible mechanism for the suppression of NgR maturation. The knockdown of vimentinsuppressed the migration/invasion of GBM cells through the increased maturation of NgR. Finally, TCGA (The CancerGenome Atlas) analysis also supported the association of NgR and vimentin. The maturation of NgR is regulated by theinteraction of vimentin and NgR, which attenuates the invasive activity of GBM, and might be a potential therapeutictarget for brain cancer.

Extracellular vesicles (EVs) play a crucial role in cell-to-cell communication. EVs and viruses share several properties related to their structure and the biogenesis machinery in cells. EVs from virus-infected cells play a key role in virus spread and suppression using various loading molecules, such as viral proteins, host proteins, and microRNAs. However, it remains unclear how and why viruses regulate EV production inside host cells. The purpose of this study is to investigate the molecular mechanisms underlying EV production and their roles in Kaposi’s sarcoma-associated herpesvirus (KSHV)-infected cells. Here, we found that KSHV induced EV production in human endothelial cells via Rab- 27b upregulation. The suppression of Rab27b expression in KSHV-infected cells enhanced cell death by increasing autophagic flux and autolysosome formation. Our results indicate that Rab27b regulates EV biogenesis to promote cell survival and persistent viral infection during KSHV infection, thereby providing novel insights into the crucial role of Rab- 27b in the KSHV life cycle.

Purpose: To evaluate the associations between early/late age-related macular degeneration (AMD) and systemic and ocular parameters using national population-based data. Methods: From 2008 to 2012, a total of 16,897 participants of the Korea National Health and Nutrition Examination Survey underwent ophthalmologic examinations by the Korean Ophthalmological Society. Cases of early and late (neovascular and geographic atrophy) AMD were identified using digital fundus images. Risk factors were identified using logistic regression. Results: Age-standardized prevalence of early and late AMD among subjects aged 40 years and older was 5.5% and 0.6%, respectively. Increasing age was the most significant risk factor for both early and late AMD (adjusted odds ratio [aOR]: 1.09, 95% confidence interval [CI]; 1.07–1.10; aOR: 1.14, 95% CI: 1.10−1.19, respectively). Patients who slept, on average, more than nine hours per day (aOR: 0.68, 95% CI: 0.47−1.00) and who were in the lowest quintile (≤ 38.21 g/dL) of high-density lipoprotein (HDL) cholesterol (aOR: 0.68, 95% CI: 0.51−0.92) were significantly associated with a lower prevalence of early AMD. Increasing spherical equivalent (SE) [aOR: 0.53 of the first quintile of SE ≤ -1.38 diopters (D), 95% CI: 0.35−0.78; aOR: 1.41 of the fifth quintile of SE ≥ +0.69 D, 95% CI: 1.07−1.86] was associated with a higher prevalence of early AMD. Lower quartile of systolic blood pressure (≤ 110.0 mmHg) was associated with a lower prevalence of neovascular AMD (aOR: 0.25, 95% CI: 0.09−0.67). Conclusions: Aging was the most significant risk factor for both early and late AMD. Lower level of HDL cholesterol, longer duration of sleep, and myopia had protective associations with early AMD, while lower systolic blood pressure had a protective association with neovascular AMD.

Purpose: To report incidence rates of primary congenital glaucoma in Korea and evaluate comorbidity and mortality from2001 to 2015. Methods: This study is a nationwide and retrospective population-based study. We used claims data from the Korean NationalHealth Insurance Service database between 2001 and 2015. Data for all patients diagnosed with primary congenitalglaucoma were retrieved using the Korean Electronic Data Interchange and Korean Standard Classification of Diseases-6codes. Results: The number of patients with primary congenital glaucoma between 2001 and 2015 was 776, of which 437 weremale (56.31%) and 339 were female (43.69%). The annual prevalence demonstrated a general decreasing trend since 2011,but this was not significant. Over the total survey period, the incidence rate was 11.0 per 100,000 births, with 12.0 casesamong males and 10.0 among females. The incidence according to age was 518 (68.78%) patients at age 0, 112 (13.66%) at 1year, 70 (8.39%) at 2 years, and 76 (9.17%) at 3 years. Of the 776 patients diagnosed with primary congenital glaucoma in thestudy population, 27 died. The observed mortality per 100,000 people is about 10 times higher than that of the general infantand child population under the age of 4 years. Visual impairment was the most common accompanying disability, followed bybrain lesion. Conclusions: Our study’s estimates of the nationwide population-based incidence of primary congenital glaucoma in a Koreanpopulation will expand our understanding of the disease and allow healthcare systems to plan for primary congenitalglaucoma.
