본 연구는 도서지역의 용수공급을 위한 대안으로 제시되고 있는 해수의 담수화 방안, 상수도 이용방안 및 우수의 생활용수 이용방안 중에서 경제적, 지형학적으로 이들 지역에 가장 적합한 대체 수자원으로서 우수이용 시스템을 제시하고 재래적인 우수이용방법을 개선하는 것을 연구의 목적으로 하고 있다. 이를 위하여 제주도지역을 대상지역으로 선정하고 그 대상지역의 실제 강우 및 생활용수자료를 개별 우수이용 시스템과 공동 우수이용 시스템에 대한 시뮬레이션을 통하여 본 시스템을 설계하였고 Bayes-Markov분석을 통하여 대상지역 강우량의 사후확률을 결정하여 운영자의 의사결정 기준으로 제공함으로써 안정적으로 시스템을 운영할 수 있도록 하였다. The purpose of this study is to propose rainwater harvesting system that is the most appropriate with the economical and topographical features among the usage of sea water, reuse sewage, and rainwater. Improving the inefficient domestic usage of rainwater in island is the anther purpose of this study. To accomplish this purposes, Cheju island was chosen as case study and the design for the roof area and tank size of rainwater harvesting system were performed by the result of the simulation of the private rainwater harvesting system and the public rainwater harvesting system with the actual precipitation and domestic water data. The posterior probabilities of rainfall at study area have been carried out by the Bayes-Markov analysis to offer the standard of decision making.

This papaer was undertaken to do morphological analysis of wear particles for sliding members hydrauric rotary acuator. The lubricating wear test was performed under different experimental conditions using the wear test device and wear specimens of the pin on disk type was rubbed in parafinic base oil by three kinds of lubricating materials, varying applied load, sliding distance. The four shape parameters (50% volumetric diameter, aspect, roundness and reflectivity) are used for morphological analysis of wear particles. The results showed that the four shape parameters of wear particles depend on a kind of the lubricating materials. It was capable of presuming wear volume for three kinds of materials on driving time.

The wear particles is released from the moving surfaces in gear systems of transmission and its morphology is directly related to the damage and failure to gear systems from which the particles originated. It is the effective method to diagnosis damage process of gear systems of automobile transmission to observe wear particle in gear oil. We tested with new transmission and took out gear oil according to drive distance. To be applied to damage diagnosis of gear system in transmission of automobile, 4 shape parameters of wear particles in gear oil were calculated and wear volume were presumed with the image processing system.
Roxythromycin-tylosin-furaltadone 합제의 닭 유래 세균에 대한 시험관내 항균효과

In vitro antimicrobial susceptibility of roxythromycin (Rx), tylosin (Tyl), erythromycin (EM) and furaltadone (Fur) was studied, either by the single type or the combinated type of Rx-Tyl-Fur (RTF) against E. co1i (30 strains), Staph. aureus (20 strains) and Mycoplasma gallisepticum (5 strains) that are the major pathogens in poultry farm. The paper disk diffusion and MIC tests were applied. In screening test by paper disk diffusion method, the single types of antibiotics showed the sensitivity against 55 strains of bacteria in order of Fur, EM, Rx and Tyl. The combinated RTF showed the higher rate of sensitivity (80∼83.3% at 64㎍/disk, 60-75% at 32㎍/disk) and wider range of growth inhibiting zone (19.6±3.7-24.8±4.7mm) than those of the single drugs. In MIC test, the sensitivity against the 55 strains appeared in order of Fur. EM, Rx and Tyl as similar to those of paper disk diffusion method. The MIC ranges of RTF were 0.125∼16㎍/ml for E. coli, 0.25∼16㎍/ml for Staph. aureus and 1-8㎍/ml for Myco. gallisepticum, showing evidently lower than those of the single types. The MIC_(50) and MIC_(90) of RTF were estimated l∼4㎍/ml and 8∼16㎍/ml, respectively, and remarkably lower than those of the single drugs.
Mutant p53-Notch1 Signaling Axis Is Involved in Curcumin-Induced Apoptosis of Breast Cancer Cells

Notch1 has been reported to be highly expressed in triple-negative and other subtypes of breast cancer. Mutant p53 (R280K) is overexpressed in MDA-MB-231 triple-negative human breast cancer cells. The present study aimed to determine whether the mutant p53 can be a potent transcriptional activator of the Notch1 in MDA-MB-231 cells, and explore the role of this mutant p53-Notch1 axis in curcumin-induced apoptosis. We found that curcumin treatment resulted in an induction of apoptosis in MDA-MB-231 cells, together with downregulation of Notch1 and its downstream target, Hes1. This reduction in Notch1 expression was determined to be due to the decreased activity of endogenous mutant p53. We confirmed the suppressive effect of curcumin on Notch1 transcription by performing a Notch1 promoter-driven reporter assay and identified a putative p53-binding site in the Notch1 promoter by EMSA and chromatin immunoprecipitation analysis. Overexpression of mutant p53 increased Notch1 promoter activity, whereas knockdown of mutant p53 by small interfering RNA suppressed Notch1 expression, leading to the induction of cellular apoptosis. Moreover, curcumin-induced apoptosis was further enhanced by the knockdown of Notch1 or mutant p53, but it was decreased by the overexpression of active Notch1. Taken together, our results demonstrate, for the first time, that Notch1 is a transcriptional target of mutant p53 in breast cancer cells and suggest that the targeting of mutant p53 and/or Notch1 may be combined with a chemotherapeutic strategy to improve the response of breast cancer cells to curcumin.

Aims: We aimed to evaluate the efficacy and safety of Tenofovir disoproxil fumarate (TDF) based therapy in naive and treatment experienced chronic hepatitis B (CHB) patients for 96 weeks in Korean real life practice. Methods: 209 CHB patients who had prescription for TDF at Seoul and Daejeon St. Mary’s hospital were enrolled from December, 2012 to October, 2014. We compared the virological responses and renal safety of naive and treatment experienced patients Results: Overall complete virological response (CVR) showed 80.4% and 84.6% of patients at week 48 and 96, respectively. In subgroup analysis, CVR at week 96 were 88.4%, 75.0 %, 75.5%, and 83.3%, in the lamivudine-resistant (LAM-R) group, Adefovir-resistant (ADV-R) group, multidrug-resistant (MDR) group, and suboptimal response group, respectively. In a multivariate analysis, ADV-R, MDR, HBV DNA, and HBeAg were independent predictors for CVR. In renal safety, Diabetes mellitus (DM), cirrhosis, and initial low estimated glomerular filtration rate were independent factors affecting Cr elevation (≥ 0.5mg/dL). Moreover, we note that 2 patients with DM and cirrhosis have experienced TDF-related Fanconi syndrome. Conclusions: TDF-based therapy demonstrated sustained viral suppression and a favorable safety throughout 2 years. Compared to the naive group, the LAM-R and suboptimal response group showed comparable efficacy, while the ADV-R group and MDR group both showed a relatively low efficacy. Also, the ADV-R group and MDR group were significantly associated with a low CVR. Close monitoring of renal function may be recommended in CHB patients receiving TDF, especially with DM and cirrhosis

<P>Highly angiogenic bone marrow mononuclear cell-derived spheroids (BM-spheroids), formed by selective proliferation of the CD31(+) CD14(+) CD34(+) monocyte subset via three dimensional (3D) culture, have had robust angiogenetic capacity in rodent syngeneic renal subcapsular islet transplantation. We wondered whether the efficacy of BM-spheroids could be demonstrated in clinically relevant intraportal islet transplantation models without increasing the risk of portal thrombosis. The thrombogenic potential of intraportally infused BM-spheroids was compared with that of mesenchymal stem cells (MSCs) and MSC-derived spheroids (MSC-spheroids). The angiogenic efficacy and persistence in portal sinusoids of BM-spheroids were examined in rodent syngeneic and primate allogeneic intraportal islet transplantation models. In contrast to MSCs and MSC-spheroids, intraportal infusion of BM-spheroids did not evoke portal thrombosis. BM-spheroids had robust angiogenetic capacity in both the rodent and primate intraportal islet transplantation models and improved post-transplant glycemic outcomes. MRI and intravital microscopy findings revealed the persistence of intraportally infused BM-spheroids in portal sinusoids. Intraportal cotransplantation of allogeneic islets with autologous BM-spheroids in nonhuman primates further confirmed the clinical feasibility of this approach. In conclusion, cotransplantation of BM-spheroids enhances intraportal islet transplantation outcome without portal thrombosis in mice and nonhuman primates. Generating BM-spheroids by 3D culture prevented the rapid migration and disappearance of intraportally infused therapeutic cells.</P>
