Activated sludge samples were collected from a municipal sewage treatment plant and used for enrichment of microbial consortia with aniline as the sole carbon and nitrogen source. A bacterium which had excellent aniline degradability was isolated for this work. Biodegradation of industrial organic solvents such as aniline, benzoate, or p-hydroxybenzoate by Delftia sp. JK-2 was examined in the media containing the target substrates in Erlenmeyer flasks, respectively. The JK-2 cells degraded 10 mM aniline, 5 mM benzoate or 5 mM p-hydroxybenzoate completely within 36 hours, 48 hours or 40 hours, respectively. Microscopic examination of the strain revealed Gram-negative and short rod cells. Physiological analysis using BIOLOG GN2 MicroPlate™ system was performed to identify the strain JK-2. As the results of this identification processes, the strain JK-2 could be assigned to genus Delftia, and designated as Delftia sp. JK-2
지각과민억제제 적용이 수종 접착제의 미세인장결합강도에 미치는 영향

This study evaluated the influence of a desensitizer(MS coat) on microtensile bond strength of different adhesives:a three step adhesive(All Bond 2), a two step adhesive(Single Bond), a one step adhesive(One up Bond F). Non-caries extracted human molars were used. Dentin surface was obtained by horizontal section on mid-portion of crown using a water cooled low speed diamond saw. Teeth were randomly divided into 6 group. AMO(MS coat + All Bond), SMO(MS coat + Single Bond) and OMO(MS coat + One up Bond F) dentin surface were treated with 17% EDTA before bonded adhesive. AMX, SMX and OMX dentin surface were bonded with All-Bond 2, Single Bond and One-up Bond F, respectively. with no previous treatment with MS coat and 17% EDTA. About 1cm high resin composite(Z 250) were incrementally build-up on the treated surface. The specimens for the microtensile test were serially sectioned perpendicular to the adhesive layer to obtain 0.7 × 0.7 mm sticks. 30 sticks were prepared from each group. After that, tensile bond strength for each stick was measured with Microtensile Tester at a 1mm/min crosshead speed. Fractured dentin surfaces were observed under the SEM. The results were statistically analysed by using a One way ANOVA and Tukey's test(p<0.05). Value in MPa were: AMO 44.35±13.21; SMO 39.35±13.32; OMO 31.07±1O.25; AMX 49.22± 16.38; SMX 56.02±13.35; OMX 72.93±16.19. Application of MS coat reduced microtensile bond strengths of both Single Bond and One up Bond F, whereas microtensile bond strengths of All Bond 2 were not affected significantly.

Approximately, 7-10 million people in the world suffer from Parkinson``s disease (PO), Recently, increasing evi-dence has suggested the protective effect of estrogens against nigrostriatal dopaminergic damage in PD. In this study, we investigated whether estrogen affects l-methyl-4-phenyl-l,2,3,6-tetrahydropyridine (MPTP)-induced behavioral impairment in estrogen receptor alpha (ERa)-deficient mice. MPTP (15 mg/kg, four times with l.5-h interval)-induced dopaminergic neurodegeneration was evaluated in ERα wild-type (WT) and knockout (KO) mice. Larger dopamine depletion, behavioral impairments (Rotarod test, Pole test, and Gait test), activation of mi-croglia and astrocytes, and neurointlammation after MPTP injection were observed in ERα KO mice compared to those in WT mice.Immunostaining for tyrosine hydroxylase (TH) after MPTP injection showed fewerTH-positive neurons in ERα KO mice than WT mice. Levels of dopamine and 3,4-<lihydroxyphenylacetic acid (OOPAC, metab-olite of dopamine) were also lowered in ERα KO mice after MPTP injection. Interestingly, a higher immunoreac-tivity for monoamine oxidase (MAO) B was found in the substantia nigra and striatum of ERα KO mice after MPTP injection. We also found an increased activation of p38 kinase (which positively regulates MAO B expression) in ERα KO mice. In vitro estrogen treatment inhibited neuroinflammation in l-methyl-4-phenyl pyridium (MPP +)-treated cultured astrocyte cells; however, these inhibitory effects were removed by p38 inhibitor. These results indicate that ERα might be important for dopaminergic neuronal survival through inhibition of p38 pathway.
소목(Caesalpinia sappan) 추출물의 Streptococcus mutans에 대한 성장, 산생성, 부착 및 비수용성 글루칸 합성 억제에 미치는 영향

Streptococcus mutans is known as the causative bacterial playing the most important role in forming plaque and it is being noticed as major causative bacteria of dental caries. Therefore, development of more effective, substantial and safe preventive agent against dental caries and periodontal disease is honestly required. The present study was designed to investigate the effect of chloroform extract(CE), n-butanol extract(BE),methanol extract(ME) and water extract(WE) of Caesalpinia on the growth, acid production, adhesion, and insoluble glucan synthesis of Streptococcus mutans. The CE, BE, ME and WE decreased the growth and acid production of S.mutans than that of control, especially, the ME was more effective than other extracts. The effect of CE, BE, ME and WE on the adherences to saliva-coated hydroxyapatite bead(S-HA) were evaluated. At the concentration of 2,000 ㎍/㎖, the CE showed 32% rate of bacterial adherence to S-HA, whereas, other extracts of the same concentration producedgreater than 50% rate. The CE and BE were significantly decreased the reduction rates of water insoluble glucan formation from sucrose by crude glucosyltransferase of S.mutans. Thus, the application of C. sappan can be considered a useful and a practical method for the prevention of dental caries.
Glycoalkaloids content in tuber peel and cortex of 24 potato cultivars of Korea

Potato glycoalkaloids(PGAs) are potentially toxic to humans at high levels and current safety regulations have recommended that PGAs content in tubers of potato cultivars should not exceed 20 mg/100g·FW. Accordingly, it is important to determine the PGAs composition and levels on potato cultivars for food safty and the breeding for new cultivars with low levels of PGAs. The main aim of this study was to evaluate α-chaconine, α-solanine and total PGAs content in the peel and cortex portions in 24 cultivars including ‘Haryoung’, ‘Goun’, ‘Hongyoung’ and ‘Jayoung’, recently released by Highland Agricultural Research Institute. The total PGAs ranged from 3.1 to 10.1 mg/100g·FW. 75-94% of total PGAs was existed in the peel part of all cultivars. We selected two cultivars, which can be eaten wth the skin on tubers, and so used for soy sauce braised potatoes and baby potatoes for the rest area. These results will provide consumers and breeders with fundamental information about the content of PGAs in Korea major cultivars.

<P>Docosahexaenoic acid (DHA) is an omega-3 fatty acid that is considered to have applications in cancer prevention and treatment. The beneficial effects of DHA against cancer metastasis are well established; however, the mechanisms underlying these effects in breast cancer are not clear. Cell invasion is critical for neoplastic metastasis, and involves the degradation of the extracellular matrix by matrix metalloproteinase (MMP)-9. The present study investigated the inhibitory effect of DHA on MMP-9 expression and cell invasion induced by 12-<I>O</I>-tetradecanoylphorbol-13-acetate (TPA) in the MCF-7 breast cancer cell line. DHA inhibited the TPA-induced activation of mitogen-activated protein kinase (MAPK) and the transcription of nuclear factor (NF)-κB, but did not inhibit the transcription of activator protein-1. DHA increased the activity of peroxisome proliferator-activated receptor (PPAR)-γ, an effect that was reversed by the application of the PPAR-γ antagonist GW9662. In addition, combined treatment with GW9662 and DHA increased NF-κB-related protein expression. These results indicate that DHA regulates MMP-9 expression and cell invasion via modulation of the MAPK signaling pathway and PPAR-γ/NF-κB activity. This suggests that DHA could be a potential therapeutic agent for the prevention of breast cancer metastasis.</P>

In the present study, we investigated anti-cancer effect of snake venom activated NK cells (NK-92MI) in lung cancer cell lines. Weused snake venom (4 mg/ml) treated NK-92MI cells to co-culture with lung cancer cells. There was a further decrease in cancer cellgrowth up to 65% and 70% in A549 and NCI-H460 cell lines respectively, whereas 30-40% was decreased in cancer cell growthby snake venom or NK-92MI alone treatment. We further found that the expression of various apoptotic proteins such as that Bax,and cleaved caspase-3 as well as the expression of various death receptor proteins like DR3, DR4 and Fas was also further increased. Moreover, consistent with cancer cell growth inhibition, the DNA binding activity of NF-kB was also further inhibited aftertreatment of snake venom activated NK-92MI cells. Thus, the present data showed that activated NK cells could further inhibitlung cancer cell growth.

This study was carried out to investigate preference and purchase factors of seaweed in some regional residents of Korea. Subjects were residents (n=1,218) whose residential area was divided into inland and coastal region and the survey was done during December 2007. Especially, purchase factors of seaweeds was conducted only in married females (n=353). The subjects are composed of 46.5% male and 53.5% female. Regional distribution of subjects was found to be 16~17%, with highest ratio in the age bracket of 20~29 years old. Proportion of students, at 29.8%, was the highest ranking occupation of the subjects. Preference score of seaweeds by region was highest for laver followed by brown seaweed and sea tangle. In terms of preference by gender, female subjects displayed higher preference score for green laver (p<0.01), seaweed fusiforme, brown seaweed, sea tangle (p<0.001) than the male subjects. Considerations being made when purchasing seaweeds for each region were in the order of freshness, taste and nutrition. In comparing the inland and coastal region, scores of freshness, convenience, design and color for the coastal region was higher, displaying significant difference.
