      • Increased 11β-hydroxysteroid dehydrogenase type 1 contributes to barrier dysfunction in aged skin

        Background: 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) converts inactive cortisone into cortisol, an active form, and is expressed by several tissues including the skin. Excessive active glucocorticoid (GC) deteriorates skin barrier function. Objectives: To find out if 11β-HSD1 affects on the barrier function in aged skin. Methods: We have performed human and in vivo studies. We measured cortisol in stratum corneum (SC) and oral epithelium of the elderly and young. Hairless mice were used for 11β-HSD1 immunohistochemistry staining of skin and measuring skin barrier function and serum cytokines. 11β-HSD1 knock-out (KO) mice and its wild-type were used for measuring lipid synthesis related enzyme. Results: Cortisol levels were elevated in SC and oral epithelium of the elderly rather than young. The 11β -HSD1 expression was increased in immunohistochemistry stain of aged mice skin. Aged mice showed decreased transepidermal water loss (TEWL) and SC hydration, and increased SC integrity than young. Serum inflammatory cytokines including interleukin-1α, -4, -10, -31 and tumor necrosis factor-α were increased in aged mice than young. The expressions of lipid synthesis related enzymes in epidermis were elevated in KO mice and topical 11β -HSD1 inhibitor applied mice. Conclusion: 11β-HSD1 expression is elevated in aged skin. It increases active GC and then deteriorates skin barrier function.

      • 종양 치료를 위한 siRNA 생체 투여 : In Vivo Delivery of siRNA for Cancer Therapy

        Cancer cells have distinctive phenotypes different from normal cells and these phenotypes are mediated by the expression of various genes. Molecular targeting, such as down regulation of certain gene or inhibiting certain protein, is needed for cancer therapy since overexpression and continuous activation of specific gene are responsible for transformation of normal cells. Since microRNA was described as new regulation mechanism of gene expression in mammalian cells, use of siRNA mediating RNA interference is not only powerful tool in gene discovery and protein function delineation but also useful therapeutic modality for genetic disease such as cancers because of its gene silencing effects through specific RNA degradation. In order to achieve the knockdown by siRNAs in vivo, many delivery systems of siRNAs based on carrier systems have been proposed since there are many obstacles to in vivo delivery of siRNAs. This review focuses on recent reports about siRNA in vivo delivery systems and the results from in vivo therapy experiments using targeted siRNA in cancer xenograft mouse models. 종양 세포의 형질은 정상 세포와 구별되는 뚜렷한 차이가 있고, 이러한 형질들은 다양한 유전자들에 의해서 발현된다. 특정 유전자의 과발현 또는 활성 증가를 종양형질을 만드는 중요한 분자생물학적인 기제로 볼 때 이러한 분자적 표적을 밝히고 이를 억제하는 것이 분자 표적치료이다. 최근 새로운 유전자 발현 조절기전으로 밝혀진 microRNA는 대부분의 포유동물 세포가 가지고 있는 중요한 생명현상의 조절 기제이며 특히, RNA 간섭 현상은 인위적으로 합성된 겹 사슬 RNA를 이용하여 mRNA를 파괴 또는 억제하여 원하는 특정 유전자의 발현을 일시적으로 차단 또는 감소시킬 수 있다는 점에서 중요한 의미를 가진다. RNA 간섭 현상에서 중요한 역할을 하는 siRNA를 이용하여 특정 유전자의 기능을 역으로 연구할 수 있고, 또한 특정 유전자의 억제를 유전적 질병의 치료에 적용할 수 있다는 점에서 많은 연구가 이루어지고 있다. 종양을 일종의 유전자 발현 조절장애 특히, 종양형질을 유도하는 특정 유전자의 과발현으로 본다면, 선택적으로 유전자의 기능을 억제할 수 있는 siRNA를 이용한 RNA 간섭 기술은 실로 임상적 이용가치가 매우 높다고 할 것이다. 그러나, 핵산 물질인 겹 사슬 RNA 가닥을 생체 세포로 전달하는 방법이 개발되지 않으면 치료에 응용하기가 어려울 것이다. 다양한 생체 투여 방법과 그 효과가 연구되고 있고, 특정 siRNA의 생체 투여로 원하는 유전자를 억제하여 종양 성장을 감소시키는 연구들이 진행되고 있다. 이에 저자는 특정 유전자를 표적으로 한 siRNA 생체 투여 방법과 종양 동물모델을 이용한 치료실험 결과를 토대로 지금까지 연구된 치료적 siRNA 생체 투여에 대해 논하고자 한다.

        This paper aims to investigate the syntax of causative verb clausal complements of make, have, let, and help in English within the minimalist framework outlined in Chomsky (1999, 2000, 2001), by emphasizing their constituency and categorial status. Generally, those syntactic causative verbs select bare infinitives as their complements, even though help also exceptionally takes another type of complements : to-infinitives. I put forward the proposals: i ) such causative verb clausal complements are single constituents, by examining a variety of constituency tests : coordination, reflexives, and expletives, and ii ) their categories are TPs whose head T is defective, lacking ∮ -complete.

      • 에탄올-물 혼합용매내에서 2,4,6,N-Tetramethyl Pyridinium Iodide의 회합에 대한 압력효과

        부피 퍼센트 95%의 에탄올-물 혼합용매내에서 2,4,6,N-테트라메틸 피리디늄요오드(TeMPI)의 이온회합 상수(K)를 수정한 자외선분광 및 전기 전도도 혼용법으로 온도범위 25∼50℃, 압력범위 1∼2,000bars에서 결정하였다. K값은 압력이 증가함에 따라서 증가하였고 40℃에서 최대값을 나타내었다. 부분몰부피변화(ΔV)는 비교적 작은 음의 값이 었으며 ΔV의 절대값은 40℃에서 최소값을 보여주었다. TeMPI의 이온 크기 변수(a)는 40℃에서 최대값을 가졌다. ΔH˚값은 40, 25℃ 그리고 50℃에서 각각 영, 음수 및 양수로 나타났으며 엔트로피(ΔS˚)와 자유에너지(ΔG˚)와 같은 다른 열역학변수 값도 계산하였다. 이와 같은 실험결과로부터 우리들은 TeMPI가 압력증가에 대하여 안정화되고, 40℃까지는 온도증가에 따라서도 안정화됨을 보여주었다. 그러나 40℃에서는 TeMPI 두 분자에 있는 8개 메틸기의 분자간 소수성 상호작용으로 인하여 약한 이량체를 형성하고 50℃ 이상에서는 다시 열적으로 분해된다는 결론을 얻게 되었다. The ionic association constants(K) of 2,4,6,N-tetramethyl pyridinium iodide(TeMPI) in 95 volume percentage ethanol-water mixture were determined by a modified UV and conductance method at 25℃ to 50℃ under 1 to 2,000 bars. The K values increase with increasing pressure and have maximum value at 40℃. The partial molar volume change (ΔV) has relatively small negative value and the absolute values of Δ are minimum at 40℃. The ion size parameter(a) of TeMPI have maximum value at 40℃. ΔH˚ values are zero, positive and negative at 40℃, 25℃ and 50℃ respectively. Other thermodynamic parameters such as the changes of standard entropy (ΔS˚) and free energy(ΔG˚) were evaluated. From these experimental results, we came to conclusion that TeMPI is stabilized by the elevation of pressure and that of temperature below 40℃ but weakly dimerized at 40℃ because of the intermolecular hydrophobic interaction of eight methyl groups of two molecules. And it thermally decomposed above 50℃.

      • 개에서 발생한 척수질환에 대한 추적조사(70예)

        This study was performed to evaluate the prevalence of spinal disorders in dogs, as well as their breed, age, gender and body weight distributions. Seventy cases of caine spinal disoreders presented in the period 2001 and 2006 were reviewed. The diagnosis was obtained by survey radiographs, myelogram, CSF analysis, CT and MRI. The most frequent breed was Maltese(15 cases), and followed by Yorkshire Terrier (10 cases), Pekingese (9 cases). Canine spinal disorders occurred from 7 month to 17 year old dog, mean age was 5.3±3.7 year-old. Canine spinal disorders occurred more often in females(59.7%) than males. Mean body weight was 5.87±7.02 kg. Degenerative (39 cases, 43.3%) and anomalous (24 cases, 26.7%) disorders were dominant in 90 specific disease distribution. This results can be a reference data for the further study.

        고속가공을 위한 검사시편에 관한 연구

        The properties of a machine tool greatly affect machining quality since a machine tool has large variance in its features. Machine tool makers want to find best machining condition with the one that they have built. Machine builders need to develop test specimen since it helps finding characteristics of machine tools when the machining properties of the specimen are analyzed. This paper develops test specimen to identify features of the main spindle, the feeding device, and the frame of a machine tool. The specimen is machined with a high speed machine and the features of the machine are analyzed with test items. They are surface roughness, overshoot in axial movement, errors in circular movement, feeding with small movement, and compensational error. This work can improve usability for a machine tool in machining practice.

        Home Health Care Classification(가정간호 분류체계)의 한글 명명화에 대한 타당성 연구

        The purpose of this study was to develop and test the validity of standardized Korean nomenclature of the Home Health Care Classification(HHCC), developed by Saba at the University of Georgetown. The four phases of the study were : (1) Two professors and 15 graduate students, who were taking a nursing intervention course, translated the HHCC into Korean. (2) 12 nurses with various clinical backgrounds reviewed each nomenclature taking into consideration of the definition and activities, and the Delphi method was used to determine the best appropriate nomenclature for each term; (3) 20 academic and clinical experts in nursing were given a questionnaire to rate the validity of each Korean nomenclature using a 5 point Likert scale ranging from very inappropriate to very appropriate : (4) Five members of the Korean Nurses Association Research Committee reviewed the survey results and determined the most appropriate Korean nomenclature for each term of the HHCC. Most of the nomenclatures had a score of more than 4.0, but several terms had a score lower than 4.0. They are Ambulation therapy(3.11), Anticipatory grieving(3.11), Sexual pattern alteration(3.17), Nursing care coordination(3.22), Physical therapist service(3.22), Regular diet(3.33), and Instrumental activities of daily living(3.44). These research results will be included in the nursing vocabulary dictionary to be published by the Korean Nurses Association.

        Temperature-dependent immune response of olive flounder (Paralichthys olivaceus) infected with viral hemorrhagic septicemia virus (VHSV)

        <P>Olive flounder (Paralichthys olivaceus) is one of the most economically important aquaculture fish. However, its production is often affected by various diseases, especially viral hemorrhagic septicemia virus (VHSV) that cause serious economic losses. In this study, we sequenced the whole transcriptome of the P. olivaceus using Illumina RNA-sEq. De novo assembly of control and virus-infected cDNA libraries of head kidney at 13 and 20 A degrees C was accomplished with 2,007,532,438 raw reads, resulting in 244,578 unigenes with an average length of 533 bp and found 65,535 candidate coding unigenes with homology to other species by BLAST analysis. DEG analysis among control and virus-infected head kidney samples of 13 and 20 A degrees C revealed that 1290 up-regulated and 162 down-regulated genes (p ae<currency> 0.01), linked to metabolism, virulence factors, adhesion and immune-response. We constructed an expressed gene catalog for the P. olivaceus to serve as a resource for marine environmental genomic and immuno-genetic/genomic studies focused on uncovering the molecular mechanisms underlying the responses of P. olivaceus to VHSV under different temperature.</P>
