        Herbal medicine Radix Scutellariae quality improved by exposure of the fresh root to high temperature

        Approximately 1200 years ago, the close association between the quality of medicine and the producing area was well known in China. Recent work has shown conclusively that the quality of herbal medicine is influenced by unfavorable conditions, but the exact cause remains unclear and is a difficult problem in research on herb resources. Fresh roots of S. baicalensis were exposed to temperatures of 35 °C ~ 55 °C, and the H2O2 level remained relatively inflexible. The superoxide dismutase (SOD) and catalase (CAT) activities remained stable at 35 °C and 45 °C but decreased on day 1 at 55 °C. The excess ROS due to the high temperatures must be removed by another pathway. Our study reported that the phenylalanine ammonia-lyase (PAL) activities rose. The fresh roots were exposed to 55 °C for 2 days, the baicalin increased by 38.5%, the wogonoside by 24.1%, and the baicalein by 37.0%. This method can substantially improve the quality of Radix Scutellariae.

        The research in spatial data processing mainly focus on two or multi-dimension data processing, and spatiotemporal data processing on moving objects are now more highlight. Spatial data structure are complicated, so the operations on spatial data will spend more cost. Therefore the optimization on spatial query is the key-point in application, and spatial index structures become the bottle-neck in spatial application, many scholars have done their best and given many methods on the problems. The paper present the review on the development of spatial index methods in recently years, and give a perspective in the spatial index and spatiotemporal index technology.

      • SOFTWARE PERFORMANCE TESTING BASED ON LINUX KERNEL

        Linux has created a huge new playing field for enterprises, academia, and governments. But lack and instability of applications have stopped many users from strategic, wholesale migrations to Linux. With more and more applications running on Linux system, for example mozilla fire fox, openoffice, GCC, etc. the software performance testing is becoming important on Linux platform. The kernel of an operating system has a significant impact on the overall performance of a computer system [1]. To test a software performance based on system kernel can give us a unique insight into the software, provide us useful information in determining the bottlenecks and weaknesses of the software, and thus enable us to improve it. This paper discusses the methodologies of software performance testing based on Linux kernel, which include design workload, get system parameter, time measurement and CPU utilized. We have done experiments on a computer running Linux; experiment results show that the methodologies of software performance testing based on Linux kernel is completely realizable and efficacious. Even though this paper is not intended to provide a comprehensive software test suite, the methodologies presented here can be extended to build such programs.

        Genome editing in potato plants by agrobacterium-mediated transient expression of transcription activator-like effector nucleases

        Genome editing (also known as targeted mutation) has promise for molecular breeding. Compared with the CRISPR/Cas9 system, the transcription activator-like effector nucleases (TALENs) have likely a lesser off-target rate in genome editing. Both a rapid test system for the functionality of designed TALENs and an effective delivery system for introducing the TALENs to plants are critical for successful target mutation. TALENs have usually been tested in protoplasts or introduced to plants with viral vectors. However, plant regeneration from protoplast culture can generate extensive somatic variation. Viral vectors are not always available, and plants edited by these vectors usually require virus elimination. Here, we used a nonviral, Agrobacterium-mediated transient expression approach, to serve both rapid test and effective delivery of TALENs into two vegetatively propagated potato cultivars, Solanum tuberosum Russet Burbank and Shepody. Two TALENs with different molecular weights (22 and 27 aarepeat modules) were expressed to target two endogenous genes (starch branching enzyme and an acid invertase) by Agrobacterium-mediated infiltration (agroinfiltration) into leaves of potato plants. The infiltrated leaf DNA was analyzed using restriction site loss assay and subsequent DNA sequencing. Deep sequencing of these tetraploid cultivars was also conducted to determine the zygosity at the targeted chromosomal loci. TALENs, with different molecular weights, successfully agroinfiltrated and induced mutations at both targeted loci.
