Effect of magnesium and calcium phosphate coatings on osteoblastic responses to the titanium surface

PURPOSE. The aim of this study was to evaluate the surface properties and in vitro bioactivity to osteoblasts of magnesium and magnesium-hydroxyapatite coated titanium. MATERIALS AND METHODS. Themagnesium (Mg) and magnesium-hydroxyapatite (Mg-HA) coatings on titanium (Ti) substrates were prepared by radio frequency (RF) and direct current (DC) magnetron sputtering.The samples were divided into non-coated smooth Ti (Ti-S group), Mg coatinggroup (Ti-Mg group), and Mg-HA coating group (Ti-MgHA group).The surface properties were evaluated using scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS). The surface roughness was evaluated by atomic force microscopy (AFM). Cell adhesion, cell proliferation and alkaline phosphatase (ALP) activity were evaluated using MC3T3-E1 cells. Reverse transcription polymerase chain reaction (RT-PCR) analysis was performed. RESULTS. Cross-sectional SEM images showed that Mg and Mg-HA depositionson titanium substrates were performed successfully. The surface roughness appeared to be similaramong the three groups. Ti-MgHA and Ti-Mg group had improved cellular responses with regard to the proliferation, alkaline phosphatase (ALP) activity, and bone-associated markers, such as bone sialoprotein (BSP) and osteocalcin (OCN) mRNA compared to those of Ti-S group. However, the differences between Ti-Mg group and Ti-MgHA group were not significant, in spite of the tendency of higher proliferation, ALP activity and BSP expression in Ti-MgHA group. CONCLUSION. Mg and Mg-HAcoatings could stimulate the differentiation into osteoblastic MC3T3-E1 cells, potentially contributing to rapid osseointegration.

<P>Ribosomal protein S3 (rpS3), a multi-functional protein, has been known to participate in DNA repair mechanism. In this study, we investigated changes in rpS3 immunoreactivity and its protein levels in the sub-regions of the gerbil hippocampus following subacute and chronic restraint stress. Serum corticosterone levels were increased in both the subacute and chronic-stress-groups compared to the control-group: the level in the subacute-stress-group was much higher than that in the chronic-stress-group. We could not find any neuronal damage in all the sub-regions of the hippocampus after both the subacute and chronic restraint stress. In the subacute-stress-group, rps3 immunoreactivity was not different compared to the control-group. However, rps3 immunoreactivity in the chronic-stress-group was decreased compared to the subacute-stress-group: especially, the immunoreactivity was markedly decreased in the pyramidal cells of the hippocampus proper (CA1-CA3 region) and granule cells of the dentate gyrus. In addition, western blot analysis also showed that rpS3 protein levels in the chronic-stress-group were significantly decreased compared to those in the subacute-stress-group. These findings indicate that chronic stress, not subacute stress, can decrease rpS3 immunoreactivity.</P>

<P>A new type of composite filler mechanically treated with multi-walled carbon nanotubes (MWNTs) and (BT) particles was prepared to produce higher dielectric properties in the composite. The hybrid film fabricated by incorporating these composite fillers in an epoxy matrix had a high dielectric constant and similar dielectric loss as compared to the composite which contained neat BT particles. The dielectric properties of these hybrid films were found to be dependent on both the content of MWNTs and mechanical processing time. Results suggest that this novel hybrid film composed of the composite filler and the epoxy matrix can be used for embedded capacitor material.</P>

<P>Ischemic preconditioning (IPC) is a condition of sublethal transient global ischemia and exhibits neuroprotective effects against subsequent lethal ischemic insult. We, in this study, examined the neuroprotective effects of IPC and its effects on immunoreactive changes of antioxidant enzymes including superoxide dismutase (SOD) 1 and SOD2, catalase (CAT) and glutathione peroxidase (GPX) in the gerbil hippocampal CA1 region after transient forebrain ischemia. Pyramidal neurons of the stratum pyramidale (SP) in the hippocampal CA1 region of animals died 5 days after lethal transient ischemia without IPC (8.6% (ratio of remanent neurons) of the sham-operated group); however, IPC prevented the pyramidal neurons from subsequent lethal ischemic injury (92.3% (ratio of remanent neurons) of the sham-operated group). SOD1, SOD2, CAT and GPX immunoreactivities in the sham-operated animals were easily detected in pyramidal neurons in the stratum pyramidale (SP) of the hippocampal CA1 region, while all of these immunoreactivities were rarely detected in the stratum pyramidale at 5 days after lethal transient ischemia without IPC. Meanwhile, their immunoreactivities in the sham-operated animals with IPC were similar to (SOD1, SOD2 and CAT) or higher (GPX) than those in the sham-operated animals without IPC. Furthermore, their immunoreactivities in the stratum pyramidale of the ischemia-operated animals with IPC were steadily maintained after lethal ischemia/reperfusion. Results of western blot analysis for SOD1, SOD2, CAT and GPX were similar to immunohistochemical data. In conclusion, IPC maintained or increased the expression of antioxidant enzymes in the stratum pyramidale of the hippocampal CA1 region after subsequent lethal transient forebrain ischemia and IPC exhibited neuroprotective effects in the hippocampal CA1 region against transient forebrain ischemia.</P>

<B>Background.</B><P>Papillary thyroid microcarcinoma (PMC; ≤1 cm) is thought to take a benign course during the lifetime. However, recent studies showed high recurrence rates for PMC.</P><B>Methods.</B><P>We analyzed the clinicopathological features, long-term prognosis, and some molecular characteristics including BRAF<SUP>V600E</SUP> mutation by retrospectively reviewing the records of 1150 patients with papillary thyroid carcinoma (PTC), 278 with PMC, and 868 with PTC >1 cm.</P><B>Results.</B><P>The prevalence of extrathyroidal invasion (52.2%) and initial nodal metastasis (34.9%) in patients with PMC was surprisingly high and almost as high as that for patients with PTC (72.4% and 51.8%, respectively). The rate of recurrent or persistent disease did not differ between patients with PMC and PTC (recurrent or persistent disease, 6.1% vs 14.1%; 53.4- vs 84.2-month follow-up; n = 98 vs 647; corrected p = .112). The frequency of BRAF<SUP>V600E</SUP> mutation was similar in patients with PMC and PTC (65.6% vs 67.2%). Immunohistochemical staining showed no different expression pattern according to the tumor size.</P><B>Conclusion.</B><P>These results suggest that PMC is not an occult cancer and it can act like larger PTC. This study investigates the physical characteristics of indoor rest zones using the Space Syntax Method. A conceptual framework is based on analysis of design works presented in competitions(544 rest zones of 24 buildings on 7 campuses). The analysis on the unit type of rest zones showed that they were mostly available as open type, followed by semi-open type and outdoor rest places. On the other hand, by classifying the shape of whole building into traffic lines and nodal points, it was found that the buildings with 3 or more traffic lines have lower availability than others, because the buildings required more nodal points along a little higher complexity and zoning formed at increased nodal points between building masses. The analysis on the location of rest zones on traffic lines showed that hall type was most excellent in the level of availability. Finally, the analysis on the floor height showed that hall-like open rest places and outdoor places like rooftop garden functioned as a factor to enhance the integration of different spaces. In conclusion, this research will help the follow-up studies make spatial creation planning that may enhance the spatial efficiency and availability of Indoor rest zones on the campus.
