Dietary factors play a crucial role in the management of type 2 diabetes mellitus (T2DM) by reducing cardiovascular disease (CVD) risk. Therefore, we aimed to examine the associations between habitual green tea consumption and risk factors of CVD among T2DM patients. A total of 1013 patients with T2DM were included in a community-based cross-sectional study. Data on dietary habits, including tea consumption, were collected using a food frequency questionnaire. A multivariable logistic regression model was used to analyze the associations. In men, as compared with nongreen tea drinkers, odds ratios (ORs) (95% confidence interval [CI]) of nonalcoholic fatty liver disease (NAFLD) were 2.06 (95% CI, 1.20–3.55) for those with green tea consumption of once per day and 2.45 (95% CI, 1.31–4.58) for more than or equal to twice per day (P-trend = .004); ORs (95% CI) of general obesity were 2.19 (95% CI, 1.02–4.68) and 2.70 (95% CI, 1.18–6.21), respectively (P-trend = .021); whereas no such association was found in women. Sensitivity analysis according to self-awareness of their T2DM status revealed that the positive association between green tea consumption and general obesity was not reliable. Higher intake of green tea was still positively associated with NAFLD, but it only persisted in participants aged ≥52 years or the lower dietary quality subgroup in further analyses. Our findings suggest that tea consumption was associated with an increased risk of NAFLD among male T2DM patients aged 52 years or older, and those with lower dietary quality, which needs to be confirmed in future prospective studies.
Overexpression of ENA1 from Yeast Increases Salt Tolerance in Arabidopsis
( Xiang Qiang Kong ),( Xiu Hua Gao ),( Wei Huan Li ),( Ji Qiang Zhao ),( Yan Xiu Zhao ),( Hui Zhang ) 한국식물학회 2008 Journal of Plant Biology Vol.51 No.2
In yeast, the plasma membrane Na+/H+ antiporter and Na+ -ATPase are key enzymes for salt tolerance. Saccharomyces cerevisiae Na+ -ATPase (Ena1p ATPase) is encoded by the ENA1/PMR2A gene; expression of ENA1 is tightly regulated by Na+ and depends on ambient pH. Although Ena1p is active mainly at alkaline pH values in S. cerevisiae, no Na+ -ATPase has been found in flowering plants. To test whether this yeast enzyme would improve salt tolerance in plants, we introduced ENA1 into Arabidopsis (cv. Columbia) under the control of the cauliflower mosaic virus 35S promoter. Transformants were selected for their ability to grow on a medium containing kanamyin. Southern blot analyses confirmed that ENA1 was transferred into the Arabidopsis genome and northern blot analyses showed that ENA1 was expressed in the transformants. Several transgenic homozygous lines and wild-type (WT) plants were evaluated for salt tolerance. No obvious morphological or developmental differences existed between the transgenic and WT plants in the absence of stress. However, overexpression of ENA1 in Arabidopsis improved seed germination rates and salt tolerance in seedlings. Under saline conditions, transgenic plants accumulated a lower amount of Na+ than did the wild type, and fresh and dry weights of the former were higher. Other experiments revealed that expression of ENA1 promoted salt tolerance in transgenic Arabidopsis under both acidic and alkaline conditions.

In this study, porous Si3N4 ceramics were obtained by a slip casting method which would be easier applied in industry. The most stable and dispersing aqueous suspension was obtained from a solid slurry content of 56.2%, a carboxymethyl cellulose concentration of 0.3% as the adhesive and 0.05% glycerol as the defoamer. Petroleum coke was used as pore-former, and at the same time, it was helpful for the stability of the suspension. The burning-out of the petroleum coke seemed to be an economical way for increasing the porosity of porous Si3N4. After sintering at 1,800℃ for 2 h, samples with a pore size about 10 μm, porosities of 45-60%, and flexural strength of 52.0-112.1MPa were obtained. Background: Chronic inflammation could affect the occurrence and development of malignant tumors. To explore the levels of tumor necrosis factor ${\alpha}$ (TNF-${\alpha}$) and C-reactive protein (CRP) in patients accompanied by impaired glucose tolerance (IGT) and their clinical significance. Materials and Methods: A total of 210 patients hospitalized in Affiliated Hospital of Taishan Medical University from Jun., 2013 to Dec., 2014 were selected, in which 92 cases were accompanied by IGT. Meanwhile, 80 randomly-selected healthy people by physical examination were as the control. The levels of routine biochemical indexes, plasma TNF-${\alpha}$ and CRP in all subjects were measured. Results: Both systolic and diastolic pressures in hypertension group and hypertension plus IGT group were significantly higher than in control group (p<0.01), but there was no statistical significance between these two groups (p>0.05). The levels of fasting plasma glucose (FPG) and blood glucose 2 h after taking glucose in hypertension plus IGT group were markedly higher than other groups (p<0.01). Homeostasis model assessment-insulin resistance (HOMA-IR), TNF-${\alpha}$ and CRP contents were on the progressive increase in control, hypertension and hypertension plus IGT groups, but significant differences were presented among each group (P<0.01). Hypertension accompanied by IGT had a significantly-positive association with CRP, TNF-${\alpha}$, FPG and blood glucose 2h after taking glucose. Conclusions: The levels of plasma TNF-${\alpha}$ and CPR in patients with hypertension accompanied by IGT increase significantly, indicating that inflammatory reaction in these patient increases, thus suggesting that these patients should be focused regarding cancer prevention.
CircZNF609 Aggravated Myocardial Ischemia Reperfusion Injury via Mediation of miR-214-3p/PTGS2 Axis
Wen-Qiang Tang,Feng-Rui Yang,Ke-Min Chen,Huan Yang,Yu Liu,Bo Dou 대한심장학회 2022 Korean Circulation Journal Vol.52 No.9
Background and Objectives: Circular RNAs were known to play vital role in myocardial ischemia reperfusion injury (MIRI), while the role of CircZNF609 in MIRI remains unclear. This study was aimed to investigate the function of CircZNF609 in MIRI. Methods: Hypoxia/reoxygenation (H/R) model was established to mimic MIRI in vitro. Quantitative polymerase chain reaction was performed to evaluate gene transcripts. Cellular localization of CircZNF609 and miR-214-3p were visualized by fluorescence in situ hybridization. Cell proliferation was determined by CCK-8. TUNEL assay and flow cytometry were applied to detect apoptosis. Lactate dehydrogenase was determined by commercial kit. ROS was detected by DCFH-DA probe. Direct interaction of indicated molecules was determined by RIP and dual luciferase assays. Western blot was used to quantify protein levels. In vivo model was established to further test the function of CircZNF609 in MIRI. Results: CircZNF609 was upregulated in H/R model. Inhibition of CircZNF609 alleviated H/R induced apoptosis, ROS generation, restored cell proliferation in cardiomyocytes and human umbilical vein endothelial cells. Mechanically, CircZNF609 directly sponged miR-214-3p to release PTGS2 expression. Functional rescue experiments showed that miR-214-3p/PTGS2 axis was involved in the function of circZNG609 in H/R model. Furthermore, data in mouse model revealed that knockdown of CircZNF609 significantly reduced the area of myocardial infarction and decreased myocardial cell apoptosis. Conclusions: CircZNF609 aggravated the progression of MIRI via targeting miR-214-3p/PTGS2 axis, which suggested CircZNF609 might act as a vital modulator in MIRI.
Superenhancers as master gene regulators and novel therapeutic targets in brain tumors
Zhuang Hai-Hui,Qu Qiang,Teng Xin-Qi,Dai Ying-Huan,Qu Jian 생화학분자생물학회 2023 Experimental and molecular medicine Vol.55 No.-
Transcriptional deregulation, a cancer cell hallmark, is driven by epigenetic abnormalities in the majority of brain tumors, including adult glioblastoma and pediatric brain tumors. Epigenetic abnormalities can activate epigenetic regulatory elements to regulate the expression of oncogenes. Superenhancers (SEs), identified as novel epigenetic regulatory elements, are clusters of enhancers with cell-type specificity that can drive the aberrant transcription of oncogenes and promote tumor initiation and progression. As gene regulators, SEs are involved in tumorigenesis in a variety of tumors, including brain tumors. SEs are susceptible to inhibition by their key components, such as bromodomain protein 4 and cyclin-dependent kinase 7, providing new opportunities for antitumor therapy. In this review, we summarized the characteristics and identification, unique organizational structures, and activation mechanisms of SEs in tumors, as well as the clinical applications related to SEs in tumor therapy and prognostication. Based on a review of the literature, we discussed the relationship between SEs and different brain tumors and potential therapeutic targets, focusing on glioblastoma.
The Antiferromagnetic Cross-coupled Spin Ladder: Quantum Fidelity and Tensor Networks Approach
Xi-Hao Chen,Sam Young Cho,Huan-Qiang Zhou,Murray T. Batchelor 한국물리학회 2016 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.68 No.9
We investigate the phase diagram of the cross-coupled Heisenberg spin ladder with antiferromagnetic couplings. For this model, the results for the existence of the columnar dimer phase, which was predicted on the basis of weak coupling field theory renormalization group arguments, have been conflicting. The numerical work on this model has been based on various approaches, including exact diagonalization, series expansions and density-matrix renormalization group calculations. Using the recently-developed tensor network states and groundstate fidelity approach for quantum spin ladders, we find no evidence for the existence of the columnar dimer phase. We also provide an argument based on the symmetry of the Hamiltonian, which suggests that the phase diagram for antiferromagnetic couplings consists of a single line separating the rung-singlet and the Haldane phases.
