      • 배추 시스테인 단백질 분해 효소 억제 유전자 BCPI-1의 발현과 종자 발아 및 유묘 생장과의 연관성

        Phytocystatins are protein inhibitors of cysteine proteinases of the papain family that have been identified in both monocot and dicot plants. A cDNA encoding a phytocystatin, BCPI-1 (Brassica Cysteine Proteinase Inhibitor-1) has been isolated from Chinese cabbage (Brassica campestris L. ssp. pekinensis) flower buds. Here, we tested whether BCPI-1 transcription is regulated by hormones, and could be involved in regulating cysteine proteinases during and after germination. BCPI-1 was sensitive to exogenous GA_(3) and ABA, which are important factors controlling seed germination, suggesting that the expression of BCPI-1 is hormonally regulated. We introduced a recombinant plasmid containing the full-length BCPI-1 cDNA under the control of cauliflower mosaic virus 35S promoter into rice embryogenic calli using the particle delivery method, and regenerated a number of transgenic rice plants. Constitutively over-expressed BCPI-1 caused changes in overall plant growth and development, including reduced germination and seedling growth. These data support the role of the BCPI-1 in the regulation of endogenous proteinases during both seed germination and subsequent seedling development. Phylocystatins은 papain계열의 cysteine 단백질 분해 효소 활성을 특이적으로 억제하는 억제자로 다양한 식물 종으로부터 분리되었다. 본고에서는, 배추 화아 cDNA library로부터 분리된 phytocystatin인 BCPI-1(Brassica Cysteine Proteinase Inhibitor-1)의 생체 내 기능에 대해 연구하였다. 먼저, BCPI-1 전사체는 발아 조절에 중요한 영향을 미치는 GA₃와 ABA에 의해 예민하게 증가, 혹은 감소되는 반응을 보임으로써, BCPI-1이 식물 호르몬의 영향을 받으며, 특히 발아나 유묘의 생장 조절에 관련이 있음을 알 수 있었다. Particle bombardment 방법을 통하여 BCPI-1을 벼의 배 형성 세포 내에 도입, 재분화 시켜 형질 전환 벼를 생성하였다. CaMV 35S promoter의 조절에 의해 지속적인 BCPI-1 발현을 보이는 형질 전환 벼의 경우, 발아와 유묘 생장이 현저히 지연되었다. 위의 결과들을 바탕으로, BCPI-1이 식물체 내에서 생성되는 cysteine 계열의 단밸질 분해 효소 활성을 조절하여 종자의 발아와 유묘의 생장에 영향을 미치는 것으로 추측한다.

        This study is aimed to introduce portfolio performance assessment that discussed recently in the geographic section of elementary social studies, to investigate theoretically and to find performance assessment method. Portfolio performance assessment evaluates inner-change and development process through individuals products that are collected themselves, and it's objectives are related to social studies objectives. Social studies objectives is compose to the understanding and application of concept, ability, attitude and conviction. For the direction of portfolio performance assessment, Social studies objectives is investigated. With them, the meaning and value of portfolio performance assessment is built in the geographic section, Portfolio assessment become interdependence studying through self-assessment, peer-assessment and teacher support-assessment. The method of portfolio performance assessment is suggested. And portfolio performance assessment instrument is developed into five section in according to geographic studying situation.

        초등지리교육에서 인터넷 활용 학습을 통한 국제이해교육

        인터넷 활용은 오늘날 가장 중요한 매체로서 우리 생활에 필수적인 정보원으로 자리잡고 있으며, 많은 교육적인 효과를 가지고 있다. 초등학교 수준에 알맞은 인터넷 자료를 선별하여 사회과 국제이해교육의 교수·학습 자료로 활용하여 국제이해를 증진시키고 간접 체험을 하게 하여 국제 사회 문제에 대한 합리적인 판단력을 길러, 세계 시민으로서 올바른 태도를 기르고자 한다. 국제이해교육의 개념과 지리교육에서의 국제이해교육, 인터넷 활용과 국제이해교육에 관한 문헌 연구와 초등학교 6학년 사회과 국제이해교육 내용을 분석하고 인터넷 자료를 추출하여 학습자료로 활용하여 탐구식 국제이해교육 수업에 적용하여 그 효과를 검증하였다. 인터넷 활용 학습을 통한 국제이해교육은 세계 시민적 자질에 올바른 가치관 형성에 유용하였고 학업 성취도면에서도 유의미한 차이가 있었다. This research is designed to demonstrate the effectiveness of Internet-based teaching and learning as a way to facilitate elementary school students ability not only to understand international issues, but to internalize appropriate and reasonable attitudes as international members in the decision-making process by guiding them into making the most use of pre-cautiously selected teaching-learning materials and experience the real world through up-to-the-minute information presented on Internet .For the above-mentioned purpose, some pre-studies and a practical experiment were conducted. The contents are as follows: (1) the definition of international understanding, especially in the teaching of geography, (2) previous research on Internet-based learning of international understanding, (3) the analysis of the contents of the sixth grade textbook in social studies focusing on international understanding, (4) the actual experiment, and its implications. while leading them to form positive and appropriate attitudes towards those international issues and concerns. In T-test based on the result of the posttest, it is evidenced that there are significant differences placed between the two approaches.

        The purpose of this study is in developing locally-oriented teaching-learning materials to motivate students to effectively take part in activities using map and in improving the field work of social studies to plan a map learning and to give them the ability of practice. It represents the right order involved in teaching the activities using map after doing preceding learning : classroom, playground, and the environs of the school including to some learning methods such as a puzzle map, a correlative of learning. Field trip study planning and lesson-plan are designed in case of Sewage disposal plant, Duknam filtration plant, Bonchon industrial complex relating to field trip study. There are some effects expected which follow through this study. First, teachers are able to draw learner's interest in finding learning sources directly through the community where they live. Second, learners have active participation in class, motivation, drawing their interests through map learning by doing preceding study of learner-centered. Third, teachers can make use of it as useful tools to get high achievement goal with self-direct learning.

      • 무증상으로 발견된 담낭-담관 누공을 동반한 Mirizzi 증후군 1예

        Mirizzi's syndrome is a rare complication of the gallstone disease, characterized by narrowing of the common bile duct (CBD) due to chronic extrinsic compression of an impacted gallstone in the cystic duct or the neck of the gallbladder. The impacted gallstone may erode into the bile duct, causing cholecystocholedochal fistula. The patients generally have history of repeated attacks of jaundice and abdominal pain. But there is no report about asymptomatic Mirizzi syndrome with cholecystocholedochal fistula which were found incidentally. So herein we present our clinical experience with a case of asymptomatic Mirizzi syndrome with cholecystocholedochal fistula in a young female.

      • 불소가 첨가된 실리콘 산화막의 다층금속절연 특성에 관한 연구

        The characteristics of fluorinated silicon oxide (SiOF) films prepared by plasma enhanced chemical vapor deposition(PECVD) were discussed. The deposition of these films was carried out in the temperature range of 80 - 220℃ in a conventional parallel plate plasma reactor with 〈 ±3% uniformity by flowing 2 sccm of disilane (Si₂H6), 100 sccm of nitrous oxide (N₂O), and 20 sccm of tetrafluoromethane (CF₄). AS the deposition temperature increased from 80 to 220℃, the deposition rate of the films increased from 16.7 to 18.4 nm/min; meanwhile, the etch rate decreased from 2.69 to 1.48 nm/sec. The refractive index was 1.46 regardless of the deposition temperature. The Fourier transform infrared (FTIR) spectra of the films showed decreasing Si-O stretching wave number with increasing full width at half maximum (FWHM) as deposition temperature increased. The high frequency capacitance-voltage (C-V) measurements of the metal-oxide-semiconductor (MOS) capacitors fabricated using the films showed increasing effective oxide charge density and decreasing dielectric constant with increasing deposition temperature. Deposition temperature of 180℃ resulted in films with the smallest dielectric constant of 3.75. These films showed an average breakdown strength of 9.14 MV/cm with 77.5% of the MOS capacitors having breakdown field strength ≥9.5 MV/cm
