Majorized proof of arithmetic-geometric-harmonic means inequality

As we all know, arithmetic -geometric mean inequalities are the most basic and important inequalities. To seek different method to prove them has been one of the study focuses and they have been proven by more than a hundred ways By using methods based on the theory of majorization, the arithmetic-geometric-harmonic means inequality is proved in a new way.

Drosophila suzukii (Diptera: Drosophilidae) is one of the very few Drosophila species which are able to lay eggs and feed on healthy ripening fruit. Adults and larvae are all able to obtain the nutrition in the decaying food, which will generate many microorganisms. However, the relationship between D. suzukii and microorganism is poorly understood. In this study, 13 species of fungi were identified in an artificial diet fed by D. suzukii. Short-term and continuously life table experiments were conducted to determine the impact of three of these fungus species including Geotrichum candidum, Talaromyces minioluteus and Actinomucor elegans on the growth and development of Drosophila suzukii. Results revealed that, compared to the control, G. candidum, T.minioluteus and A. elegans increased the mortality of D. suzukii adults in the short time, while extended the developmental time of pupal D. suzukii by 18.00%, 16.22% and 26.44%, respectively in the life table experiment. T. minioluteus reduced the total longevity of D. suzukii by 15.52%, while A. elegans enhanced the total longevity by 25.96%. G. candidum and A. elegans increased the fecundity bymore than two folds. T. minioluteus elongated the mean generation time (T) by 31.34%, whereas G. candidum shortened it by 15.26% but increased the net reproductive rate (R0) by 217.76%, intrinsic rate of increase (r) by 88.89% and finite rate of increase (λ) by 9.17%. It was concluded that G. candidumand A. elegans significantly had the beneficial effect on the growth and reproduction of D. suzukii, however, T. minioluteus had the negative effect on D. suzukii. Our results could provide a new integrated pest management strategy for D. suzukii which would be discussed in this study.

All-benzocyclobutene functionalized polycarbosilanes and the derived copolymers were successfully synthesized by ring-opening polymerization (ROP) of 1,1-dibenzocyclobutene-1-silacyclobutane (DBCBSCB) and 1,1-diphenylsiletane (DPSCB) using Karstedt’s catalyst. Adjusting the copolymerization ratio of DBCBSCB and DPSCB could afford to control the ultimate properties of the cured resins. The linear polycarbosilanes containing dibenzocyclobutene were directly cross-linked upon high temperature. With increasing the copolymerization ratio of DBCBSCB and DPSCB, the 5% weight loss temperature (T5%) of cured resins is increased from 450 °C to 506 °C under a nitrogen atmosphere, while the dielectric constant (k) is decreased from 2.60 to 2.42 at 10 MHz. The excellent thermostability and low-dielectric performance make such thermosets have potential application in the microelectronic industry.
The m6A reader IGF2BP3 promotes acute myeloid leukemia progression by enhancing RCC2 stability

N6-methyladenosine (m6A) is the most abundant posttranscriptional modification of mRNA in eukaryotes. Recent evidence suggests that dysregulated m6A-associated proteins and m6A modifications play a pivotal role in the initiation and progression of diseases such as cancer. Here, we identified that IGF2BP3 is specifically overexpressed in acute myeloid leukemia (AML), a subtype of leukemia associated with poor prognosis and high genetic risk. IGF2BP3 is required for maintaining AML cell survival in an m6A-dependent manner, and knockdown of IGF2BP3 dramatically suppresses the apoptosis, reduces the proliferation, and impairs the leukemic capacity of AML cells in vitro and in vivo. Mechanistically, IGF2BP3 interacts with RCC2 mRNA and stabilizes the expression of m6A-modified RNA. Thus, we provided compelling evidence demonstrating that the m6A reader IGF2BP3 contributes to tumorigenesis and poor prognosis in AML and can serve as a target for the development of cancer therapeutics.
Synthesis of S-adenosyl-L-methionine in Escherichia coli

S-adenosyl-L-methionine (SAM) is an importantphysiological metabolite in vivo and may be useful inmedicines. SAM is produced from L-methionine and ATPcatalyzed by S-adenosyl-L-methionine synthetase (SAMS)in vivo. In this study, the gene encoding SAMS was clonedand a genetically engineered Escherichia coli (E. coli)BL21(pET-28a-SAMS) was constructed. The recombinantSAMS with a molecular mass of approximately 46 kDawas expressed by inducing the engineered E. coli usingisopropyl-β-D-1-thiogalactopyranoside (IPTG) as an inducer. To produce SAM using a low-cost, nontoxic and highperformanceexpression system, lactose was used as asubstitute for IPTG to induce BL21(pET-28a-SAMS). Byoptimizing the expression conditions, the concentration ofSAM produced by the engineered E. coli was 48 mg/L in theculture medium supernatant. To increase the concentrationof SAM produced, a coupled system was constructedconsisting of E. coli BL21(pET-28a-SAMS) and Saccharomycescerevisiae (S. cerevisiae) JM-310. In this coupled system,ATP generated from S. cerevisiae was provided to E. colifor producing a higher concentration of SAM. The SAMconcentration in the coupled system reached 1.7 g/L. SAMwas purified by a weak acid cationic exchange resin D113,and a simple and economical purification procedure forSAM isolation was achieved. SAM was confirmed byHigh Performance Liquid Chromatography-tandem MassSpectrometry analysis. Our study provides a feasible andconvenient approach to produce SAM.

The CKLF-like MARVEL transmembrane domain-containing family (CMTM) is a novel family of proteins linking classical chemokines and the transmembrane 4 superfamily (TM4SF). Our earlier studies indicated several CMTM members (such as CKLF1 and CMTM2) have a secreted form. This is the first report of the secreted form of CMTM5-v1, the major RNA splicing form of CMTM5, which is produced as small vesicles (<100 nm diameter) and floats at a peak density of 1.19 g/ml on continuous sucrose gradients. CMTM5-v1 has no obvious co-localization with CD63 or Golgi complex. In addition, brefeldin A but not wortmannin can inhibit the secretion of CMTM5-v1. Our results suggest that CMTM5-v1 might be secreted via a different vesicle-mediated secretory pathway, which will be helpful for the studies of vesicle-mediated secretion and MARVEL domain-containing proteins. B-32 is one of a panel of monoclonal anti-idiotypic antibodies to growth hormone (GH) that we developed. To characterize and identify its potential role as a novel growth hormone receptor (GHR) agonist, we determined that B-32 behaved as a typical Ab2β based on a series of enzyme-linked immunosorbent assay assays. The results of fluorescence-activated cell sorting, indirect immunofluorescence and competitive receptor binding assays demonstrated that B-32 specifically binds to the GHR expressed on target cells. Next, we examined the resulting signal transduction pathways triggered by this antibody in primary porcine hepatocytes. We found that B-32 can activate the GHR and Janus kinase (2)/signal transducers and activators of transcription (JAK2/STAT5) signalling pathways. The phosphorylation kinetics of JAK2/STAT5 induced by either GH or B-32 were analysed in dose-response and time course experiments. In addition, B32 could also stimulate porcine hepatocytes to secrete insulin-like growth factors-1. Our work indicates that a monoclonal anti-idiotypic antibody to GH (B-32) can serve as a GHR agonist or GH mimic and has application potential in domestic animal (pig) production.
