        A STUDY ON NUTRITIONAL STATUS OF TRACE MINERALS OF CATTLE IN JAVA IN INDONESIA

        The nutritional status of trace minerals in cattle of Java in Indonesia was investigated by evaluating Cu, Fe, Mo, Zn and Mn concentrations in diets and livers, and Cu and Zn concentrations in blood plasma. Investigations were conducted on Jonggol (West Java), Malang (East Java) and Mojokerto (East Java) in both the rainy and the dry seasons in 1988. In Jonggol, low Cu concentrations in diets showing 7.1 mg/kg in the rainy season and 10.9 mg/kg in the dry season were observed and all plasma samples showed Cu concentrations below the critical level ($0.65{\mu}g/ml$). Thirty percent of the liver samples in Malang and 54% of those in Mojokerto showed lower Cu concentrations than the critical level (75 mg/kg on a dry matter basis). Fe concentrations in diets from the three regions showed a wide variation of values ranging from 249 to 30,000 mg/kg. A large amount of Fe was accumulated in livers from Malang and Mojokerto, giving average concentrations of 498 mg/kg. Zn concentration in diet and plasma samples were close to the borderline and some Zn concentration in these samples showed deficient levels. Mo and Mn concentrations in diets and livers showed normal levels.

        A STUDY ON NUTRITIONAL STATUS OF MACRO MINERALS OF CATTLE IN JAVA IN INDONESIA

        Nutritional status of cattle in Java in Indonesia was investigated by evaluating P, Ca, Mg, K and Na concentrations in forage and plasma inorganic phosphorus (Pi), Ca and Mg, hematocrit and hemoglobin concentrations of cattle during both rainy and dry seasons in 1988. Investigations were conducted at Jonggol (West Java), Malang (East Java) and Majokerto (East Java). Low P content in forage averaging 0.11% on a dry matter basis, high Ca:P ratio in forage in the rainy season (7.7:1) and low plasma Pi concentrations averaging 4.71 mg/dl were observed in Jonggol. In all three places, Ca content in forages was higher than the requirement according to the NRC standard and plasma Ca concentrations were above the critical level. During the rainy season forty percent of cattle in Malang and Mahokerto showed plasma Mg concentrations below 2mg/dl in spite of normal Mg and K content in forage. Forage samples from each place showed a wide variety in Na content, and Na content of two thirds of forage samples was lower than the requirement according to the NRC standard. Hematocrit and hemoglobin concentrations in Malang and Majokerto showed higher values than the normal range.

        Development of Bovine Specific Leptin Radioimmunoassay and Relationship of Plasma Leptin with Vitamin A and Age of Wagyu

        Leptin is produced by adipocytes and its role in the regulation of lipid metabolism, feed intake, productive and reproductive performance of domestic animal species has been greatly stressed and extensively investigated in recent years. This study was conducted to develop a radioimmunoassay (RIA) for the estimation of plasma bovine leptin and to determine plasma leptin concentration in fattening Japanese Black cattle (Wagyu) and its crossbreds at commercial farms. Relationships of plasma leptin with plasma vitamin A and age of crossbred cattle were also determined. Recombinant bovine leptin (rbleptin) was produced by the E. coli overexpressed leptin as a GST (glutathione S-transferase)-fusion protein. Then antiserum against bovine leptin was obtained by its immunization in rabbits. Using this antiserum, a bovine specific RIA was developed and plasma leptin level was determined in 120 crossbred fattening cattle (WagyuHolstein, 50:50) at commercial farms. The plasma leptin level increased with the age of cattle and its level was greater in the crossbred heifers than in the steers. Plasma vitamin A level was negatively correlated with plasma leptin level in crossbred heifers and steers. This relationship was stronger in heifers than in steers. Plasma leptin was gradually increased with advancing age in fattening Wagyu cattle. In conclusion, development of a bovine specific RIA to estimate plasma leptin will contribute to better understanding of the role of leptin in cattle.

        MINERAL CONCENTRATION IN RICE STRAW AND SOIL IN KYONGBUK PROVINCE, KOREA

        A field survey was carried out in Korea to assess the mineral composition of rice straw since it is a cheap and available cattle feedstuff. Forage and soil samples were collected in 4 localities in the Kyongbuk province. Soil analysis were also carried out in order to establish the relationships between soil composition and mineral content in the rice straw. Based on NRC tables, the rice straw samples provided adequate amounts of Mg, Ca, K, S, Mo, Mn and Zn. Percentages of samples deficient in P, Na, Cu and Se were 83, 50, 67 and 83 respectively. Soil samples, whose mean pH was 5.8 and mean organic matter content was 3.99%, were comparatively acidic and had high organic matter content. They also had high amounts of extractable Fe and Mn. Correlation coefficients between mineral content in soil and rice straw were low, i.e., 0.42 for Me (p < 0.05), and 0.37 for Mo (p < 0.05). The low or nonexistent correlation between soil and forage composition indicates the difficulty of establishing appropriate methods of mineral availability to the plants.

        Changes in Serum Biochemical Markers of Bone Cell Activity in Growing Thoroughbred Horses

        We studied the changes in biochemical markers of bone metabolism in growing Thoroughbred horses. Serum osteocalcin (OC), as a marker for bone formation, and carboxy-terminal propeptide of type-I collagen (PICP), as a marker for bone formation, carboxy-terminal telopeptide of type-I collagen (ICTP), as a marker for bone resorption, were determined in nine clinically healthy horses from 3 d to 17 mo of age. The BW and withers height (WH) increased during the study. On the other hand, a rapid reduction in body weight gain (BWG) was observed between 1 mo and 9 mo of age and a rapid reduction in withers height gain was observed between 1 mo and 5 mo of age. The serum markers decreased significantly with increasing age. In particular, dramatic changes in serum markers occurred between 3 d to 1 wk and 5 to 7 mo of age in these horses, which suggests that bone turnover rapidly decreased after birth. On the other hand, the ratio of PICP to ICTP decreased through the experiment. This result suggests that the reduction in bone formation exceeded that of bone resorption. There was a significant correlation between markers and growth parameters, except for the correlation between PICP and BWG on single linear regression analysis. Serum OC and ICTP were affected by the WH in multiple linear regression analysis. These results indicated that the age-related variation in serum biochemical markers of bone metabolism reflected bone growth, but neither BW nor BWG. Therefore, we consider that changes in bone modeling are the major factor affecting the levels of serum biochemical markers by 17 mo of age in horses.

        Changes of Serum Mineral Concentrations in Horses during Exercise

        We investigated the exercise-induced changes in the serum concentration of several minerals in horses. Four welltrained Thoroughbred horses performed exercise for 5 d. The blood hemoglobin (Hb) concentration increased during exercise, recovered to the pre-exercise level immediately after cooling down and did not change again up till the end of experiment. The changes in serum zinc (Zn) and copper (Cu) concentrations were similar to those of blood Hb during the experiment. The serum magnesium (Mg), inorganic phosphorus (Pi) and iron (Fe) concentrations also increased during exercise. Though the serum Pi concentration recovered to the pre-exercise level immediately after the cooling down, it decreased further before the end of the experiment. The serum Mg concentration was lower immediately after cooling down than its pre-exercise level but gradually recovered from the temporal reduction. The recovery of the serum Fe concentration was delayed compared to that of other minerals and recovered 2 h after cooling down. The serum calcium (Ca) concentration did not change during exercise but rapidly decreased after cooling down. As a result, it was lower immediately after cooling down than its pre-exercise level. It recovered, however, to the pre-exercise level 2 h after cooling down. The temporal increase in the serum concentrations of all minerals except Ca is considered to result from hemoconcentration induced by exercise and the stable concentration of the serum Ca during exercise is possibly due to its strict regulation of homeostasis. These results indicate that the serum concentration of each mineral responds differently to exercise in horses, which may be due to the difference in metabolism among these minerals.

        Considerable number of endocrine disruptors (7 compounds) are known today (Japan Environmental Agency, 1998). Among them, phthalates are considered to be in most abundance in our environment. The production of phthalates in Japan was 474 thousand tones in 1998 (Kasozai Kogyo Kai, 1999), around 56% of which was di(2-ethylhexyl)phthalate (DEHP) uesd mainly as plasticizers in polyvinyl chloride (PVC) production, is widely used in PVC, nitrocellulose polyvinyl acetate, painting inks, adhesives and lacquers. PVC contains up to 40% phthalates by weight wuthout covalently binding to other ingredients in the PVC(Needham et al., 2000). Thus, significant amounts of DEHP and DBP are easily released from the plastics into the environment during production, used and disposal, resulting in the pollution of food air, water, soil and homes.

        Evaluation Technique for Reliability in Low-Temperature poly-Si

        Analysis of the degradation in n-ch TFT under dynamic stress by using pico-second timeresolved emission microscopy was performed. We have successfully detected an emission at the pulse fall edge. Emission intensity increased with decreasing pulse fall time. As the degradation depended on the pulse fall time, this dependence clearly illustrates that hot electrons are the dominant cause of the degradation under dynamic stress. Based on these dependence, we proposed model considering electron traps in poly-Si. Further, we analyzed thermal degradation in low temperature poly-Si thin film transistors by using infrared thermal imaging microscopy. Non uniform distribution was observed in the saturation region along the gate length. The increase of temperature was remarkable at large gate width, therefore, large voltage shift was observed. A universal relationship was obtained, independent of crystallinity of poly-Si. This curve suggests that we should take the degradation of gate oxide such as electron traps into account.
