        Immunomodulatory Properties of Lactobacillus plantarum NC8 Expressing an Anti-CD11c Single-Chain Fv Fragment

        The lactic acid bacteria species Lactobacillus plantarum (L. plantarum) has been used extensively for vaccine delivery. Considering to the critical role of dendritic cells in stimulating host immune response, in this study, we constructed a novel CD11c-targeting L. plantarum strain with surface-displayed variable fragments of anti-CD11c, single-chain antibody (scFv-CD11c). The newly designed L. plantarum strain, named 409-aCD11c, could adhere and invade more efficiently to bone marrow-derived DCs (BMDCs) in vitro due to the specific interaction between scFv-CD11c and CD11c located on the surface of BMDCs. After incubation with BMDCs, the 409-aCD11c strain harboring a eukaryotic vector pValac-GFP could lead to more efficient expression of GFP compared with wild-type strains shown by flow cytometry analysis, indicating the enhanced translocation of pValac-GFP from L. plantarum to BMDCs. Similar results were also observed in an in vivo study, which showed that oral administration resulted in efficient expression of GFP in both Peyer’s patches (PP) and mesenteric lymph nodes (MLNs) within 7 days after the last administration. In addition, the CD11c-targeting strain significantly promoted the differentiation and maturation of DCs, the differentiation of IL-4+ and IL-17A+ T helper (Th) cells in MLNs, as well as production of B220+ IgA+ B cells in the PP. In conclusion, this study developed a novel DC-targeting L. plantarum strain which could increase the ability to deliver eukaryotic expression plasmid to host cells, indicating a promising approach for vaccine study.

        Study on Supervision System of Chinese Specialized Farmers Cooperatives: Experience of the Korean NongHyup

        Purpose - The Chinese government's policies and funds enable specialized farmers cooperatives to develop vigorously. However, the non-systematic supervision system affects the interests of farmer-members of these cooperatives, which are similar to the Korean NongHyup in many aspects but differ in their supervision systems such as distribution. Therefore, this paper aims to identify the problems of specialized farmers cooperatives, and obtain some insights from NongHyup. Research design, data, and methodology - Data were collected from farmers, the government, and cooperatives in northern China's Shandong Province (the cities of Jinan, Qingdao, Weifang, Linyi, and Heze) through a literature survey, case analysis, and comparative analysis in each city. Results - 1) Specialized farmers cooperatives should establish a transparent regulatory mechanism and be subject to dual supervision from both the Chinese government and farmers. 2) The Chinese government and civil society should offer more support to the cooperatives, and strive to change farmers' backward attitude through education and training. Conclusions - Small cooperatives could merge into large ones and undertake social responsibility through the establishment of labor unions.

        Consumption of Oxidized Soybean Oil Increased Intestinal Oxidative Stress and Affected Intestinal Immune Variables in Yellow-feathered Broilers

        This study investigated the effect of oxidized soybean oil in the diet of young chickens on growth performance and intestinal oxidative stress, and indices of intestinal immune function. Corn-soybean-based diets containing 2% mixtures of fresh and oxidized soybean oil provided 6 levels (0.15, 1.01, 3.14, 4.95, 7.05, and $8.97meqO_2/kg$) of peroxide value (POV) in the diets. Each dietary treatment, fed for 22 d, had 6 replicates, each containing 30 birds (n = 1,080). Increasing POV levels reduced average daily feed intake (ADFI) of the broilers during d 1 to 10, body weight and average daily gain at d 22 but did not affect overall ADFI. Concentrations of malondialdehyde (MDA) increased in plasma and jejunum as POV increased but total antioxidative capacity (T-AOC) declined in plasma and jejunum. Catalase (CAT) activity declined in plasma and jejunum as did plasma glutathione S-transferase (GST). Effects were apparent at POV exceeding $3.14meqO_2/kg$ for early ADFI and MDA in jejunum, and POV exceeding $1.01meqO_2/kg$ for CAT in plasma and jejunum, GST in plasma and T-AOC in jejunum. Relative jejunal abundance of nuclear factor kappa B ($NF-{\kappa}B$) P50 and $NF-{\kappa}B$ P65 increased as dietary POV increased. Increasing POV levels reduced the jejunal concentrations of secretory immunoglobulin A and cluster of differentiation (CD) 4 and CD8 molecules with differences from controls apparent at dietary POV of 3.14 to $4.95meqO_2/kg$. These findings indicated that growth performance, feed intake, and the local immune system of the small intestine were compromised by oxidative stress when young broilers were fed moderately oxidized soybean oil.

        Proteomic analysis of chicken peripheral blood mononuclear cells after infection by Newcastle disease virus

        Characteristic clinical manifestations of Newcastle diseaseinclude leukopenia and immunosuppression. Peripheral bloodmononuclear cells (PBMCs) are the main targets of Newcastledisease virus (NDV) infection. To survey changes in proteomicexpression in chicken PBMCs following NDV infection,PBMC proteins from 30 chickens were separated using twodimensionalelectrophoresis (2-DE) and subjected to massspectrometry analysis. Quantitative intensity analysis showedthat the expression of 78 proteins increased more thantwo-fold. Thirty-five proteins exhibited consistent changes inexpression and 13 were identified as unique proteins bymatrix assisted laser desorption ionization-time of flight massspectrometer/mass spectrometer including three that weredown-regulated and 10 that were up-regulated. These proteinswere sorted into five groups based on function: macromolecularbiosynthesis, cytoskeleton organization, metabolism, stressresponses, and signal transduction. Furthermore, Westernblot analysis confirmed the down-regulation of integrin-linkedkinase expression and up-regulation of lamin A production. These data provide insight into the in vivo response of targetcells to NDV infection at the molecular level. Additionally,results from this study have helped elucidate the molecularpathogenesis of NDV and may facilitate the development ofnew antiviral therapies as well as innovative diagnostic methods.
