      • MiRNA Synergistic Network Construction and Enrichment Analysis for Common Target Genes in Small-cell Lung Cancer

        Background: Small-cell lung cancer (also known as SCLC) is an aggressive form and untreated patients generally die within about 3 months. To obtain further insight into mechanism underlying malignancy with this cancer, an miRNA synergistic regulatory network was constructed and analyzed in the present study. Method: A miRNA microarray dataset was downloaded from the NCBI GEO database (GSE27435). A total of 546 miRNAs were identified to be expressed in SCLC cells. Then a miRNA synergistic network was constructed, and the included miRNAs mapped to the network. Topology analysis was also performed to analyze the properties of the synergistic network. Consequently, we could identified constitutive modules. Further, common target genes of each module were identified with CFinder. Finally, enrichment analysis was performed for target genes. Results: In this study, a miRNA synergistic network with 464 miRNAs and 2981 edges was constructed. According to the topology analysis, the topological properties between the networks constructed by LC related miRNAs and LC unrelated miRNAs were significantly different. Moreover, a module cilque0 could be identified in our network using CFinder. The module included three miRNAs (hsa-let-7c, hsa-let-7b and hsa-let-7d). In addition, several genes were found which were predicted to be common targets of cilque0. The enrichment analysis demonstrated that these target genes were enriched in MAPK signaling pathways. Conclusions: Although limitations exist in the current data, the results uncovered here are important for understanding the key roles of miRNAs in SCLC. However, further validation is required since our results were based on microarray data derived from a small sample size.

      • <i>Haloactinopolyspora alkaliphila</i> sp. nov., and emended description of the genus <i>Haloactinopolyspora</i>

        <P>A facultatively alkaliphilic actinomycete strain, designated EGI 80088<SUP>T</SUP>, was isolated from a saline-alkali soil sample from Xinjiang province, north-west China, and subjected to a polyphasic taxonomic characterization. Strain EGI 80088<SUP>T</SUP> formed fragmented aerial hyphae and short spore chains, and rod-like spores aggregated at maturity. Whole-cell hydrolysates of the isolate contained <SMALL>ll</SMALL>-diaminopimelic acid as the diagnostic diamino acid, and glucosamine, mannose, galactose, glucose and rhamnose as the marker sugars. The major fatty acids identified (>5 %) were anteiso-C<SUB>15 : 0</SUB>, iso-C<SUB>15 : 0</SUB>, summed feature 4 (iso-C<SUB>17 : 1</SUB>I/anteiso-C<SUB>17 : 1</SUB>B), iso-C<SUB>16 : 0</SUB> and anteiso-C<SUB>17 : 0</SUB>. The predominant menaquinone was MK-9(H<SUB>4</SUB>). The G+C content of the genomic DNA of strain EGI 80088<SUP>T</SUP> was 70.6 mol%. EGI 80088<SUP>T</SUP> showed the highest 16S rRNA gene sequence similarity to its closest phylogenetic neighbour <I>Haloactinopolyspora alba</I> YIM 93246<SUP>T</SUP> (98.5 %). The DNA–DNA relatedness value of the strain EGI 80088<SUP>T</SUP> and <I>H. alba</I> YIM 93246<SUP>T</SUP> was 59.3±5.2 %. On the basis of morphological, chemotaxonomic and phylogenetic characteristics and DNA–DNA hybridization data, strain EGI 80088<SUP>T</SUP> represents a novel species of the genus <I>Haloactinopolyspora</I>, for which the name <I>Haloactinopolyspora alkaliphila</I> sp. nov. (type strain EGI 80088<SUP>T</SUP> = BCRC 16946<SUP>T</SUP> = JCM 19128<SUP>T</SUP>) is proposed. The description of the genus <I>Haloactinopolyspora</I> has also been emended.</P>

        Intravenous Tenecteplase for Acute Ischemic Stroke Within 4.5–24 Hours of Onset (ROSE-TNK): A Phase 2, Randomized, Multicenter Study

        Background and Purpose Intravenous tenecteplase (TNK) efficacy has not been well demonstrated in acute ischemic stroke (AIS) beyond 4.5 hours after onset. This study aimed to determine the effect of intravenous TNK for AIS within 4.5 to 24 hours of onset. Methods In this pilot trial, eligible AIS patients with diffusion-weighted imaging (DWI)-fluid attenuated inversion recovery (FLAIR) mismatch were randomly allocated to intravenous TNK (0.25 mg/kg) or standard care within 4.5–24 hours of onset. The primary endpoint was excellent functional outcome at 90 days (modified Rankin Scale [mRS] score of 0–1). The primary safety endpoint was symptomatic intracranial hemorrhage (sICH). Results Of the randomly assigned 80 patients, the primary endpoint occurred in 52.5% (21/40) of TNK group and 50.0% (20/40) of control group, with no significant difference (unadjusted odds ratio, 1.11; 95% confidence interval 0.46–2.66; <i>P</i>=0.82). More early neurological improvement occurred in TNK group than in control group (11 vs. 3, <i>P</i>=0.03), but no significant differences were found in other secondary endpoints, such as mRS 0–2 at 90 days, shift analysis of mRS at 90 days, and change in National Institutes of Health Stroke Scale score at 24 hours and 7 days. There were no cases of sICH in this trial; however, asymptomatic intracranial hemorrhage occurred in 3 of the 40 patients (7.5%) in the TNK group. Conclusion This phase 2, randomized, multicenter study suggests that intravenous TNK within 4.5–24 hours of onset may be safe and feasible in AIS patients with a DWI-FLAIR mismatch.

        Reliable control of magnetic vortex chirality in asymmetrically optimized magnetic nanodisk

        Magnetic vortex has attracted attention in the field of information storage because their topological spin structures with chiral bistable states. If the vortex core polarity and vortex circulation sense can be controlled simultaneously in a nanodisk, which will be more beneficial to realize the multi-bit ultrahigh density storage. In this paper, a reliable control scheme for magnetic vortex chirality is proposed by optimizing the structure of Pac- Man-like nanodisk. The results show that the polarity and circulation of the vortex can be controlled simultaneously by changing the direction of the global magnetic field, and even the chiral states of the vortex can be determined by detecting the stray field distribution on the surface of the nanodisk. The optimized Pac-Man-like nanodisk provide an experimental method for the control and detection of magnetic vortex chirality, which will be beneficial to the realization of multi-bit magnetic storage or magnetic logic technology in the future.

        Stability analysis of rock slopes using strength reduction adaptive finite element limit analysis

        The main aim of this study is to propose an effective approach to conduct the stability analysis of rock slopes. Based on the strength reduction technology, the recent developed adaptive finite element limit analysis is adopted to investigate the stability of rock slopes for the first time. The rock mass strength is described by the latest generalized Hoek-Brown failure criterion. To better estimate the rock slope stability, the rock mass disturbance D and slope angle β are also incorporated into the determination of factor of safety by implementing the disturbance weighting factor ζ and the slope angle weighting factor fβ, respectively. The factors of safety obtained from this study are validated by those from existing works based on two real slope cases. For practical use, the factors of safety for the cases of β=45° in undisturbed rock masses are provided in the form of design charts. Finally, a set of design charts for ζ and a fitting formula for fβ are presented and discussed.

        <P>Matricellular protein SPON2 acts as an HCC suppressor and utilizes distinct signaling events to perform dual functions in HCC microenvironment.</P><P>Tumor-associated macrophages (TAM) represent key regulators of the complex interplay between cancer and the immune microenvironment. Matricellular protein SPON2 is essential for recruiting lymphocytes and initiating immune responses. Recent studies have shown that SPON2 has complicated roles in cell migration and tumor progression. Here we report that, in the tumor microenvironment of hepatocellular carcinoma (HCC), SPON2 not only promotes infiltration of M1-like macrophages but also inhibits tumor metastasis. SPON2-α4β1 integrin signaling activated RhoA and Rac1, increased F-actin reorganization, and promoted M1-like macrophage recruitment. F-Actin accumulation also activated the Hippo pathway by suppressing LATS1 phosphorylation, promoting YAP nuclear translocation, and initiating downstream gene expression. However, SPON2-α5β1 integrin signaling inactivated RhoA and prevented F-actin assembly, thereby inhibiting HCC cell migration; the Hippo pathway was not noticeably involved in SPON2-mediated HCC cell migration. In HCC patients, SPON2 levels correlated positively with prognosis.         Manipulator-actuated Adaptive Integrated Translational and Rotational Stabilization for Spacecraft in Proximity Operations with Control Constraint

        This paper tackles the problem of integrated translation and rotation stabilization of the spacecraft in proximity operations by proposing a novel manipulator actuation strategy. To do so, by theoretically integrating the attitude/position motion of the spacecraft and the joint motion of the manipulator, a coupled translational and rotational kinematics of the spacecraft with a single space manipulator mounted is formulated, where system unknown parameters and residual system momentum are taken into account and analyzed. Taking the joint motion as the control input, a projection-based adaptive control scheme is then developed such that the translation and rotation of the spacecraft can be robustly stabilized with the manipulator-based actuation. The closed-loop asymptotic stability is guaranteed within Lyapunov framework. Meanwhile, considering the constrained joint motion of the manipulator, the resulting control constraint issue is handled by developing an optimization based bound analysis method, which also facilitates the determination of control parameters. Two scenario numerical simulations demonstrate the effect of the designed control scheme.

        Transient propagation of BmLV and dysregulation of gene expression in nontarget cells following BmLV infection

        Bombyx mori latent virus (BmLV), a novel positive-strand RNA virus was first identified in the B. mori cultured BmN cell line. Whether the infectivity of BmLV to silkworm larvae and non-silkworm cells is connected with dysregulation of gene expression are not well understood. A complete sequence of BmLV genomic RNA was identified and revealed that a fragment with 495 nt in length was deleted from the RNA-dependent RNA poly merase (RdRp) gene in some BmLV genomic RNAs. Studies on the infectivity of BmLV to nontarget cells showed that BmLV can infect silkworm larvae, Spodoptera frugiperda Sf-9 and H1299 lung cancer cells with transient propagation. The dysregulation of gene expression of Sf-9 cells followed by BmLV infection was analyzed. Out of 743 differentially expressed genes (DEGs), 300 were upregulated and 443 were downregulated. Gene Ontology (GO) analysis indicated the DEGs were enriched into oxidoreductase activity for CH-NH 2 group donors, gluta mate biosynthetic process, response to stress and proteasome core complex. KEGG enrichment analysis showed that DEGs were mainly enriched into sulfur metabolism, RNA degradation, proteasome, pentose and glucuronate interconversions. Undesirable nutrients and temperature factors contributed to the propagation of BmLV in Sf-9 cells. Additionally, the Imd and RNAi pathways were activated by BmLV infection without stimulating Toll and JAK-STAT pathways. Therefore, it is suggested that BmLV is originated from plants, which can enter nontarget cells with transient propagation. The transient infection of BmLV may not only be regulated by Imd and RNAi immune pathways but also mediated by dysregulation of gene expression.

      • Lack of any Prognostic Value of Body Mass Index for Patients Undergoing Chemoradiotherapy for Esophageal Squamous Cell Carcinoma

        Background: The relationship between body mass index(BMI) and outcomes after chemoradiotherapy(CRT) has not been systematically addressed. The purpose of this study was to evaluate the effect of BMI on survival in patients with esophageal squamous cell carcinoma (ESCC). Materials and Methods: Sixty ESCC cases were retrospectively reviewed in this study. Patient overall survival(OS) and disease-free survival (DFS) were compared between two groups (BMI< $24.00kg/m^2$ and $BMI{\geq}24.00kg/m^2$). Results: There were 41 patients in the low/normal BMI group (BMI< $24.00kg/m^2$) and 19 in the high BMI group ($BMI{\geq}24.00kg/m^2$). No significant differences were observed in patient characteristics between these. We found no difference in 2-year OS and DFS associated with BMI (p=0.763 for OS; p=0.818 for DFS) using the Kaplan-Meier method. Univariate analysis revealed that higher clinical stage was prognostic for worse 2-year OS and DFS, metastasis for 2-year OS, lymph node status for 2-year DFS, while age, gender, smoking, drinking, tumor location and BMI were not prognostic. There were no differences in the 2-year OS (hazard ratio=1.117; p=0.789) and DFS(hazard ratio=1.161; p=0.708) between BMI groups in multivariate analysis, whereas we found statistical differences in the 2-year OS and DFS associated with clinical stage, gender and tumor infiltration (p<0.04), independent of age, smoking, drinking, tumor location, the status of lymph node metastases and BMI. Conclusions: BMI was not associated with survival in patients with ESCC treated with CRT as primary therapy. BMI should not be considered a prognostic factor for patients undergoing CRT for ESCC.

        Comparative transcriptome analysis of wing discs from Bombyx mori and Bombyx mandarina

        The insect wing is developed from the wing imaginal disc which is designed from the embryonic ectoderm. To get insight into gene expression profiles in wing discs of Bombyx mori during metamorphosis, we compared the gene expression in the wing between B. mori and B. mandarina moth through RNA-seq. Out of total valid reads identified from the 5th day of 5th instar larvae of silkworm (L5), 7th day of pupae (P7), 1st day of moth (M1) and 1st day of wild silkworm moth (WM1), 20,092,004, 29,251,647, 24,654,695 and 19,753,089 reads were mapped to the mRNA reference sequences of silkworm, respectively. 9229, 7048, 9268 and 6701 differentially expressed genes (DEGs) were respectively recorded in P7 vs L5, M1 vs P7, M1 vs L5 and WM1 vs M1. Further, the peroxisome, ribosome, endocytosis and oxidative phosphorylation pathways were significantly regulated in the metamorphosis of the silkworm. Our study identified 16 orthologous genes with a positive selection from M1, which might be subjected to artificial selection in the domestication of B. mori and would play vital roles in the flight of B. mandarina.
