Crack identification in a rotor with an open crack

A finite element (FE) model, which is based on a transfer matrix analysis and local flexibility theorem, is introduced for crack identification of a static (non-rotating) rotor with an open crack. Through numerical simulation, the effects of crack location and crack depth on the mode shapes and the changes in the eigenfrequencies of the cracked rotor are investigated. A crack identification algorithm that makes use of the translations of the first mode at two symmetric points and the contour diagram of crack location versus crack depth for the first two given normalized eigenfrequencies is proposed to estimate the crack location and depth in the rotor. Two illustrative examples are demonstrated and compared for availability and validity of the proposed algorithm.

Quaternary ammonium salt modified montmorillonite (QASMMT) nanoplatelets were well dispersed in poly(ether ketone ketone) (PEKK) polymers to make pleasing fifth generation (5G) substrates. Substantially smaller dielectric properties and linear thermal expansion coefficient (LCTE) were found for each PEKKa xQASMMTy film series filled with small and appropriate QASMMT loadings. The dielectric constant (εr) and dielectric loss (tan δ) values acquired for each PEKKa xQASMMTy film series reduced to a lowest value as QASMMT loadings approached 3wt%. Satisfactory εr (2.59, 2.71, and 2.80 at 1 MHz), tan δ (0.0032, 0.0038, and 0.0042 at 1 MHz) and LCTE (~36.8×10-6/℃, 38.8×10-6/℃ and 40.5×10-6/℃) for 5G communication were acquired for PEKKa 97QASMMT3 films filled with only 3wt% optimum loading of QASMMT nano-platelets. In the meantime, the onset degradation temperatures acquired for each PEKKa xQASMMTy film series increased substantially with increasing QASMMT loadings. All free volume properties, such as, the radius of the free-volume-cavity or free-volume-cavity numbers per unit volume evaluated for every PEKKa xQASMMTy film sequence enlarged to a largest value, as QASMMT loadings approached an appropriate value of 3wt%. Substantial smaller εr and tan δ were acquired for PEKKa and PEKKa xQASMMTy with larger free volume properties. Possible reasons accounting for these substantially diminished dielectric and LCTE properties of PEKKa xQASMMTy films are proposed.

This study describes the use of quantitative PCR (qPCR) to determine gene copy number in Beauveria bassiana genome. This procedure has been improved by generating a reference strain to calibrate PCR amplification efficiency and using a γ-tubulin gene as internal control. The reference strainwas constructed by introducing one copy of phosphinothricin resistance gene and two copies of green fluorescent protein gene into the wild-type strain. Finally, this improved procedurewas validated via determining the gene copy number in the previously characterized transformants, and displays a great potential in rapid detection of gene copy number in the genome of transformant.

The yellow mealworm beetle, Tenebrio molitor (Coleoptera: Tenebrionidae), is a common insect pest that damages stored grain. The insect olfactory co-receptor (Orco) is a transmembrane protein localized on the surface of the dendrites of olfactory sensory neurons and plays an essential role in the olfactory perception. In this study, an Orco ortholog (named as Tmol\Orco) inT.molitorwas characterized. The deduced amino acid sequence of Tmol\Orco shared significant high identities with Orcos in other insect species from distinct Orders. Reverse transcription quantitative PCR (RT-qPCR) analysis showed that Tmol\Orco was predominantly expressed in the antennae of both male and female adults, and the difference between male antennae and female antennae was not significant. Double-stranded RNA (dsRNA) directly against the Tmol\Orco mRNA was synthesized and injected into the T. molitor male adults, RT-qPCR results demonstrated that the transcription level of Tmol\Orco was significantly reduced at 6 d after the injection. The mate-seeking behavior of Tmol\Orco-silenced beetles was significantly inhibited. Compare to the control individuals, the percentage of Tmol\Orco-silenced beetles successfully find mateswas significantly decreased and the average time they spent for searching were significantly increased. These results provided first direct evidence that suppression of Tmol\Orco transcription contributes to the decline in mate recognition of T. molitor

Pemetrexed is an antifolate agent which has been used for treating malignant pleural mesothelioma and non small lung cancer in the clinic as a chemotherapeutic agent. In this study, pemetrexed inhibited cell growth and induced G1 phase arrest in the A549 cell line. To explore the molecular mechanisms of pemetrexed involved in cell growth, we used a two-dimensional polyacrylamide gel electrophoresis (2-DE) proteomics approach to analyze proteins changed in A549 cells treated with pemetrexed. As a result, twenty differentially expressed proteins were identified by ESI-Q-TOF MS/MS analysis in A549 cells incubated with pemetrexed compared with non-treated A549 cells. Three key proteins (GAPDH, HSPB1 and EIF4E) changed in pemetrexed treated A549 cells were validated by Western blotting. Accumulation of GAPDH and decrease of HSPB1 and EIF4E which induce apoptosis through inhibiting phosphorylation of Akt were noted. Expression of p-Akt in A549 cells treated with pemetrexed was reduced. Thus, pemetrexed induced apoptosis in A549 cells through inhibiting the Akt pathway.

The yttria-stabilized zirconia (YSZ) nanocrystals with uniform size, high purity, and high degree of crystallinity, were prepared by ultrasonic–microwave-assisted method. The structure, optical properties and morphologies of YSZ nanocrystals were characterized by X-ray powder diffraction (XRD), Raman spectroscopy, UV–vis absorption, scanning electron microscope (SEM) and transmission electron microscopy (TEM). The SEM and TEM images of the YSZ nanocrystals indicate that the product is a mono-dispersion structure with an average particle size of about 25 nm.

Immunological functions of heat shock proteins (HSPs) have long been recognized. In this study we aimed to efficiently purify HSP70 from renal cell carcinoma and test it as a tumor antigen for pulsing dendritic cells in vitro. HSP70 was purified from renal cell carcinoma specimens by serial column chromatography on Con A-sepharose, PD-10, ADP-agarose and DEAE-cellulose, and finally subjected to fast protein liquid chromatography (FPLC). Dendritic cells derived from the adherent fraction of peripheral blood mononuclear cells were cultured in the presence of IL-4 and GM-CSF and exposed to tumor HSP70. After 24 hours, dendritic cells were phenotypically characterized by flow cytometry. T cells obtained from the non-adherent fraction of peripheral blood mononuclear cells were then co-cultured with HSP70-pulsed dendritic cells and after 3 days T cell cytotoxicity towards primary cultured renal cell carcinoma cells was examined by Cell Counting Kit-8 assay. Dendritic cells pulsed in vitro with tumor-derived HSP70 expressed higher levels of CD83, CD80, CD86 and HLA-DR maturation markers than those pulsed with tumor cell lysate and comparable to that of dendritic cells pulsed with tumor cell lysate plus TNF-${\alpha}$. Concomitantly, cytotoxic T-lymphocytes induced by HSP70-pulsed dendritic cells presented the highest cytotoxic activity. There were no significant differences when using homologous or autologous HSP70 as the tumor antigen. HSP70 can be efficiently purified by chromatography and induces in vitro dendritic cell maturation in the absence of TNF-${\alpha}$. Conspecific HSP70 may effectively be used as a tumor antigen to pulse dendritic cells in vitro.

Objectives: To explore the impact of low- vs conventional-dose chemotherapy via transcatheter arterial chemo-embolization (TACE) on serum fibrosis indicators and treatment efficacy of hepatocellular cancer patients (HCC). Materials and Methods: Patients fulfilling the eligibility criteria were assigned to TACE in Group A (with low-dose chemotherapy) or Group B (conventional-dose chemotherapy). Four serum fibrosis related indicators, hyaluronic acid(HA), human pro-collagen type-III (hPC-III), laminin (LN), and collagen type-IV(IV-C) before TACE were compared with the values 7 days after TACE. The response rate and survival time were also compared between the two groups. Results: Fifty patients with HCC were enrolled in this study, including 25 in Group A and 25 in Group B. No significant differences were detected between the two groups in the four indicators before TACE. After TACE, the value of the four serum indicators increased significantly in Group B. However, no significant differences regarding these four indicators were found in Group A after TACE. Significant differences were demonstrated between the two groups after TACE, but median survival time and 1 or 2 year overall survival rates did not differ (P>0.05). Conclusions: Low-, compared with conventional-dose chemotherapy exerts the same impact on the variation of fibrosis related indicators and has no influence on median survival time and survival rate after TACE in HCC patients.
Experimental study of the association between sandstone size effect and strain rate effect

The association between sandstone size effect and strain rate effect were investigated experimentally with a split Hopkinson pressure bar (SHPB) system. The sandstone samples with Φ50 mm and different lengths were loaded under the constant ratio of punch velocity to sample length to study their size effects. Sandstone samples with constant length of 25 mm were taken as the reference to study their strain rate effects. Results indicate that, under the same velocity of the punch, strain rate of each sandstone sample is inversely proportional sample length; dynamic strength of sandstone increases with the strain rate and the length to diameter ratio (L/D), and presents a quadratic curvilinear relation with strain rate while presenting a cubic curvilinear relation with sample L/D; the reasonable L/D of Φ50 mm sandstone samples ranges from 0.5 to 0.8; that dissipated energy can present a fixed proportional relation with punch kinetic energy is unrelated to sample length.
