        Effect of Glutamate on the Vestibulo-Solitary Projection after Sodium Nitroprusside-Induced Hypotension in Conscious Rats

        Orthostatic hypotension is most common in elderly people, and its prevalence increases with age. Attenuation of the vestibulo-sympathetic reflex (VSR) is commonly associated with orthostatic hypotension. In this study, we investigated the role of glutamate on the vestibulo-solitary projection of the VSR pathway to clarify the pathophysiology of orthostatic hypotension. Blood pressure and expression of both pERK and c-Fos protein were evaluated in the nucleus tractus solitarius (NTS) after microinjection of glutamate into the medial vestibular nucleus (MVN) in conscious rats with sodium nitroprusside (SNP)-induced hypotension that received baroreceptor unloading via sinoaortic denervation (SAD). SNP-induced hypotension increased the expression of both pERK and c-Fos protein in the NTS, which was abolished by pretreatment with glutamate receptor antagonists (MK801 or CNQX) in the MVN. Microinjection of glutamate receptor agonists (NMDA or AMPA) into the MVN increased the expression of both pERK and c-Fos protein in the NTS without causing changes in blood pressure. These results indicate that both NMDA and AMPA receptors play a significant role in the vestibulo-solitary projection of the VSR pathway for maintaining blood pressure, and that glutamatergic transmission in this projection might play a key role in the pathophysiology of orthostatic hypotension.

        Numerical analysis and fluid-solid coupling model test of filling-type fracture water inrush and mud gush

        The geological conditions surrounding the Jijiapo Tunnel of the Three Gorges Fanba Highway project in Hubei Province are very complex. In this paper, a 3-D physical model was carried out to study the evolution process of filling-type fracture water inrush and mud gush based on the conditions of the section located between 16.040 km and 16.042 km of the Jijiapo Tunnel. The 3-D physical model was conducted to clarify the effect of the self-weight of the groundwater level and tunnel excavation during water inrush and mud gush. The results of the displacement, stress and seepage pressure of fracture and surrounding rock in the physical model were analyzed. In the physical model the results of the model test show that the rock displacement suddenly jumped after sustainable growth, rock stress and rock seepage suddenly decreased after continuous growth before water inrushing. Once water inrush occured, internal displacement of filler increased successively from bottom up, stress and seepage pressure of filler droped successively from bottom up, which presented as water inrush and mud gush of filling-type fracture was a evolving process from bottom up. The numerical study was compared with the model test to demonstrate the effectiveness and accuracy of the results of the model test.

        Foxl2 of the Hong Kong catfish (Clarias fuscus): cDNA cloning, tissue distribution and changes in gene expression towards methyltestosterone, estradiol and letrozole exposure of the fries during gonadal differentiation

        Winged helix/forkhead transcription factor gene 2 (Foxl2) plays a crucial role during early ovarian development in fish. Sex steroids and aromatase inhibitors can regulate gonadal differentiation in teleosts. To address the role of Foxl2 in gonadal differentiation, foxl2 was isolated and characterized from the Hong Kong catfish, Clarias fuscus. Tissue distribution analysis by quantitative real-time PCR (qPCR) showed that the foxl2 mRNA was highest in the ovaries; moderate in the female pituitary and hypothalamus; but weak in the forebrain, liver, testis and male pituitary, with a sexually dimorphic pattern in which there was higher expression overall in females versus males. Treatment of the fries during the period of gonadal differentiation [2–30 days post hatching (dph)] with 17amethyltestosterone (MT), an aromatase inhibitor (letrozole) and 17b-estradiol (E2) affected the expression of foxl2. The expression of foxl2 was highest before gonadal differentiation (12 dph), but decreased significantly after gonadal differentiation (18-30 dph). In addition, MT and letrozole were able to down-regulate foxl2, but E2 up-regulated foxl2 during gonadal morphological differentiation. These results indicate the significant roles of Foxl2 in the gonadal differentiation of C. fuscus.

        Osteosarcoma is a common malignant tumor of bone, but mechanisms underlying its development are still unclear. At present, it is believed that the inhibition of normal apoptotic mechanisms is one of the reasons for the development of tumors, so specific stimulation of tumor cell apoptosis can be considered as an important therapeutic method. Livin, as a member of the newly discovered inhibitor of apoptosis proteins (IAPs) family, has specifically high expression in tumor tissues and can inhibit tumor cell apoptosis through multiple ways, which can become a new target for malignant tumor treatment (including osteosarcoma) and might of great significance in the clinical diagnosis of tumors and the screening of anti-tumor agents and carcinoma treatment.

        Role of peripheral vestibular receptors in the control of blood pressure following hypotension

        Hypotension is one of the potential causes of dizziness. In this review, we summarize the studies published in recent years about the electrophysiological and pharmacological mechanisms of hypotension-induced dizziness and the role of the vestibular system in the control of blood pressure in response to hypotension. It is postulated that ischemic excitation of the peripheral vestibular hair cells as a result of a reduction in blood flow to the inner ear following hypotension leads to excitation of the central vestibular nuclei, which in turn may produce dizziness after hypotension. In addition, excitation of the vestibular nuclei following hypotension elicits the vestibulosympathetic reflex, and the reflex then regulates blood pressure by a dualcontrol (neurogenic and humoral control) mechanism. In fact, recent studies have shown that peripheral vestibular receptors play a role in the control of blood pressure through neural reflex pathways. This review illustrates the dual-control mechanism of peripheral vestibular receptors in the regulation of blood pressure following hypotension.

        Expression profiles and ligand-binding properties of two odorant-binding proteins from Grapholita molesta (Busck) (Lepidoptera: Tortricidae)

        The ligand-binding function of odorant binding proteins (OBPs) that are expressed exclusively or enriched in antennae are well-studied, whilst the ligand binding properties of relatively low expressed OBPs in insect antennae are still unclear. Here, two low expressed GmolOBPs (namely GmolOBP12 and GmolOBP16) were cloned based on the antennal transcriptome of Grapholita molesta, and then their expression profiles and binding properties were investigated via real-time quantitative PCR (qRT-PCR) and fluorescence binding assays, respectively. Multiple sequence alignment and phylogenetic tree analyses displayed that both of GmolOBP12 and GmolOBP16 possessed the typical six-cysteine motifs unique to the classic OBPs subfamily and were classified as such. Although the abundance of transcripts of GmolOBP12 and GmolOBP16 were relatively low compared to that of many other OBPs in the antennal transcriptome of G. molesta, qRT-PCR results indicated that the transcript levels of GmolOBP12 and GmolOBP16 in antennae were significantly higher than those in other tissues. GmolOBP12 displayed higher expression level in male antennae than in female antennae, while the transcript level of GmolOBP16 in antennae was similar for both sexes. Both recombinant GmolOBP12 and GmolOBP16 exhibited strong binding affinities to the sex pheromone (Z)-8-dodecenyl alcohol and host-plant volatile pear ester. Besides, rGmolOBP12 showed outstanding binding affinities to (E)-2-hexenal, benzaldehyde, 1-hexanol, and (Z)-3-hexenyl acetate with K i values of 4.21, 6.81, 4.68 and 4.68 μM, respectively. rGmolOBP16 had moderate binding abilities with hexanal, decanal, 1-hexanol, methyl myristate and benzonitrile. We speculated that GmolOBP12 may have dual functions in recognition of green leaf volatiles and sex pheromone components and GmolOBP16 may participate in the detection of host-plant volatiles in chemoreception.

        CpG methylated ribosomal RNA genes in relation to wing polymorphism in the rice pest Sogatella furcifera

        Sogatella furcifera is amajor rice pest that exhibits wing dimorphism. The methylation-sensitive representational difference analysis (MS-RDA) is widely used in plants and some animals to identify methylation differences in genomic DNA regions. However the method has been applied to insects very less. The objective of the current research was tomonitor differential cytosine methylations at CCGG sequences inmacropterous and brachypterous adults of female S. furcifera to determine the association between gene methylation and wing phenotypes using MS-RDA. After the second subtractive hybridization, two differentially methylated DNA fragments were obtained. These DNA fragments were then cloned into the pGEM-T vector and sequenced. One fragment fromthe positive hybridization was 464 bp, and highly similar to the 18S ribosomal RNA (rRNA) genes from some animals. Another fragment from the reverse hybridization was 453 bp, and homologous to the 28S rRNA gene of S. furcifera with a similarity rate as high as 99%. These results provide potential evidence that DNA methylation of rRNA genes may be related to wing phenotype variations in S. furcifera, and will facilitate future studies on the epigenetic mechanisms of insect wing polymorphism.

      • Validation of prognostic scores to predict short-term mortality in patients with HBV-related acute-on-chronic liver failure: The CLIF-C OF is superior to MELD, CLIF SOFA, and CLIF-C ACLF

        ABSTRACT: Acute-on-chronic liver failure (ACLF) in chronic hepatitis B (CHB) patients has a high short-term mortality. Identification of effective models to predict the short-term mortality may enable early intervention and improve patients’ prognosis. We aim to assess the performance of the CLIF Consortium Organ Failure score (CLIF-C OFs), CLIF sequential organ failure assessment score (CLIF-SOFAs), CLIF Consortium ACLF score (CLIF-C ACLFs), ACLF grade, and model for end-stage liver disease score (MELDs) in predicting the short-term mortality in CHB patients with ACLF.Among the 155 consecutive adult patients with liver failure as a discharge diagnosis were screened, and all the patients were treated at the Department of Infectious Diseases, Huashan Hospital, Fudan University (Shanghai, China) from January 2010 to February 2016. The diagnosis of ACLF was based on the criteria formalized by the ACLF consensus recommendations of the Asian Pacific Association for the Study of the Liver (APASL). Diagnostic accuracy for predicting short-term (28-day) mortality was calculated for CLIF-C OFs, CLIF-SOFAs, CLIF-C ACLFs, ACLF grade, and MELDs in all patients.One hundred fifty-five consecutive adult liver failure patients were screened and 85 patients including 73 males and 12 females were enrolled. Overall, the 28-day transplant-free mortality was 32% in all patients, and 100% in those with severe early course (ACLF-3). The area under the receiver operating characteristic curve (AUROC) of CLIF-C OFs (AUROC: 0.906, P = .0306, compared with MELDs) was higher than those of CLIF-SOFAs (AUROC: 0.876), CLIF-C ACLFs (AUROC: 0.858), ACLF grade (AUROC: 0.857), and MELDs (AUROC: 0.838) for predicting short-term mortality. The cut-point for baseline CLIF-C OFs in predicting death was 8.5, with 67% sensitivity, 90% specificity, and AUROC of 0.906 (95% CI: 0.8450–0.9679).The results indicate that short-term mortality is high in patients with ACLF and CLIF Consortium Organ Failure score is superior to MELD, CLIF SOFA, and CLIF-C ACLF in predicting its short-term mortality.
