      • Research on Tourism Policies and Laws for Revitalizing Regional Tourism Industry: Taking the Offshore Duty-Free Policy of China Hainan International Tourism Island as an Example

        This study is conducted to analyze the improvement and progress of regional tourism vitality by explaining the changes of tourism policies and laws in Hainan Province of China. Focusing on the offshore duty-free policy of Hainan International Tourism Island, it has collected data from relevant laws and regulations and explores the impact of policies on the tourism industry. Combined with the domestic strategic tendency, it also makes a comprehensive comparison and in-depth interpretation of the policies. By analyzing the implementation of the offshore duty-free policy, it proposes feasible and necessary suggestions for the adjustment of the offshore duty-free policy.

        RO 및 NF막에서의 부식질에 의한 Fouling 및 미량오염물질의 제거

        This study investigated the phenomena of membrane fouling by NOM and the effect of the fouling on removal of micro-pollutants. NOM has a great effect on decline of permeate flux. Permeate flow rate was reduced by 88% in RO and 34.8% in NF for 323hr operation period. Removal rate of UV_(254), is 87.4% in RO and 78.5% in NF and removal rate of DOC is 42.7% in RO and 32.9% in NF for 2㎎/l humic acid. Removal efficiency of the micmpollutants by the RO and NF membranes fouled by humic acid was mostly lower than that by the new membrane. The concentration polarization which affects the flux and the rejection was thought to occur in the active layer of the membrane, as the membrane was getting fouled.

        Identification and expression profiles of putative chemosensory protein genes in Cnaphalocrocis medinalis (Lepidoptera: Pyralidae)

        Insect chemosensory proteins (CSPs) are small, water-soluble proteins which can bind and transport hydrophobic odorants through sensillum lymph to activate odorant receptors, thus play important roles in the olfactory recognition. In the present study, by searching the transcriptome data sets, a total of 22 candidate CSP genes were identified from Cnaphalocrocis medinalis, a serious lepidopteran rice pest in Asia. CSP proteins encoded by these genes showed typical characteristics: an N-terminal signal peptide, four conserved cysteine residues and the pattern of cysteine spacing. The expression profiles of these putative CSP genes were investigated using real-time quantitative PCR. The results showed that five genes (CmedCSP4, CmedCSP8, CmedCSP11, CmedCSP18 and CmedCSP21) were expressed primarily in antennae, suggesting their involvement in olfactory processes; whilst other genes are mainly expressed in non-olfactory tissues, such as abdomen and legs, indicating a broader physiological function for these CSPs. The findings will lead to a better understanding of the molecular mechanisms of chemoreception in C. medinalis.

      • 5-Aza-2'-deoxycytidine Induces Hepatoma Cell Apoptosis via Enhancing Methionine Adenosyltransferase 1A Expression and Inducing S-Adenosylmethionine Production

        In hepatocellular cancer (HCC), lack of response to chemotherapy and radiation treatment can be caused by a loss of epigenetic modifications of cancer cells. Methionine adenosyltransferase 1A is inactivated in HCC and may be stimulated by an epigenetic change involving promoter hypermethylation. Therefore, drugs releasing epigenetic repression have been proposed to reverse this process. We studied the effect of the demethylating reagent 5-aza-2'-deoxycitidine (5-Aza-CdR) on MAT1A gene expression, DNA methylation and S-adenosylmethionine (SAMe) production in the HCC cell line Huh7. We found that MAT1A mRNA and protein expression were activated in Huh7 cells with the treatment of 5-Aza-CdR; the status of promoter hypermethylation was reversed. At the same time, MAT2A mRNA and protein expression was significantly reduced in Huh7 cells treated with 5-Aza-CdR, while SAMe production was significantly induced. However, 5-Aza-CdR showed no effects on MAT2A methylation. Furthermore, 5-Aza-CdR inhibited the growth of Huh7 cells and induced apoptosis and through down-regulation of Bcl-2, up-regulation of Bax and caspase-3. Our observations suggest that 5-Aza-CdR exerts its anti-tumor effects in Huh7 cells through an epigenetic change involving increased expression of the methionine adenosyltransferase 1A gene and induction of S-adenosylmethionine production.

        Thermal-hydraulic research on rod bundle in the LBE fast reactor with grid spacer

        The research on the flow and heat transfer characteristics of lead bismuth(LBE) is significant for the thermal-hydraulic calculation, safety analysis and practical application of lead-based fast reactors(LFR). In this paper, a new CFD model is proposed to solve the thermal-hydraulic analysis of LBE. The model includes two parts: turbulent model and turbulent Prandtl, which are the important factors for LBE. In order to find the best model, the experiment data and design of 19-pin hexagonal rod bundle with spacer grid, undertaken at the Karlsruhe Liquid Metal Laboratory (KALLA) are used for CFD calculation. Furthermore, the turbulent model includes SST k u and k ε; the turbulent Prandtl includes Cheng-Tak and constant (Prt ¼1.5,2.0,2.5,3.0). Among them, the combination between SST k u and Cheng-Tak is more suitable for the experiment. But in the low Pe region, the deviation between the experiment data and CFD result is too much. The reason may be the inlet-effect and when Pe is in a low level, the number of molecular thermal diffusion occupies an absolute advantage, and the buoyancy will enhance. In order to test and verify versatility of the model, the NCCL performed by the Nuclear Thermal-hydraulic Laboratory (Nuthel) of Xi'an Jiao tong University is used for CFD to calculate. This paper provides two verification examples for the new universal model

      • Expression of Tumor Necrosis Factor-alpha-induced Protein 8 in Pancreas Tissues and its Correlation with Epithelial Growth Factor Receptor Levels

        Tumor necrosis factor (TNF)-alpha-induced protein 8 (TNFAIP8 or TIPE) is a recently identified protein considered to be associated with carcinogenesis. To investigate its expression pattern in pancreatic cancer patients and to analyse its correlation with clinicopathological significance and the expression levels of epithelial growth factor receptor (EGFR), immunohistochemistry was performed to detect the TNFAIP8 and EGFR proteins in pancreatic cancers, pancreatitis tissues, and healthy controls. The results showed stronger staining of TNFAIP8 protein in pancreatic cancer tissues compared with normal pancreas tissue. Furthermore, in 56 patients with pancreatic cancer, the expression levels of TNFAIP8 in patients with low tumor stage was higher than that with high tumor stage, and correlated with tumor staging and lymph node metastasis (P<0.05). Furthermore, TNFAIP8 expression positively correlated with EGFR levels (r=0.671135, P<0.05). These results indicate that TNFAIP8 may play important roles in the progression of pancreatic cancer.

        Identification and expression profiles of putative cytochrome P450 monooxygenase genes from Cnaphalocrocis medinalis (Lepidoptera: Pyralidae)

        In insects, cytochrome P450 monooxygenases (P450s) have received considerable attention for their roles in the detoxification of harmful chemicals and insecticide resistance. The rice leaf‐folder, Cnaphalocrocis medinalis, is a severe lepidopteran rice pest. In this research, 36 putative P450 genes were identified from the transcriptome dataset of C. medinalis. Phylogenetic analysis showed that these genes fell into four clans, including the mitochondrial clan (6 genes), CYP2 clan (5 genes), CYP3 clan (16 genes) and CYP4 clan (9 genes), and were classified into 19 families and 30 subfamilies. Since the CYP6 P450 family in insects is the major family related to the detoxification of xenobiotics, the expression profiles of five genes in the CYP6 family were determined by real‐time quantitative PCR. These genes were differentially expressed in various larval tissues, including the midgut, Malpighian tubules and fat body. Of these, CYP6AW1 and CYP6CV1 were mainly expressed in the midgut, while CYP6AE76 was highly expressed in the fat body. CYP6AB62 and CYP6AE28 were ubiquitously expressed. The transcription of CYP6AE28 was significantly induced by abamectin at LD20 dose. Taken together, the results provided useful information for the characteristics of P450s in C. medinalis and for future functional studies.

        Molecular characterization of a delta class glutathione S-transferase gene from the black cutworm Agrotis ipsilon

        In insects, glutathione S-transferases (GSTs) play essential roles in the detoxification of xenobiotic toxins and elimination of oxidative stress induced by toxic compounds. In the present study, a delta class GST gene (AiGSTd) was identified and characterized in the black cutworm, Agrotis ipsilon (Lepidoptera: Noctuidae). The deduced protein sequence of AiGSTd contained highly conserved features of GST enzymes and shared high identities with its orthologs from other lepidopteran species. Recombinant AiGSTD protein was expressed in Escherichia coli and purified. The protein displayed the GSH-dependent conjugating activity toward the substrate 1-chloro-2,4-dinitrobenzene (CDNB). Moreover, AiGSTD had the ability to protect DNA from oxidative damage, and the E. coli cells overexpressing AiGSTD showed long-term resistance to oxidative stress. The AiGSTd transcripts were most abundant in the larval midgut. Exposure to chlorpyrifos and lambda-cyhalothrin increased lipid peroxidation in larvae and significantly upregulated AiGSTd expression levels. This study is the first report of molecular characterization of a GST in A. ipsilon, and the present study suggest that AiGSTD might be involved in protecting against the oxidative stress induced by insecticides.

      • Semaphoring mAb: a New Guide in RIT in Inhibiting the Proliferation of Human Skin Carcinoma

        Semaphoring is a transmembrane receptor which participates in many cytokine-mediated signal pathways that are closely related to the angiogenesis, occurrence and development of carcinoma. The present study was designed to access the effect of mono-antibody (mAb) guided radioimmunotherapy (RIT) on skin carcinoma and investigate the potential mechanisms. Semaphoring mAb was acquired from mice (Balb/c), purified with rProtein A column; purity, concentration and activity were tested with SDS-PAGE and indirect ELISA; specificity and expression on the cutanuem carcinoma line and tissue were tested by Western blotting; morphology change was assessed by microscopy. MTT assay and colony inhibition tests were carried out to test the influence on the proliferation of tumor cells; Western blotting was also carried out for expression of apoptosis-associated (caspase-3, Bax, Bcl-2) and proliferation-related (PI3K, p-Akt, Akt, p-ERK1/2, ERK1/2) proteins and analyse the change in signal pathways (PI3K/Akt and MEK/ERK). The purity of purified semaphorin mAb was 96.5% and the titer is about $1{\times}10^6$. Western blotting showed semaphoring mAb to have specifically binding stripes with semaphoring b1b2 protein, B16F10, and A431 cells at 39KDa, 100KDa and 130KDa, respectively. Positive expression was detected both in cutanuem carcinoma line and tissue and it mostly located in cell membranes. MMT assay revealed dose-relate and time-relate inhibitory effect of semaphorin mAb on A431 and B16F10. Colony inhibition tests also showed dose-relate inhibitory effects. Western blotting demonstrated the expression of apoptosis and proliferation-related protein and changes in signal pathway. In conclusion, we demonstrated that semaphorin is highly expressed on the tumor cell-surfaces and RIT with semaphorin mAb has effect in i nhibiting proliferation and accelerating apoptosis of tumor cells.
