        Substituting oat hay or maize silage for portion of alfalfa hay affects growth performance, ruminal fermentation, and nutrient digestibility of weaned calves

        Objective: The impact of forage feeding strategy on growth performance, ruminal fermentation and nutrient digestibility in post-weaning calves was investigated. Methods: Forty-five female Holstein calves (body weight [BW] = 79.79±0.38 kg) were enrolled in the 35-d study at one week after weaning and randomly assigned to one of three dietary treatments. All diets were fed as total mixed ration containing 60% (dry matter [DM] basis) of basal starter feed and 40% (DM basis) of forage, but varied in composition of forage source including i) alfalfa (40% DM, AH); ii) alfalfa hay (26.7% DM)+oat hay (13.3% DM; OH); iii) alfalfa hay (26.7% DM)+corn silage (13.3% DM; WS). Results: Dry matter intake was not different among treatment groups (p>0.05). However, BW (p<0.05) and average daily gain (p<0.05) of calves fed AH and OH were greater than WS-fed calves, whereas heart girth was greater in OH-fed calves than those fed AH and WS (p<0.05). Ruminal fermentation parameters including proportion of butyric acid, acetated-to-propionate ratio, concentration of total volatile fatty acid, protozoal protein, bacterial protein, and microbial protein in rumen were the highest in OH (p<0.05) and the lowest in WS. Compared with the AH and WS, feeding oat hay to postweaning calves increased crude protein digestibility (p<0.05), and decreased duration of diarrhea (p<0.05) and fecal index (p<0.05). Conclusion: Our results suggested that partially replacing alfalfa hay with oat hay improved ruminal fermentation, nitrogen utilization, and reduced incidence of diarrhea in post-weaning dairy calves.

      • Prognostic Significance of Hes-1, a Downstream Target of Notch Signaling in Hepatocellular Carcinoma

        Background: Hairy and enhancer of split 1 (Hes-1) protein is a downstream target of Notch signaling and is a basic helix-loop-helix transcriptional repressor. However, definitive evidence for a role in hepatocellular carcinoma (HCC) cells has not been reported. Here, Hes-1 was revealed to an important component of the Notch signaling cascade in HCC cell lines possessing different potential for lung metastasis. Materials and Methods: RNAi mediated by plasmid constructs was used to analyze the role of Hes-1 in MHCC-97L HCC cells by assessing proliferation, apoptosis, cell migration and matrigel invasion following transfection. Hes-1 protein expression analysis in HCC tissue was also conducted by immunohistochemistry. Results: Our studies revealed that Hes-1 was decreased in HCC cell lines with higher lung metastasis potential at both the mRNA and protein levels. Down-regulation of the Hes-1 gene in MHCC-97L cells resulted in increased cell proliferation, reduced apoptosis and increased migration and invasion. Conclusions: Hes-1 has potential prognostic value in post-surgical HCC patients and may be an independent prognostic indicator for overall survival and tumor recurrence. These findings have important implications for understanding the mechanisms by which Hes-1 participates in tumor proliferation and invasion.

      • Expression of TS, RRM1, ERCC1, TUBB3 and STMN1 Genes in Tissues of Non-small Cell Lung Cancer and its Significance in Guiding Postoperative Adjuvant Chemotherapy

        Background: To explore the expression of TS, RRM, ERCC1, TUBB3 and STMN1 genes in the tissues of patients with non-small cell lung cancer (NSCLC) and its significance in guiding the postoperative adjuvant chemotherapy. Materials and Methods: Real time polymerase chain reaction (PCR) was applied to detect the expression of TS, RRM, ERCC1, TUBB3 and STMN1 genes in the tissues of NSCLC patients so as to analyze the relationship between the expression of each gene and the clinical characteristics and to guide the postoperative individualized chemotherapy according to the detection results of NSCLC patients. Results: Expression of TS gene was evidently higher in patients with adenocarcinoma than those with non-adenocarcinoma (P=0.013) and so was the expression of ERCC1 (P=0.003). The expression of TUBB3 gene was obviously higher in NSCLC patients in phases I/II and IV than those in phase III ($P_1=0.021$; $P_2=0.004$), and it was also markedly higher in patients without lymph node metastasis than those with (P=0.008). The expression of STMN1 gene was apparently higher in patients in phase I/II than those in phase IV (P=0.002). There was no significant difference between the rest gene expression and the clinical characteristics of NSCLC patients (P>0.05). Additionally, the diseasefree survival (DFS) was significantly longer in patients receiving gene detections than those without (P=0.021). Conclusions: The selection of chemotherapeutic protocols based singly on patients' clinical characteristics has certain blindness. However, the detection of tumor-susceptible genes can guide the postoperative adjuvant chemotherapy and prolong the DFS of NSCLC patients.

        Baicalin attenuates TNBS-induced colitis in rats by modulating the Th17/Treg paradigm

        Baicalin, a flavonoid, has a wide range ofpharmacological properties, including immunomodulation. The objective of this study was to investigate the effect ofbaicalin on the balance of T helper 17 (Th17) and regulatoryT (Treg) cells in a colitis model. The rat colitis modelwas induced by 2,4,6-trinitrobenzene sulfonic acid(TNBS). Baicalin (10 ml/kg, each) or mesalazine (positivecontrol) was then administered orally for 7 days. Inflammatoryand immunological responses were evaluated bypathology, enzyme-linked immunosorbent assay, real-timepolymerase chain reaction, western blot analysis, and flowcytometry. Our study showed that baicalin not only significantlyattenuated TNBS-induced colitis by reducing thedisease activity index as well as macroscopic and microscopicscores, but it also improved the weight loss andshortening of the colon. Baicalin treatment also induced asignificant decrease in the levels of inflammatory mediators,including the myeloperoxidase activity, the levels oftumor necrosis factor a, IL-1b, and Th1-related cytokinesIL-12 and IFN-c. Furthermore, the beneficial effects ofbaicalin seem to be associated with regulation of the Th17and Treg paradigm. We found that administration ofbaicalin significantly downregulated the number of Th17cells and the levels of Th17-related cytokines (IL-17 andIL-6) and retinoic acid receptor-related orphan receptor ct. In contrast, there was an increase in Treg cells numbers,Treg-related cytokines transforming growth factor-b andIL-10, and forkhead box P3. Our results suggest that theanti-inflammatory effect of baicalin may be linked tomodulation of the balance between Th17 and Treg cells inTNBS-induced ulcerative colitis.

        Effect of Diallyl Trisulfide on the Pharmacokinetics of Dipyridamole in Rats

        This study was aimed to evaluate the effect of diallyl trisulfide (DATS), a major component derived from garlic used to inhibit platelet thromboxane formation, on the pharmacokinetics of dipyridamole. Pharmacokinetic parameters of dipyridamole were determined in rats following intragastric (80 mg/kg suspension or 40 mg/kg solution) or intravenous (3 mg/kg) administration of dipyridamole with coadministration (20 mg/kg) and long-term pretreatment of DATS (10 or 20 mg/kg/day for 15 consecutive days). In addition, everted gut sac models were used to assess transepithelial transport of dipyridamole and the effect of DATS on the intestinal absorption of dipyridamole. After coadministration and long-term pretreatment of DATS, significantly lower C_max and AUC_(0–240 h were observed for intragastric administration of dipyridamole,whereas little change was noted after intravenous dipyridamole administration. After adding DATS (10 and 50 μg/mL) in the everted gut sacs, absorption of dipyridamole was remarkably decreased in the ileum and jejunum (p < 0.01). In conclusion, DATS reduced the oral exposure of dipyridamole in rats likely by the modification of the dissolution rate and intestinal absorption of dipyridamole, indicating that combined use of DATS or DATS-containing supplements with dipyridamole may require caution as low plasma concentrations of dipyridamole may lead to a subtherapeutic effect of this agent.

        Significant Approaches to Promote Post-Cure Reaction of Bulky Polyimides with Pendant Phthalonitrile Unit

        Approaches to promote the post-cure reaction of polyimide with pendant phthalonitrile unit (CN-PI) werefirst realized by introducing of hydroxyl groups via chemical copolymerization or physical blending. The post-curereaction of the CN-PI was performed at 250 or 300 oC, and monitored by various techniques, such as infrared spectroscopy(IR), differential scanning calorimetry (DSC), wide-angle X-ray diffraction (WAXD), scanning electronmicroscopy (SEM), and thermogravimetric analysis (TGA). A significant result was found in that the trend of theincreasing glass transition temperature Tg with nitrile conversion followed DiBenedetto’s equation. This provided achance to deeply discuss the relationship between molecular structure, segmental mobility, and cure behavior of CNPI. Chemical structure changes and morphology evolution of the polymer during post-cure process were investigatedby IR, WAXD, and SEM. The thermal and mechanical properties of polymer films showed it to be a thermallystable and strong material.

        Pharmacokinetics of Carbamazepine Polymorphs and Dihydrate in Rats, Related to Dogs and Humans

        Species differences in the oral pharmacokinetics and absolute bioavailability (F_abs) of carbamazepine polymorphs (form I and form III) and dihydrate were studied. The pharmacokinetics of each form was investigated in rats following a single oral/intravenous administration of 10 mg/kg and an oral dose of 80 mg/kg, which were compared with the published data obtained from dogs and humans. No significant differences were found in their C_max, T_max, AUC_(0-∞) and F_abs among the forms at the low dose. However, significant differences were observed at the high dose. The F_abs of each form was markedly reduced with increasing of doses in species (e.g. F_abs in rats ranged from > 82% to 38.4% - 56.0%). At a comparable dose, the C_max, and AUC_(0-∞)of rats and humans were about 3-10 times higher than in dogs. The absorption rate of form III in rats exhibited a similar trend to that in humans, and was far higher in dogs. A multi-peak phenomenon in plasma curves was observed in rats and humans, but not in dogs. In conclusion, rats appear to be a better predictor of carbamazepine polymorphs absorbed in humans,and form III may be more suitable as a pharmaceutical crystal.

        Knockdown of lncRNA PVT1 Inhibits Vascular Smooth Muscle Cell Apoptosis and Extracellular Matrix Disruption in a Murine Abdominal Aortic Aneurysm Model

        This study was designed to determine the effects of the long non-coding RNA (lncRNA) plasmacytoma variant translocation 1 (PVT1) on vascular smooth muscle cell (VSMC) apoptosis and extracellular matrix (ECM) disruption in a murine abdominal aortic aneurysm (AAA) model. After injection of PVT1-silencing lentiviruses, AAA was induced in Apolipoprotein E-deficient ($ApoE^{-/-}$) male mice by angiotensin II (Ang II) infusion for four weeks. After Ang II infusion, mouse serum levels of pro-inflammatory cytokines were analysed, and aortic tissues were isolated for histological, RNA, and protein analysis. Our results also showed that PVT1 expression was significantly upregulated in abdominal aortic tissues from AAA patients compared with that in controls. Additionally, Ang II treatment significantly increased PVT1 expression, both in cultured mouse VSMCs and in AAA murine abdominal aortic tissues. Of note, the effects of Ang II in facilitating cell apoptosis, increasing matrix metalloproteinase (MMP)-2 and MMP-9, reducing tissue inhibitor of MMP (TIMP)-1, and promoting switching from the contractile to synthetic phenotype in cultured VSMCs were enhanced by overexpression of PVT1 but attenuated by knockdown of PVT1. Furthermore, knockdown of PVT1 reversed Ang II-induced AAA-associated alterations in mice, as evidenced by attenuation of aortic diameter dilation, marked adventitial thickening, loss of elastin in the aorta, enhanced aortic cell apoptosis, elevated MMP-2 and MMP-9, reduced TIMP-1, and increased pro-inflammatory cytokines. In conclusion, our findings demonstrate that knockdown of lncRNA PVT1 suppresses VSMC apoptosis, ECM disruption, and serum pro-inflammatory cytokines in a murine Ang II-induced AAA model.

        Influence of different parameters on nonlinear friction-induced vibration characteristics of water lubricated stern bearings

        To investigate the mechanism of friction-induced vibration and noise of ship water lubricated stern bearings, a two-degree-of-freedom (2-DOF) nonlinear self-excited vibration model is established. The novelty of this work lies in the detailed analysis of influence of different parameters on the stability and nonlinear vibration characteristics of the system, which provides a theoretical basis for the various friction vibration and noise phenomenon and has a very important directive meaning for low noise design of water lubricated stern bearings. The results reveal that the change of any parameter, such as rotating speed of shaft, contact pressure, friction coefficient, system damping and stiffness, has an important influence on the stability and nonlinear response of the system. The vibration amplitudes of the system increase as (a) rotating speed of shaft, contact pressure, and the ratio of static friction coefficient to dynamic friction coefficient increase and (b) the transmission damping between motor and shaft decreases. The frequency spectrum of the system is modulated by the first mode natural frequency, which is continuous multi-harmonics of the first mode natural frequency. The response of the system presents a quasi-periodic motion.
