Seventeen compounds derived from traditional Chinese medicines (TCMs) were tested for their antiviral activity against porcine reproductive and respiratory syndrome virus (PRRSV) in vitro. Visualization with the cytopathologic effect (CPE) assay and the 3-(4, 5-dimethyithiazol- 2-yl)-2,5-diphenyltetrazolium bromide test were used to determine the 50% cytotoxic concentration (CC50) and 50% effective concentration (EC50) in cultured Marc-145 cells. Among the tested compounds, chlorogenic acid and scutellarin showed potential anti-PRRSV activity. The EC50 values were 270.8 ± 14.6 μg/ml and 28.21 ± 26.0 μg/ml and the selectivity indexes were >5.54 and 35.5, respectively. The time-of-addition and virucidal assay indicated that the anti-PRRSV activity of the two compounds could be due to their inhibiting the early stage of virus replication and/or inactivating the virus directly. The inhibition of the virus attachment was not observed in the adsorption inhibition assay. The inhibition ratios of chlorogenic acid and scutellarin were, respectively, 90.8% and 61.1% at the maximum non-cytotoxic concentrations. The results have provided a basis for further exploration of their antiviral properties and mechanisms in vivo. We believe that the chlorogenic acid and scutellarin have a great potential to be developed as new anti-PRRSV drugs for clinical application.
Energy Generation Coupled to Azoreduction by Membranous Vesicles from Shewanella decolorationis S12

Previous studies have demonstrated that Shewanella decolorationis S12 can grow on the azo compound amaranth as the sole electron acceptor. Thus, to explore the mechanism of energy generation in this metabolism, membranous vesicles (MVs) were prepared and the mechanism of energy generation was investigated. The membrane, which was fragmentized during preparation, automatically formed vesicles ranging from 37.5-112.5 nm in diameter under electron micrograph observation. Energy was conserved when coupling the azoreduction by the MVs of an azo compound or Fe(III) as the sole electron acceptor with H2, formate, or lactate as the electron donor. The amaranth reduction by the vesicles was found to be inhibited by specific respiratory inhibitors, including Cu2+ ions, dicumarol, stigmatellin, and metyrapone, indicating that the azoreduction was indeed a respiration reaction. This finding was further confirmed by the fact that the ATP synthesis was repressed by the ATPase inhibitor N,N`-dicyclohexylcarbodiimide (DCCD). Therefore, this study offers solid evidence of a mechanism of microbial dissimilatory azoreduction on a subcell level.

Microorganisms determine the overall biofilter performance under specific operating conditions. The toluene removal and process robustness of a laboratoryscale,ceramisite-based biotrickling filter inoculated with Burkholderia sp. strain T3 (BTFb) were compared with those of another biotrickling filter inoculated with activated sludge (BTFa) for 3 months under various operating conditions. Denaturing gradient gel electrophoresis was applied to visualise the bacterial community of the BTFa and BTFb. Real-time polymerase chain reaction was performed to determine the genes coding for toluenedegrading enzymes. Burkholderia sp. strain T3, which possesses the major toluene-degrading genes in BTFb, was traced in the BTFb bacterial community. The strain was found to stabilize the relative quantity steadily at higher than 60% during toluene biofiltration. Thus, BTFb performed more efficiently than BTFa as evidenced by achieving 98.86% toluene removal efficiency (RE) on 3 day, critical elimination capacity (EC) of 234.23 ± 10.54 g/m3/h, and rapid restoration of the initial RE and EC levels within 3 day of reoperation, even after 1 month of shutdown. The efficiency of BTFb is also evident by the stabilised RE and EC levels within a wide temperature range and a gradually decreasing system pH. Maintaining the pressure drop levels below 150 Pa during prolonged operation also contributed to the efficiency of BTFb. Thus, based on the study results,we propose that Burkholderia sp. strain T3 is a highly efficient and applicable inoculum for toluene biofiltration.
Transmembrane Protein 166 Expression in Esophageal Squamous Cell Carcinoma in Xinjiang, China

Objective: Transmembrane protein 166 (TMEM166) expression in esophageal squamous cell carcinoma (ESCC) and remote normal esophageal tissues was examined to assess any role in tumour biology. Methods: TMEM166 mRNA expression in 36 cases with ESCC (36 tumour samples, 36 remote normal esophageal tissue samples) was detected by RT-PCR. TMEM166 protein expression was analysed in paraffin-embedded tissue samples from the same cases by immunohistochemistry. Results: Semi-quantitative analysis showed TMEM166 mRNA expression in ESCCs to be significantly lower than in remote normal esophageal tissues ($0.759{\pm}0.713$ vs. $2.622{\pm}1.690$, P=0.014). TMEM166 protein expression was also significantly reduced (69.4% vs. 94.4%, P<0.01). Conclusion: TMEM166 mRNA and protein expression demonstrated significant reduction in ESCCs compared with remote esophageal tissues, albeit with no correlation with tumour size, differentiation, stage, and lymph node metastasis, suggesting a role in regulating autophagic and apoptotic processes in the ESCC.
Model reduction of a multibody system including a very flexible beam element

An optimum method to reduce the system size of a multibody system including a very flexible beam element is presented. In this paper,the absolute nodal coordinate formulation (ANCF) is used to describe the motion of the flexible beam in this system. Based on continuummechanics approach, the bending stiffness matrix of the system is assumed to be a constant matrix when the axial strain is small. Thus, free-interface method, in which modal coordinate vector is free from the boundary region, is used to reduce the degrees of freedomof a flexible beam with sliding joints. Finally, two numerical examples are presented to verify the efficiency and accuracy of this method. The results show that the present method can reduce the computing time with precision. Meanwhile, a deformation energy criterion ofmode truncation is established.

Hydrochar was produced from hydrothermal treatment of corn straw black liquor. Response surface methodology (RSM) and the central composite design (CCD) were employed for determination of optimal char with maximum Pb2+ removal capacity. The operational parameters such as hydrothermal temperature (oC), duration (min) and solid liquid ratio (LSR) were chosen as independent variables in CCD. The statistical analysis indicates that the effects of hydrothermal temperature, duration, LSR and combined effect of hydrothermal temperature and duration are all significant for the Pb2+ removal capacity. The optimal condition for achieving the maximum Pb2+ adsorption capacity is obtained as the following: hydrothermal temperature (205 oC), duration (28min), LSR (12) with Pb2+ removal capacity reaching 47mg/g. The BET specific surface area of char prepared at optimal conditions could reach 85m2/g.

Background: Soft tissue sarcomas (STS) are a heterogeneous group of tumors, and approximately 40-50% of patients with STS develop metastatic disease. The median overall survival of those patients was 12 months and their 5-year survival rate was 8%. Therefore, study on more effective treatment, especially the targeting therapies, is urgently needed. Objective: To evaluate the efficacy and safety of Endostar$^{(R)}$ combined with chemotherapy in patients with advanced STS. Methods: A retrospective case-series study was conducted in Cancer Institute of PLA, Xinqiao Hospital. A total of 71 patients suffering from advanced STS (IIB - IV) were included, of whom 49 cases treated with chemotherapy alone were defined as the control group and the rest 22 cases treated with the traditional chemotherapy combined with Endostar$^{(R)}$ were defined as the test group. The short-term therapeutic effects including objective response rate (ORR), disease control rate (DCR) and safety were evaluated in the two groups. In the follow-up, progression-free survival (PFS) and overall survival (OS) were also observed. Results: In the test and control groups, the ORR was 18.2% and 12.2%, respectively (P=0.767), and the DCR was 86.4% and 61.2%, respectively (P=0.034). The median time to progression in the test and control groups was 120 days and 70 days with significant difference (P = 0.017), while the median overall survival was 452 days and 286 days without significant difference (P=0.503). The one-year survival rate in the test group and control group was 56.2% and 35.4%, respectively, while the two-year survival rate was 30.2% and 26.5%, respectively. No significant difference in the side effects was found between the two groups. Conclusions: Endostar$^{(R)}$ combined with chemotherapy resulted in a higher DCR and longer PFS in the patients with advanced STS, and the toxicity was tolerable.

Acute graft-versus-host disease (GVHD) following liver transplantation is a rare but fatal complication. The correct diagnosis and management of GVHD after liver transplantation are still major challenges. Herein, we reported successful salvage treatment of acute GVHD by withdrawal of immunosuppression in a patient who presented with fever, skin rashes, and decreased blood cell counts after liver transplantation. This case highlights the need for awareness of drug-induced liver injury if liver function tests are elevated during treatment, especially in patients taking multiple potentially hepatotoxic drugs, such as broad-spectrum antibiotics. When occurs, an artificial liver support system is a useful tool to provide temporary support of liver function for the patient in the event of drug-induced liver injury.
