Seventeen compounds derived from traditional Chinese medicines (TCMs) were tested for their antiviral activity against porcine reproductive and respiratory syndrome virus (PRRSV) in vitro. Visualization with the cytopathologic effect (CPE) assay and the 3-(4, 5-dimethyithiazol- 2-yl)-2,5-diphenyltetrazolium bromide test were used to determine the 50% cytotoxic concentration (CC50) and 50% effective concentration (EC50) in cultured Marc-145 cells. Among the tested compounds, chlorogenic acid and scutellarin showed potential anti-PRRSV activity. The EC50 values were 270.8 ± 14.6 μg/ml and 28.21 ± 26.0 μg/ml and the selectivity indexes were >5.54 and 35.5, respectively. The time-of-addition and virucidal assay indicated that the anti-PRRSV activity of the two compounds could be due to their inhibiting the early stage of virus replication and/or inactivating the virus directly. The inhibition of the virus attachment was not observed in the adsorption inhibition assay. The inhibition ratios of chlorogenic acid and scutellarin were, respectively, 90.8% and 61.1% at the maximum non-cytotoxic concentrations. The results have provided a basis for further exploration of their antiviral properties and mechanisms in vivo. We believe that the chlorogenic acid and scutellarin have a great potential to be developed as new anti-PRRSV drugs for clinical application.

Hepatocellular carcinoma (HCC) is a major cause of cancer-related deaths, but its molecular mechanisms are not yet well characterized. Long noncoding RNAs (lncRNAs) play crucial roles in tumorigenesis, including that of HCC. However, the role of homeobox A11 antisense (HOXA11-AS) in determining HCC stem cell characteristics remains to be explained; hence, this study aimed to investigate the effects of HOXA11-AS on HCC stem cell characteristics. Initially, the expression patterns of HOXA11-AS and HOXA11 in HCC tissues, cells, and stem cells were determined. HCC stem cells, successfully sorted from Hep3B and Huh7 cells, were transfected with short hairpin or overexpression plasmids for HOXA11-AS or HOXA11 overexpression and depletion, with an aim to study the influences of these mediators on the self-renewal, proliferation, migration, and tumorigenicity of HCC stem cells in vivo. Additionally, the potential relationship and the regulatory mechanisms that link HOXA11-AS, HOXA11, and the Wnt signaling pathway were explored through treatment with Dickkopf-1 (a Wnt signaling pathway inhibitor). HCC stem cells showed high expression of HOXA11-AS and low expression of HOXA11. Both HOXA11-AS silencing and HOXA11 overexpression suppressed the self-renewal, proliferation, migration, and tumorigenicity of HCC stem cells in vivo, as evidenced by the decreased expression of cancer stem cell surface markers (CD133 and CD44) and stemness-related transcription factors (Nanog, Sox2, and Oct4). Moreover, silencing HOXA11-AS inactivated the Wnt signaling pathway by decreasing the methylation level of the HOXA11 promoter, thereby inhibiting HCC stem cell characteristics. Collectively, this study suggested that HOXA11-AS silencing exerts an antitumor effect, suppressing HCC development via Wnt signaling pathway inactivation by decreasing the methylation level of the HOXA11 promoter.

Objective: To investigate the effects of stellate ganglion block (SGB) on the peri-operative vasomotor cytokine content and intrapulmonary shunt in patients with esophagus cancer who underwent thoracotomy. Materials and Methods: Forty patients undergoing elective resection of esophageal cancer patients who had I~II American Society of Anesthesiologist (ASA) were randomly divided into total intravenous anesthesia group (group N, n=20) and total intravenous anesthesia combined with SGB group (group S, n=20, 0.12 mL/kg 1% lidocaine was used for SGB 10 min before induction). Heart rate (HR), mean arterial pressure (MAP), central venous pressure (CVP), mean pulmonary arterial pressure (MPAP) and continuous cardiac output (CCO) were continuously monitored. The blood from internal jugular vein was drawn respectively before induction ($T_0$), and 30 min ($T_1$), 60 min ($T_2$) and 120 min ($T_3$) after one-lung ventilation (OLV), and 30 min (T4) after two-lung ventilation. The contents of plasma endothelin (ET), nitric oxide (NO) and calcitonin gene-related peptide (CGRP) were detected with enzyme linked immunosorbent assay (ELISA). Meanwhile, arterial and mixed venous blood samples were collected for determination of blood gas and calculation of intrapulmonary shunt fraction (Qs/Qt). Results: During OLV, ET contents were increased significantly in two groups (P<0.05), and no significant difference was presented (P>0.05). NO content in group S was obviously higher than in group N at T3 (P<0.05), whereas CGRP content in group N was markedly lower than in group S at each time point (P<0.05). Qs/Qt was significantly increased in both groups after OLV, but there was no statistical significant regarding the Qs/Qt at each time point between two groups. Conclusions: Total intravenous anesthesia combined with SGB is conducive to regulation of perioperative vasomotor cytokines in thoracotomy, and has little effect on intrapulmonary shunt at the time of OLV.

Based on the development of new media technology and the background of COVID-19, this study aims to analyze the influence of console game characteristics on the intention of continuous use. The research summarizes the characteristics of console games into three factors: interaction, presence and nostalgia, and verifies the influence of console game characteristics on intention of continuous use, as well as the mediating effects of game flow, intimate relationship and brand attitude. The results show that the characteristics of console games are an important factor affecting game flow, which can improve the willingness of the intention of continuous use in console games. Second, game flow, intimate relationship, and brand attitude have positive effects on intention of continuous use, and there is a mediating effect between game characteristics and intention of continuous use. This research provides theoretical and practical implications for the development and operation of future console games.
Adaptive Motion Vector Smoothing for Improving Side Information in Distributed Video Coding
Guo, Jun,Kim, Joo-Hee Korea Information Processing Society 2011 Journal of information processing systems Vol.7 No.1
In this paper, an adaptive motion vector smoothing scheme based on weighted vector median filtering is proposed in order to eliminate the motion outliers more effectively for improving the quality of side information in frame-based distributed video coding. We use a simple motion vector outlier reliability measure for each block in a motion compensated interpolated frame and apply weighted vector median filtering only to the blocks with unreliable motion vectors. Simulation results show that the proposed adaptive motion vector smoothing algorithm improves the quality of the side information significantly while maintaining low complexity at the encoder in frame-based distributed video coding.
Anti-Hero as Promise of a Future New World: Interpreting the Image of Bloom in Ulysses
( Guo Jun ) 한국제임스조이스학회 2021 제임스조이스저널 Vol.27 No.2
James Joyce’s Ulysses is a mock epic that parodies the Homeric epic Odyssey, therefore, not only its style is no longer grand, its content is also trivial. What is most striking is that its protagonist Leopold Bloom, the Irish Ulysses, compared with his prototype Odysseus, is a radical anti-hero, devoid of any heroic quality, but just an apparently most common man. But at the same time, he is also a most uncommon man not only because of his deviation from an epic hero but especially because of his great difference from the stereotypical Dubliners of his time. He is thus a heteroglossia among his fellow citizens and provides Joyce a critical perspective both to deconstruct the status quo of that time and to imagine a new future for his own nation.

This study was carried out to investigate the relationship between endogenous polyamines (PAs) and floral bud differentiation in chrysanthemum (Chrysanthemum morifolium). In this study, PA content (free, bound, and conjugated) in apical buds, leaves, and roots changed appreciably during floral bud differentiation. PAs accumulated during series of processes such as floral induction, differentiation of floret primordia, and crown formation in apical buds; changes in PAs in apical buds may have a relationship with those in leaves and roots. The levels of free PAs and conjugated PAs [putrescine (Put) and spermine (Spm)] in apical buds rapidly increased during the initiation stage of floral bud differentiation, while free and conjugated spermidine (Spd) reached their highest levels at the stage of floret primordium differentiation. In the free, conjugated, and bound PA fractions, the changes in Spm content were negligible compared to those of Put and Spd throughout the experiment. These findings indicate that PAs participate in regulating the process of flower bud differentiation in chrysanthemum.

Background: Novel prognostic biomarkers or therapeutic molecular targets for laryngeal squamous cell carcinoma (LSCC) are an urgent priority. We here sought to identify multiple novel LSCC-associated genes. Methods: Using high-density microarray expression profiling, we identified multiple genes that were significantly altered between human LSCCs and paired normal tissues. Potential oncogenic functions of one such gene, DCUN1D5, were further characterized in vitro. Results: Our results demonstrated that DCUN1D5 was highly expressed in LSCCs. Overexpression of DCUN1D5 in vitro resulted in 2.7-fold increased cellular migration, 67.5% increased invasive capacity, and 2.6-fold increased proliferation. Endogenous DCUN1D5 expression was decreased in a time-dependent manner after genotoxic stress, and silencing of DCUN1D5 by siRNA decreased the number of cells in the S phase by 10.2% and increased apoptosis by 11.7%. Conclusion: Our data suggest that DCUN1D5 in vitro might have vital roles in DNA damage response, but further studies are warranted to assess its significance in vivo.
Control strategy for the substructuring testing systems to simulate soil-structure interaction
Jun Guo,Zhenyun Tang,Shicai Chen,Zhenbao Li 국제구조공학회 2016 Smart Structures and Systems, An International Jou Vol.18 No.6
Real-time substructuring techniques are currently an advanced experimental method for testing large size specimens in the laboratory. In dynamic substructuring, the whole tested system is split into two linked parts, the part of particular interest or nonlinearity, which is tested physically, and the remanding part which is tested numerically. To achieve near-perfect synchronization of the interface response between the physical specimen and the numerical model, a good controller is needed to compensate for transfer system dynamics, nonlinearities, uncertainties and time-varying parameters within the physical substructures. This paper presents the substructuring approach and control performance of the linear and the adaptive controllers for testing the dynamic characteristics of soil-structure-interaction system (SSI). This is difficult to emulate as an entire system in the laboratory because of the size and power supply limitations of the experimental facilities. A modified linear substructuring controller (MLSC) is proposed to replace the linear substructuring controller (LSC).The MLSC doesn\'t require the accurate mathematical model of the physical structure that is required by the LSC. The effects of parameter identification errors of physical structure and the shaking table on the control performance of the MLSC are analysed. An adaptive controller was designed to compensate for the errors from the simplification of the physical model in the MLSC, and from parameter identification errors. Comparative simulation and experimental tests were then performed to evaluate the performance of the MLSC and the adaptive controller.
A new dynamic updating key strategy for Wireless Mesh Networks
Jun-Guo Liao,Ming-Yan Wang 한국정보통신학회 2014 2016 INTERNATIONAL CONFERENCE Vol.6 No.1
In the security protocols of EMSA, the key updating strategy is an effective method to ensure the security of communication. For the existing programs of automatic updates, the PTK is updated through the complex 4-way handshake to produce each time. Once the update frequency of the PTK is greater, it will have a greater impact on throughput and delay of the network. On this basis, we propose a new strategy of dynamic key updating to ensure the safety and performance of wireless Mesh network. In the new strategy, when PTK updates, it only updates the TK communication key. The key updating is based on the initial seed, and then generate a new key by random function both sides at the same time. The analysis of performance with existing programs showed that the dynamic key updating strategy proposed in this paper have a larger increase in delay and throughput of the network.
