        Limiting Concentrate during Growing Period Affect Performance and Gene Expression of Hepatic Gluconeogenic Enzymes and Visfatin in Korean Native Beef Calves

        This study elucidated the effects of limited concentrate feeding on growth, plasma profile, and gene expression of gluconeogenic enzymes and visfatin in the liver of Hanwoo beef calves. The purpose of this study was to test that reducing the amount of concentrate would partially be compensated by increasing the intake of forage and by altering the metabolic status. The study utilized 20 Korean native beef calves (Hanwoo; 60 to 70 d of age) divided into two groups of 10 calves each for 158 d. Control group calves received the amount of concentrate as per the established Korean feeding standards for Hanwoo, whereas calves in the restricted group only received half the amount of concentrate as per standard requirements. Good quality forage (Timothy hay) was available for ad libitum consumption to both groups. Since calves were with their dam until 4 months of age in breeding pens before weaning, the intake of milk before weaning was not recorded, however, the concentrate and forage intakes were recorded daily. Body weights (BW) were recorded at start and on 10 d interval. Blood samples were collected at start and at 50 d interval. On the final day of the experiment, liver biopsies were collected from all animals in each group. The BW was not different between the groups at all times, but tended to be higher (p = 0.061) only at final BW in control than restricted group. Total BW gain in the control group was 116.2 kg as opposed to 84.1 kg in restricted group that led to average BW gain of 736 g/d and 532 g/d in respective groups, and the differences were significant (p<0.01). As planned, the calves in the control group had higher concentrate and lower forage intake than the restricted group. The plasma variables like total protein and urea were higher (p<0.05) in control than restricted group. The mRNA expressions for the gluconeogenic enzymes such as cytosolic phosphoenol pyruvate carboxykinase (EC 4.1.1.32) and pyruvate carboxylase (EC 6.4.1.1), and visfatin measured by quantitative real-time PCR in liver biopsies showed higher expression (p<0.05) in restricted group than control. Overall, restricting concentrate severely reduced the growth intensity and affected few plasma indices, and gene expression in liver was increased indicating that restricting concentrate in the feeding schemes during early growth for beef calves is not advocated.

        염분 수준이 육계 병아리의 생산성과 생리적인 변화에 미치는 영향

        본 연구는 사료와 음수에 함유된 염분 수준이 육계 초기 병아리에 미치는 영향을 조사하기 위하여 실시하였다. 1일령 초생추(Hubbard)를 공시하여 14일 동안 시험을 수행하였다. 시험 1에서, 사료내 염분 함량은 0, 0.25, 0.5, 0.75, 1.0, 2.0, 3.0, 4.0%로 조절하여 8처리구 처리구당 3반복, 반복당 12수씩 총 288수에 급여하였고, 시험 2에서, 음수내 염분 함 량은 0, 0.1, 0.2, 0.5, 1.0%로 조절하여 5처리구 처리구당 3반복, 반복당 12수씩 총 180수에 공급하였다. 시험 1의 경우에, 사료 섭취량과 증체량은 사료내 염분 함량이 증가할수록 감소하는 경향이었다. 특히 4% 처리구에서 각각 481 g과 168 g으로 가장 낮았으며, 폐사율은 75%로 가장 높았다. 시험 2의 경우에는, 음수 1% 처리구에서 사료 섭취량과 증체량은 각각 427 g과 162 g이었고, 폐사율은 77.8%로 가장 높았다. 염 중독 증상을 나타낸 닭들은 갈증, 식욕부진, 기립불능, 졸림, 경련 등을 나타내었고, 부검 소견에서는 복수증, 심낭수종, 심비대, 각종 장기의 출혈 등이 나타났다. 이상의 결과로 육계 초기에서 사료내 염분 함량이 3%, 음수내 염분 함량은 1% 이상에서 뚜렷한 생산성의 저하는 물론 생리 해부학적인 염분중독 증상이 나타남을 알 수 있다. To investigate the effects of salt contents in diet and water on the growth of broiler chicks, 288 heads of Hubbards strain chicks, were fed with com-soybean meal diets containing 0, 0.25, 0.5, 0.75 1.0, 2.0, 3.0, or 4.0% salt from 1d to l4d posthatching. And 180 chicks were also supplemented with salt to their water at 0, 0.1, 0.2, 0.5 or 1.0%. Feed intake and weight gain tended to decrease as the salt levels in diets increased and those were the lowest in 4% group as 481 g and 168 g, however, mortality was the highest in 4% group as 75.0%. As for the salt levels in water, feed intake and weight gain were low as 427 g and 162 g in 1% group, while mortality was 77.8%. Salt poisoning chicks were shown with thirsty, appetite deficiency, rising impossibility, sleepiness, or convulsions, and in autopsy, were shown with ascites, hydropericardium, cardiac hypertrophy, hemorrhage of eyelid, hemorrhage of trachea, hemorrhage of intestine, etc. Conclusively, salt content above 4% in diet or 1% in water could reduce the performance in broiler chicks.

      • Submergence-inducible and circadian rhythmic basic helix-loop-helix protein gene in Nicotiana tabacum

        Submergence stress leads to diverse changes in transcription and translation of genes involved in developmental and physiological metabolisms of plants. The basic helix-loop-helix (bHLH) protein family is one of the largest transcriptional factor families in plants, and has been shown to play pivotal roles in diverse biological responses. However, there has been no report on bHLH protein related to submergence stress response. In this study, a novel bHLH gene, NtbHLH, was isolated from tobacco (Nicotiana tabacum) by differential screening of a submergence-stress-induced cDNA library. NtbHLH cDNA is 1027bp in length, with an open reading frame (ORF) of 702 nucleotides encoding 233 amino acid residues that contain the bHLH domain. RNA-blot analyses showed that transcription of NtbHLH was induced by submergence stress, while cold, heat shock, and drought decreased its expression. The gene expression was down-regulated by gibberellins, but ABA and ethylene seemed not to affect it. It was also apparent that NtbHLH expression follows circadian rhythmicity. The electrophoretic mobility shift and chemical cross-linking assays showed that NtbHLH specifically binds to G-box and forms homo-dimers.

        Energy Distributions and Yields of Secondary Electrons from a Clean Au Target Bombarded by High-Energy H+, D+, He+, O2+, and Cl2+ ~ 5+

        The energy distributions and the yields of secondary electrons from a Au target with a clean surface were measured under bombardment with energetic ions such as H+, D+, He+, O2+, and Cl2+5+. A concentric spherical analyzer was manufactured to obtain the secondary electron yield. The retarded eld method was applied to measure the energy spectra of the secondary electrons. The secondary electron yield was founded to depend on the stopping power of the projectile in the target material, the charge states of the projectile particles, and the incident angle of the projectile particles; however, the shapes of energy distributions were found to be similar to for the ions used in this research.

        Korean Byungkyul - Citrus platymamma Hort.et Tanaka flavonoids induces cell cycle arrest and apoptosis, regulating MMP protein expression in Hep3B hepatocellular carcinoma cells

        <P>Citrus platymamma Hort.et Tanaka is an indigenous fruit of Jeju island in Korea. In this study the bioactivity of C. platymamma flavonoids were evaluated on human hepatoma Hep3B cell lines. Eleven flavonoids were identified from the peels of C. platymamma Hort.et Tanaka through high-performance liquid chromatography-Tandem mass spectrometry and the anticancer effect of these C. platymamma flavonoids on human hepatoma Hep3B were studied. Chromatin condensation was observed in Hep3B cells treated with C. platymamma flavonoids. DNA fragmentation was confirmed through agarose gel electrophoresis and TUNEL assay. An increase in the total apoptotic cells and G2/M cell cycle arrest with decreased protein expression of CDC25C, CDK1, cycl in B1 and p21 were observed in Hep3B cells treated with flavonoids of C. platymamma. Further, protein expression of Bcl-XL, Bax, caspase-3 and -9 were also modulated by C. platymamma flavonoids treatment indicating that cell death is through intrinsic apoptotic pathway. Moreover, C. platymamma flavonoids also regulated the phosphorylation of MAPKs, PI3K, and Akt in Hep3B cells. Relevant to inhibiting metastasis, C. platymamma treatment reduced wound closure of Hep3B cells and the protein expression of matrix metalloproteinase-2 and -9 were reduced in C. platymamma treated cells. The results show that C. platymamma flavonoids induce cell cycle arrest and apoptosis following activation of MAPKs and suppression of PI3K/Akt pathway which eventually inhibits cell migration in Hep3B cells. The finding provides evidence on biochemical activities of C. platymamma Hort.et Tanaka, which would be an essential agent for hepatocellular carcinoma (HCC) treatment.</P>

        한우 경제형질에 미치는 Mitochondrial DNA D-loop 영역의 염기서열 변이효과

        This study was performed to analyse the sequences of variations of mtDNA D-loop and their effects on carcass traits in Hnawoo(Korean cattle). The resulting sequences were compared with perviously published sequences for other cattle breeds(GenBank J01394). The PCR was used to amplify a total of 964 bp between nucleotide 15758 and 383 within D-loop region of mtDNA using specific primers. Twenty five polymorphic sites by nucleotide substitution were found in mtDNA of Hanwoo. The frequencies of positions at 169, 16042, 16093. 16119, 16255 and 16302 nt with high levels of sequence polymorphism were 0.891, 0.117, 0.109, 0.182, 0.197 and 0.117, respectively. The substitution effect at 169 and 16119 nt was found significant on marbling score. Also substitution effect at 169 and 16042 nt was highly significant(p<0.01) on backfat, thickness. Polymorphisrn of mtDNA sequence in D-loop region could be useful for the analysis of cytoplasmic genetic variation and associations with the other economically important traits and maternal lineage analysis in Hanwoo.

      • Intracellular imaging of zinc ion in living cells by fluorescein based organic nanoparticles

        <P><B>Abstract</B></P> <P>A fluorescein based organic nanoparticles were prepared by using synthesized <I>N</I>-(3′,6′-dihydroxy-3-oxospiro[isoindoline-1,9′-xanthene]-2-yl)-2-hydroxybenzamide(FB). The interaction of series of metal ions with the FB nanoparticles (FBNPs) was investigated by means of fluorescence measurement. The selective and sensitive fluorescence enhancement was observed to FBNPs by the addition of Zn<SUP>2+</SUP> ion in an aqueous medium. The interaction of FBNPs and Zn<SUP>2+</SUP> introduced chelation enhanced fluorescence that was unaltered even addition of other metal ion. The formation of excited state complexation of FBNPs-Zn<SUP>2+</SUP> was supported by UV–vis absorption titration, fluorescence lifetime and zeta particle size of FBNPs with and without addition of Zn<SUP>2+</SUP>. The mode of binding and interaction was confirmed by results procured through NMR and IR spectroscopy. The stoichiometry and binding constant estimated by Job’s plot and Benesi-Hildebrand plot. The limit of detection (LOD) for Zn<SUP>2+</SUP> was 0.0011 μg/mL. The fascinating feature of present nano probe (FBNPs) is that outstanding intracellular Zn<SUP>2+</SUP> sensor in MDA-MB-231 living cells with nominal toxicity.         The incremental sheet forming (ISF) method is known as valuable tools to make a rapid prototype which can be observed the forming shape and also able to re-design the part based on the requirements. In this study, we have applied the ISF to prototype of walkway and stair step used in factories and architecture fields. CAD model of walkway and stair step is initially designed and imported to CAM software to model and generate G-code files. These files are imported to CNC machine to make a female die and subsequently producing the prototype of walkway and stair step by using the ISF. The major safety requirement is to avoid permanent deflection due to low stiffness and the slip of passenger while walking on the metal walkway. If the part is not able to satisfy given requirements, then the design should be modified. After we got experience with more trial, the best shape, which was obtained by incremental sheet forming process, will be applied to actual manufacturing of walkway and stair step by punching and stamping process.
