Identification and characterization of heat shock proteins in a parasitic wasp Chouioia cuneae (Hymenoptera: Eulophidae)

        Li‐Na Pan,Feng-ZhuWANG,Xin-Yue ZHANG,Yan-Ni ZHAO,Geng-Ping ZHU,Min LI 한국곤충학회 2018 Entomological Research Vol.48 No.3

Heat shock proteins (HSPs) are known to be induced in response to various stress factors. Although HSPs have been studied in a number of insects, not much is known about HSPs in the natural enemies of insects, especially parasitoids. In this study, we identified and characterized five full‐length HSP genes (Cchsp40, Cchsp60, Cchsp70, Cchsp83, and Cchsp90) from an endoparasitic chalcid wasp, Chouioia cunea, which parasitizes the fall webworm, Hyphantria cunea pupae, a worldwide pest. The expression of Cchsps in response to temperature, pesticide stresses and UV radiation were also investigated by quantitative real‐time polymerase chain reaction (RT‐qPCR). The results showed that all five Cchsps were induced in response to hot and cold temperatures. Four pesticides induced the abundant expression of Cchsp70, Cchsp83 and Cchsp90 while ultraviolet radiation up‐regulated Cchsp40, Cchsp70, Cchsp83 and Cchsp90. These results indicate the different transcriptional profiles of the five different Cchsps in response to various abiotic stresses. The findings of this study provide insights into the response of C. cunea to abiotic stresses and insight into the use of this parasitoid in biological control strategies.

Improving the Capture-range Problem in Phase-diversity Phase Retrieval for Laser-wavefront Measurement Using Geometrical-optics Initial Estimates

        Li Jie Li,Wen Bo Jing,Wen Shen,Yue Weng,Bing Kun Huang,Xuan Feng 한국광학회 2022 Current Optics and Photonics Vol.6 No.5

To overcome the capture-range problem in phase-diversity phase retrieval (PDPR), a geometricaloptics initial-estimate method is proposed to avoid a local minimum and to improve the accuracy of laser-wavefront measurement. We calculate the low-order aberrations through the geometrical-optics model, which is based on the two spot images in the propagation path of the laser, and provide it as a starting guess for the PDPR algorithm. Simulations show that this improves the accuracy of wavefront recovery by 62.17% compared to other initial values, and the iteration time with our method is reduced by 28.96%. That is, this approach can solve the capture-range problem.

Genetic Diversity and Geographical Differentiation of Desmodium triflorum (L.) DC. in South China Revealed by AFLP Markers

        Mao-feng Yue,Ren-chao Zhou,Ye-lin Huang,Guo-rong Xin,Su-hua Shi,Li Feng 한국식물학회 2010 Journal of Plant Biology Vol.53 No.2

High levels of genetic variation enable species to adapt to changing environments and provide plant breeders with the raw materials necessary for artificial selection. In the present study, six AFLP primer pairs were used to assess the genetic diversity of Desmodium triflorum (L.)DC. from 12 populations in South China. A high percentage of polymorphic loci (P=76.16%) and high total gene diversity (HT=0.310) were found, indicating that the genetic diversity of D. triflorum is high at the species level. Genetic diversity was also relatively high at the population level (P=55.85%, He=0.230). The coefficient of gene differentiation among populations (GST) was 0.255, indicating that while most genetic diversity resided within populations, there was also considerable differentiation among populations. AMOVA also indicated 24.29% of the total variation to be partitioned among populations (ΦST=0.243). UPGMA clustering analysis based on genetic distances showed that the 12 populations could be separated into three subgroups: an eastern, a western, and a central-southern subgroup. However, a Mantel test revealed no significant correlation (r=0.286, p=0.983)between the geographical distances and genetic distances separating these populations; mountain barriers to gene flow and human disturbance may have confounded these correlations. The present study has provided some fundamental genetic data that will be of use in the exploitation of D. triflorum.

The role of CTNNB1 and LEF1 in feather follicles development of Anser cygnoides and Anser anser

        Yue Sun,Yuxuan Zhou,Petunia Msuthwana,Jing Liu,Chang Liu,Cornelius Tlotliso Sello,Yupu Song,Ziqiang Feng,Shengyi Li,Wei Yang,Yunpeng Xu,Xiaomin Yan,Chuanghang Li,Yujian Sui,Jingtao Hu,Yongfeng Sun 한국유전학회 2020 Genes & Genomics Vol.42 No.7

Background Wingless-types/beta-catenin (Wnt/β-catenin) signaling pathway is one of the most extensively studied transcriptional cascades involved in various types of organogenesis including embryonic and postnatal development. Downy feather quantity is primarily affected by follicular development and gene regulations. Objective This research was aimed to investigate the role of catenin beta-1(CTNNB1) and lymphoid enhancerbinding factor-1 (LEF1) on feather follicles development at different developmental stages. Methods Fluorescence quantitative PCR, Western-blot and immunohistochemical methods were used in Anser cygnoides and Anser anser embryos (E12, E13 E18, and E28) and after birth gosling stages (G18, G48, G88) for gene expression analysis. Results CTNNB1 and LEF1 genes were expressed in Anser cygnoides and Anser anser at different embryonic and after-birth gosling developmental stages and the expression levels were significantly different in different stages (p < 0.05). The mRNA expression of CTNNB1 and LEF1 genes reached the highest level at D88 in Anser cygnoides, while the highest expression levels were at D18 and D88 in Anser anser, and the expression levels of CTNNB1 genes at D88 in all embryonic stages were significantly lower than after-birth stages. CTNNB1 and LEF1 protein expression were the highest at E12 and E28 for Anser cygnoides feather follicles development. While at a similar stage for Anser anser, the expression of CTNNB1 and LEF1 protein was the highest at D48 and D18. Protein expression at embryonic stages was in the epidermis (E) and the hair basal plate (P), the expression site for after-birth stages was in the dermal papilla (DP). Conclusion Our study illustrated that CTNNB1 and LEF1 has an impact on Anser cygnoides and Anser anser feather follicles growth and development.

Intracerebral Microdialysis Technique and its Application on Brain Pharmacokinetic- Pharmacodynamic Study

        Yue-fang Pan,Jian Feng,Qiao-yuan Cheng,Fan-zhu Li 대한약학회 2007 Archives of Pharmacal Research Vol.30 No.12

Intracerebral microdialysis (IC-MD) has been developed as a well-validated and powerful technique for decades. As a practical sampling tool, it can gain the continuous dialysates of endogenous and exogenous substances in extracellular fluid (ECF) of awake freely moving animals. Also, variform IC-MD probes (IC-MDPs) have grown more exquisite. The implantation of the IC-MDP in certain tissue of brain allows monitor drug distribution and measure drug and corresponding neurotransmitters levels in brain ECF after administration for brain pharmacokineticpharmacodynamic (B-PK-PD) study. So it is suitable for IC-MD to B-PK-PD study (IC-MD/BPK- PD). The performance of IC-MD/B-PK-PD can not only elevate the degree of precision and accuracy of experimental data, minimize the individual difference by reduced number of animals, but also give important information for the prediction and optimization of drug effective dose in preclinical study. In this review, we have discussed various IC-MD/B-PK-PD studies of analgesic, antiepileptic and antidepressant drug. The role of IC-MD/B-PK-PD in confirming and assessing the drug effect before clinic trials is highlighted.

Recent insights into autophagy and metals/nanoparticles exposure

        Li Qiong,Feng Yajing,Wang Ruike,Liu Rundong,Ba Yue,Huang Hui 한국독성학회 2023 Toxicological Research Vol.39 No.3

Some anthropogenic pollutants, such as heavy metals and nanoparticles (NPs), are widely distributed and a major threat to environmental safety and public health. In particular, lead (Pb), cadmium (Cd), chromium (Cr), arsenic (As), and mercury (Hg) have systemic toxicity even at extremely low concentrations, so they are listed as priority metals in relation to their significant public health burden. Aluminum (Al) is also toxic to multiple organs and is linked to Alzheimer’s disease. As the utilization of many metal nanoparticles (MNPs) gradually gain traction in industrial and medical applications, they are increasingly being investigated to address potential toxicity by impairing certain biological barriers. The dominant toxic mechanism of these metals and MNPs is the induction of oxidative stress, which subsequently triggers lipid peroxidation, protein modification, and DNA damage. Notably, a growing body of research has revealed the linkage between dysregulated autophagy and some diseases, including neurodegenerative diseases and cancers. Among them, some metals or metal mixtures can act as environmental stimuli and disturb basal autophagic activity, which has an underlying adverse health effect. Some studies also revealed that specific autophagy inhibitors or activators could modify the abnormal autophagic flux attributed to continuous exposure to metals. In this review, we have gathered recent data about the contribution of the autophagy/mitophagy mediated toxic effects and focused on the involvement of some key regulatory factors of autophagic signaling during exposure to selected metals, metal mixtures, as well as MNPs in the real world. Besides this, we summarized the potential significance of interactions between autophagy and excessive reactive oxygen species (ROS)-mediated oxidative damage in the regulation of cell survival response to metals/NPs. A critical view is given on the application of autophagy activators/inhibitors to modulate the systematic toxicity of various metals/MNPs.
