        민간투자사업과 재정사업 성과에 관한 연구 - 전라북도 민간투자사업을 중심으로

        전통적 재정사업의 단점을 극복하고자 민간투자사업제도가 도입되었고, 지방자치단체 역시 다양한 민간투자사업을 시행해왔다. 2020년도 기준 전라북도 민간투자현황을 분석한 결과, 전라북도의 민간투자 사업은 대부분 하수관거정비임대형 사업에 해당하고 시설임대료 대부분이 국가 및 도비 부담으로 운영되고 있음을 알 수 있다. 이러한 사업 유형은 이론적으로 민간자본 투입을 통하여 사업효율성이 확보될 수 있는 장점이 있으나 장기적인 자본투입과 낮은 수익성으로 사실상 민간자본의 참여 유도가 쉽지 않다. 더욱이 전라북도 민간투자사업 상환금 세부내역을 살펴본 결과, 향후 전북지역 재정에 큰 부담으로 작용할 수 있는 우려가 있다. 민간투자사업의 성과를 높이는 것은 곧 정부 재정을 절감하고 재정운용의 효율성을 제고할 수 있는 길이라는 점에서 민간투자사업의 성과와 재정사업의 성과는 동전의 양면과 같다. 민간투자사업이 성과를 거두기 위해서는 무엇보다 사업추진 단계에서 경제성 분석을 통한 수요예측과 정확한 비용 산정이 중요하다. 과거 사업추진 단계에서 무산된 전주 경전철 사례를 되풀이하지 않기 위해서는 철저한 사전 분석 결과를 주민에게 공개하고 민간투자사업에 대한 주민의 여론을 수렴할 필요가 있다. The PPP system was introduced to overcome the shortcomings of traditional financial projects, and local governments have also implemented various PPP projects. As a result of analyzing the current state of Jeollabuk-do private investment as of 2020, it can be seen that most of the private investment projects in Jeollabuk-do are sewer pipe maintenance rental-type projects, and most of the facility rental fees are operated by the government and the provincial government. Although this type of business has the theoretical advantage that business efficiency can be secured through private capital input, it is difficult to induce private capital participation in reality due to long-term capital input and low profitability. Moreover, as a result of examining the details of the repayment of the private investment project in Jeollabuk-do, there is a concern that it may act as a heavy burden on the Jeollabuk-do region's finances in the future. The performance of the PPP project and the performance of the financial project are two sides of the same coin in that improving the performance of the PPP project is a way to reduce the government finances and improve the efficiency of financial management. For a PPP project to be successful, it is most important to predict demand and accurately calculate costs through economic analysis in the project implementation stage. In order not to repeat the case of the Jeonju Light Rail Transit, which failed in the past project promotion stage, it is necessary to disclose the results of thorough preliminary analysis to the residents and collect public opinion on the private investment project.

        경력개발과 번영감, 그리고 잡크래프팅 간의 관계

        최근 국내에서도 조직행동분야에서 번영감에 대한 논의가 점차 관심을 받고 있다. 번영감이란 조직 구성원이 업무수행과정에서 지각하는 활력감, 학습감이 결합된 개념을 말한다. 번영감의 긍정적인 영향력 때문에 연구자와 실무에서 번영감에 관한 관심이 높고, 이에 따라 관련 연구가 지속되고 있으나, 여전히 선행요인과 결과요인, 그리고 메커니즘을 탐색하기 위한 연구가 요구되고 있다. 이에 따라 본 연구는 선행연구에서 다루지 않았던 번영감의 선행 요인과 결과영향을 탐색하고자 한다. 본 연구는 경력관리행동과 조직경력개발지원을 번영감의 선행요인으로 잡크 래프팅을 번영감의 결과요인으로 연구모형을 설계하고 실증하였다. 실증분석을 위해 국내 기업에서 근무하는 477 명의 종업원을 대상으로 설문조사를 실시하였다. 실증분석결과는 다음과 같다. 첫째, 경력관리행동은 번영감에 정 (+)의 영향을 미치는 것으로 나타났다. 둘째, 조직경력개발지원은 번영감에 정(+) 영향을 주는 것으로 확인되었다. 셋째, 번영감이 잡크래프팅에 정(+)의 영향을 미쳤다. 넷째, 번영감은 경력관리행동과 잡크래프팅 간의 관계와 조직경력개발지원과 잡크래프팅 간의 관계를 매개하는 것으로 나타났다. 본 연구의 실증 결과를 바탕으로 연구의 시사점과 한계 및 향후 연구방향을 논의하였다 Recently, ‘thriving at work’ has been receiving attention in the field of organizational behavior. Thriving at work refers to a concept that combines the sense of vitality and learning that employees perceive in the process of performing tasks. Researchers and practitioners are interested in ‘thriving at work’ and related research is con-tinuing because of the positive impact of it. However, there is still a need for research to explore the anteced-ents, consequences, and mechanisms. Thus, this study intends to explore the antecedents and consequences of ‘thriving at work’ that were not addressed in previous studies. Specifically, this study designed and demonstrated a research model in which career management behavior and organizational career development support were antecedents for thriving at work and job crafting was a consequence of it. For empirical analysis, a survey was conducted with 477 employees working in domestic companies. The empirical analysis results are as follows. First, career management behavior was found to have a positive effect on the thriving at work. Second, it was confirmed that organizational career development support had a positive effect on the thriving at work. Third, thriving at work had a positive effect on job crafting. Fourth, thriving at work was found to mediate the rela-tionship between career management behavior and job crafting, and between support for organizational career development and job crafting. Based on the empirical results of the study, the implications and limitations of the study and future research directions were discussed.

        Clinical utilization of radiation therapy in Korea between 2017 and 2019

        Purpose: This study aimed to evaluate the clinical infrastructure and utilization of radiotherapy (RT) services in Korea between 2017 and 2019. Materials and Methods: We extracted the data of patients who underwent RT between 2017 and 2019 from the Health Insurance Review and Assessment Service. We further analyzed this data according to the diagnosis and treatment modalities of patients diagnosed with International Classification of Disease 10 (ICD-10) diagnostic codes C00–C97 and D00–D48. In addition, we collected statistics on RT facilities in Korea using a nationwide survey. Results: The total number of patients who received RT in 2017, 2018, and 2019 were 77,901, 81,849, and 87,460, respectively. The number of patients diagnosed with ICD 10 C- and D-codes in 2019 was 86,339, of whom 39,467 were men and 46,872 women. The rate of utilization of RT among cancer patients was 30.4% in 2017 and 2018 and 30.9% in 2019. In 2019, the most common types of cancers treated with RT were breast, lung, prostate, colorectal, and liver cancers. Regarding the RT infrastructure in Korea, there were 95 radiation oncology centers, 237 megavoltage (MV) teletherapy units, 35 brachytherapy units, and two proton accelerators in 2019. There were 4.5 MV teletherapy machines per million. Conclusion: The number of patients treated with RT has increased consistently from 2017 to 2019. As the number of patients with cancer increases, it is expected that the RT infrastructure will be further expanded in Korea.

        CRISPR/Cas9-mediated gene knockout screens and target identification via whole-genome sequencing uncover host genes required for picornavirus infection

        <P>Several groups have used genome-wide libraries of lentiviruses encoding small guide RNAs (sgRNAs) for genetic screens. In most cases, sgRNA expression cassettes are integrated into cells by using lentiviruses, and target genes are statistically estimated by the readout of sgRNA sequences after targeted sequencing. We present a new virus-free method for human gene knockout screens using a genome-wide library of CRISPR/Cas9 sgRNAs based on plasmids and target gene identification via whole-genome sequencing (WGS) confirmation of authentic mutations rather than statistical estimation through targeted amplicon sequencing. We used 30,840 pairs of individually synthesized oligonucleotides to construct the genome-scale sgRNA library, collectively targeting 10,280 human genes (<I>i.e.</I> three sgRNAs per gene). These plasmid libraries were co-transfected with a Cas9-expression plasmid into human cells, which were then treated with cytotoxic drugs or viruses. Only cells lacking key factors essential for cytotoxic drug metabolism or viral infection were able to survive. Genomic DNA isolated from cells that survived these challenges was subjected to WGS to directly identify CRISPR/Cas9-mediated causal mutations essential for cell survival. With this approach, we were able to identify known and novel genes essential for viral infection in human cells. We propose that genome-wide sgRNA screens based on plasmids coupled with WGS are powerful tools for forward genetics studies and drug target discovery.</P>

      • Differential effects on sodium current impairments by distinct <i>SCN1A</i> mutations in GABAergic neurons derived from Dravet syndrome patients

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>We investigated how two distinct mutations in <I>SCN1A</I> differentially affect electrophysiological properties of the patient-derived GABAergic neurons and clinical severities in two Dravet syndrome (DS) patients.</P> <P><B>Materials and Methods</B></P> <P>We established induced pluripotent stem cells from two DS patients with different mutations in <I>SCN1A</I> and subsequently differentiated them into forebrain GABAergic neurons. Functionality of differentiated GABAergic neurons was examined by electrophysiological recordings.</P> <P><B>Results</B></P> <P>DS-1 patient had a missense mutation, c.4261G > T [GenBank: NM_006920.4] and DS-2 patient had a nonsense frameshift mutation, c.3576_3580 del TCAAA [GenBank: NM_006920.4]. Clinically, contrary to our expectations, DS-1 patient had more severe symptoms including frequency of seizure episodes and the extent of intellectual ability penetration than DS-2 patient. Electrophysiologic recordings showed significantly lower sodium current density and reduced action potential frequency at strong current injection (>60 pA) in GABAergic neurons derived from both. Intriguingly, unique genetic alterations of <I>SCN1A</I> differentially impacted electrophysiological impairment of the neurons, and the impairment’s extent corresponded with the symptomatic severity of the donor from which the iPSCs were derived.</P> <P><B>Conclusion</B></P> <P>Our results suggest the possibility that patient-derived iPSCs may provide a reliable <I>in vitro</I> system that reflects clinical severities in individuals with DS.</P>

        Trends in Regional Disparity in Cardiovascular Mortality in Korea, 1983–2019

        Background and Objectives: Despite remarkable reduction in cardiovascular disease (CVD) mortality, the burden has remained the leading cause of death. Since little research has focused on regional disparity in CVD mortality, this study aims to investigate its spatiotemporal trends in Korea from 1983 to 2019. Methods: Using the causes of death statistics in Korea, we analyzed the geographic variation in deaths from CVDs from 1983 to 2019. The sex and age-standardized mortality rate was calculated according to the 17 administrative regions. The analyses include all diseases of the circulatory system (International Classification of Diseases-10 codes, I00–I99), along with the following 6 subcategories which were not mutually exclusive: total heart disease (I00–I13 and I20–I51), hypertensive heart disease (I10–I13), ischemic heart disease (I20–I25), myocardial infarction (I21–I23), heart failure (I50), and cerebrovascular disease (I60–I69). Results: Overall, heart failure death rate increased across all regions, and other CVD death rates showed a decreasing trend. Regional disparity in mortality was substantial in the early 1980s but converged over time. In all types of cardiovascular mortality, Busan, Ulsan and Gyeongnam remained the highest, although they showed a downward trend like other regions. Jeju continued to have a relatively low CVD mortality rate. Conclusions: The regional disparity substantially decreased compared to the 1980s. However, the relatively high burden of CVD mortality in the southeastern region has not been fully resolved.

      • The Antioxidant and Anti-Inflammatory Activities of 8-Hydroxydaidzein (8-HD) in Activated Macrophage-Like RAW264.7 Cells

        <P>8-Hydroxydaidzein (8-HD) is a daidzein metabolite isolated from soybeans. This compound has been studied for its anti-proliferation, depigmentation, and antioxidant activities. However, the anti-inflammatory activities of 8-HD are not well-understood. Through its antioxidant effects in ABTS and DPPH assays, 8-HD reduces the production of sodium nitroprusside (SNP)-induced radical oxygen species (ROS). By triggering various Toll-like receptors (TLRs), 8-HD suppresses the inflammatory mediator nitric oxide (NO) without cytotoxicity. We examined the regulatory mechanism of 8-HD in lipopolysaccharide (LPS)-induced conditions. We found that 8-HD diminishes inflammatory gene expression (e.g., inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2, and tumor necrosis factor (TNF)-α) by regulating the transcriptional activities of nuclear factor (NF)-κB and activator protein 1 (AP-1). To find the potential targets of 8-HD, signaling pathways were investigated by immunoblotting analyses. These analyses revealed that 8-HD inhibits the activation of TAK1 and that phosphorylated levels of downstream molecules decrease in sequence. Together, our results demonstrate the antioxidant and anti-inflammatory actions of 8-HD and suggest its potential use in cosmetics or anti-inflammatory drugs.</P>

        Digenome-seq: genome-wide profiling of CRISPR-Cas9 off-target effects in human cells

        Although RNA-guided genome editing via the CRISPR-Cas9 system is now widely used in biomedical research, genome-wide target specificities of Cas9 nucleases remain controversial. Here we present Digenome-seq, in vitro Cas9-digested whole-genome sequencing, to profile genome-wide Cas9 off-target effects in human cells. This in vitro digest yields sequence reads with the same 5′ ends at cleavage sites that can be computationally identified. We validated off-target sites at which insertions or deletions were induced with frequencies below 0.1%, near the detection limit of targeted deep sequencing. We also showed that Cas9 nucleases can be highly specific, inducing off-target mutations at merely several, rather than thousands of, sites in the entire genome and that Cas9 off-target effects can be avoided by replacing 'promiscuous' single guide RNAs (sgRNAs) with modified sgRNAs. Digenome-seq is a robust, sensitive, unbiased and cost-effective method for profiling genome-wide off-target effects of programmable nucleases including Cas9.
