A muscle-specific lipase gene of the bumblebee Bombus ignitus was cloned and characterized. This gene, which we named Bi-Lipase, consists of seven exons encoding 317 amino acid residues. Bi-Lipase possesses all the features of lipases, including GXSXG consensus motif and Ser-Asp-His catalytic triad. Expressed as a 37-kDa polypeptide in baculovirus-infected insect Sf9 cells, recombinant Bi-Lipase showed an optimal pH of 9.0 and exhibited its highest catalytic activity at 40℃. Furthermore, through the addition of tunicamycin to the recombinant virus-infected Sf9 cells, recombinant Bi-Lipase was found to be N-glycosylated. Northern and western blot analyses indicated that Bi-Lipase was expressed in the wing, thorax, and leg muscles. These results show that Bi-Lipase is a muscle-specific lipase, suggesting a possible role of Bi-Lipase in the utilization of lipids for muscular activity in B. ignitus.

Fibulin-5 has recently been considered as a potential tumor suppressor in human cancers. Several studies have shown that it is down-regulated in a variety of tumor types and inhibits tumor growth and metastasis. This study was aimed to investigate the clinical significance of fibulin-5 in glioma and its role in cell proliferation and invasion. We found that the expression of fibulin-5 in glioma tissues was significantly lower than those in normal brain (NB) tissues. Negative expression was significantly correlated with advanced clinical stage (grade III+IV). Furthermore, Fibulin-5 negative expression was correlated with a shorter overall survival of glioma patients. Multivariate Cox repression analysis indicated that fibulin-5 was an independent factor for predicting overall survival of glioma patients. Overexpression obviously inhibited cell proliferation in U251 and U87 cells. Furthermore, it significantly reduced the number of migrating and invading glioma cells. In conclusion, impaired expression of fibulin-5 is correlated with the advanced tumor stage in glioma. Otherwise, Fibulin-5 is an independent prognostic marker for predicting overall survival of glioma patients. Mechanistically, it may function as a tumor suppressor via inhibiting cell proliferation and invasion in gliomas.

Microbial community shifts, associated with performance data, were investigated in an anaerobic batch digester treating high-solid food waste under mesophilic conditions using, a combination of molecular techniques and chemical analysis methods. The batch process was successfully operated with an organic removal efficiency of 44.5% associated with a biogas yield of 0.82 L/g VSremoval. Microbial community structures were examined by denaturing gel gradient electrophoresis. Clostridium and Symbiobacterium organisms were suggested to be mainly responsible for the organic matter catabolism in hydrolysis and acidogenesis reactions. The dynamics of archaeal and methanogenic populations were monitored using real-time PCR targeting 16S rRNA genes. Methanosarcina was the predominant methanogen, suggesting that the methanogenesis took place mainly via an aceticlastic pathway. Hydrogenotrophic methanogens were also supported in high-solid anaerobic digestion of food waste through syntrophism with syntrophic bacterium. Microbial community shifts showed good agreement with the performance parameters in anaerobic digestion, implying the possibility of diagnosing a high-solid anaerobic digestion process by monitoring microbial community shifts. On the other hand, the batch results could be relevant to the start-up period of a continuous system and could also provide useful information to set up a continuous operation.
Synthesis of nonsharp distillation sequences via genetic programming

This paper addresses the application of Genetic Programming (GP) to the synthesis of multicomponent product nonsharp distillation sequences. Combined with the domain knowledge of chemical engineering, some evolutionary factors are improved, and a set of special encoding method and solving strategy is proposed to deal with this kind of problem. The system structural variable is optimized by GP and the continuous variable is optimized by the simulated annealing algorithm simultaneously. Because GP has an automatic searching function, the optimal solution can be found including distillation, splitting, blending and bypassing operations automatically without any superstructures of nonsharp distillation sequences. Three illustrative examples are presented to demonstrate the effective computational strategies.

Considering the influence of the visible load-induced transverse cracks on chloride ion diffusion in cracked concrete of the in-service reinforced concrete (RC) elements under marine environment, a typical concrete volume with one transverse crack was taken to establish the governing equation. Assuming the crack widths at the outer lateral and bottom surfaces were equal, a model to predict the equivalent chloride diffusion coefficient in cracked concrete was put forward to consider the influence of the average transverse crack width, crack spacing and crack extending lengths in beam height and width directions on chloride diffusion. Results show that the proposed model can better reflect the variation trend of the equivalent chloride diffusion coefficient with different crack widths in in-service RC elements under marine environment.

Fully dense THCN) based carbonitride ceramics were fabricated by pressureless sintering. During sintering, solid solutions were formed from the ceramic ingredients. The ceramics exhibited microvickers hardness of 1560-2050kgf/mm2, fracture toughness of 3.0-5.4 MPa $m^(1/2)$, and three point flexural strength of 645-1072 MPa. Some of the ceramics were shaped in a cutting tool, and the cutting performance was evaluated. In case of cutting SCM440 alloy steel, the ceramics showed better performance than the commercially available alumina-titanium carbide ceramic cutting tool. Considering the excellent productivity of pressureless sintering compared with other densification methods and their cutting performance, this new class of ceramics are very promising for wear resistant applications.

This paper reports the results of a seismic performance study of a precast shear wall with a new horizontal connection. The new connection is the rabbet-unbonded horizontal connection, which is composed of rabbets and unbonded rebar segments. The rabbets are used to improve the shear capacity and prevent slippage of the connection, and the unbonded rebar segments are used to improve the ductility and energy dissipation. Three specimens were tested with different parameters under cyclic quasi-static loading. The test results showed that the specimen with a larger unbonded level had a richer hysteresis curve, larger ductility, larger energy dissipation, and slightly smaller bearing capacity. Moreover, in relation to the stiffness degradation, in the initial stage, the specimen with a larger unbonded level had a smaller stiffness, whereas in the last stage, the stiffnesses were similar regardless of the unbonded level. A parameter analysis using a finite element model proved that the ductility and energy dissipation of a shear wall with the rabbet-unbonded horizontal connection increased with the unbonded length and level. In addition, when the axial compression ratio increased, the bearing capacity increased, but the load-displacement curves decreased more rapidly. It was concluded that the unbonded length and unbonded level could effectively improve the ductility and energy dissipation of a shear wall. However, they should not be too large under high pressure, and the design suggestions for the new connection need further research considering other factors.
