A conditional version of the classical central limit theorem is derived rigorously by using conditional characteristic functions, and a more general version of conditional central limit theorem for the case of conditionally independent but not necessarily conditionally identically distributed random variables is established. These are done anticipating that the field of conditional limit theory will prove to be of significant applicability.

Purpose Colon cancer-associated transcript 1 (CCAT1) was identified as an oncogenic long non-coding RNA (lncRNA) in a variety of cancers. However, there was a lack of understanding of the mechanism by which CCAT1 conferred cisplatin (also known as DDP) resistance in ovarian cancer cells. Materials and Methods Cell viability of A2780, SKOV3, A2780/DDP, and SKOV3/DDP cells upon cisplatin treatment was monitored by MTT assay. Quantitative reverse transcription polymerase chain reaction (qRT-PCR) detected the expression levels of CCAT1 and miR-454. The effect of sh-CCAT1 on cisplatin response was investigated in xenografts study. Bioinformatic analysis, luciferase reporter assay and qRT-PCR were conducted to validate the direct interaction among CCAT1, miR-454, and survivin. Apoptosis was determined by flow cytometry after dual staining of Annexin-V-FITC/propidium iodide, and the expression of apoptosis-related proteins Bcl-2, Bax and survivin were detected by qRT-PCR and Western blotting. Xenograft study was conducted to monitor in vivo tumor formation. Results CCAT1 was highly expressed in cisplatin-resistant ovarian cancer cell line A2780/DDP and SKOV3/DDP. Knockdown of CCAT1 restored sensitivity to cisplatin in vitro and in vivo. Our data revealed that silencing of CCAT1 promoted cisplatin-induced apoptosis via modulating the expression of pro- or anti-apoptotic proteins Bax, Bcl-2, and survivin. CCAT1 directly interacted with miR-454, and miR-454 overexpression potentiated cisplatin-induced apoptosis. Survivin was identified as a functional target of miR-454, restoration of survivin attenuated the effect of miR-454 on cisplatin response. In addition, miR-454 inhibitor or overexpression of survivin was found to abolish sh-CCAT1–induced apoptosis upon cisplatin treatment. Conclusion CCAT1/miR-454/survivin axis conferred cisplatin resistance in ovarian cancer cells.

Thee kinds of explants, young spikes, immature and mature embryos, was compared in callus induction, embryogenic callus formation, differentiation and plant regeneration of Russian wildrye. Callus induction efficient of young spikes is highest. The imature embryos formed more somatic embryogenesis, and with highest frequency of embryogenic callus.
Hot-Stamping Die-Cooling System for Vehicle Door Beams

The need for lightweight automobiles has promoted the rapid development of hot stamping. The cooling system of the hotstamping die determines the performance of hot-stamped workpieces. By establishing a simplified hot-stamping model, we determined that the temperature–time curve of a workpiece is an exponential function. Our research results showed that the depth from the die surface to the cooling pipes has the biggest impact on the cooling rate and uniformity of the workpiece; the correct depth must be ensured to obtain a uniform quenching rate. By studying the cooling buffer flumes, we discovered that the water flow in the pipes becomes more uniform with increasing flume width. With an increase in the flume height, the water flow uniformity in the pipes first increased, and then decreased. In addition, augmenting the pipe diameter near the inlet improved the water flow uniformity in the pipes.
Integrin-linked Kinase Functions as a Tumor Promoter in Bladder Transitional Cell Carcinoma

The aim of this study was to elucidate the role of the integrin-linked kinase (ILK) gene in development of human bladder transitional cell carcinoma (BTCC). Expression of ILK protein and ILK mRNA in 56 cases of human BTCC tissue and in 30 cases of adjacent normal bladder tissue was detected by immunohistochemistry S-P and reverse transcription polymerase chain reaction (RT-PCR), respectively. Four specific miRNA RNAi vectors targeting human ILK were synthesized and transfected into BIU-87 cells by liposome to obtain stable expression cell strains. The influence of ILK on proliferation of BTCC was detected by MTT, FCM on athymic mouse tumorigenesis. The positive rate of ILK protein in BTCC tissue (53.6%) was much higher than adjacent normal bladder tissue (10.0%) (p<0.05). Similarly, expression of ILK mRNA in BTCC tissue ($0.540{\pm}0.083$) was significantly higher than in adjacent normal bladder tissue ($0.492{\pm}0.070$) (p<0.05). MTT showed that the proliferation ability of miRNA-ILK transfected group was clearly decreased (p<0.05), the cell cycle being arrested in G0/G1-S, an tumorigenesis in vivo was also significantly reduced (p<0.05). ILK gene transcription and protein expression may be involved in the development of BTCC, so that ILK might be the new marker for early diagnosis and the new target for gene treatment.

Optimal design of high temperature vacuum furnace using response surface method

A new method using the response surface method and optimization technique has been developed instead of the original method based on trial and error. In order to construct a response surface, thermal analysis was performed under the condition of using the calculated thermal conductivity of the insulator in a previous study. In order to set up the response surface, the D-Optimal method was used in the process of selecting experimental points. Using a weighting factor, an optimization study was carried out under the condition of satisfying user requirements. Finally, the merits and drawbacks of the new method were described by comparing with the optimal design method based on the thermal analysis database which was developed in a previous study. The optimal results show that the developed method can be used to design an energy efficient, low manufacturing cost, high temperature vacuum furnace with avoiding unnecessary iterative manufacturing, and anticipating the performance before manufacturing.

Telomerase extends the proliferation and prevent replicative senescence in most somatic cells. Whether it has similar function in human epidermal stem cells remains to be determined. In this study, the human telomerase reverse transcriptase (hTERT) cytalytic subunit was introduced into epidermal stem cells derived from human fetal skins. The expression of hTERT mRNA, protein and telomerase activity in the transduced cells was observed by real time PCR (RT-PCR) techniques, western blot analysis and telomeric repeat amplification protocol enzyme-linked immunosorbent assay (TRAP-ELISA) assay, respectively. The proliferation of the transduced cells was examined. The results showed the introduction of hTERT into the epidermal stem cells upregulated the expression of the hTERT gene and protein. And also the hTERT-transduced epidermal stem cells exhibited significantly elevated telomerase activity and proliferation. These works establish the base of further gene modification, monoclone screening and cell differentiation mechanism. The ability to maintain the biological characteristics and proliferation of epidermal stem cells could have important applications in wound healing and skin tissue engineering.
