        Study of compensatory growth based on different nutrition conditions of Bombyx mori

        Organisms achieve compensatory growth after a period of nutrient restriction followed by recovering the nutrient status. However, the underlying mechanisms associated with such growth acceleration remain unclear. The silkworm Bombyx mori is a lepidopteran model insect. This study aimed to investigate the physiological characteristics and the underlying mechanisms in B. mori fed on mulberry leaves (MG), artificial diet (AG), and artificial diet + mulberry leaves (AG-MG), respectively. Silkworms in AG-MG which fed on artificial diet from 1st to 3rd instars followed by feeding on mulberry leaves from 4th to 5th instars exhibited a higher weight gain rate than that in MG and AG, indicating that compensatory growth occurred as a result of the switch in the silkworm food regime. Trypsin and lipase activities of silkworms in AG-MG were shown to be up-regulated at 72 h after changing food. Digital gene expression profiling (DGE) analysis revealed that genes related to metabolism and development in silkworm midguts were differentially expressed. The quantitative real-time PCR (qRT-PCR) re sults showed that the expression levels of IIS/PI3K-AKT pathway genes including INR, IRS, AKT, PI3K60 and PI3K110 of silkworms in AG were down-regulated compared with that in MG at 0 h. Whereas AKT, PI3K and PI3K60 of silkworms were significantly increased by 1.68-, 1.49-, 1.67-fold, respectively, at 72 h after switched to mulberry leaves than the same instar fed on artificial diet. PDK’s expression of silkworms in AG was higher than that in other two groups at each timepoint. Compared with MG and AG, PTEN and IRS were down-regulated at 48 and 72 h in AG-MG. Collectively, these results indicate that compensatory growth in B. mori is regulated by IIS/ PI3K-AKT signaling pathway.

      • Protective Effects of Scutellaria barbata Against Rat Liver Tumorigenesis

        Scutellaria barbata D. Don (S. barbata), a traditional Chinese medicine, is used to treat cancers, inflammation, and urinary diseases. This study aimed to determine any protective effects of S. barbata crude extract (CE-SB) against rat liver tumorigenesis induced by diethylnitrosamine (DENA). Liver malfunction indices in serum were measured by biochemical examination. Hematoxylin and eosin staining was performed to examine liver pathology. Contents of malondialdehyde (MDA) and superoxide dismutase (SOD) were measured in liver homogenates to evaluate oxidative stress. The levels of liver malfunction indices in the CE-SB groups, especially in the CE-SB high dose group, were lower than that of the model group (P<0.05). The results from histological examination indicated that the number of liver nodules in the CE-SB groups decreased compared with the model group (P<0.05). Content of MDA determined in liver was significantly decreased, and level of SOD elevated by CE-SB. CE-SB can inhibit experimental liver tumorigenesis and relieve hepatic injury in rats.

        Genome-wide identification of the ARRs gene family in tobacco (Nicotiana tabacum)

        Background The growth of axillary buds determines the shoot branching and morphology of plants, and its initiation and development are regulated by a series of hormonal signals, such as cytokinin. Arabidopsis response regulators (ARRs) can regulate the growth and development, disease resistance and stress resistance of plants by participating in cytokinin signaling. Objective To explore the distribution and expression pattern of ARR members in tobacco. Methods The identifcation, isoelectric points, molecular weights, protein subcellular localization prediction, multiple sequence alignment, phylogenetic analysis, protein motifs and structures, chromosome distributions of deduced ARR proteins were conducted. The gene expression profling of various tissues in response to topping, low temperature and drought were analyzed by RNA-seq and qRT-PCR. Results 59 ARR genes from cultivated tobacco (Nicotiana tabacum) were identifed, namely NtARRs, including 21 type A NtARRs and 38 type B NtARRs. The 59 NtARRs were expressed mainly in all organs except the fruits. Some representative NtARRs may participate in axillary bud initiation and development, as well as in stress resistance through cytokinin signal transduction. Conclusion Understanding the roles of NtARRs in the molecular mechanisms responsible for axillary bud growth and stress tolerance could aid in targeted breeding in crops.

      • Robustness-Improvement Strategy and Modeling under Uncertainty of Chinese Catering Service Supply Chain

        Catering Service Supply Chain (CSSC) is a new supply chain, of which Catering Service Integrator (CSI) is in the center accompany with Functional Catering Service Provider (FCSP). The core objective of CSSC is to provide different catering enterprises with supply chain services. Instability, non-standardization, specialty and diversity characteristics of the Chinese catering industry inevitably lead to demand uncertainty of CSSC, directly resulting in the uncertainty of operation and supply side. The uncertainty of supply chain system is the basis of robust optimization study, under the influence of uncertainty, robustness is extremely important to ensure stable income and continuous operation of supply chain. This research classifies various uncertainty scenarios, based on robust optimization method. Two strategies, service coordination with widely knowledge sharing, and synchronous decision with intelligent inducement, are designed to induce and integrate personalized demand to get the scale effect and the coordination of the entire supply chain. From a point of quantitative view, this research builds up robust optimization model to improve robustness of CSSC, deposing the target into two coordinated parts. The coordination is of great sense, including two objectives simultaneously, the first one aims to maximize the delivery rate of order demand, the second one aims to optimize profit of CSI and FCSP. According to data examination, the coordinative strategies and robust optimization model can effectively control the uncertainty of the whole supply chain, and then improve the robustness of CSSC.

        Effect of Light with Different Wavelengths on Nostoc flagelliforme Cells in Liquid Culture

        The effects of lights with different wavelengths on the growth and the yield of extracellular polysaccharides of Nostoc flagelliforme cells were investigated in a liquid cultivation. N. flagelliforme cells were cultured for 16 days in 500 ml conical flasks containing BG11 culture medium under 27 μmol·m-2·s-1 of light intensity and 25oC on a rotary shaker (140 rpm). The chlorophyll a, phycocyanin, allophycocyanin, and phycoerythrin contents in N. flagelliforme cells under the lights of different wavelengths were also measured. It was found that the cell biomass and the yield of polysaccharide changed with different wavelengths of light. The biomass and the yield of extracellular polysaccharides under the red or violet light were higher than those under other light colors. Chlorophyll a, phycocyanin, and allophycocyanin are the main pigments in N. flagelliforme cells. The results showed that N. flagelliforme, like other cyanobacteria, has the ability of adjusting the contents and relative ratio of its pigments with the light quality. As a conclusion, N. flagelliforme cells favor red and violet lights and perform the complementary chromatic adaptation ability to acclimate to the changes of the light quality in the environment.

        Inhibitory effects of piceatannol on human cytomegalovirus (hCMV) in vitro

        Human cytomegalovirus (hCMV) is a ubiquitous herpesvirus, which results in the establishment of a latent infection that persists throughout the life of the host and can be reactivated when the immunity is low. Currently, there is no vaccine for hCMV infection, and the licensed antiviral drugs mainly target the viral enzymes and have obvious adverse reactions. Thus, it is important to search for compounds with antihCMV properties. The present study aimed to investigate the suppressive effects of piceatannol on hCMV Towne strain infection and the putative underlying mechanisms using human diploid fibroblast WI-38 cells. Piceatannol supplementation prevented the lytic changes induced by hCMV infection in WI-38 cells. Furthermore, piceatannol suppressed the expression of hCMV immediate-early (IE) and early (E) proteins as well as the replication of hCMV DNA in a dose-dependent manner. Moreover, hCMV-induced cellular senescence was suppressed by piceatannol, as shown by a decline in the senescence-associated β-galactosidase (SA-β-Gal) activity and decreased production of intracellular reactive oxygen species (ROS). p16INK4a, a major senescence-associated molecule, was dramatically elevated by current hCMV infection that was attenuated by pre-incubation with piceatannol in a dose-dependent manner. These results demonstrated that piceatannol suppressed the hCMV infection via inhibition of the activation of p16INK4a and cellular senescence induced by hCMV. Together, these findings indicate piceatannol as a novel and potent anti-hCMV agent with the potential to be developed as an effective treatment for chronic hCMV infection.

        Three new glycoalkaloids, N-formyl-asimilobine-2-O-b-D-glucoside (1), (-)-1-O-b-D-glucoside-8-oxotetrahydropalmatine(2), and 1-N-monomethylcarbamateargentinine-3-O-b-D-glucoside (3) were isolated fromtubers of Stephania succifera. The structures were establishedbased on spectroscopic analysis, and the antimicrobialactivities of the three glycoalkaloids are reported.

        Isoindolin-1-ones from the stems of Nicotiana tabacum and their antiviral activities

        In previous studies, several isoindolin-1-oneanalogs that exhibited signifi cant anti-tobacco mosaic virus(anti-TMV) activities were isolated from Nicotiana tabacum . Since gene-editing mutants provide a new sample for thediscovery of active metabolites, we focused on the stems ofYN-18–23 (a mutant N. tabacum for gene editing with thealkaloid metabolic pathway cultivated by Yunnan TobaccoCompany), which led to the isolation of four new ( 1–4 )and four known ( 5–8 ) isoindolin-1-ones. To the best of ourknowledge, nicindole C ( 3 ) is the fi rst subclass of isoindolin-1-one bearing a pentacyclic ketone, while nicindole D ( 4 )is the fi rst example of isoindolin-1-one bearing a methylpyridin-2-(1 H )-one moiety. Compounds 1–4 were testedfor their anti-TMV activities, and the results revealed thatcompounds 1 , 3 , and 4 exhibited high anti-TMV activities atconcentrations of 20 μM with inhibition rates of 48.6, 42.8,and 71.5%, respectively. These rates are higher than the inhibitionrate of the positive control (33.2%). The mechanisticstudy of compound 4 , which had the highest anti-TMV activityrevealed that increased potentiation of defense-related enzyme activities and downregulation of expression of theNtHsp70 protein may induce resistance in tobacco againstthe viral pathogen TMV. Molecular docking studies alsorevealed that the isoindolin-1-one substructure is fundamentalfor anti-TMV activity. The methyl-pyridin-2-(1 H )-onemoiety in compound 4 and the 2-oxopropyl groups in compounds1 and 3 at the N -2 position may increase inhibitoryactivities. This study of the structure–activity relationshipis helpful for fi nding new anti-TMV activity inhibitors. Tostudy whether the isoindolin-1-ones have broader antiviralactivities, compounds 1–4 were also tested for their antirotavirusactivities. Compound 4 exhibited high anti-rotavirusactivity with a therapeutic index (TI) value of 20.7. This TI value is close to that of the positive control (20.2).
