        Comparison of Proliferative Activity in Each Histological Subtypes of Benign and Atypical Intracranial Meningiomas by PCNA and Ki-67 Immunolabeling

        목 적 : 양성 뇌수막종에 비하여 이형성 및 악성 뇌수막종이 나쁜 임상적인 예후 및 양상을 보이는 것은 잘 알려져 있으나, 양성 뇌수막종에 있어서 각각의 병리조직학적 아형에 따른 생물학적 양상의 차이에 대해서는 잘 알려지지 않거나 일부 논란이 되고있다. 본 연구에서는 이형성 뇌수막종 및 양성 뇌수막종의 각각의 병리조직학적 아형에 따른 증식능의 차이여부를 알아보고자 PCNA와 Ki-67표지지수를 분석하였다. 방 법 : 본원에서 뇌수막종으로 수술을 시행하여 얻은, 재발을 보여 재수술을 시행한 2례를 포함하여, 파라핀에 포매시킨 27개의 조직을 대상으로 병리학적인 증식능을 분석하기 위해, PCNA에 대한 단일항체 및 MIB-1 단일항체를 이용한 면역조직화학적 염색을 시행하였다. 조직학적 분류상 meningothelial type이 8례, transitional type이 9례, fibroblastic type이 5례였으며, 이형성 수막종이 5례였다. 결 과 : PCNA표지지수의 평균값은 양성 수막종에서 meningothelial type이 $4.82{\pm}5.10%$, transitional type이 $9.01{\pm}4.25%$, fibroblastic type이 $5.66{\pm}5.32%$를 보였으나 이형성 수막종에서는 $27.62{\pm}19.67%$의 높은 지수를 나타냈고, Ki-67 표지지수의 평균값은 양성 수막종의 아형에서 각각 $0.43{\pm}0.85%$, $0.44{\pm}1.08%$, $0.24{\pm}0.18%$를 보이고, 역시 이형성 수막종에서는 $0.84{\pm}0.59%$의 높은 지수를 보였다. 즉, 양성 수막종에서 각각의 아형에 따른 PCNA 및 Ki-67 표지지수는 통계학적으로 의미있는 차이는 없었으나(p>0.05), 이형성 수막종에서는 의미있는 높은 표지지수를 보여(p<0.05) 양성 수막종에서 보다 높은 증식능을 보임을 알 수 있었다. 결 론 : PCNA 및 Ki-67 표지지수를 이용한 증식능의 비교결과, 양성 뇌수막종에서는 각각의 아형에 따른 생물학적 양상이나 예후는 차이가 없을것으로 생각되나, 이형성 수막종에서는 높은 증식능을 보여 이에 대한 예후를 예상할 수 있을것으로 생각되며, 또한 이러한 표지지수가 병리조직학적으로 양성과 이형성의 감별에 많은 도움이 될것으로 사료된다. Objective : The clinical prognosis and biological behavior of atypical and especially malignant meningiomas are well known to be worse than benign meningioma, but the degree of biological aggressiveness in each classical subtypes of benign meningioma is controversy. This study was performed to see whether there is a difference in the proliferative activity between each different histological subtypes of benign meningioma as well as atypical meningioma. Methods : Paraffin-embedded surgical specimens of 27 meningiomas, including two recurrent tumors, were studied to evaluate proliferative activity by immunohistochemical method with monoclonal antibodies to proliferating cell nuclear antigen(PCNA) and MIB-1. The specimens consisted of 8 cases of meningothelial, 9 cases of transitional, 5 cases of fibroblastic subtypes and 5 cases of atypical meningiomas. Results : Mean PCNA labeling indices of meningothelial, transitional and fibroblastic meningiomas were $4.82{\pm}5.10%$, $9.01{\pm}4.25%$ and $5.66{\pm}5.32%$, but that of atypical meningiomas was $27.62{\pm}19.67%$, noting a higher value compared to all three subtypes of benign meningiomas. Mean Ki-67 labeling indices of the above 3 subtypes were $0.43{\pm}0.85%$, $0.44{\pm}1.08%$ and $0.24{\pm}0.18%$, and that of atypical meningiomas was also revealed to be of higher value ($0.84{\pm}0.59%$). PCNA and Ki-67 labeling indices were not statistically different between histological subtypes of benign meningioma(p>0.05), but the differences of both immunolabeling between benign and atypical meningiomas were statistically significant(p<0.05). Conclusion : Immunolabeling of PCNA and Ki-67 in intracranial meningiomas reveals no prognostic difference between meningothelial, transitional and fibroblastic subtypes in classical benign meningiomas by measuring expression of PCNA and Ki-67, but it seems to be helpful in differentiating benign and atypical meningioma, later showing more proliferative activity and biological aggressiveness.

        We performed theoretical simulation for directional coupler using BPM program. Simulation results showed that the optimum optical waveguide width and the gap between the guides were 4μm. Using the parametes of n₁=2.25, n₂=2.25, n₃=2.2, gap=4μm, W=30μm, Z=0.1μm, λ=1.5m, we decided other design parametes. Proton exchange of proton diffusion method was applized to pattern a waveguide on a LiNbO₃ substrate.

      • Bifidobacterium angulatum으로부터 α-galactosidase의 정제 및 특성

        α-galactosidase는 (α-D-galactoside galactohydrolase: EC 3.2.1.22)는 다당류나 올리고당인 melibiose, raffinose, stachyose와 guar gum 등의 α-galactosidic 결합을 하고 있는 당을 분해하는 효소이다. 장내세균중 우익균이며 우세균주인 Bifidobacteria로부터 α-galactosidase의 효소학적 특성을 알아보기 위하여 배양여액으로부터 ammonium sulfate분획, 핵산의 제거, Sephadex G-200 gel filtration 및 DEAE-Sephadex A-50 ion exchange chromatography 등의 4단계 정제과정을 거쳐 정제한 결과 약 8배로 정제되어 단일 단백질로 분리하였다. 또한, 효소분해산물을 알아보기 위하여 TLC에 의하여 당분해 및 올리고당의 생성을 알아보았다. 정제효소의 활성 최적온도는 37℃, 최적 pH는 7.0이었다. 효소활성이 K^+, Zn^2+과 같은 금속이온과 8-Hydroxyquinoline, Sodium Bisulfite 등에 의해선 그다지 저해되지 않았으나, Hg^+, Cu^2+과 p-Chloromercuribenzoic acid에 의해선 저해되었다. 효소의 분자량이 301, 995, 합성기질인 PNPG에 대한 K_m은 0.069mM, V_max는 347.22μmol/min.mg protein이었다. To elucidate enzymatic properties of α-galactosidase (EC 3.2.1.22) from Bif.angulatum. α-galactosidase was purified to homogeneity by ammonium sulfate fractionation, protamine sulfate precipitation, sephadex G-200 gel filtration, and DEAE-sephadex A-50 ion exchange chromatography. The purified α-galactosidase was found tobbe homogeneous by SDSpolyacrylamide gel electrophoresis. The molecular weight of this enzyme, estimated by sepadex G-200 gel filtration, was about 301,995. The optimum conditions for the enzyme reaction was pH 6.5 to 7.0 at 37℃. The purified enzyme was stable at 45℃ or below and in buffer at pH 6 to 7.0. The activity was inhibited by mercury, copper ion, and p-chloromercuribenzoic acid. The kinetics of this enzyme, with p-nitrophenyl-α-D-galactoside as substrate, was determined : K_m was about 0.069mM and V_mas was 347.22 μmoles/min mg protein.

      • 축구선수의 무산소운동시 에너지대사 관련 효소에 대한 연구

        The goal of sport science is to identify the variables which influence of improving the athletic performance of a particular sport or event. Many studies have been made to investigate the appropriate variables for effectively performing the various sport activities. The subjects selected for this study were 14 National delegates, 13 collegiate soccer players and untrained collegiate students. The purposes of this study were to analyze the contributions of energy metabolism-related enzymes during rest, immediate and recovery periods after 10-second anaerobic exercise in highly trained soccer players and to offer the basic scientific informations for the new talent selection and useful anaerobic exercise prescription programs for training the excellent soccer players. The findings of this study were as follows; 1. The enzyme levels during rest were significantly higher in National delegates and collegiate players than collegiate students, and CPK were significantly higher in National delegates than collegiate players. 2. The enzyme levels at immediate period after 10-second exercise were significantly higher in National delegates and collegiate players than collegiate students, and CPK and GOT were significantly higher in National delegates than collegiate players. 3. The enzyme levels during recovery periods were significantly higher in National delegates and collegiate players than collegiate students, and CPK, LDH, GOT and GPT were significantly higher in National delegates than collegiate players.

      • LiNbO₃기판을 사용한 광 도파로의 설계 및 분석

        In this paper, we calculate refractive index change of optical waveguide and electric field to simulate y-cut LiNbO₃optical waveguide at He-Ne laser (λ=0.6328[㎛]) by BPM mechanism. At waveguide width of 4[㎛], buffer layer of 0.02[㎛] of LiNbO₃ substrate (XI 55[㎛] x ZI 5000[㎛], we observed electric field Ex, Ey in x, y-direction. By applying these parameters of single waveguide to simulate a X-switch, we have chosen index change of 0.002, width of 3[㎛] and angle of 0.4。∼0.6°of optical waveguide and under these conditions, optical beam propagates cross-side at 0.4° When applied switching vlotage of 25[V], optical beam of X-switch turns cross-side to bar-side at intersection angle 0.4° index change of 0.002, wavegulde width of 3[㎛], electrode gap 2[㎛]. By the above results, we can obtain the basis design conditions of X-switch optical modulator. And optical single waveguide is fabricated on the y-cut LiNbO₃ single crystal substrate by Ti-indiffusion method. The line-width of the waveguide was made from 4[㎛] to 6[㎛]that observed single mode at 4[㎛] as a result of propagation of He-Ne(0.6328[㎛]) laser beam.

      • 왕방산 남사면의 식물상에 관한 연구

        This study was to establish the floristic composition of resources vascular plants of the southern slope area at Mt. Wangbang(736.3 m; 127˚ 08' 11" -127˚ 12' 19" E, 3 7˚ 51' 29" ~37˚ 55' 32" N). This work was investigated during the period of 1996 & 2002. Resources vascular plants were composed of 100 families, 301 genera, 409 species, 47 varieties and 9 formae, Totaling 465 taxa. Among the investigated resources plants, 7 taxa(l.2%) were Korean endemic species. The special plants based on floral region by Ministry of Environment(1999) were 27 taxa; Ⅳ rank species 2 taxa, Ⅲ rank species 4 taxa, Ⅱ rank species 4 taxa, Ⅰrank species 17 taxa. A naturalized plants were 44 species, correspond to 20.2% of totaling 218 species appeared in South Korea. Life form spectra was H-D₄-R_(5)-e type and, useful resources plants are as follows; edible source(45.2%), pasture source (41.9%), medicinal source(34.0%) and ornamental source(l6.1%) in the investigated region.

        Toluene, Xylene, Trichloroethylene 투여가 흰쥐의 시상하부와 뇌하수체의 GnRH, GnRH Receptor, Pit-1 유전자 발현에 미치는 영향

        The workplace exposure of chemicals has steadily increased, therefore the concern for subsequent effect on reproductive outcome has been an important issue in occupational medicine. In previous studies, higher rates of spontaneous abortion, reduced fertility and menstrual disorder among women, and an impairement of sperm quantity and quality among men have been associated with a wide variety of chemical agents. This study was conducted to evaluate the effects of toluene, xylene and trichloroethylene(TCE) injection on the mRNA levels of GnRH, GnRH receptor and Pit-1 genes in male rats hypothalamus and pituitary and the effects on the plasma levels of FSH, LH, prolactin and testosterone. Sprague-Dawley male rats were divided into five groups of five each according to concentration of toluene, xylene and TCE. The rats were injected subcutaneously to 0, 50, 100, 200, 400 mg/kg body weight/day of toluene, xylene and TCE, respectively for 6 days. Rat brains were excised and hypothalamus and pituitary were separated. Reverse transcription-polymerase chain reaction (RT-PCR) and RNase protection assay(RPA) were used to evaluate the GnRH, GnRH receptor and Pit-1 mRNA levels. Plasma concentrations of FSH, LH, prolactin and testosterone were assayed by radioimmunoassay(RIA). The results were as follows; 1. GnRH, GnRH receptor and Pit-1 mRNA levels in toluene and xylene injected groups, and GnRH receptor mRNA levels in TCE injected group were lowered dose-dependently. Especially, GnRH receptor and Pit-1 mRNA levels in 200mg/kg of toluene injected group, and GnRH, GnRH receptor and Pit-1 mRNA levels in 400mg/kg of toluene injected group were significantly lower than control group (p<0.05). GnRH receptor and Pit-1 mRNA levels in 400mg/kg of xylene injected group, and GnRH receptor mRNA levels in 400mg/kg of TCE injected group were significantly lower than control group(p<0.05). 2. The plasma levels of prolactin and testosterone in 400mg/kg of toluene injected group, and LH in 100, 200 and 400mg/kg of xylene injected group, and testosterone in 400 mg/kg of TCE injected group were significantly lower than control group(p<0.05). In conclusion, we speculated that toluene and xylene affected reproductive system secondarily through hypothalamus-pituitary axis, and TCE affected directly through steroidogenesis. And we recommended that further study for assessment of the reproductive toxicity of mixed organic solvent exposures should be conducted.

      • 大學生의 學校 및 社會에 대한 情報追求行爲 : 부산 대학생을 중심으로

        The study attempted to investigate the relationship between information-seeking behavior about the university life and society in general and background variables including family structure, class size and departmental employment rate. In this study, alienation and self-esteem were treated as intervening variables. For the analysis, 523 questionnaires were collected from undergraduate students at the Busan National University. A path analysis showed that respondent's self-esteem played a significant role in affecting student's information-seeking behavior and alienation. Among the background variables, class size was the only significant variable related to self-esteem. However, alienation which was assumed to be the major variable influencing information-seeking behavior among students did not show a significant effect on information-seeking behavior. It was assumed that one of the reasons for the very low relationship between these two variables was the structural constraints created by the university system such as extreme competition and anxiety for the among lower grade students.
