        마로 부터 분리한 Acetylmannan의 Paraquat 독성 억제 효과

        Paraquat is a useful nonselective herbicide widely used throught the world. However accidental or intentional ingestion of the herbicide cause fatal pulmonary injuring. But there is not suitable antidote of paraquat intoxication and therapeutic agents now be used are not effective. So, in this study we intended to evaluate the inhibitory effects of acetylmannan from Dioscorea balolas on paraquat toxicity. 100mg/kg acetylmannan from wild or cultured Dioscorea batatas was administered orally to male SD rats for 3 days and the administration time interval was 24 hours. After one hour of final administration, 50 mg/kg paraquat was administered intraperitonially. After 24 hours, the biochemical parameters of blood and tissues were examined. In paraquat treated groups, sGPT, BUN, creatinine, ALP levels were increased by 2 to 4 times of normal values. However in acetylmannan from wild Dioscorea batatas treated groups, sGPT, BUN, creatinine, ALP levels in blood and lung tissue were significantly decreased to normal levels. In acetylmannan from cultured Dioscorea batatas treated groups, BUN, creatinine were significantly decreased to normal values, but not in sGPT, ALP levels. Therefore, we concluded that acetylmannan from wild Dioscorea batatas can be used as an antidote of paraquat toxicity.

        Paraquat 독성에 대한 Schizandrin의 억제효과

        Paraquat is a useful nonselective herbicide widely used throughout the world. However, accidental or intentional ingestion of the paraquat cause fetal pulmonary injuring. But there is not suitable antidote of paraquat intoxication and therapeutic agents now be used are not effective. So, in this study we intended to evaluate the inhibitory effects of DDB(dimethyl-4, 4'-dimethoxy-5, 6, 5', 6'-dimethylene dioxyphenyl-2, 2'-dicarboxylate) on paraquat toxicity. DDB (100 mg/kg) was administered orally to SD rats 1hr after paraquat(50mg/kg) injection. After 24 hours, the biochemical parameters of blood and tissues were examined. In paraquat treated groups sGPT, sGOT, BUN, creatinine, MDA and alkaline phosphatase levels in blood and MDA, glucose-6-phosphatase activity in tissues were elevated by 2 to 5 times of normal values. However in Schizandrin treated groups, sGPT, sGOT, MDA and alkaline phosphatase activity in blood and MDA and glucose-6-phosphatase activity were significantly decreased to notmal levels but not in biochemical parameters of nephrotoxicity, BUN and creatinine levels. Therefore, we concluded that schizandrin can be used as an antidote of pulmono, hepatotoxicity of paraquat.

      • Cis-Diaminocyclohexane을 배위자로 하는 백금(Ⅱ)착체의 선택적 세포독성

        The use of cisplatin is limited by severe side effects such as renal toxicity. Our platinum-base drug discovery is aimed at developing drugs capable of diminishing toxicity and improving antitumor activity. We synthesized new Pt (II) complex analogue [Pt (cis-DACH)(DPPP)]ㆍ2NO₂,(PC) containing cis- l,2-diaminocyclohexane as a carrier ligand and 1, 3-bis(diphenylphosphino) propane as a leaving group. Furthermore, nitrate was added to improved the solubility. In this study, its structure was determined and its antitumor activity against SKOV-3 and NIH-OVCAR-3 human ovarian adenocarcinoma, and in nitro cytotoxicity was determined against primary cultured rabbit kidney proximal tubular and renal cortical cells of human kidney using colorimetric MTT assay. PC demonstrated acceptable antitumor activity against SKOV-3 and NIH-0VCAR-3 human ovarian adenocarcinoma and significant activity as compared with that of cisplatin. The toxicity of PC was found quite less than that of cisplatin using MTT and ³H-thymidine uptake tests in rabbit proximal tubular cells and human kidney cortical cells. PC was used for human cortical tissue in 7 weeks hitoculture by the glucose-consumption tests. We determined that the new platinum drug has lower nephrotoxicity than cisplatin. Based on these results, this novel platinum (Ⅱ) complex compound (PC) represent a valuable lead in the development of a new anticancer chemotherapeutic agent capable of improving antitumor activity and low nephrotoxicity.

        비복신경이식을 이용한 긴 거리의 말초신경 결손부 수복

        The surgery of oral and maxillofacial area poses the risk of cranial nerve damage such as trigeminal nerve or facial nerve. Inferior alveolar nerve is prone to damage in the third molar extraction, implant installation, orthognathic surgery, open reduction and rigid fixation, and tumor ablation surgery. On the other hands,facial nerve is likely to be damaged or sacrificed with trauma or parotidectomy. In case of inferior alveolar nerve injury, the incidence is reported to be about 1.3%. The nerve function will almost recover in minimal damage, but it won't recover at last in total damage of a part of nerve unit. In latter cases, nerve regeneration in intended by allograft as nerve substitute or various route of merve condit. But the recovery with autograft is believed to be most relialbe mrthod in the rapair of long-span(longer than 15㎜)nerve defect. We have performed autologous sural nerve graft in the repair of nerve defect, which is caused by resection of benign or malignant tumor. Hereby we report the method of nerve harvesting, recovery of defected peripheral nerve and the complications of donor site with the discussion of sural nerve anatomy.

      • Berberinephenolbetaine 誘導體合成 및 活性硏究[Ⅲ]

        Nucleophilic additon of allyl magnesium bromide to berberine chloride resulted in the 8-allyl-9, 10-dimethoxy-2, 3-methylenedioxy-13, 13a-dehydroberbine[Ⅰ]. And oxidation of [Ⅰ] with m-chloroperbenzoic acid in a stream of Argon at-20∼-30℃ afforded the 8-allylberberinephenolbetaine [Ⅱ], and cycloaddition of [Ⅱ] with dialkylacetylenedicarboxylate(R; a=CH_3, b=C_2H_5) in THF gave 8-allyl-9, 10-dimethoxy-2, 3-ethylendioxy-13-oxo-8, 14-ethenodimethyldicarboxyberbine[Ⅲ a] and 8-allyl-9, 10-dimethoxy-2, 3-methylenedioxy-13-oxo-8, 14-ethenodiethyl-dicarboxy-berbine[Ⅲb]. Heating of 8, 14-ethenoberbines[Ⅲa and Ⅲb] in ethanol effected smooth isomerization leading to the 3, 4-[benzo(3",4"-dimethoxy)]-1, 8-[dihydre(2',3'-methylenedioxy) naphthalene]-5-allyl-6, 7-dimethylcarboxy-2-oxo-azocine[Ⅳa] and 3, 4-[benzo(3", 4"-dimethoxy)]-1, 8-dihydro(2', 3'-methylenedioxy)naphthalene]-5-allyl-6, 7-diethylcarboxy-2-oxo-azocine[Ⅳb]. Irradiation of berberine chloride in methanol containing rose bengal and sodium methoxide with O_2 stream afforded the 8, 9, 10, 14-tetramethoxy-2, 3-methylenedioxy-13-oxoberberine[Ⅳ], and recrystallization of [Ⅳ] from methanol gave 8-methoxyberberinephenolbetaine[Ⅶ]. And cycloadditon of [Ⅶ] with dimethyl-acetylenedicarboxylate gave 8, 9, 10-trimethoxy-2, 3-methylene-dioxy-13-oxo-8, 14-ethenodimethyldicarboxyberbine[Ⅷ], and heating of [Ⅷ] in ethanol effected smooth isomerization leading to 3, 4-[benzo(3" 4"-dimethoxy)]-1, 8-[dihydro(2',3'-methylene-dioxy) naphthalene]-5-methoxy-6, 7-dimethylcarboxy-2-oxo-azocine[Ⅸ]. The in vitro biological evaluation showed that compounds[Ⅷ] and [Ⅸ] had antibacterial effect against Pseudomonas aeruginosa, and compounds [Ⅰ], [Ⅱ], [Ⅲb] and [Ⅶ] completely had anticancer activity to P-388 mouse lymphocyle leukemia at concentration of 100㎍, and [Ⅰ] almost had anticancer activity at concentration of 20㎍.

      • 개발한 제3세대 항암제의 아만성 독성연구

        The purpose of this study was to characterise the subacute toxic potency of i, v. administered KHPC-005 and 006. Few test compounds-related toxic effects were observed in body weight gain, clinical signs, urinalysis, hematological parameters and serum biochemical values. Gross necropsy and histopathology revealed no evidance specific toxicity. Our data indicated that no-observed effect level of KHPC-005 and 006 were estimated to be 10mg/kg and 4mg/kg in male rats, and 10mg/kg and 1.33mg/kg in female rats, respectively.

      • 새로운 항암성 백금(Ⅱ) 착체류의 in vivo 독성연구

        The general toxicological study of new platinum(Ⅱ) compounds, KHPC-002, KHPC-005 and KHPC-006 were investigated in rats. The effects of these Pt(Ⅱ) complexes on renal, hematopoietic and hepatic system in rats showed lower toxicity compared with cisplatin. In the consideration of the maximal dose of these Pt(Ⅱ) complexes using in this experiment is 4-8 times higher than cisplatin, these novel compounds will have the less general toxicity in vivo.

        In the head and neck area, there are so many sensory nerves, which are sometimes injuried iatrogenically or inadvertently so that involved patients complained of the loss of sensations. In such cases, it is important to judge the degree of injuries and regeneration of nerve for better diagnosis and treatment. Seddon and Sunderland's classification, which is commomly used, is focused on histological change and nerve conduction. As times goes by, it is difficult to access patient's sensory disturbance by this method. Until now, so many methods such as contract threshold, direction, two-point discrimination, pin prick, thermal discrimination and current perception threshold have been introduced for sensory evaluation. However, there hasn't been enough information regarding each methodology nor integrated standard methodology for the measurement. the purpose of this study is to get Korean adult normative sensory values of lower lip,chin and tongue using modified methods of contact thershold, ditection, two point discrimination, pin prick, thermal discrimination and assess degree of regeneration of sensory nerve damage.

      • 암치료에 있어서 체질과 항암효과에 관한 한의학적 연구

        As one of the chemical and experimental study on cancer treatment in Korea oriental medical hospital. Furthermore, we carried out the cytotoxicity test of extracts of herbal medicine. The results are as follows : About Clinical Aspects of the Oriental Medical Cancer Treatment - (1) Clinical experience is that 420 Oriental medical hospital were surveyed. Number of cancer patient treated by one oriental medical doctor was under 10(about 60%) or from 10 to 20(about 30%). (2) Diagnostic Procedure is that more than 80% of cancer patients were diagnosed by medical doctor of western hospital. (3) Therapeutic Method is that combination of oriental and western medical treatment was used in oriental medical hospital. (4) Relationship between cancer and constitutions is that to the relationship between cancer and constitutions, about 75% responded and most were considered to be Um. About Experimental Study - to evaluate the direct cytotoxicity of hot water and methanol extracts of 34 oriental medical presciptions on stomach cancer cell, SRB assay was carried out. (1) The IC50 value of water extract of Samsoum was 19㎍/㎖ and that of methanol extract was 15㎍/㎖, so that we confirmed Samsoum has a strong direct cytotoxicity. (2) The IC50 values of water extract of Coptidis Rhizoma, methanol extract of Caesalpiniae Lignum and Draba Semen were 10㎍/㎖, 19㎍/㎖ and 12.5㎍/㎖ respectively, so that we confirmed Coptidis Rhizoma, Caesalpiniae Lignum and Draba Semen have a strong direct cytototoxicity. (3) The IC50 value of combinatory prescription of Rhei Rhizoma, Forsythie Fructus and Caesalpiniae Lignum was about 40㎍/㎖.
