      • 유전자 증폭법(PCR)을 이용한 methicillin-registant Staphylococcus 균속의 신속동정에 관한 연구

        In order to indentify rapidly the methicillin-resistant genus Staphylococcus (MRS) which is known as a causative bacteria of nosocomial infection, we performed the polymerase chain reaction (PCR) method. In this study, we used 44 strains which were consist of 39 starins of Staphylococcus aureus, 2 strains of Staphylococcus epidermidis. 1 strain of Staphylococcus warneri, 1 strain of Staphylococcus cohnii isolated from clinical specimens and 1 type strain of Staphylococcus aureus KCTC 1928 for PCR experiments. DNA was extracted directly from the bacterial colony. 22 mer-oligonucleotides were used as primer which complements to the mecA region of structural gene coding low affinity binding pro-tein (PBP2a). A single band of 533 bp length from MRSA, MRSE and MRSW were confirmed. Also, an antibiotic susceptibility test by agar dilution method of NCCLS was performed for the strains isolat-ed from clinical specimens. The results obtained by this method were compared with the PCR results. In the result, most of the strains which have strong resistance to methicillin were amplified for the 533 bp fragment of the mecA gene in PCR experiment. However, 4 strains (MRA1-10, MRW2-3. MRE1-15. MRE1-16) that were classified as a sensitive group in the antibiotic susceptibility test also was showed mecA gene fragment in PCR. This suggested that the specificity of mecA gene is not always consistant with that of MRSA. In order to confirm the fragment in a region of mecA gene, a southern blot hybridization process was performed using the PCR products as probe. As a result, a single hybrid band was detected at about 4 Kb region of Hind III and Hae III-digested DNA fragment in MRS. Conclusively for the diagnosis of MRS, the confirmation of mecA gene by PCR seems to be more accurate and rapid than the antibiot-ic sensitivity test.

        이동장애인을 위한 山林浴路의 계획 및 설계 : 노선형식, 거리, 경사 및 노폭을 중심으로

        This study was carried out to plan and design an accessible forest aromatic trail for the disabled people. In order to achieve the object, environmental characteristics of forest aromatic bath trails, the actual condition of the disabled people, and guideline of disability were investigated. The results of the study are classified into two categories; First, the exiting forest aromatic bath trails have some environmental problems in relation with slope, road surface conditions and regulational defects especially in forest recreational system. Thus, it is too difficult for the disabled people to use these trails. However, it is convenient to access there, because several forest recreational areas are widely located both in suburban and urban areas. If the trails are environmentally suitably improved, they will be good accessible recreation facilities and satisfy the public interest. Second, the trail type is designed as a mixed trail system, which is a combination of the loop and the line form. Its distance should be 150m~560m and the required moving time is about 30~60 minutes. The standard for running slope should be 1/8(12.5%) maximum, in length for 3m maximum, and cross slope cannot exceed 1/20(5%) maximum. The surface slope of resting space Should be 1/50(2%) maximum. The clear tread width of trail shall be 90~120cm(one-way) and 150~180cm(two-way). If the gutter cross slope was over 10%, the length of vertical curve should be applied. The surface of trails should be firm and stable.

      • 분산 루프 시스템에서 결함진단 재구성 알고리즘

        분산 시스템의 설계에서 중요시 되는 것은 바로 결함 허용이며, 이 시스템들은 중복 또는 여분의 프로세서들(노드들)이나 중복 통신 링크들에 의해서 결함 허용이 이뤄진다. 결함 허용에는 4가지 기본에 의해서 처리과정이 요구된다. 즉, 결함탐지, 결함진단, 시스템 재구성, 시스템 복구이다. 다중 프로세서 네트워크의 주요한 설계 목적 중에 하나인 노드 (프러세서)에 대한 재구성은 집중이나 분산 방식으로 각각 이루어질 수 있는데 본 논문에서는 분산 루프 시스템을 위한 시스템 모델및 분산 방식에 따른 동적 재구성 알고리즘을 제시하고 그 성능을 평가한다. 제시된 알고리즘은 재구성 수행시 후보 로컬 슈퍼바이저의 사용없이도 로컬 슈퍼바이저의 propagation 방향을 동적으로 결정함으로서 propagation 단계를 줄여 재구성 시간을 단축시킬 수 있도록 하였으며 이를 위하여 시스템 구성시 여분 노드를 동적으로 배치하였다.

      • 중공사막 모듈에 의한 Na의 분리에 관한 연구

        By use of hydrophobic hollow fiber membrane module, this study examines the value of Kw(overall mass transfer coefficient) on Na^+, and used Di-2-ethylhexylphosphonic acid mono-2-ethylhexyl ester(EHPNA), Dodecyl-1,1,3,3,5,5-hexa-methyl hexylamine(LA-2), as extractant. We also shown the effect of additive(Cl^-, C_8H_17OH) and mixed extractant(Tri-n-octylphosphineoxide(TOPO)). LA-2 separated well by increased concentration of HCl and EHPNA decreased. LA-2 shown better separation ratio at compare EHPNA with LA-2. The effect of Kw vs. count ion(Cl^-) was shown that Kw increased by increased concentration of Cl^-, in effect of C_8H_17OH was weak.

        수침고목재의 보존을 위한 PEG, Sucrose, Lacitol 처리 및 습도조건에 따른 상태변화 실험

        광주 신창동 저습지유적에서 출토된 수침목재유물에 적합한 보존처리 방법을 설정하고자 동일유적에서 출토된 2종의 자연목재를 PEG(Poly-Ethylene Glycol), Sucrose, Lactitol로 처리하고 약제별 치수안정성 및 처리목재의 습윤↔건조 특성을 상호 비교하였다. 그 결과 상수리나무류는 저분자의 PEG#200(MW:200)과 고분자량의 PEG#4000(MW:3,350)을 이용한 2단계 PEG처리(PEG-2Step)에서, 단풍나무류는 PEG#4000만을 단독으로 적용한 처리에서 가장 높은 치수안정성을 나타내었다. 또한 처리 후 습윤↔건조 실험에서는 2단계 PEG처리한 시험재의 치수변화가 가장 작게 나타났다. 반면 Sucrose와 Lactitol의 경우 약제 확산속도가 빠르지만 Sucrose처리재는 84%의 높은 습도조건에서 흡습량이 급격히 증가하면서 약제가 다량 용출되었고, Lactitol은 습윤↔건조 후 미세한 할렬과 함께 처리재가 팽창되는 현상이 발생되었다. 또한 습윤↔건조 실험에서 2단계 PEG처리를 제외한 모든 시험재에서 갈라짐이 발생하였다. 이번 실험결과 신창동 저습지 출토수침목재는 2단계 PEG처리법을 적용한 경우에 치수안정성이 가장 높고 습도변화에 대한 적응성도 양호한 것으로 나타났다. In order to studies proper conservation treatment condition of waterlogged archaeological wood excavated from wetland in Shinchang-dong, Gwangju, 2 kinds of wooden objects were treated with PEG(Poly-Ethylene Glycol), sucrose and lactitol and their size stability and relative humidity were analyzed and compared each other. The result showed that Quercus spp. had the highest size stability in 2 Step PEG treatment using PEG#200(MW:200) and PEG#4000 (MW:3,350) and Acer spp. was the highest in treatment using only PEG#4000. In relative humidity test after treatment 2 Step-PEG treatment showed the lowest size stability. In the meantime, sucrose and lactitol-treated sample was fast for penetration, sucrose-treated sample showed a sharp increase for penetration in as high as 84% humidity condition and medicine flew out a lot and lactitol-treated sample got enlarged with fine cracking(splitting) in relative humidity test. In relative humidity test, the samples showed cracking(splitting) in all treatment materials except for 2 Step-PEG treatment. This study showed that waterlogged archaeological wood excavated from Shinchang-dong had the highest size stability and highest adaptation to humidity change in case of treatment with 2 Step-PEG.

        An analysis of the cure kinetics of two different bifunctional epoxy resins and aromatic diamine was performed. A series of isothermal differential scanning calorimetry (DSC) runs provided information about the kinetics of cure in the temperature range, 110∼125℃. All kinetics parameters of the curing reaction, including the reaction rate order, activation energy and frequency factor were calculated and reported. Glass transition temperature and extent of the residual exotherm were dependent on temperature.

        고장으로 인한 대구경 계량기의 불감률

        Leaks and meter under registrations are suspected as primary components of unaccounted-for water in Korea. Roughly, one third of the unaccounted-for water is believed to be due to meter under registration. However, no justification has been given to prove the statistics. In this study the rate of meter under registration due to failures of large-diameter meters is investigated. Replacement records of over 2600 consumer sites are analyzed. The minimum, the maximum and the probabilistic distribution of meter under registration rate are computed.

        Vocal cord dysfunction (VCD) is a respiratory disorder characterized by the abnormal adduction of the vocal cords upon inspiration, expiration, or both, leading to a diverse array of acute obstructive airway signs and symptoms (wheezing, chest tightness, shortness of breath, and cough). VCD frequently mimics or confounds asthma. Misdiagnosis as asthma has led to inappropriate treatment, most notably with high-dose corticosteroids. Mistaken for anaphylaxis or angioedema, VCD has led to intubation and tracheostomy. VCD should be suspected in patients who are treated aggressively for asthma but continue to experience dyspnea. The diagnosis of VCD is made from direct visualization of the vocal cords during an attack. Inspiratory, anterior vocal cord closure with a posterior glottic chink is seen. On pulmonary function testing, a pattern of "saw-toothing", or fluttering of the inspiratory limb, representing fluctuations in the abnormal cord motion is seen in VCD patient. Treatment includes discussion of the diagnosis with the patients, discontinuing unnecessary medications, using of a helium-oxygen mixture in acute attack. Speech therapy and psychotherapy are the mainstays of long-term treatment. Here in we report a case of VCD in company with asthma.

      • Late Quaternay Paleoceanography as Recorded by Planktonic Foraminifera in the Ulleung Basin, East Sea

        Paleoceanographic history of the East Sea is reconstructed based on several environmental parameters (coarse fraction content, planktonic foraminifera/benthic foraminifera ratio, fragmentation and assemblages of planktonic foraminifera, and coiling ratio of Neogloboquadrina pa-chyderma, etc,) of the late Quaternary sediments obtained from the Ulleung Basin. N. pa-chydeyma and Globigerina bulloides are dominant species (greater than 90% in abundance)among the total planktonic foraminifera assemblages in the late Quaternary sediments. The benthic foraminifera rarely occurred throughout the cores. Sinistrally-coiled specimens of N. pa-chyderma representing cold water temperature are observed more abundantly than dextrally-coil-ed ones. In addition, the sinistrally-coiled N, pachydeyma showed more the amount at the lower part of the cores than at the upper part suggesting the restriction of the Tsushima Warm Current into the East Sea during glacial period. G. bulloides, a species representative of upwelling condition, shows more abundant occurrence in the sediments of Core 941013 than those of Core 941006. This implies that Core 941013 is more influenced by upwelling than Core 941006. The upper part of the two cores contain more fragmentation of planktonic foraminifera suggesting significant dissolution by corrosive bottom wafer. Ascending CCD also played an important role for the absence of planktonic foraminifera at the upper part of the cores.

        Expression Analysis of Lily Type Lectin Isotypes in the Rock Bream, Oplegnathus fasciatus: in the Tissue, Developmental Stage and Viral Infection

        Lectins belong to the pattern-recognition receptors (PRRs) class and play important roles in the recognition and elimination of pathogens via the innate immune system. Recently, it was reported that lily-type lectin-1 is involved when a pathogen attacks in the early immune response of fish. However, this study is limited to information that the lectin is involved in the innate immune response against viral infection. In the present study, the lily-type lectin-2 and -3 of Oplegnathus fasciatus (OfLTL-2 and 3) have been presented to be included B-lectin domain and two D-mannose binding sites in the amino acid sequence that an important feature for the fundamental structure. To investigate the functional properties of OfLTLs, the tissue distribution in the healthy rock bream and temporal expression during early developmental stage analysis are performed using quantitative real-time PCR. OfLTL-2 and 3 are predominantly expressed in the liver and skin, but rarely expressed in other organ. Also, the transcripts of OfLTLs are not expressed during the early developmental stage but its transcripts are increased after immune-related organs which are fully formed. In the challenge experiment with RBIV (rock bream iridovirus), the expression of OfLTLs was increased much more strongly in the late response than the early, unlike previously known. These results suggest that OfLTLs are specifically expressed in the immune-related tissues when those organs are fully formed and it can be inferred that the more intensively involved in the second half to the virus infection.
