      • Infinitely many solutions for equations of <i>p</i>(<i>x</i>)-Laplace type with the nonlinear Neumann boundary condition

        <P>We investigate the following nonlinear Neumann boundary-value problem with associated <I>p</I>(<I>x</I>)-Laplace-type operator</P><P/><P>where the function <I>φ</I>(<I>x, v</I>) is of type |<I>v</I>|<SUP>p(<I>x</I>)−2</SUP><I>v</I> with continuous function <I>p</I>: <I>→</I> (1,<I>∞</I>) and both <I>f</I> : <I>Ω ×</I> ℝ <I>→</I> ℝ and <I>g</I> : <I>∂Ω ×</I> ℝ <I>→</I> ℝ satisfy a Carathéodory condition. We first show the existence of infinitely many weak solutions for the Neumann problems using the Fountain theorem with the Cerami condition but without the Ambrosetti and Rabinowitz condition. Next, we give a result on the existence of a sequence of weak solutions for problem (P) converging to 0 in <I>L</I><SUP><I>∞</I></SUP>-norm by employing De Giorgi's iteration and the localization method under suitable conditions.</P>

        ADHD 아동의 공격성과 반항성 및 모-자 상호작용을 위한 치료놀이 사례연구

        본 연구는 치료놀이가 주의력 결핍 과잉행동 장애(Attention Deficit Hyperactivity Disorder) 아동의 공격성과 반항성 및 모-자 상호작용에 미치는 영향을 알아보는데 목적이 있다. 연구대상은 ADHD 진단을 받은 만 9세 초등학교 3학년 남아로 ADHD 증상으로 인해 감정조절이 어렵고 모-자녀 및 또래 관계 문제를 보이는 아동이었다. 치료놀이는 2014년 3월부터 2015년 2월에 걸쳐 주 1회 50분씩 총 44회 진행되었다. 연구도구로는 아동·청소년행동평가척도(K-CBCL), 마샥상호작용평가(MIM), MIM 평정척도, 마샥행동평정척도를 실시하였고, 질적분석으로는 각 회기의 모든 활동과 행동 변화를 치료자가 관찰, 기록하고 분석하였다. 본 연구를 통해 얻은 결론은 다음과 같다. 첫째, 치료놀이가 ADHD 아동의 공격성 및 반항성 감소에 효과가 있는 것으로 나타났다. 둘째, 치료놀이가 ADHD 아동의 모-자 상호작용의 긍정적인 효과가 있는 것으로 나타났다. 이상의 결과를 통해 치료놀이가 ADHD 아동의 공격성과 반항성을 감소시키고 모-자 상호작용을 증진시키는데 긍정적인 효과가 있음을 입증하였다. This study investigated the effects of theraplay for Aggression and Oppositional Defiant problem of children with Attention Deficit Hyperactivity Disorder(ADHD) and mother-child interaction. The participant of this study was a boy (9 years old) in the 3rd grade of elementary school with emotional regulation problem, mother-child relationship problem and peer relationship problem due to ADHD. A total of 44 sessions were given from February, 2014 to March, 2015. Each session was fifty minute long, one session a week. For analysis, Korean-Child Behavior Checklist(K-CBCL) and Marschak Interaction Method(MIM), Marschak Interaction Method Rating System, Marschak Behavior Rating Scale. For Qualitative analysis, all activities of each session are observed, recorded, and analyzed by therapist. The results obtained are as follows, First, theraplay is effective on reducing Aggression and Oppositional Defiant problem. Second, theraplay is effective on improving mother-child interaction. As a result of this study, theraplay has a positive effect on reducing aggression and oppositional defiant problem and improving mother-child interaction.

        SARS-CoV-2 mRNA Vaccine Elicits Sustained T Cell Responses Against the Omicron Variant in Adolescents

        Vaccination against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been acknowledged as an effective mean of preventing infection and hospitalization. However, the emergence of highly transmissible SARS-CoV-2 variants of concern (VOCs) has led to substantial increase in infections among children and adolescents. Vaccineinduced immunity and longevity have not been well defined in this population. Therefore, we aimed to analyze humoral and cellular immune responses against ancestral and SARSCoV- 2 variants after two shots of the BNT162b2 vaccine in healthy adolescents. Although vaccination induced a robust increase of spike-specific binding Abs and neutralizing Abs against the ancestral and SARS-CoV-2 variants, the neutralizing activity against the Omicron variant was significantly low. On the contrary, vaccine-induced memory CD4+ T cells exhibited substantial responses against both ancestral and Omicron spike proteins. Notably, CD4+ T cell responses against both ancestral and Omicron strains were preserved at 3 months after two shots of the BNT162b2 vaccine without waning. Polyfunctionality of vaccine-induced memory T cells was also preserved in response to Omicron spike protein. The present findings characterize the protective immunity of vaccination for adolescents in the era of continuous emergence of variants/subvariants.

      • Effects of myeloid sirtuin 1 deficiency on hypothalamic neurogranin in mice fed a high-fat diet

        <P><B>Abstract</B></P> <P>Hypothalamic inflammation has been known as a contributor to high-fat diet (HFD)-induced insulin resistance and obesity. Myeloid-specific sirtuin 1 (SIRT1) deletion aggravates insulin resistance and hypothalamic inflammation in HFD-fed mice. Neurogranin, a calmodulin-binding protein, is expressed in the hypothalamus. However, the effects of myeloid SIRT1 deletion on hypothalamic neurogranin has not been fully clarified. To investigate the effect of myeloid SIRT1 deletion on food intake and hypothalamic neurogranin expression, mice were fed a HFD for 20 weeks. Myeloid SIRT1 knockout (KO) mice exhibited higher food intake, weight gain, and lower expression of anorexigenic proopiomelanocortin in the arcuate nucleus than WT mice. In particular, KO mice had lower ventromedial hypothalamus (VMH)-specific neurogranin expression. However, SIRT1 deletion reduced HFD-induced hypothalamic neurogranin. Furthermore, hypothalamic phosphorylated AMPK and parvalbumin protein levels were also lower in HFD-fed KO mice than in HFD-fed WT mice. Thus, these findings suggest that myeloid SIRT1 deletion affects food intake through VMH-specific neurogranin-mediated AMPK signaling and hypothalamic inflammation in mice fed a HFD.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Myeloid surtuin1 deficiency increases food intake and body weight in normal diet (ND)-fed mice. </LI> <LI> Myeloid surtuin1 deficiency decreases hypothalamic POMC and neurogranin in ND-fed mice. </LI> <LI> High-fat diet (HFD) increases hypothalamic neurogranin in mice. </LI> <LI> Myeloid surtuin1 deficiency reduces HFD-induced neurogranin, p-AMPK, and parvalbumin in the hypothalamus. </LI> </UL> </P>

        Effects of caloric restriction on the expression of lipocalin-2 and its receptor in the brown adipose tissue of high-fat diet-fed mice

        Obesity causes inflammation and impairs thermogenic functions in brown adipose tissue (BAT). The adipokine lipocalin 2 (LCN2) has been implicated in inflammation and obesity. Herein, we investigated the protective effects of caloric restriction (CR) on LCN2-mediated inflammation and oxidative stress in the BAT of high-fat diet (HFD)-fed mice. Mice were fed a HFD for 20 weeks and then either continued on the HFD or subjected to CR for the next 12 weeks. CR led to the browning of the white fat-like phenotype in HFD-fed mice. Increased expressions of LCN2 and its receptor in the BAT of HFD-fed mice were significantly attenuated by CR. Additionally, HFD+CR-fed mice had fewer neutrophils and macrophages expressing LCN2 and iron-positive cells than HFD-fed mice. Further, oxidative stress and mitochondrial fission induced by a HFD were also significantly attenuated by CR. Our findings indicate that the protective effects of CR on inflammation and oxidative stress in the BAT of obese mice may be associated with regulation of LCN2.

        Enhanced tumor retention of radioiodinated anti-epidermal growth factor receptor antibody using novel bifunctional iodination linker for radioimmunotherapy

        <P>Radioimmunotherapy (RIT) uses an antibody labeled with a radionuclide to deliver cytotoxic radiation to a target tumor cells. Radioiodine is most commonly employed to prepare radiolabeled proteins (antibodies, peptides) for <I>in vitro</I> and <I>in vivo</I> applications. A major shortcoming of radioiodinated proteins prepared by direct labeling methods is their deiodination <I>in vivo</I>. For the preparation of more stable radioiodinated antibodies, we developed a new linker (N-(4-isothiocyanatobenzyl)-2-(3-(tributylstannyl)phenyl) acetamide (IBPA). This study evaluated the usefulness of IBPA as a linker for the stable radioiodinated internalizing antibody, cetuximab. Directly labeled cetuximab ([<SUP>125</SUP>I]-cetuximab) was prepared by the chloramine T method. To prepare indirectly labeled cetuximab using IBPA ([<SUP>125</SUP>I]-IBPA-cetuximab), IBPA was radioiodinated using chloramine-T to give N-(4-isothiocyanatobenzyl)-2-(3-[<SUP>125</SUP>I]phenyl)acetamide ([<SUP>125</SUP>I]-IBPA), which was purified by high performance liquid chromatography. [<SUP>125</SUP>I]-IBPA was then conjugated to cetuximab. <I>In vitro</I> target binding and internalizing assays were performed in PC9, LS174T, and FaDu cell lines. <I>In vivo</I> planar images were obtained using an Inveon SPECT scanner 3, 24, 48, and 168 h after i.v. injection of [<SUP>125</SUP>I]-cetuximab or [<SUP>125</SUP>I]-IBPA-cetuximab in athymic mice bearing LS174T tumor xenografts. Specific binding and internalized radioactivity of [<SUP>125</SUP>I]-IBPA-cetuximab were higher than those of [<SUP>125</SUP>I]-cetuximab in PC9, LS174T, and FaDu cell lines. In planar images scant radioactivity was evident in thyroid glands after injection of [<SUP>125</SUP>I]-IBPA-cetuximab, while a high level of radioactivity was present in thyroid glands after injection of [<SUP>125</SUP>I]-cetuximab. Tumor uptake value of [<SUP>125</SUP>I]-IBPA-cetuximab was higher than that of [<SUP>125</SUP>I]-cetuximab for up to 168 h. [<SUP>125</SUP>I]-IBPA-cetuximab is stable and resistant to deiodination <I>in vivo</I>. IBPA is a promising bi-functional linker for radioiodination of internalizing monoclonal antibodies for <I>in vivo</I> applications including radioimmunotherapy.</P>

        대한민국인재상 발전방향 탐색

        본 연구는 우리나라 대한민국인재상 제도의 검토와 아울러 해외의 유사제도의 비교를 통하여 현행 대한민국인재상 제도의 문제점을 파악하고, 개선점을 제시하고자 한다. 이를 위해 대한민국인재상과 관련한 정책 자료, 법규, 사례 등 문헌을 분석해, 제도 운영 현황과 문제점을 파악했다. 이와 함께 국내·외 유사한 정책을 심층적으로 조사해, 대한민국인재상과 비교한 후 개선점을 도출했다. 다음으로, 대한민국인재상 수상자들을 인터뷰했으며, 대한민국 인재상 심사과정의 실무 담당자를 대상으로 FGI를 진행했다. 또한, 대한민국인재상 수상자들을 대상으로 설문조사를 실시하여 ‘현재 직업, 대한민국인재상에 대한 인식 및 만족도, 사후관리에 대한 만족도’를 조사했다. 그 결과를 ‘기술통계분석, 빈도분석, t-test’ 등으로 정리했다. 본 연구를 통해 대한민국 인재상 제도가 대한민국을 이끌어나갈 청년 우수인재를 발굴·시상하여 미래 국가의 주축으로 성장하도록 지원하는데 기초자료를 제공하고자 하였다. This study aims to identify problems of the current system of Talent Award of Korea through the review on it by comparing to the similar overseas systems to present the points to be improved. For this, this study figured out the present conditions of system operation and problems identified by analyzing policy materials, regulations and cases related to the Talent Award of Korea. In addition, by researching similar domestic and overseas policies in-depth and comparing with the Talent Award of Korea to draw ideas for improvement. Next, the Korean Talent Award winners were interviewed, and the FGI was conducted for the person in charge of the Korean Talent Award screening process. And a survey targeting the winners of the Talent Award of Korea was conducted on “the current job, the level of awareness and satisfaction with the Talent Award of Korea, and the level of satisfaction with follow-up service. And the result was arranged with descriptive statistical analysis, frequency analysis, t-test, and etc. This study intended to provide the basic date for the Korean Talent Award System to support the growth of the future nation by discovering and awarding outstanding youth talent who will lead the Republic of Korea.
