      • 마우스 단핵 탐식 세포에서 Nitric oxide 생성의 조절 기전에 관한 연구

        ADP-ribosylation may be involved in the process of macrophage activation. Nitric oxide (NO) has emerged as an important intracellular and intercellular regulatory molecule with function as diverse as vasodilation, neural communication or host defense. NO is derived from the oxidation of the terminal guanidino nitrogen atom of L-arginine by the NADPH-dependent enzyme, nitric oxide synthase (NOS) which is one of the three different isomers in mammalian tissues. Since NO can exert protective or regulatory functions in the cell at a low concentration while toxic effects at higher concentrations, its role may be tightly regulated in the cell. Therefore, this paper was focused on signal transduction pathway of NO synthesis, role of endogenous TGF-β in NO production, effect of NO on superoxide formation. Costimulation of murine peritoneal macrophageswith interferon-gamma (IFN-γ) and phorbol 12-myristate 13-acetate (PMA) increased both NO secretion and mRNA expression of inducible nitric oxide synthase (iNOS) when PMA abolished costimulation. Pretreatmnet of the cells with PMA abolished costimuation effects due to the depletion of protein kinase C (PKC) activities. The involvement of PKC in NO secretion could be further confirmed by PKC inhibitor, staurosporine, and phorbol ester derivative, phorbol 12,13-didecanoate. Addition of actinomycine D in IFN-γ plus PMA stimulated cells inhibited both NO secretion and mRNA expression of iNOS indication that PMA stabilizes mRNA of iNOS. Exogenous TGF-β reduced NO secretion in IFN-γ stimulated murine macrophages. However addition of antisense oligodeoxynucleotide (ODN) to TGF-β to this system recovered the ability of NO production and inhibited mRNA expression of TGF-β. ACAS interactive laser cytometry analysis showed that transportation of FITC-abeled antisense ODN complementary to TGF-β mRNA could be observed within 5 min and reached maximal intensity in 30 min in the murine macrophage cells. NO released by activated macrophages inhibits superoxide formation in the same cells. This inhibition may be related on NO-induced auto-adenosine diphosphate (ADP)-ribosylation. In addition, ADP-ribosylation may be involved in the process of macrophage activation.

        합성피혁제조업체 근로자들에 있어서 N-methylformamide 배설속도에 관한 연구

        This study was conducted to examine the excretion rate of dimethylformamide(DMF) from the workers exposed to DMF. The study was done at two synthetic leather factories located in Kyeonggi-do from the period of May 2 to 30, 1996. N-methyl-formamide (NMF) concentrations in urine were measured and compared by the three exposure level of DMF in air. The mean concentration of the Low (dry and winder part), Moderate (rinsing part) and High (mixing and coating part) exposure group were 3.99±3.54, 10.19±5.69 and 32.10±7.87㎎/㎥ during workshift of 8 hours, respectively. The mean concentration of urinary N-methylformamide (NMF) were 2.13±2.58, 11.16±4.98 and 26.24±7.35㎎/g creatinine, respectively. The concentration of NMF in urine could reach to maximum in 3 hours and was reduced nearly to zero in about 18 hours after exposure to DMF.

        A new device, muscle clamping system was developed to facilitate exact quatifying technique A strabismus surgery, and reduce the risk of complications. The device is composed of a lower fixing body with three jaws and an upper supporting body. They are used to clip an extraocular muscle and fix it to the sclera with a single bite. Superior rectus recession on 16 rabbit eyes were performed with this new device. Conjunctival injection, muscle adhesion strength, and light microscopic findings were examined at 1, 2, 4 and 8 weeks postoperatively. The Conjunctival injection were minimal, adhesion power ranged from 420 to 600 gram gravity, which is sufficient in withstand the normal pull of human extraocular muscle. In microscopic exam, some Inflammatory cells and fibrosis were found. The new device was technically easy, fast, and accurate, so it may be useful in stabismus surgery.

        Trichloroethyene 처리한 흰쥐의 이물질 대사효소의 활성도에 관한 연구

        Trichloroethylene(TRI) is one of the most widely used organic solvent in many industries. It is used a degreasing agent, extract and dillution solvents. TRI is metabolized by liver microsomal cytochrome P450 to reactive intermediates, which are considered responsible for its hepatotoxicity. Cytochrome P450 is a key enzyme in the first step of TRI metabolism resulting in chloral hydrate(CH) formation, which is a rate-limiting step in TRI metalbolism. This study was performed to find change of hepatic cytochrome P$%) levels and of electron transport system for P-450 dependent reaction. Injections of TRI were done to the male Sprague Dawley rats(mean body wights, 170±10g) in corn the dosage of 150,300,600 ㎎/㎏ for 2 days. The results of experiments are following: 1. The contents of microsomal protein, cytochrome P450 and b5 are tended to be decreased with increasing amount of TRI, but not signifcantlly(p>0.05). 2. The activity of NADPH-cytochrome P450 reductase is slowly decreased by increasing amount of TRI, but not significantlly(p>0.05). On the other hand, the activity of NADH-b5 reductase is increased with increasing amount of TRI(p<0.05). 3. The activity of 7-ethoxyresolufin O-deethylase did not show any critical changes(P>0.05). This result suggests that the inductive pattern of TRI may not be related to P 450IA1 and P450IIB.

      • 골프 스트로크중 칩샷의 운동학적 분석

        본 연구의 목적은 8번 아이언으로 30M 거리를 칩 샷을 통한 3차원적 운동학적 분석을 통해서 칩 샷의 원리와 근거를 제시하여 골퍼들의 기초 자료를 제공하고자 하는데 그 목적을 두었다. 1. 선운동의 변인 스윙 소요시간에서는 숙련자가 1.083±0202sec.로 비숙련자는 1.217sec. 유의한 차이를 보였다(p<.05). event별로는 event 1에서만 유의하게 숙련자가 빠르게 나타났지만(p<.05), event 2,3에서는 차이를 보이지 않았다. 클럽헤드 속도에서는 event 3(p<.001), 4(p<.01)에서 비숙련자가 숙련자보다 빠르게 나타나 통계적으로 유의한 차이를 보였지만, event 1, 2에서는 차이를 보이지 않았다. 신체중심변위에서는 event 3에서만 숙련자가 비숙련자보다 중심이 낮게 형성되었고(p<.05). 나머지는 event 1, 2, 4에서는 차이를 보이지 않았다. 2. 관절운동의 변인 코킹각 변위에서는 event 2(p<.05), 3(p<.01)애소 통계적으로 유의한 차이를 보였고, event 1, 4에서는 차이를 보이지 않았다. 양 슬관절 각 변위에서는 오른쪽 슬관절의 event 1을 제외하고는 통계적으로 유의한 차이가 없었다. 고관절의 각 변위에서는 집단간의 통계적으로 유의한 차이가 없었다. This study is designed to present the principles and foundation of chip shots through a chip-shot three-dimension kinematic analysis of 30-meter distance with the 8th iron and to offer the reader basic materials for golfers. 1. The variations of Lineal Exercise There is a significant difference between the time required for a swing by skillful golfers(1.083±0.02sec.) and the time required for a swing by unskillful golfers(1.217sec.). Although event 1 shows that the former turns out to be faster(p<.05), event 1 and even 2 show no difference. In terms of the velocities of the clubhead, event 3(p<.001) and event 4(p<.01) turn out for the latter to be faster than the former, which means that there is a significant difference. But there is no difference in event 1 and 2. In terms of the displacement of the body center, the former forms the body center lower(p<.05) than the latter in event 3, while there is no difference in event 1, 2, and 4. 2. The Variations of the Joint Exercise In terms of the displacement of the left wrist, event 2(p<.05) and event 3(p<.01) show a statistically significant difference, and event 1 and event 4 show no difference. In terms of each displacement of both the knee joints, event 1 of right knee joint only shows statistically significant difference. In terms of each displacement of the coxal articulations, there is no statistically significant difference between groups.

        Locoregional recurrence is the most common type of recurrence in surgical operation of gastric adenocarcinoma, and peritoneal dissemination is one of the most difficult problems in advanced gastric adenocarcinoma treatment. Because the peritoneal cavity is the most common site of the first recurrence after gastric cancer resection, intraperitoneal chemotherapy seems a logical choice for cancer chemotherapy. In this study, Mitomycin C (MMC)-activated charcoal (CH)-alginate (ALG) beads were prepared by the mixtures of CH particles adsorbed with MMC as an anti-cancer drug and aqueous alginate solution. The alginate is recognized as biodegradable, nontoxic, and biocompatible. The release of MMC from the beads in 0.1 M Tris buffer was stable and continuous until about 1 week. The MMC-CH-ALG beads can be applied in the peritoneal cavity for intraperitoneal chemotherapy since they provide a good adhesiveness on the tissue and controlled release pattern of the drugs.

      • 생체분해성 고분자의 특성 및 의학적 응용

        "Biomaterials" are nonviable materials used in medical devices, intended to interact with biological systems. They should have "biocompativility", which is defined as the ability of materials to perform with an appropriate host response in a specific application. It includes many materials that are often classified as biomaterials. Metals, ceramics, polymers, glasses, carbons, and composite materials are some examples. Among them, biodegradable polymers are discussed in this revies article. Since a biodegradable polymeric implant does not have to be removed surgically once it is no longer needed, biodegradable polymers are of value in short-term applications that require only the temporary presence of a polymeric implant. Main short-term medical applications of biodegradable polymers include the temporary tissue scaffold, the temporary adhesion barrier, the drug delivery device, and the multifunctional implant. In this article, the properties, processing methods, and medical applications of the biodegradable polymers are reviewed.

      • 백서의 복강내에 투여한 Mitomycin-C-Polyglycolic acid 제형이 장기에 미치는 영향에 관한 연구

        To evaluate the in-vivo effects of mitomycin-C-polyglycolic acid composite(MMC-PGA) the disk: shaped composite were implanted into the peritoneal cavity of the Sprague-Dawley rats. The pathologic changes were examined at various time points up to 12 weeks. Initially the soft tissue around the inserted disks showed capillary congestion at 3 days. Inflammatory infiltrates with foreign body giant cells appeared from the 2nd week and reached peak response at 6-8 weeks. These reaction diminished prominently at 12 weeks. No specific pathologic change was found in the liver, the kidneys, and the heart. The above results suggest that the MMC-PGA composite can serve as a new device for intraperitoneal chemotherapy of various types of cancers.

        Biodegradable retinal fixation devices obtain mechnical fixation of the retina with desirable chorioretinal scarring and with the potential for local, sustained release of antimetabolites and steroids to inhibit proliferative vitreoretinopathy. We manufactured a biodegradable retinal tack with barb that was designed in order to prevent intrusion from implantation of retinal tacks. This study was carried to evaluate the efficacy for retinal fixation and the capability for sustained release of drugs with a newly designed biodegradable retinal tack Biodegradable retinal tacks were made of polymers of glycolic acids and were designed with barbs in a shape to prevent the disinsertion. Biodegradale retinal tacks are divided into 3 parts, a conical portion that is inserted into the sclera, a cylinder portion that remains in the vitreous, and a neck portion between the pin and the cylinder. The tapered conical end was manufactured to allow easy insertion through the retina and choroid into the sclera. A cylinder portion was manufactured with a tapered angle that fixes firmly into the orifice of 19 gauge spinal needle. A neck portion, 0.4 mm in diameter, was designed to prevent disinsertion from following implantation of retinal tack. The applicator was a 19 gauge spinal needle and its orifice was prepared to 15°angle to accept the tapered cylinder portion of the retinal tack. The retinal tacks, secured in the needles, were passed through the formed vitreous and inserted into the retina, choroid, and sclera and were released by pushing the internal needle, usually within 2-3mm of the medullary ray of the posterior rabbit retina A retinal tack was placed in each of 8 pigmented rabbit eyes. Slit-lamp biomicroscopy, indirect ophthalmoscopy and fundus photography were performed periodically from 1 day to 8 weeks after surgery. Eight eyes were enucleated and studied by light microscopy at 8 weeks. Biomicroscopic evaluation of the animals revealed edemas adjacent to the retinal surfaces immediately after insertion of the biodegradable retinal tacks in all the animals. These edemas disappeared after 1 week. The first noticeable change in the size of retinal tacks was shown after 2weeks. The size of the retinal tacks gradually got smaller, decreasing to about one-half at 4 weeks and about one-third at 8 weeks. All retinal tacks remained in inserted places without any movement for an 8 week period. On light microscopy, epiretinal proliferations were seen to extend into the vitreous cavity. Cellular capsules that lined the inner aspect of the scleral defect caused by tack insertion were found. However the adjacent retina had a normal cytologic appearance and architecture in all specimens. We manufactured a biodegradable retinal tack that is designed to prevent intrusion from implantation of retinal tacks. All biodegradable retinal tacks reduce in size with time, but no retinal tacks extruded from the inserted place. The newly designed biodegradable retinal tack can be used for retinal fixation and may be used as a vehicle for the introduction of pharmacologic agents to prevent the cellular events that promote proliferative vitreoretinopathy.
