        The feasibility of detecting endometrial and ovarian cancer using DNA methylation biomarkers in cervical scrapings

        Objective: We hypothesized that DNA methylation of development-related genes may occur in endometrial cancer (EC)/ovarian cancer (OC) and may be detected in cervical scrapings. Methods: We tested methylation status by quantitative methylation-specific polymerase chain reaction for 14 genes in DNA pools of endometrial and OC tissues. Tissues of EC/normal endometrium, OC/normal ovary, were verified in training set using cervical scrapings of 10 EC/10 OC patients and 10 controls, and further validated in the testing set using independent cervical scrapings in 30 EC/30 OC patients and 30 controls. We generated cutoff values of methylation index (M-index) from cervical scrapings to distinguish between cancer patients and control. Sensitivity/specificity of DNA methylation biomarkers in detecting EC and OC was calculated. Results: Of 14 genes, 4 (PTGDR, HS3ST2, POU4F3, MAGI2) showed hypermethylation in EC and OC tissues, and were verified in training set. POU4F3 and MAGI2 exhibited hypermethylation in training set were validated in independent cases. The mean M-index of POU4F3 is 78.28 in EC and 20.36 in OC, which are higher than that in controls (6.59; p<0.001 and p=0.100, respectively), and that of MAGI2 is 246.0 in EC and 12.2 in OC, which is significantly higher that than in controls (2.85; p<0.001 and p=0.480, respectively). Sensitivity and specificity of POU4F3/MAGI2 were 83%–90% and 69%–75% for detection of EC, and 61% and 62%–69% for the detection of OC. Conclusion: The findings demonstrate the potential of EC/OC detection through testing for DNA methylation in cervical scrapings.

        Risk factor analysis of coexisting endometrial carcinoma in patients with endometrial hyperplasia: a retrospective observational study of Taiwanese Gynecologic Oncology Group

        Objective: To evaluate the clinical outcome and parameters related to coexisting endometrial carcinoma in women with tissuediagnosed endometrial hyperplasia. Methods: Between January 1991 and December 2009, three hundred and eighty-six patients with the presumptive diagnosis of endometrial hyperplasia were retrieved. Among these, one hundred and twenty-five patients were identified as having coexisting endometrial carcinoma in hysterectomy specimens. The three hundred and eighty-six patients were divided into two groups: the hyperplasia-benign group (261 cases) and the hyperplasia-malignant group (125 cases). Several clinical parameters including age, menopausal status, history of abnormal uterine bleeding, obstetrical history, medical history of diabetes and hypertension, BMI, and preoperative pathologic results were investigated. Results: Age ≥53 (odds ratio [OR], 2.40; 95% confidence interval [CI], 1.26 to 4.57), menopausal status (OR, 2.07; 95% CI, 1.14to 3.76), diabetes history (OR, 7.33; 95% CI, 2.79 to 19.26), abnormal uterine bleeding (OR, 3.99; 95% CI, 1.22 to 13.02), atypical endometrial hyperplasia (OR, 7.38; 95% CI, 4.03 to 13.49), and body mass index ≥27 (OR, 3.24; 95% CI, 1.76 to 5.97) were independent risk factors for prediction of endometrial hyperplasia coexisting with endometrial carcinoma. The diagnostic efficacy of atypical endometrial hyperplasia to predict the endometrial hyperplasia coexisting with endometrial carcinoma was better than or similar to those of other independent factors and combinations of these factors. Conclusion: Coexisting malignancy should be considered when examining endometrial hyperplasia patients with the related risk factors, especially atypical endometrial hyperplasia.

        Dry Etching Characteristics of Zinc Oxide Thin Films in Cl_2-Based Plasma

        We investigated the etching characteristics of zinc oxide (ZnO) and the effect of additive gases in a Cl2-based inductively coupled plasma. The inert gases were argon, nitrogen, and helium. The maximum etch rates were 44.3,39.9, and 37.9 nm/min for Cl2(75%)/Ar(25%), Cl2(50%)/N2(50%), and Cl2(75%)/He(25%) gas mixtures, 600 W radiofrequency power, 150 W bias power, and 2 Pa process pressure. We obtained the maximum etch rate by a combination of chemical reaction and physical bombardment. A volatile compound of Zn-Cl. achieved the chemical reaction on the surface of the ZnO thin films. The physical etching was performed by inert gas ion bombardment that broke the Zn-O bonds. The highly oriented (002) peak was determined on samples, and the (013) peak of Zn_2SiO_4 was observed in the ZnO thin film sample based on x-ray diffraction spectroscopy patterns. In addition, the sample of Cl_2/He chemistry showed a high full-width at half-maximum value. The root-mean-square roughness of ZnO thin films decreased to 1.33 nm from 5.88 nm at Cl_2(50%)/N_2(50%) plasma chemistry.

        Anti-Wear and Friction Properties of Nanoparticles as Additives in the Lithium Grease

        TiO2/CuO nano-particles were dispersed into lithium grease to improve the lubricating properties of the greases. Then, the processof sliding friction was tested. The friction force of the contact interface between a ball and a flat surface with TiO2/CuO nano-greasewas measured for a ball on a flat sliding wear tester. The worn surface was examined by a surface roughness tester and scanningelectron microscopy (SEM). The tested lithium grease was synthesized with 0.5wt.%, 1.0wt.%, 1.5wt.% and 2.0wt.% TiO2/CuO nanoparticles. Results indicate that the TiO2/CuO nano-particle additives can improve the wear resistance of lithium grease and decreasethe friction coefficient. In addition, the results show that a 1.0 wt% content of TiO2 nano-particles is the most efficient in reducingfriction and wear, and can significantly reduce the friction coefficient of the rubbing interface by about 40%, according to the frictiontests. The wear of the lithium grease with a 2.0wt.% content of CuO nano-particles added can reduce wear about by 60% incomparison with lithium grease. Furthermore, the SEM analysis results demonstrate that the addition of tribo-film forms and TiO2/CuO nano-particles on the worn surface is responsible for the decrease in friction and wear.

      • UNDERSTANDING TOURIST SERVICE QUALITY PERCEPTIONS AND LOYALTY INTENTIONS IN THE INTERNATIONAL HOTEL SECTOR: APPLICATION OF DATA MINING METHODS AND KANO’S MODEL

        This study used the methods of decision tree analysis, association rule analysis, and Kano’s model to explore the behavior patterns of mainland China tourists staying at the international tourist hotels in Taiwan. To this end, the data of their demographics, travel variables, overall satisfaction with the lodging experience, different service quality perceptions, and loyalty intentions were included. The decision tree analysis showed that a tourist’s overall satisfaction with the lodging experience, satisfaction with the quality of core intangible services, and certain demographic characteristics are three important determinants of tourist loyalty towards the hotels. In terms of the effect of demographics, the customers’ monthly income and length of stay at the hotel are two main determinants in this study. In addition, if the customer perceptions of different parts of hotel service quality are taken into account, among the five hotel service quality domains, core intangible services from the receptionist, housekeeping personnel, and food & beverage personnel are found to be important influences on hotel customer loyalty intention. In other words, high quality intangible services are important for luxury hotels to demonstrate their unique ability to help customers experience the service quality that creates loyalty intentions. With regard to the association rule analysis, the results showed that core intangible service aspects from the receptionist, housekeeping personnel, and food & beverage personnel are strongly associated with customer loyalty intentions, as are the tangible aspects of the reception and hotel room facilities. The former indicated that reception in the hotel lobby could be considered one of the most important servicescapes because of its impact in forming many of the first impressions of hotel guests, while the latter is treated as core offerings in hotels that would be encountered by most hotel customers. If the tourists are mainly from package tours, the intangible services and tangible facilities of these areas are the important areas to create customer satisfaction. However, if the tourists are mainly independent tourists because they have more time and free choice to stay at the hotel longer than the package tour tourists, the intangible services and tangible facilities of the entertainment or business centers would be even more important to these tourists than to the package tour tourists. With regard to Kano’s model analysis, the results showed that, based on mainland China tourists’ perceptions, most of the service elements fit into the category of one-dimensional quality attributes. This means that these service elements are positively and linearly related to customer satisfaction, and the greater fulfillment of the attribute results in a greater degree of satisfaction. This also means that hotels should make more effort to innovate their intangible services and tangible facilities to create business advantages in the market.

        Enhanced Cellulolytic Enzyme Production by the Synergism between Trichoderma reesei RUT-C30 and Aspergillus niger NL02 and by the Addition of Surfactants

        Two improvement approaches comprising of a mixed culture of Trichoderma reesei and Aspergillus niger and the addition of surfactants were employed in this study in order to enhance cellulolytic enzyme production as well as to improve the composition. Different delay times of A. niger inoculation (0, 24, and 48 h) and inoculum ratios of T. reesei versus A. niger (1:1 and 5:1) derived six mixed culture forms, which were 0 h/1:1, 0 h/5:1, 24 h/1:1, 24 h/5:1,48 h/1:1, and 48 h/5:1. It was found that the form 48 h/5:1allowed the highest FPA, 3.30 ± 0.34 IU/mL, and a relatively high BGA, 1.01 ± 0.25 IU/mL, thereafter being selected for the subsequent improvement step addition of surfactants. Among the three surfactants, including Triton X-100, CHAPS and sodium taurocholate, the third one was found to be the best one giving rise to the highest FPA and BGA, 5.02 ± 0.40 and 1.48 ± 0.28 IU/mL, respectively. Differently sourced cellulases were compared in the enzymatic hydrolysis of steam-exploded corn stover (SECS). Moreover, the cellulase produced by the mixed culture form 48 h/5:1 using SECS as a substrate showed the highest yield at 80.93 ± 2.04%, indicating that the composition of this cellulase was improved by the mixed culture of T. reesei and A. niger. The results validate that these two improvement approaches are efficient and applicable in cellulase production.

        Damage on the Surface of Zinc Oxide Thin Films Etched in Cl-based Gas Chemistry

        We investigated the etching characteristics of zinc oxide (ZnO) thin films deposited by the atomic layer deposition method. The gases of the inductively coupled plasma chemistry consisted of $Cl_2$, Ar, and $O_2$. The maximum etch rate was 40.3 nm/min at a gas flow ratio of $Cl_2$/Ar=15:5 sccm, radio-frequency power of 600 W, bias power of 200 W, and process pressure of 2 Pa. We also investigated the plasma induced damage in the etched ZnO thin films using X-ray diffraction (XRD), atomic force microscopy and photoluminescence (PL). A highly oriented (100) peak was present in the XRD spectroscopy of the ZnO samples. The full width at half maximum value of the ZnO sample etched using the $O_2/Cl_2$/Ar chemistry was higher than that of the as-deposited sample. The roughness of the ZnO thin films increased from 1.91 nm to 2.45 nm after etching in the $O_2/Cl_2$/Ar plasma chemistry. Also, we obtained a strong band edge emission at 380 nm. The intensities of the peaks in the PL spectra from the samples etched in all of the chemistries were increased. However, there was no deep level emission.

        Malignant transformation of ovarian mature cystic teratoma into squamous cell carcinoma: a Taiwanese Gynecologic Oncology Group (TGOG) study

        Objective: The malignant transformation (MT) of ovarian mature cystic teratoma (MCT)to squamous cell carcinoma (SCC) is very rare. This study analyzed cases from multiplemedical centers in Taiwan to investigate the clinicopathologic characteristics, treatment, andprognostic factors of this disease and reviewed related literature. Methods: Pathological reports of 16,001 patients with primary ovarian cancer who weretreated at Taiwan medical centers from 1990 to 2011 were reviewed. In total, 52 patients withMT of MCT to SCC were identified. Results: Among all ovarian MCTs, the incidence of MT to SCC is 0.2%. The median age ofpatients was 52 years (range, 29–89 years), and the mean tumor size was 10.5 cm (range, 1–40cm). We analyzed the patients in our study and those in the literature and determined thatearly identification and complete surgical resection of the tumor are essential for long-termsurvival. In addition, adjuvant chemotherapy or concurrent chemoradiotherapy can be usedto treat this malignancy. Old age, large tumor size (≥15.0 cm), and solid components in MCTsare suitable indicators predicting the risk of MT of MCT to SCC. Conclusion: Similar to general epithelial ovarian cancers, the early detection of MT of MCTto SCC is critical to long-term survival. Therefore, older patients with a large tumor or those with a tumor containing a solid component in a clinically diagnosed MCT should beevaluated to exclude potential MT to SCC.
