        혈중 갑상선자극홀몬 측정의 진단적 의의에 관한 연구

        저자들은 혈중 TSH의 방사면역측정을 정상인과 각종 갑상선질환 환자들에게 실시하여 의미있는 결과를 얻었기에 이에 보고하는 바이다. 환자들은 1978년 1월부터 1980년 8월까지 국립경찰병원 내과에 내원한 각종 갑상선환자 41예와 정상인 22예를 대상으로 하였다. 1) 정상인 22예의 혈충 TSH치는 평균 4.2∼1.7μU/ml이고, 그 범위는 1.9∼7.4μU/ml이었다. 2) 원발성 갑상선기능저하증 9예에 있는 혈중 TSH의 평균치가 97.1±116.4μU/ml이고 그 범위는 14.0±300μU/ml였다. 3) Graves씨병 17예는 혈중 TSH치의 평균이 1.5±0.6μU/ml였다. 4) 갑상선염 8예는 갑상선기능상태에 따라 혈중 TSH치의 변화가 많았으며 그 범위는 1.6∼220μU/ml이었다. 5) 갑상선결절 7예는 혈중 TSH치의 평균이 2.4±2.0μU/ml이었다. 6) Graves씨병에 Taoazole치료후 TSH치가 상승하는 것을 볼 수 있었고, 이것이 병경과를 판정하는데 아주 유용하였다. 이상의 결과로써 혈중 TSH의 측정은 갑상선질환의 진단뿐 아니라 치료경과를 판정하는데도 좋은 수단이 될 수 있다고 하겠다. The radioimmunoassay of TSH (human thyrotropin) was performed by utilizing anti-h-TSH antibody and purified human thyrotropin supplied from Daiichi from Jan. 1978 through Aug. 1980 the serum concentration of TSH was measured on 41 cases with various thyroid diseases, and 22 normal persons. Among 41 cases, 9(22%) were primary hypothyroitlism, 17(41%) Graves' disease, 8(20%) subacute or chronic lymphocytic thyroiditis, and 7(17%) nodular goiter. The results were as follows:1) The normal values of serum TSH in 22 cases of control group were 4.2±1.7μU/ml, (1.9∼7.4μU/ml), which were within normal range in kit used in this study. 2) The serum TSH concentration in 9 cases with primary hypoehroidism were 97.1±116.4μU/ml(14. 0∼300μU/ml), which were significantly elevated as compared, with normal control values. 3) The serum TSH concentration in 17 cases with Graves' disease were 1.5±0.6μU/ml(1.0∼2.5μU/ml), which were below than normal control. 4) The serum TSH concentration in 8 cases with subacute or chrotic thyroiditis revealed wide ranges(1.6∼220μU/ml), according to the state of thyroid function. 5) The serum TSH values in 7 cases with nodular goiters were 2.3±2.0μU/ml, which were strictly within normal levels. 6) The serum TSH levels were elevated during prolonged treatment with Tapazole(Methimazole) without serial check of the serum TSH concentration in Graves' disease, so the serial measurement of serum TSH concentration was considered of available index of thyroid states.

      • 이온 보존 반응법에 의하여 표면처리된 Polyimide(PI) 표면과 구리박막의 접착력 향상

        Polyimide films are modified by Ar^+ion beam at 1 kV in an oxygen environments. Amounts of ions changed from 5 x 10^(15) to 1 x 10^(17) ions /㎠ and amounts of blowing oxygen from 0 to 8 sccm ml /min. The wettabilities and the surface free energies of modified polyimide were measured by a contact angle meter and the chemical state of the modified polyimide surface was measured by x-ray photoelectron spectroscopy. The wetting angles between water and polyimide films modified by Ar^+ ion without oxygen blowing decrease from 67 to 40 degrees and surface free energies increase from 46 to 64 dyne /㎠. The wetting angle of polyimide films modified by Ar^+ ion in an oxygen environments decrease to 12 degree and surface free energy increase to 72 dyne /㎠. Polyimide surface was modified with various gas environments and ions. Lowest wetting angle was obtained by oxygen ion irradiation in an oxygen gas environment and its value was 8°. In the case of polyimide film modified by Ar^+ ions in an oxygen environment, the wetting angle increase up to 65° when it kept in air and that increase up to 46° when it kept in water after 110 hour. In the case of polyimide film modified by O^+ ions in an oxygen environment, however, the wetting angle of polyimide film dose not increase. From the x-ray photoelectron analysis, it is found that the chemical bonds between polyimide components are severed by ion irradiation and hydrophilic groups such as (C=O)-(ON)-, COH and (C=O)-C are formed by the reaction between newly formed radicals and blowing oxygen. I t was found that the adhesion between Cu and polyimide modified by ion assisted reaction was improved, and the main reason of the enhanced adhesion is due to the reaction between Cu and C-O groups formed by ion assisted reaction on the polyimide surface.

      • 장티푸스의 Cefotaxime의 치료효과

        Cefotaxime은 반합성 Cephalosporin 제제로서 광범위항생제이며 β-lactamase에 안정성을 갖고 있으며 S. typhi에 대해서는 최저저지농도가 0.04㎍/dl로서 매우 유요한 것으로 나타나 있다. Mekendrik은 2예의 장티푸스환자에서 Cefotaxime 4.0gm/day 치료로 임상적인 효과를 못보았다고 한다. 그 이유는 Cefotaxime이 포유동물의 세포내로 침투하기 어려운 때문이라고 하였으며 또다른 임상연구에서는 Cefotaxime 투여 24시간후 혈액배양에서 S. typhi가 자란다고 한다. 본 연구에서는 45명의 많은 환자에서 Cefotaxime을 투여한 결과 비교적 양호한 치료효과를 얻었다. Table2에서 보는 바와 같이 45명중 31명이 7일이내에 치료 효과를 보였다. 평균 치료기간이 6.9일로서 Chloram-phenicol 투여시의 5.4일 보다는 길으나 임상적으로는 치료제로서 사용할만한 약제로 볼 수 있다. Santiago 에서의 연구결과는 Chloramphenicol 5.1일, Ampicillin은 6.7일이었으며 Co-trimoxazole은 7일이었다. 31명의 환자에서 Cefotaxime 2.0 gm/day의 용량으로는 해열이 되지않으므로 1 일 4.0 gm으로 중량하여 평균 3.2일만에 해열이 되었다. 그러나 초기치료부터 1일 4.0 gm 으로 시작한 자료가 없으므로 용량간에 효과를 비교할만한 근거는 없다. 3예의 재발과 5예의 단기보균자수는 Amoxycillin 치료시와 유사하다. 이들은 Co-trinoxazole로 치료하였다. 3예의 장출혈환자는 Cefortaxime의 부작용은 아니며 장티푸스환자에서 통상적으로 볼 수 있는 합병증이라고 사료된다. Cefotaxime 투여시 약간의 피부발진 이외의 부작용을 발견할 수 없었다. Cefotaxime은 가격이 비싸고 경구적 투여라는 단점이 있으나 골수장애를 일으킬 수 있는 Chloramphenicol에 대신해서 장티푸스치료에 사용할 수 있는 약제다.

        Synthesis, spectroscopy and thermal behavior of new lead(II) complexes derived from S-methyl/benzyldithiocarbazates (SMDTC/SBDTC): X-ray crystal structure of [Pb(SMDTC)(NO<sub>3</sub>)<sub>2</sub>]

        Lead(II) complexes of S-methyldithiocarbazate (SMDTC), [Pb(SMDTC)(NO<SUB>3</SUB>)<SUB>2</SUB>] (1) and S-benzyldithiocarbazate (SBDTC), [Pb(SBDTC)(NO<SUB>3</SUB>)<SUB>2</SUB>] (2) have been synthesized for the first time and characterized by elemental analysis, IR and TGA techniques. The complexes were obtained by addition of the appropriate ligand to an aqueous ethanolic solution of lead(II) nitrate in 1:1 molar ratio. The X-ray crystal structure of complex 1 has been determined by single crystal X-ray diffractometry. In complex 1, lead(II) is in a nine coordinated sphere with seven oxygen atoms of the nitrate groups and thione sulfur, β-nitrogen of neutral bidentate NS chelating ligand. Three nitrate groups act as bidentate chelating whereas the fourth nitrate group is coordinating to the central lead(II) and at the same time it bridges with neighboring lead(II) atom. Coordination geometry of the central lead(II) atom has a tricapped trigonal prismatic arrangement with streochemically inactive lone pair. The lead atoms are linked into polymeric chains and these chains form twin polymeric ribbons linked through bridging oxygen atoms. The N-H...O hydrogen bond network between N<SUB>SMDTC</SUB> and O<SUB>nitrate</SUB> atom leads to self-assembled molecular conformation and stabilizes the crystal structure. The complex 2 with similar spectral and thermal behavior is expected to have a tricapped trigonal prismatic structure. The thermal behavior studies shows that the complexes start to decompose at relatively low temperature (ca. 110<SUP>o</SUP>C) to give PbS residue.

        소의 동결수정란이 수태에 미치는 영향;3. 오단계부유에 의한 그리세롤제거란의 외과적이식의 영향

        Frozen embryos with highest genetic-potency were imported from America in cooperation with the Holstein-Friesian of America and transferred to the recipients in the National Animal Breeding Institute. The estrus synchronization were induced with 64 cows by intramuscular injections of 25㎎ PGF₂α between 11 days interval. The frozen embryos thawed at 32℃ were rehydrated within the medium at 5 steps in order to remove glycerol. The embryos were transferred to 30 recipients with surgical method and the results were as follows: 1. Among 64 cows, heat inducing rate after 1st and 2nd injections of PGF₂α was 53.1 and 76.6%, respectively. 2. Most of synchronized estrus was induced within 2-4 days after PGF₂α injections, however prevalent one was within 3 days. 3. Heat inducing rate after 1st and 2nd injections of PGF₂α was 35.5 and 77.4% in the heifer and 69.7 and 75.8% in the cow. The rate after 2nd injection was much higher than 1st injection and the cow showed much higher rate than the heifer in 1st injection. 4. Among 30 recipients, 20 cows (66.7%) has corpus luteum in right ovary and 10 cows (33.3%) in the left. Three cows in 16 cows with A-class CL, 7 cows in 12 cows with B-class CL and 1 cow in 2 cows with C-class CL showed highest conception rate. 5. The frozen embryos staged from late morula to expended blastocysts were transferred and concepted. Conception was possible even in the C-class embryo. 6. Among 30 recipients, 11 cows (36.7%) were concepted and the cows (37.5%) showed slightly higher conception rate than the heifer (36.7%). 7. The conception rate of frozen embryo was 1 S.8% in first trial with 6 steps glycerol equilibration, 16.7% in 2nd trial with 2 steps sucrose equilibration and 36.7% in this 3rd trial with S steps glycerol equilibration. Third trial showed highest conception rate. 8. Among 79 recipients, 26 cows (32.9%) were concepted and farrowed. The conception rate of fresh embryos (55.6%) was higher than the frozen (26.2%) and that of the surgical (35.0%) was slightly higher than the nonsurgical (30.8%).

        Under cyclic loading, the fatigue failures of welded joints occur at weld toes which induce stress concentration by weld shape. So we need to obtain the peak stress and the S-N curve to assess the fatigue strength of welded joints. However the measurement of peak stress is of high uncertainty and low reproducibility, so we use nominal stress instead in fatigue tests of welded joints. In this study, fatigue tests to obtain S-N curves am FE analyses to obtain stress concentration factors were conducted for the two types of cruciform fillet welded joints, that is, load-carrying and non load-carrying types. Then we changed the obtained S-N curves to that based on peak stress using the bot-spot stress concept. From the analyses of the S-N curves obtained, we have concluded that there is a need to develop a new method to evaluate the fatigue life.

        Novel anti-apoptotic mechanism of A20 through targeting ASK1 to suppress TNF-induced JNK activation

        The zinc-finger protein A20 has crucial physiological functions as a dual inhibitor of nuclear factor-κB (NF-κB) activation and apoptosis in tumor necrosis factor (TNF) receptor 1 signaling pathway. Although the molecular basis for the anti-NF-κB function of A20 has been well elucidated, the anti-apoptotic function of A20 is largely unknown. Here, we report a novel mechanism underlying the anti-apoptotic function of A20: A20 blocks TNF-induced apoptosis through suppression of c-jun N-terminal kinase (JNK) by targeting apoptosis signal-regulating kinase1 (ASK1). First, the ectopic expression of A20 drastically inhibits TNF-induced JNK activation and apoptosis in multiple cell types including those deficient of NF-κB activation. Unexpectedly, the blunting effect of A20 on TNF-induced JNK activation is not mediated by affecting the TNFR1 signaling complex formation. Instead, A20 interacts with ASK1, an important MAPKK kinase in the JNK signaling cascade. More importantly, overexpression of wild-type A20, but not of mutant A20 (ZnF4; C624A, C627A), promotes degradation of the ASK1 through the ubiquitin-proteasome system. Taken together, the results from this study reveal a novel anti-apoptotic mechanism of A20 in TNF signaling pathway: A20 binds to ASK1 and mediates ASK1 degradation, leading to suppression of JNK activation and eventually blockage of apoptosis.

        Evaluation of Manganese Ion on Controlling Harmful Microorganisms In vitro and In vivo for the Early-Weaned Pig

        Two experiments were conducted to determine the effects of $MnSO_4$ on controlling harmful microorganisms in vitro and in vivo. The in vitro experiment was conducted to examine the effects of manganese sulfate $(MnSO_4)$ on the reduction of Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) by growth stimulation of Pediococcus acidilactici (P. acidilactici; lactic acid bacteria). Manganese ion (0.003 %) was found to stimulate the growth of P. acidilactici in the In Vitro system. When E. coli and S. aureus were grown in a mixture with P. acidilactici, their numbers were reduced. This may be the result of a reduction of pH in the medium as a result of better growth of P. acidilactici due to stimulation by the Mn ion. The in vivo experiment was conducted to determine the effects of $MnSO_4$ in diets on controlling harmful microorganisms in fecal samples of pigs. There were no significant differences for the microbial numbers (i.e., total microorganisms, E. coli, lactic acid bacteria and S. aureus) in feces of pigs fed $MnSO_4$ compared to feces of pigs fed the control diet through 7 days. However, on day 7 of experiment, the pH of feces in pigs fed $MnSO_4$ (0.1%) decreased faster than pigs fed the control diet.

        To estimate the fatigue life of flange-shaft assembly, fatigue test for flange material and bending fatigue test for flange-shaft assembly were conducted. Also, finite element analysis for flange-shaft assembly was conducted. Then, we have changed the obtained P-N curve to S-N curve using the finite element analysis results which were stress values at the location of fracture. The S-N curve of flange material itself was almost consistent with that of flange-shaft assembly, so it seems that the fatigue life of flange-shaft assembly could be estimated by using S-N curve for flange material and the stress at the location of fracture calculated by finite element methods.
