        扶正抗癌湯의 抗腫瘍效果에 關한 實驗的 硏究

        腫瘍으로 인한 死亡率은 多樣한 名種 治療에도 不拘하고 循環器疾患 다음으로 2위를 차지하고 있다. 이에 著者는 韓醫學과 西醫學의 結合治療를 通해서 抗腫瘍藥物의 效果를 增進시키고 副作用을 줄이는 藥物의 開發을 爲하여 健脾 益氣 祛痰 補腎 杭癌效能을 갖고 잇는 扶正杭癌湯抽出液을 使用하여 Colony 形成抑制實驗, Sulforhoda-mine B(SRB) assay를 觀察하였고, 扶正抗癌湯抽出液과 抗癌化學療法劑인 Mitomycin C(MMC)를 Ehrlich carcinoma의 solid tumor와 ascites tumor에 各各 單獨 및 倂用 投與하여 抗腫瘍效果 및 Iysosomal enzymes의 活性에 대한 效果를 觀察하였다. 먼저 扶正抗癌湯抽出液을 Caki-1 cell, Hep 3B 및 A549에 투여한 후 增殖抑制作用을 colony 形成抑制實驗과 SRB assay를 통하여 觀察한 결과 濃度依存的으로 腫瘍細胞의 成長을 抑制하였다. Ascites form of Ehrlich carcinoma에 대한 抗腫瘍效果 實驗에서 扶正抗癌湯抽出液과 MMC를 병용투여한 결과, 扶正抗癌湯抽出液에 의하여 MMC의 抗腫瘍效果가 增加하였다. Solid form of Ehrlich carcinoma에 대한 抗腫瘍效果 實驗에서 扶正抗癌湯抽出液과 MMC를 병용 투여하였을 경우, MMC를 단독 투여하였을 경우보다 腫瘍의 크기가 현저하게 減少하는 결과를 보였다. Ehrlich ascites carcinoma cell에 扶正抗癌湯抽出液과 MMC를 병용 투여하였을 경우, MMC를 단독 투여하였을 경우보다 lysosomal enzymes의 活性이 强하게 나타났다. 本 實驗을 통하여 扶正抗癌湯抽出液은 MMC의 抗腫瘍效果를 증가시키는 效果를 보였다. 또한 扶正抗癌湯抽出液은 마우스에서 solid form of Ehrlich ascites carcinoma의 크기를 감소시키는 效果를 나타냈으며, 생존기간 實驗에서는 扶正抗癌湯抽出液과 MMC를 병용투여시 ascites tumor에 MMC를 단독으로 투여하였을 때와 비교하여 유의하게 생존기간을 증가시키는 결과를 보였다. 이상의 결과를 종합하여 볼 때 扶正抗癌湯抽出液은 用量에 依存的으로 直接的인 抗腫瘍效果가 있으며, 抗癌療法劑인 MMC의 效果를 亢進시키는 것으로 보아 間接的인 抗腫瘍效果도 있을 것으로 思科되나 正確한 效果를 究明하기 위해서는 보다 正確한 抗腫瘍機轉에 대한 硏究와 藥物學的 硏究 및 臨床的 硏究가 追加되어야 할 것이다. In order to investigate the effects of Bujeonghangamtang Extract(扶正抗癌湯抽出液) on antitumor effects after human cell lines(A549, Hep3B, Caki-1, Ehrlich) transplantation into the peritoneal cavity or right groin in mice induced by RPMI 1640 and DMEM etc, the extracts of its herbal medicines were orally administered for 10 or 12 days. Experimental studies were performed for measurement of antitumor effect of Mitomycin C(MMC) and lysosomal enzyme's activities using colony forming efficiency, SRB assay which were regarded as a valuable method for the measurement of antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows: 1. The change of colony-forming efficiency and SRB assay of Caki-1 cells, Hep3B and A549 Cells after exposure to the extract of Bujeonghangamtang extract depressed the growth of tumor cells by concentration of Bujeonghangamtang. 2. Antitumor activity of the ethanol extract of Bujeonghangamtang extract and MMC on ascites form of Ehrlich carcinoma in mice is slightly improved. Especially the mean of survival times in the group of 200㎎/㎏ and MMC 0.1㎎/㎏ is improved over 30.9%. 3. When Bujeonghangamtang extract and MMC are administered together, the weight of tumor is more decreased than MMC alone. 4. The lysosomal enzyme's activities of the Bujeonghangamtang extract and MMC are more significantly improved than MMC alone. According to the above result, it could be suggested that Bujeonghangamtang extract has indirect antitumor effect by the increase of MMC uptake.

      • 제주재래마 특이 표지인자 개발

        본 연구는 제주재래마의 유전적 특성파악과 이를 통한 타품종과의 유전적 특이성을 가지는 DNA 표지인자를 개발하고자 PCR-RAPD기법을 이용하여 제주재래마와 더러브렛, 가고시마의 3품종에서의 다형 band에 대한 특성파악을 실시한 결과이다. 각 품종별로 10두의 혈액을 혼합한 후 분석하였고 200개의 random primer들 중 PCR증폭양상의 재현성을 기준으로 70개의 primer를 선별하였다. 또한 선별된 primer들중 다형성을 9개의 primer를 이용하여 개체별 DNA분석을 실시하였으며 9개의 prkmer에 의해 나타난 15개의 band는 제주재래마 특이 band, 4개는 더러브렛종에 그리고 4개는 가지고시마 재래마에 품종특이 RAPD band가 나타났다. 따라서 RAPD기법에 의해 품종간의 차이를 보이는 표지인자의 개발이 가능할 뿐 아니라 품종간의 특성차이를 보이는 유전인자 개발과 특성파악이 용이한 것으로 평가되었다. This study was carried out using PCR-RAPD technique to estimate genetic characteristics of Cheju Horse (Jorangmal) and to develop the DNA marker for bread identification in horse. Each three horse breeds was represented by pooled DNA from 10 individuals and tested for 200 different random primers for their reproducible PCR pattern. From them 70 primer were selected and 9 primers were used to detect the difference between breeds at the individual level. Fifteen highly polymorphic bands were observed between breeds using 9 primers . Additionally, the 15 most polymorphic bands showed great difference between breeds in their appearing frequencies, implicating their usefulness in the breed discrimination and in the characterization of genetic structure. Therefore, the polymorphism of RAPD could be used in characterizing genomic difference between breeds and some genetic mechanism that give rise to breed disctimination in horse.

      • 국내에서 유행하는 HIV의 전파 경로에 따른 Subtype 분포

        Background : Previous data have been reported that subtype B is prevalent in South Korea, but neither the extent nor the proportion of subtypes could be evaluated. This study was designed to analyze the distribution of HIV-1 subtypes, temporal instructions and transmission dynamics between epidemiological groups. Methods : 1,280 Koreans had been diagnosed as HIV seropositive during the period 1985 to 2000. Among them, 134 individuals were selected for this molecular epidemiological study. 134 DNAs were isolated from uncultured or cultured peripheral blood mononuclear cells. V3-V5 (0.7 kb) fragment of HIV-1 env gene was amplified by nested polymerase chain reaction and was sequenced. Results : HIV-1 isolates from thirty-seven homosexuals were all subtype B (100%). On the other hand, 66 isolates from 94 heterosexuals were subtype B (70%) and 28 were non B subtypes (30% : 13 A, 4 C, 2 D, 8 E , 1 G). Only subtype B strains were isolated from 73 males who were infected with HIV inside Korea while 16 B and 20 non B subtype strains were isolated from 36 males who were HIV infected outside of Korea. However, B and non B strains were isolated half and half from females who were infected inside Korea except one. Conclusion : The HIV-1 subtype B strains are prevalent in Korea from the early HIV infection until present in both homo and heterosexuals. Non B strains have been transmitted from men who were infected outside Korea to their spouses and casual partners. So, we need further study to monitor subtype B and non B HIV transmission in epidemiological groups of Korea, (Korean J Infect Dis 33:311∼318, 2001)

        EFFECT OF THE METHANOL EXTRACT OF KOREAN TRADITIONAL MEDICINE HERB OLDENLANDIAE DIFFUSAE HERBA ON PROLIFERATION AND DIFFERTATION OF CANCER CELL

        백화사설초는 여러 가지 암세포의 성장을 억제할 수 있는 항암효과가 뛰어난 약재로, 근래에 많이 암환자에 활용되어지고 있으며, 실제 실험적으로도 그 효과가 입증되어지고 있다. 본 연구에서는 백화사설초가 사람 암세포의 증식과 분화에 어떠한 효과를 미치는지를 알아보기 위하여 monocytic leukemic cell line인 U937을 백화사설초 추출물만을 처리하고, 또한 백화사설초(10 ㎍/ml)의 농도에서 활성화된 단핵세포(ODH-MNC-CM)를 사용하였다. 그 결과 백화사설초 단독으로 사용할 경우 암세포의 분화를 유도하는 작용은 가지고 있지 않았지만 백화사설초가 Leukemic U937 세포에 대한 증식억제와 분화를 유도하는 IFN-γ, TNF-α와 같은 cytokine의 생산을 야기할 수 있다는 것을 알았으며, 백화사설초 추출물의 항종양효과와 암세포분화효과는 이러한 cytokines의 생성과 관련된 것으로 생각된다. Oldenlandiae diffusae Herba is herb medicine with antitumor activity capable of suppressing the growth of some cancer cell lines. In the present study, we have isolated a methanol extraction of Oldenlandiae diffusae Herba(ODH) and investigated its effect on the proliferation and differentiation of human cancer cell line using an in vitro culture system. Our results showed that the conditioned medium from ODH(10 ㎍/ml)-stimulated blood mononuclear cells (ODH-MNC-CM) treatment induced about 50% of the cells differentiating into mature monocytes/macrophages of CD 11b, CD 14, and CD68. Yet, the differentiated U937 cells also had functions of phagocytosis and superoxide production. However, ODH alone or normal MNC-CM had no such effects. The levels of interferon (IFN)-γ, tumor necrosis factor (TNF)-α, and interleukin (IL)-1 were very low in normal MNC-CM, and they were greatly increased in MNC-CM prepared with ODH stimulation. Antibody neutralization studies further revealed that the tumoricidal and differentiating effects of ODH-MNC-CM were mainly derived from the elevated cytokines, especially IFN-γ and TNF-α. These two cytokines acted synergistically on inhibiting cell growth and inducing differentiation of the target U937 cells.

        린 6시그마 분석을 통한 도면 주기 품질 향상 방안 연구

        Purpose: The purpose of this study was to find useful solutions by analyzing causes and results about defects on mechanical drawing notes and provide an automated tool with solutions to mechanical engineers. Methods: The collected data for defects on mechanical drawing notes were from ongoing development and mass production projects. Various measurement methods were used based on the Lean 6 Sigma analysis such as Process analysis, C&E diagram and some statistical analysis. Results: The results of this study are as follows; The results of the Lean 6 Sigma analysis, the validity of the selected indicators for improving drawing notes quality was verified through the verification of cause variables. The strategy established to improve the mechanical drawing notes was reflected as an automated program, and the defects were within a manageable range and achieved target Sigma level. Conclusion: Through the application of the “Mechanical drawing notes automation tool”, it is expected to resolve the “Voice of Customer, VOC” and “Voice of Business, VOB”

        ECO2 운영프로파일 보정을 통한 에너지 사용량 비교에 관한 연구

        The building energy efficiency rating system has been in operation in Korea, and the ECO2 is used for such a purpose. The ECO2 is based on a monthly calculation method using predefined room operating conditions. Such conditions were set in a statistical manner over existing cases, but individual deviations between measured and calculated results may be attributed to those predefined data. In this work, the measured data of energy consumption are compared to the initial ECO2 results, and the room operation conditions newly observed during field surveys are took into account in the ECO2. We analyze the effects of different operation profiles on ECO2 results. Results show that corrections in operating conditions can reduce the error by 4 to 8% depending on test buildings.

      • A study on the effects of BTB HVDC for fault current reduction applied metropolitan area in KEPCO Grid

        The South Korean Power System is characterized by a large percentage of its loads, approximately 43%, being concentrated in the Seoul area, in the northwestern part of the country(metropolitan area), while the largest percentage of the generation is located in the southern part of the country. As such, most of the power typically flows from the southern areas of the grid to the northwestern area. This means long distance power transfer which has the potential to result in voltage instability problems. Therefore, when that area has transmission fault, huge fault current occurs. This problems have been solved by deploying BTB(Back To Back) HVDC. BTB HVDC can divide into two or three systems and control power flow. If BTB is installed at metropolitan area, there are many advantages. Electric power can flow between systems by BTB HVDC. And BTB HVDC can prevent from blackout of the wide area by blocking the spread of the faults. This report shows the effects of BTB HVDC for fault current reduction applied metropolitan area in KEPCO Grid.

        EU의 개정통상정책과 대응방안

        2010년 EU는 장기비전인 Europe 2020전략 달성을 위한 대외통상부문 실행전략으로 개정통상정책을 채택하였다. 앞서 시행된 2006 Global Europe 통상정책 성과평가를 바탕으로 수정 보완해 작성된 개정통상정책은 서비스교역, 해외직접투자, 공공조달, 규정장벽, 지적재산권 등을 통상이슈로 적극 제기해 EU기업의 글로벌 시장 확대를 지원하며, WTO DDA타결을 위한 노력과 함께 현재 협상중인 FTA 타결에 적극 나서는 계획을 담고 있다. 외국의 보호무역조치를 감시하기 위한 민관 합동의 모니터링 시스템을 운영하고, 통상장벽규정(TBR)등 무역방어수단을 적극 활용하는 내용도 포함하고 있다. 이러한 EU의 개정통상정책에 대응 방안으로는 첫째, 국제통상이슈의 협력을 통해 한-EU간 ‘전략적 동반자 관계’ 구축기회로 삼아야 한다. 한-EU FTA를 통해 한국은 EU에서 제기하고자하는 공격적 통상이슈에 대한 협상을 대부분 마무리했기 때문에 양측간 통상분쟁의 가능성은 낮으며 오히려 제 3의 시장을 개방시키기 위한 협력가능성이 크다는 점을 적극 활용해야 한다. 둘째, EU의 적극적 WTO DDA 협상타결 노력을 위해 협력하는 것이 필요하다. 셋째, EU의 공격적 통상정책으로 인해 한국과의 통상분쟁 가능성이 있는 분야를 점검할 필요가 있다. 특히 EU는 한-EU FTA의 실행 여부를 면밀히 모니터링하기로 하는 등 협정의 이행에 관심을 많이 둘 예정이다. 넷째, EU와 주요 통상국-특히 일본 및 중국-과의 관계를 관심있게 관찰해야 한다. 마지막으로 국제통상분야의 규정제정자인 EU통상정책을 모니터링 할 수 있는 네트워크를 확대 구축해야 할 것이다. In 2010, the trade policy of EU was revised, which was for the implementation of the foreign trade policy of the strategy “Europe 2020”. The revised trade policy was modified on the basis of the performance assessment of the Global Europe 2006 trade policy. In adopting the new policy, the EU will actively raise trade issues such as service trade, foreign direct investment, public procurement, and intellectual properties for supporting European corporations in the global market. Furthermore, the EU is planning to conclude the FTAs currently being negotiated as well as the WTO DDA as early as possible, run a public-private joint monitoring system to keep a watchful eye on non-tariff barriers in trading partners, and take full advantage of various trade defense measures including Trade Barriers Regulations (TBR). As the countermeasures to the revised trade policy of the EU, the following 5 points are of importance: ① Through the cooperation with the EU in the field of international trade issues, Korea should find more chances for strategic partnership relations between the EU and Korea. By the Korea-EU FTA, Korea has already completed negotiations for sensitive trade issues, which the EU could possibly raise. Therefore the possibility of bilateral trade dispute is very low, and the situation in reality gives Korea more bilateral cooperation possibilities for the openness of the third markets. ② Korea should take up a positive attitude for signing the WTO DDA of EU. ③ Korea should check the areas that could cause a trade dispute under the new trade policy of the EU. The EU is expected to pay special attention to the implementation of the Korea-EU FTA, closely monitoring in particular the fulfillment of the agreement. ④ Korea is to pay attention to the relationship between EU and its main trading partners - in particular Japan and China. ⑤ Korea should expand its network system to monitor the trade policy of the EU, which will have a significant influence on various regulations of international trade.
