A multiplex PCR was developed for the simultaneous detection and differentiation of Mycoplasma (M.) hyopneumoniae and M. hyorhinis in clinical samples. Improved sensitivity is advantage of this technique over the previously reported multiplex assay. It was capable of detecting as little as 125 fg genomic DNA from M. hyopneumoniae and 62.5 fg genomic DNA from M. hyorhinis. Application of this multiplex PCR method to field isolates showed that M. hyopneumoniae and M. hyorhinis were present in 29% (107 of 370) of lung specimens and no mycoplasmas were detected in 56% (208 of 370) of the slaughtered pigs' lungs. At the farm level, M. hyopneumoniae and M. hyorhinis were detected in 34 of 36 (94.4%) randomly selected farms. We conclude that this assay would prove itself a value tool for monitoring these mycoplasmal infections and both M. hyopneumoniae and M. hyorhinis have been widely spread in swine herds of Korea.

Salmonella enterica Gallinarum (SG) causes fowl typhoid (FT), a septicemic disease in avian species. We constructed deletion mutants lacking the stress sigma factor RpoS, the nitric oxide (NO)-detoxifying flavohemoglobin Hmp, and the SsrA/SsrB regulator to confirm the functions of these factors in SG. All gene products were fully functional in wild-type (WT) SG whereas mutants harboring single mutations or a combination of rpoS, hmp, and ssrAB mutations showed hypersusceptibility to H2O2, loss of NO metabolism, and absence of Salmonella pathogenicity island (SPI)-2 expression, respectively. A triple-deletion mutant, SGΔ3 (SGΔrpoSΔhmpΔssrAB), was evaluated for attenuated virulence and protection efficacy in two-week-old Lohmann layer chickens. The SGΔ3 mutant did not cause any mortality after inoculation with either 1 × 106 or 1 × 108 colony-forming units (CFUs) of bacteria. Significantly lower numbers of salmonellae were recovered from the liver and spleen of chickens inoculated with the SGΔ3 mutant compared to chickens inoculated with WT SG. Vaccination with the SGΔ3 mutant conferred complete protection against challenge with virulent SG on the chickens comparable to the group vaccinated with a conventional vaccine strain, SG9R. Overall, these results indicate that SGΔ3 could be a promising candidate for a live Salmonella vaccine against FT.

The antimicrobial susceptibility of the 41 Bordetella bronchiseptica isolates was tested by using the Kirby-Bauer agar disk diffusion method. The B. bronchiseptica isolates were found to be sensitive to amoxicillin/clavulanic acid (100%), gentamicin (100%), neomycin (100%) and amikacin (97.6%), whereas they were resistant to streptomycin (100%), trimethoprim/sulfamethoxazole (100%), penicillin (100%) and ampicillin (97.6%). All the B. bronchiseptica isolates resisted to at least 4 antimicrobial agents and totally 8 different combinations of multiple antibiotic resistance patterns were noted. All of the B. bronchiseptica isolates, except one, were simultaneously resistant to streptomycin, trimethoprim/sulfamethoxazole, penicillin and ampicillin. The observed antibiotic resistance is not plasmid mediated as plasmids were absent from all the B. bronchiseptica isolates.

<P><I>Brucella abortus</I> readily multiplies in professional or nonprofessional phagocytes <I>in vitro</I> and is highly virulent in mice. Isogenic mutants of <I>B. abortus</I> biovar 1 strain IVKB9007 lacking the ATP/GDP-binding protein motif A (P-loop) (named <I>looP</I>; designated here the IVKB9007 <I>looP</I>::Tn<I>5</I> mutant) and the ATP-binding/permease protein (<I>cydC</I>; designated here the IVKB9007 <I>cydC</I>::Tn<I>5</I> mutant) were identified and characterized by transposon mutagenesis using the mini-Tn<I>5</I>Km2 transposon. Both mutants were found to be virtually incapable of intracellular replication in both murine macrophages (RAW264.7) and the HeLa cell line, and their virulence was significantly impaired in BALB/c mice. Respective complementation of the IVKB9007 <I>looP</I>::Tn<I>5</I> and IVKB9007 <I>cydC</I>::Tn<I>5</I> mutants restored their ability to survive <I>in vitro</I> and <I>in vivo</I> to a level comparable with that of the wild type. These findings indicate that the <I>cydC</I> and <I>looP</I> genes play important roles in the virulence of <I>B. abortus</I>. In addition, intraperitoneal immunization of mice with a dose of the live IVKB9007 <I>looP</I>::Tn<I>5</I> and IVKB9007 <I>cydC</I>::Tn<I>5</I> mutants provided a high degree of protection against challenge with pathogenic <I>B. abortus</I> strain 544. Both mutants should be evaluated further as a live attenuated vaccine against bovine brucellosis for their ability to stimulate a protective immune response.</P>
Proteomic Analysis of Outer Membrane Proteins in Salmonella enterica Enteritidis

Salmonella enterica serovar Enteritidis is the predominant agent causing salmonellosis in chickens and other domestic animals. In an attempt to identify antigenic S. Enteritidis outer membrane proteins (OMPs) that may be useful for subunit vaccine development, we established a proteomic map and database of antigenic S. Enteritidis OMPs. In total, 351 and 301 spots respectively from S. Enteritidis strain 270 and strain 350 were detected by twodimensional gel electrophoresis. Fifty-one antigen-reactive spots were detected by antisera on two-dimensional immunoblots and identified as 12 specific proteins by matrix-assisted laser desorption ionization time-of-flight mass spectrometry. OmpA and DNA starvation/ stationary phase protection protein (Dps) were the most abundant proteins among the identified OMPs, comprising 22 and 12 protein species, respectively. Interestingly, we found that the Dps of S. Enteritidis is also antigenic. OmpW was also verified to have high antigenicity. These results show that OmpA, Dps, and possibly OmpW are antigenic proteins. This study provides new insights into our understanding of the immunogenic characteristicsof S. Enteritidis OMPs.
A micro-level perspective to fostering IDEAs in the classroom and beyond

Inclusion, Diversity, Equity, & Access (IDEA) spurs creativity, encourages ideas from people who have been historically ignored, and promotes an invigorating learning environment. Despite such benefits, we do precious little when it comes to practicing the same in our classrooms. Regrettably, our infrastructure and education system does not always provide enough opportunities for the younger generation to appreciate, understand and experience the significance of an inclusive society, preventing our students an opportunity to apply what they learn in the classroom for the betterment of marginalized and minoritized communities. Against such a backdrop, the author focuses on how faculty can enhance the classroom-level student-experience through a model that helps students’ understanding and appreciation of IDEA. The author believes that this research model not only plugs a significant gap in our attitude towards IDEA practice and implementation but will also spur educators and practitioners to consider adopting similar measures while designing their respective curricula.
Characteristics of Surface Urban Heat Island (SUHI) over the Gangetic Plain of Bihar, India

The rapid urbanisation impacts on environment, climate, agriculture, water resources trigger several problems to human beings. The present study is carried out to estimate intensity and trend of Urban Heat Island (UHI) as Surface UHI (SUHI) over towns/cities of the Gangetic plain of the state of Bihar, India, in which urban areas show relatively greater Land Surface Temperature (LST) than its rural surroundings especially during night times. The LST data (2001-14) of Moderate Resolution Imaging Spectroradiometer (MODIS) is used for five major towns/cities of Bihar namely, Bhagalpur, Gaya, Patna, Purnea and Muzzaffarpur. Each city is classified into Urban, Suburban and Rural zones as per land cover of the area. During winter months (January, February, November and December), UHI is more intense over all towns/cities. Mann-Kendall Test is applied on Surface Urban Heat Island Intensity (SUHII) in which MK-Test Statistic (S) shows a significant increasing trend. This trend would alarm a risk to increase in air pollution, heat related biohazards, energy demand in the region. This study shows the need of urban greening and proper town planning over the considered areas to mitigate the changes.

Background Biomass yield is an important trait for wheat breeding programs. Enhancing the yield of the aerial components of wheat cultivars will be an integral part of future wheat improvement. Aluminum (Al) toxicity is one of the main factors limiting wheat growth and production in acid soils, which occur on up to 50% of the arable lands of the world especially in tropical and subtropical regions. Objective Our objective was to identify quantitative trait loci (QTL) of plant growth characteristics and yield in wheat. Methods A recombinant inbred line (RIL) population consisting of 167 lines, derived from a cross between SeriM82 and Babax were evaluated under two Al treatments (+ Al, 800 µM of Al; −Al, 0 µM of Al) in the field based on an alpha lattice design with two replications for two consecutive crop seasons. Results A total of 40 QTLs including nine putative and 31 suggestive QTLs were found for all traits using the composite interval mapping (CIM) method. By mixed model-based composite interval mapping (MCIM) method, 42 additive QTLs and nine pairs of epistatic effects were detected for studied traits, of which 20 additive and six pairs of epistatic QTLs showed significant QTL × environment interactions. Most of the detected QTLs across environments were stable, and the highest number of stable QTLs was related to A genome. Co-localization of QTL was found on linkage groups (LGs) 2B, 4B, 6A-a, and 7A (CIM method) and 2A-d, and 6A-a (MCIM method). Conclusion These results have implications for selection strategies in biomass yield and for increasing the yield of the aerial part of wheat following further evaluations in various genetic backgrounds and environments.
