        제조합 대장균에서 과발현된 Citrobacter freundii KCTC2006 유래의 β-Tyrosinase를 이용한 3,4-Dihydroxyphenyl-L-alanine의 생산

        재조합 대장균에서 대량발현 시킨 Citrobacter freundii KCTC 2006 유래이 효소 β-tyrosinase를 이용하여 pyrocatechol, sodium pyruvate, ammonium acetate로부터 3,4-dihydroxy phenyl-L-alanine을 생산하기 위한 연구를 수행하였다. 이 효소반응에 적합한 온도 및 pH 조건은 각각 18℃와 8.5로 결정되었고, 반응액 중의 ammonium acetate와 sodium pyruvate의 농도는 각각 300 mM, 50 mM 이상으로 조절하는 것이 적합하였다. Pyrocatechol의 경우는 20 mM에서 가장 높은 반응성을 나타냈으나, 기질을 반복적으로 첨가하며 장시간 동안 효소반응을 수행하는 경우에는 pyrocatechol의 고갈을 피하기 위하여, 20 mM에서 50 mM 사이로 조절하였다. 한편, 반응액 중에 ethanol을 10% 첨가한 경우에는 반응속도가약 20% 증가하였다. 이상과 같은 효소반응특성에 기초하여 조제한 기질용액에 β-tyrosinase를 1 unit/㎖ 농도로 가하고, pyrocatechol과 pyruvate가 고갈되지 않도록 간헐적으로 첨가하면서 효소반응을 수행한 결과, 24시간 만에 85.2%의 수율로 31.6g/ℓ의 3,4-dihydroxyphenyl-L-alanine를 생산할 수 있었다. By using the β-tyrosinase of Citrobacter freundii KCTC2006, which was cloned and overexpressed in Escherichia coli, 3,4-dihydroxy phenyl-L-alanine (L-DOPA) was synthesized efficiently from pyrocatechol, sodium pyruvate, and ammonium acetate. Optimal temperature and pH for the reaction were determined to be about 18℃ and 8.5, respectively. The effects of substrate concentrations were also examined at different concentrations of ammonium acetate, sodium pyruvate, and pyrocatechol. Ammonium acetate and sodium pyruvate increased the reaction rate until the concentrations reached to 300 mM and 50 mM, respectively. Although pyrocatechol showed the optimal concentration at 20 mM, it was controlled between 20 mM and 50 mM to avoid the depletion of substrate during the enzymatic synthesis. Based on above results, a reaction medium for the production of L-DOPA was prepared and incubated with 1 unit/㎖ of β-tyrosinase. Pyrocatechol and sodium pyruvate was added to the reaction solution intermittently to avoid the substrate depletion during the enzymatic reaction. After 24 hour of reaction, 31.6 g/ℓ L-DOPA was accumulated in the reaction solution as soluble and precipitated ones and the conversion yield was about 85.2%.

        Erythropoietin Reduces Death and Neurodevelopmental Impairment in Neonatal Hypoxic-Ischemic Encephalopathy

        Purpose: Erythropoietin (EPO) is a promising neuroprotective drug. We investigated whether EPO has beneficial effects on neurodevelopmental outcomes in infants with hypoxic-ischemic encephalopathy (HIE). Methods: We retrospectively reviewed the data of 56 infants with HIE born at or after 35 weeks of gestation who were admitted to Inha University Hospital between 2012 and 2021. Patients were divided into two groups based on EPO use and compared. In the EPO group, patients were administered 1,000 U/kg of EPO on days 1, 2, 3, 5, and 7, starting within 24 hours after birth. The primary outcome was death or neurodevelopmental impairment (NDI) at the age of 12 months. Results: EPO was administered to 38 infants, and 18 did not receive EPO. Only 37.5% of patients with HIE (21/56) and 60% of patients with moderate-to-severe HIE (21/35) received therapeutic hypothermia. Among all patients with HIE, death or NDI (21.1 % vs. 50.0%; odds ratio [OR], 0.09; 95% confidence interval [CI], 0.01 to 0.78; P=0.029) and brain injury on imaging (42.1% vs. 83.3%; OR, 0.16; 95% CI, 0.03 to 0.92; P=0.040) were significantly lower in the EPO group than in the control group. Among patients with moderate-to-severe HIE, brain injury on imaging (54.2% vs. 90.9%; OR, 0.04; 95% CI, 0.002 to 0.700; P=0.027) was significantly lower in the EPO group than in the control group. Conclusion: EPO administration significantly reduced mortality and NDI in infants with HIE. EPO can be considered an adjunctive therapeutic agent for neonatal HIE.

      • Success rate of vaginal delivery after induction of labor in term nulliparous women with twin pregnancies

        Objective: Induction of labor (IOL) is the most common procedures in obstetrics, and twin pregnancy is reported to be the risk factor for failed induction. The patients need to be counselled before induction, but there were not enough data regarding success rate of IOL in twin, especially in nulliparous women. The purpose of this study was to examine the success rate of vaginal delivery after IOL in term nulliparous women with twin pregnancy. Methods: In this retrospective cohort study, 806 nulliparous women with twin pregnancy were delivered at term after IOL between 2005 and 2018. A successful induction was defined as vaginal delivery after IOL, and a failed induction was defined as cesarean delivery. Clinical characteristics were compared according to the success or failure of induction. Results: Among the study population, 78% (628/806) women were successfully delivered vaginally. Women with successful induction had lower maternal age, higher maternal height, earlier gestational age at delivery, and lower summated birthweight of both twin. Logistic regression analysis showed that maternal age, height and summated birthweight of both twin were significant associated factors with successful IOL. Conclusion: In term nulliparous women with twin pregnancy, IOL resulted in 78 % of success rate. This can be used in counselling before induction of labor in twin pregnant women.

        소켓 및 RMI 기반 자바 메시지 전달 시스템의 구현 및 성능평가

        본 논문은 자바 언어로 완성된 MPI(Message Passing Interface) 스펙인 MPJ(Message Passing in Java) 스펙을 준수하여 병렬 컴퓨팅 환경에서 메시지 통신 인터페이스를 제공하는 JMPI(Java Message Passing Interface) 라이브러리를 설계하고 구현하였다. 이 라이브러리는 간단하면서도 매우 편리한 GUI(Graphical User Interface) 도구를 제공하여, 사용자가 손쉽게 병렬 컴퓨팅 환경을 구성할 수 있다. 또한, 본 논문에서는 두 가지 전형적인 분산 시스템 통신 메커니즘인 소켓과 RMI(Remote Method Invocation)방식을 이용하여 두 가지 버전의 시스템을 구현하였고, 기존의 JPVM 시스템과의 성능을 벤치마크 애플리케이션들을 통하여 컴퓨터 대수의 증가에 따른 처리 속도를 비교해 보았다. 실험 결과로는 본 논문에서 제시한 JMPI 시스템이 JPVM시스템보다 다양한 측면에서 높은 성능을 발휘한다는 것과 컴퓨터의 가장 효율적인 처리 속도는 애플리케이션에 따라 컴퓨터의 수를 증가시킨다고 해서 일정하게 처리속도가 증가하는 것이 아니라 네트워크의 트래픽을 고려하여 컴퓨터의 수를 증가시켰을 때 얻을 수 있다는 것을 보여준다. 마지막으로 컴퓨터의 수가 증가함에 따라 RMI를 사용하여 메시지를 전달하는 것이 소켓에 부속된 객체 스트림을 사용하여 메시지를 전달하는 것보다 효과적이라는 것을 알 수 있다. This paper designs and implements a message passing library called JMPI (Java Message Passing Interface) which complies with MPJ (Message Passing in Java), the MPI standard Specification for Java language. This library provides some graphic user interface tools to enable parallel computing environments to be configured very simply by their administrators and JMPI applications to be executed very conveniently. Also in this paper, we implement two versions of systems using Socket and RPC which are both typical distributed system communication mechanisms and with three benchmark applications, compare performance of these systems with that of an existing system JPVM depending on the increasing number of the computers. Experimental results show that our systems outperform JPVM system in terms of various aspects and that the most efficient processing speedup can be obtained by increasing the number of the computers in consideration of network traffic through processing evaluation. Finally, we can see that, as the number of computers increases, using RMI to transmit a message is more effective than using object streams attached to sockets to transmit a message.

        JMPI 시스템의 통신 패턴기반 성능평가

        Although many researches on developing Java based high performance message passing systems are very actively being performed, their performance has not been evaluated systematically from various points of view. For this, our previous work developed a JMPI system which complies with MPJ(Message Passing in Java) and, in particular, is implemented in two versions based on two representative distributed system communication mechanisms, Socket and RMI. Therefore, this paper shows effectiveness and efficiency of this system by comparing and analyzing its performance of various communication patterns using systematic Ping Pong benchmarks suites. From experimental results of this paper, we can see that the performance of the Ping*-Ping pattern outperforms that of every other pattern and the RMI-based JMPI system is more efficient than the Socket-based one in various aspects.

        최근 컴퓨터의 처리속도와 네트워크의 속도가 빠르게 발전함에 따라, 통신망으로 연결된 이기종 컴퓨터들을 이용하는 병렬 컴퓨팅에 대한 많은 연구가 진행되고 있다. 본 논문에서는 자바 MPI스펙인 MPJ 표준에 따라 병렬 컴퓨팅 환경에서 메시지 통신 인터페이스를 제공하는 JMPI(Java Message Passing Interface) 라이브러리를 설계하고 구현하였다. 그리고 사용자 측면에서 좀 더 간단하게 병렬 컴퓨팅 환경을 조성하고 조성된 환경에서 JMPI 라이브러리로 구현된 프로그램을 편리하게 실행 시킬 수 있는 UI를 구현 하도록 하였다.

        건축물 CO<SUB>2</SUB> 평가의 효율성 향상을 위한 조달청 표준공사코드 기반 BIM 템플릿 구축에 관한 연구

        This study is aimed at utilizing LCA processor with BIM template, eliciting the problems of the existing CO<sub>2</sub> assessment by constructing the database for values of green buildings. With the rise of Building Information Modeling application and green building assessment, studies on assessment of CO<sub>2</sub> emissions from buildings using BIM are needed. Based on the research of construction materials, this study develops database by applying Korea Procurement Agency Item Code to BIM Template. In BIM program, while Procurement Agency Item Code of construction material information is established, application building components to its assesment is also performed. After constructing BIM materials based on database, input-output tables and Korea LCI database to standard item code of public procurement service are also used for assessment The results of this research make it possible to assess CO<sub>2</sub> emissions of materials with BIM Template and building data modeling implementation.

        사용자 편의적 자바 메시지 패싱 시스템의 설계 및 구현

        Recently, as speeds of computer processors and networks are rapidly increasing, a lot of researches are actively progressing to develop efficient and lightweight parallel computing platforms using heterogeneous and networked computers. According to this technical trend, this paper designs and implements a JMPI(Java Message Passing Interface) software library to follow the standard Message Passing Java interface model called MPJ. This library provides some graphic user interface tools to enable parallel computing environments to be configured very simply by their administrators and JMPI applications to be executed very conveniently. Especially, it is platform-independent because it is purely written in Java programing langage.
