        Identification of flutter derivatives of bridge decks using stochastic search technique

        A more applicable optimization model for extracting flutter derivatives of bridge decks is presented, which is suitable for time-varying weights for fitting errors and different lengths of vertical bending and torsional free vibration data. A stochastic search technique for searching the optimal solution of optimization problem is developed, which is more convenient in understanding and programming than the alternate iteration technique, and testified to be a valid and efficient method using two numerical examples. On the basis of the section model test of Sutong Bridge deck, the flutter derivatives are extracted by the stochastic search technique, and compared with the identification results using the modified least-square method. The Empirical Mode Decomposition method is employed to eliminate noise, trends and zero excursion of the collected free vibration data of vertical bending and torsional motion, by which the identification precision of flutter derivatives is improved.

      • Clinical Observation on Recombinant Human Endostatin Combined with Chemotherapy for Advanced Gastrointestinal Cancer

        Objective: To explore the clinical efficacy and toxic and side effects of recombinant human endostatin (rhendostatin/endostar) combined with chemotherapy in the treatment of advanced gastric cancer. Materials and Methods: A total of 70 patients with advanced gastrointestinal adenocarcioma confirmed by histopathology and/or cytological examination were divided into group A (37 patients) and group B (33 patients). Patients in group A were given intravenous drip of 15 mg endostar added into 500 mL normal saline, once every other day until the cessation of chemotherapy or patients' maximal tolerance to chemotherapy. Patients in group B received chemotherapy alone. Two groups selected the same chemotherapy regimens. FOLFIRI scheme: 90-min intravenous drip of $180mg/m^2$ irinotecan, intravenous drip of $200mg/m^2$ calcium folinate (CF) and $400mg/m^2$ 5-fluorouracil (5-Fu) on d1, and continuous intravenous pumping of 2 $400mg/m^2$ 5-Fu for 46 h. FOLFOX4 scheme: intravenous injection of $85mg/m^2$ oxaliplatin (L-OHP), $200mg/m^2$ calcium folinate (CF) and $400mg/m^2$ 5-FU on d1 for 2 h, and then continuous intravenous pumping of 2 $400mg/m^2$ 5-Fu for 46 h. XELOX scheme: oral administration of 1 $500mg/m^2$ xeloda (or tegafur 50~60 mg) in twice during d1~14 and intravenous drip of $135mg/m^2$ L-OHP on d1 for 2 h. The modified FOLFOX scheme: intravenous injection of $135mg/m^2$ L-OHP on d1 for 2 h, $200mg/m^2$ CF and 1.0 g tegafur during d1~5. Whereas, control Group B received chemotherapy regimens which were same as Group A, but no addition of endostar. Before chemotherapy, patients were given intravenous injection of 8 mg ondansetron, intramuscular injection of 10 mg metoclopramide and 20 mg diphenhydramine for prevention of vomiting, protection of liver and stomach as well as symptomatic supportive treatment. One cycle was 21 d, 4~6 cycles in total. The efficacy was evaluated every 2 cycles. Results: 32 patients in Group A could be evaluated, and the response rate (RR) and disease control rate (DCR) were 59.38% and 78.13%, respectively. 31 patients in Groups could be evaluated, and the RR and DCR were 32.26% and 54.84%, respectively. The differences between 2 groups were significant. The toxic effects include myelosuppression, gastrointestinal reaction, fatigue, cardiotoxicity and peripheral neurotoxicity. Conclusions: Preliminary observations show that endostar (once every other day) combined with chemotherapy is effective in the treatment of advanced gastrointestinal cancer, with low toxic effects, good tolerance, deserving further study.

        Cyclic AMP Responsive Element Binding Protein 3-like 4/AarF Domain Containing Kinase 5 Axis Facilitates Proliferation, Migration and Invasion of Lung Adenocarcinoma Cells by Modulating the TGFβ Pathway

        Cyclic AMP responsive element binding protein 3-like 4 (CREB3L4) has been reported as a transcription factor showing high expression in various cancers, whereas the mechanism of CREB3L4 in lung adenocarcinoma (LUAD) progression remains unclear yet. Expression levels of CREB3L4 and its downstream target gene AarF Domain Containing Kinase 5 (ADCK5) in LUAD tissues were analyzed by bioinformatics methods and were detected by real-time quantitative polymerase chain reaction at cellular level. The signaling pathways in which the downregulated ADCK5 enriched were analyzed via Gene Set Enrichment Analysis. The regulatory relationship between CREB3L4 and ADCK5 was identified by ChIP and dual luciferase reporter assay. CCK-8 and colony formation assays were utilized to evaluate LUAD cell proliferation. Transwell was utilized to measure migration and invasion of LUAD cells. Western blot was employed to check expression changes of CREB3L4, ADCK5, TGFβ pathway proteins and Epithelial- Mesenchymal Transition related proteins. Compared with paracancer tissues, CREB3L4 and ADCK5 were highly expressed in LUAD tissues, while silencing CREB3L4 could dramatically hinder invasion, proliferation and migration of LUAD cells. ADCK5 was the downstream target gene of CREB3L4, and CREB3L4 promoted the transcription of ADCK5. ADCK5 was markedly enriched in the TGFβ pathway, which stimulated the malignant development of LUAD cells by activating this pathway. In addition, the rescue assay verified that CREB3L4 regulated the TGFβ pathway by activating ADCK5, thereby accelerating LUAD cell malignant phenotypes. This study revealed the mechanism of CREB3L4/ADCK5 axis facilitating malignant phenotypes of LUAD cells, and bred new insights into the LUAD treatment.

        Preparation of Chitooligosaccharides from Chitosan using Crude Enzyme of Bacillus cereus D-11

        In order to enzymatically produce chitooligosaccharide using the crude enzyme preparation from Bacillus cereus D-11, we first studied the optimal reaction conditions. It was found that the optimal temperature for hydrolysis of chitosan was 55oC. The ratio of enzyme/substrate should not be lower than 0.13 U/mg in the reaction mixture. The enzyme activity was stable below 50oC. The products of enzymatic reaction were analyzed by both thin layer chromatography and high performance liquid chromatography. Under the appropriate condition, chitosan was hydrolyzed using the enzyme preparation. The resulting chitooligosaccharides were purified and separated by Dowex (H+) ion exchange chromatography. From 4 g soluble chitosan, 0.95 g (GlcN)2, 1.43 g (GlcN)3, and 1.18 g (GlcN)4 were recovered.

        Optimization of the Culture Condition for the Production of Chitosanase by Bacillus cereus D-11 and the Chitooligosaccharide Hydrolysis Pattern of Its Enzyme Preparation

        A chitosanase-producing bacterium, Bacillus cereus D-11, was isolated from an environmental soil sample. The optimal culture condition for chitosanase production by B. cereus D-11 was 3 days of cultivation at 30 o C in a medium composed of 0.5% inoculation concentration (1.4×10 8 CFU/mL), 0.7% colloidal chitosan, 1% yeast extract and 1% NaCl at initial pH 7.0. The highest activity achieved was 7.8 U/mL. The crude enzyme preparation from Bacillus cereus D-11 degraded the chitooligomers pentamer, hexamer, and heptamer into (GlcN)2-4, but not degraded the chitooligomers less than (GlcN)4. These results indicate that B. cereus D-11 chitosanase cleaves the oligomeric chains in the endo-splitting manner and the catalytic domain of D-11 chitosanase recognizes and needs at least 5 glucosamine units for hydrolytic catalysis of the glycosidic linkages.

        Simulation Studies on the Structural Stability and Properties of Beta Uranium

        The structural stability of beta uranium (-U) is investigated by using the projector augmentedwave (PAW) method based on the density function theory (DFT). Optimized lattice parameters arein agreement with other available data. Especially, the energy per atom and the phonon propertiesmanifest that the model described by using space group No. 102 can be stabilized vibrationallyand energetically. A reasonable agreement with the experimental data for the phonon entropycorresponding to the proper structure has been successfully achieved. Moreover, the thermodynamicfunctions are predicated at elevated temperatures.

      • Risk Factors for Cervical Cancer in Rural Areas of Wuhan China: a Matched Case-control Study

        Cervical cancer is a serious public health problem in developing countries. We investigated possible risk factors for cervical cancer in rural areas of Wuhan China using a matched case-control study with 33 women diagnosed with cervical cancer and 132 healthy women selected from the same area as matched controls. A questionnaire, which included questions about general demography conditions, environmental and genetic factors, the first sexual intercourse, first marriage age, age at first pregnancy, pregnancy first child's age, female personal health history, social psychological factors, dietary habits, smoking and alcohol status and other living habits was presented to all participants. At the same time, HPV infection of every participant was examined in laboratory testing. Results showed HPV infection (P<0.000, OR=23.4) and pregnancy first child's age (P<0.000, OR=13.1) to be risk factors for cervical cancer. Menopause (P=0.003, OR=0.073) was a protective factor against cervical cancer. However, there was no indication of associations of environmental (drinking water, insecticide, disinfectant) genetic (cancer family history), or life-style factors (smoking status, alcohol status, physical training, sleep quality), including dietary habits (intake of fruit and vegetable, meat, fried food, bean products and pickled food) or social psychological factors with cervical cancer. The results suggest that the risk of cervical cancer in Chinese rural women may be associated with HPV infection, menopause and the pregnancy first child's age.

        Promoted Growth of Maize by the Phosphate Solubilizing Bacteria Isolated from North-east China

        A strain of phosphate solubilizing bacterium was isolated from rhizosphere and identified as Burkholderia sp. by 16S-rRNA gene sequence analyses. The bacterium was found to release gluconic acid and the solubilization of hydroxyapatite in the liquid medium by a significant drop in pH to 3.7 from an initial pH 7.0. The soluble-P concentration continuously increased during the incubation periods and the total amount of soluble P released in culture filtrate was detected at 990 mg L<SUP>-1</SUP> after 10 days of inoculation. Most promoted maize growth was found in the standard NPK (240-120-120 kg ha<SUP>-1</SUP>) soil inoculation with Burkholderia sp. (Twenty milliliters/ plant, 106 CFU) and also in the absence of Burkholderia sp. inoculation, the soil amended with only 2/3 levels of P gave significant higher plant yield compared to 1/3 levels of P or without P supplementation.

        Radiation Hybrid Mapping and mRNA Expression of Chicken N- myc downregulated gene 4

        N-myc downregulated gene 4 (NDRG4) is a member of the N-myc downregulated gene family which belongs to the alpha/beta hydrolase superfamily. The protein encoded by this gene is a cytoplasmic protein that may be involved in the regulation of mitogenic signalling in vascular smooth muscle cells. To map NDRG4 gene in chicken chromosome, a 6,000 rads chicken- hamster radiation hybrid panel was used. Primers were designed according to the published human sequence for amplification of chicken NDRG4. We compared the corresponding human mRNA sequence with the predicted coding sequence of chicken NDRG4, and found that the assembled contig shared a high percentage of similarity with that of human gene. PCR of samples from ChickRH6 revealed the locations of NDRG4 to be linked to the maker PARD6G (5 cR away) with a LOD score 20.46. In addition, we detected the mRNA expression and distribution of chicken NDRG4 in various tissues by RT-PCR, and found that NDRG4 was highly expressed in chicken brain and heart, whereas lowly but detectable in thymus. The mRNA expression of this gene in chicken liver, spleen, lung and muscle was rarely detectable under present experimental conditions.
