        한국인에서 5,10-Methylenetetrahydrofolate Reductase 유전자 다형성 분석 및 임신의 결과에 미치는 영향

        목적: 본 연구는 한국인 임신 여성에서 MTHFR 유전자형 변이와 혈청내 호모시스테인 및 엽산의 농도와 이들이 임신에 미치는 영향을 알아보고자 하였다. 방법: 임신부 600명에게서 혈액을 채취하여 얻어진 DNA로 MTHFR C677T와 A1298C 유전자의 다형성을 PCR-RELP로 분석하였고 혈청내 호모시스테인은 HPLC 방법으로, 엽산은 RIA 방법으로 측정하였다. 결과: 혈청내 호모시스테인은 MTHFR C677T 유전자형이 C/C형이나 C/T인 군에 비해 T/T인 군에서 유의하게 높았으며 MTHFR A1298C 유전자형이 A/C형이나 C/C인 군에 비해 A/A인 군에서 역시 유의하게 높았다( p<0.05). 혈청 호모시스테인은 모든 MTHFR 유전자형에서 혈청 엽산농도와 역상관관계를 보였으나, 변이형 유전자형(T/T와 A/A유전자형)에서 더 큰 역 상관관계를 보였다. 15?mol/L 이상의 고호모시스테인혈증의 임신부에서 임신 주수와 출생체중이 36.1주, 3053.8 g으로 15 ?mol/L미만인 군의 38.3 주, 3215.3 g 보다 유의하게 낮았다( p<0.05). 결론: 혈청 호모시스테인의 농도는 MTHFR C677T와 A1298T 유전자의 다형성에 의해 영향을 받으며, MTHFR C677T와 A1298T 유전자의 다형성과 혈청 호모시스테인은 모두 임신주수와 신생아의 체중에 영향을 미친다. Objective: The purpose of this study was to analyze MTHFR polymorphism among the Korean population and to evaluate the relationship between serum levels of homocysteine and MTHFR polymorphism and also to investigate the effect on pregnancy outcomes. Methods: DNA was extracted from whole blood of 600 pregnant women. All samples were genotyped for the C677T and A1298C polymorphisms in MTHFR gene by PCR-RELP assay. Serum levels of homocysteine and folate were measured by high performance liquid chromatography for homocysteine and radioassay for folate. Pregnancy outcomes were estimated by gestational weeks and birth weights of newborns. Results: Serum homocysteine was higher in women with the T/T genotype than those with the C/T or C/C genotype of the MTHFR C677T polymorphism (p<0.05). And also serum homocysteine was higher in women with the A/A genotype than those with the A/C or C/C genotype of the MTHFR A1298C polymorphism (p<0.05). Serum homocysteine was negatively correlated with serum folate in all MTHFR genotypes, especially prominent in T/T genotype of MTHFR C677T polymorphism and A/A genotype of MTHFR A1298C polymorphism. Gestational age and the birth weight of infant from hyperhomocysteinemic mothers whose homocysteine levels higher than 15 μmol/L were 36.1 weeks, 3053.8 g, respectively, which were significant lower than those from normohomocysteinemic mothers (38.3 weeks, 3,215.3 g) (p<0.05). Conclusion: Serum homocysteine was influenced significantly by MTHFR C677T polymorphism and MTHFR A1298C polymorphism. MTHFR C677T and A1298C polymorphism and serum homocysteine levels affect pregnancy outcomes, although not mainly by serum folate level.

      • S-510 A case of peritoneal dialysis-associated peritonitis caused by Agromyces mediolanus

        Introduction:?Agromyces mediolanus (A.mediolanus) is an unusual pathogen in human infection. No case of peritoneal dialysis (PD)-associated peritonitis by A.mediolanus has been reported. This is the first case report of PD-associated peritonitis by A.mediolanus in the world. We describe a case of PD-associated peritonitis by A.mediolanus that was cured after catheter removal.?Case: A 59-year-old man who had been treated with CAPD was admitted to our hospital because of turbid peritoneal effluent accompanied by constant diffuse abdominal pain. The patient had an exit-site infection with purulent secretion. The laboratory findings showed PD peritonitis: the white blood cell (WBC) count of the peritoneal effluents was 1157/mm3 with a neutrophil predominance (91.3%). Cefazolin and ceftazidime were given intraperitoneally per day. The peritoneal WBC decreased, but the patient’s clinical condition didn’t improve on the 5th day after starting intraperitoneal antibiotics. Culture of the peritoneal dialysate revealed Agromyces species, which was susceptible to meropenem, imipenem, vancomycin, but resistant to ceftriaxone. Thus, we changed antibiotics to intraperitoneal vancomycin and meropenem on the 5th day. Despite the intraperitoneal antibiotics for 4 days, the patient’s abdominal pain persisted and the peritoneal WBC count became elevated. Therefore, we removed the PD catheter and the patient was switched to hemodialysis on the 9th day. Vancomycin and imipenem was continued for another 2 weeks and the patient’s clinical condition improved. The patient has been maintained on hemodialysis.?Conclusions: There is no previous report of peritonitis associated with A.mediolanus in a CAPD patient. In this case, we used intraperitoneal cefazolin and ceftazidime and changed to vancomycin and imipenem according to the result of peritoneal dialysate. However the patient improved after removal of catheter. Therefore, early removal of the PD catheter in A. mediolanus related peritonitis without prompt reaction to antibiotic therapy should be considered.

      • A Driving Method of Pixel Circuit Using a-IGZO TFT for Suppression of Threshold Voltage Shift in AMLED Displays

        <P>A driving method of pixel circuit using amorphous indium gallium zinc oxide (a-IGZO) thin-film transistor (TFT) is proposed to improve the image quality of active matrix light-emitting diode displays. The proposed pixel circuit employs a diode-connected structure to compensate for variation in threshold voltage (V-th) of the a-IGZO TFT. In addition, the proposed driving method adopts negative bias annealing to suppress the V-th shift. The annealing time is optimized based on the experimental observation of the minimum V-th shift. After a stress time of 30 000 s, the measurement results showthat the (Vth) shift is reducedby 29.6%, using an optimized annealing time of 5% of one frame time. In addition, the maximum deviation in the emission current using the proposed driving method wasmeasured to be less than 4.32% after a stress time of 30 000 s.</P>

        Background: Asiatic acid is a putative anticancer agent in several types of cancer cells. Researches of its biological role in negative regulation of cell growth have focused on the extent of induction of apoptosis in a cell-type-specific manner. Objectives: To identify the mechanisms underlying asiatic acid-induced downregulation of BCL2. Methods: MicroRNA microarray analysis was used to investigate the effect of asiatic acid treatment of A549 cells on miRNA expression profiles. We used quantitative RT-PCR to confirm the asiatic acid-induced increase in miR-1290 level with primers specific for the mature miR-1290 sequence. We also assessed the effect of asiatic acid on the levels of BCL2 in A549 cells using western blot analysis. Results: We identified an important regulator of asiatic acid-induced cell death, microRNA (miR)-1290, which sensitizes cells to Asiatic acid-induced cytotoxicity and negatively regulates BCL2 expression. Asiatic acid significantly upregulated miR-1290, and asiatic acid-induced cell death was shown to be dependent on miR-1290 activity. Molecular assays demonstrated that BCL2 mRNA is a direct target of miR-1290-mediated RNA interference. Conclusion: The findings of the present study suggest that miR-1290 suppresses cell viability and cell cycle progression. Our results provide insight into miR-1290-mediated cellular mechanisms in asiatic acid-treated A549 non-small cell lung carcinoma cells.

        Wear Characteristics of Al-Si-Mg- (Cu)/Al2O3 Composites Fabricated by Thermit Reaction

        A₂O₃ particles could be formed by a thermit reaction in an Al-12Si-4Mg-1.5Cu/Al₂O₃ composite due to the in-situ reaction between Al-12Si-4Mg-(1.5Cu) molten metal and SiO₂ particles in preform, which took place at 1173 K for 24 hours, resulting in the decomposition of SiO₂ particles and the formation of Al₂O₃particles simultaneously. The mechanically mixed layers (MMLs) consisting of a-Fe and Fe oxides existed on the subsurface layers beneath the worn surface in composites or mother alloys, which improved the wear resistance. The characteristics of wear resistance and hardening of an Al-12Si-4Mg-1.5Cu/Al₂O₃ composite are superior to those of the Al-12Si-4Mg/Al₂O₃ composite and Al-12Si-4Mg-1.5Cu alloy.

        Fluorescent chemosensor based-on the combination of julolidine and furan for selective detection of zinc ion

        A new sensor 1-[[(2-furanylmethyl)imino]methyl]-2-hydroxyjulolidine (1) based on the combination of julolidine and furan groups was designed and synthesized as a Zn<SUP>2+</SUP> selective fluorescent chemosensor. Upon treatment with zinc ions, the complexation of 1 with Zn<SUP>2+</SUP> exhibited a pronounced enhancement in the fluorescence emission in methanol, while many other ions such as Mn<SUP>2+</SUP>, Co<SUP>2+</SUP>, Ni<SUP>2+</SUP>, Cd<SUP>2+</SUP>, Hg<SUP>2+</SUP>, Na<SUP>+</SUP>, K<SUP>+</SUP>, Mg<SUP>2+</SUP>, Ca<SUP>2+</SUP>, Al<SUP>3+</SUP> and Ag<SUP>+</SUP>, had no influence. Notably, this chemosensor could distinguish clearly Zn<SUP>2+</SUP> from Cd<SUP>2+</SUP>. The 1:1 binding mode of the 1-Zn<SUP>2+</SUP> complex was drawn, based on UV-vis titration, fluorescence titration, Job plot and ESI-mass spectrometry analysis.

        Dendritic Localization and a cis-acting dendritic targeting element of Kv4.2 mRNA

        Kv4.2, a pore-forming α-subunit of voltage-gated A-type potassium channels, is expressed abundantly in the soma and dendrites of hippocampal neurons, and is responsible for somato-dendritic IA current. Recent studies have suggested that changes in the surface levels of Kv4.2 potassium channels might be relevant to synaptic plasticity. Although the function and expression of Kv4.2 protein have been extensively studied, the dendritic localization of Kv4.2 mRNA is not well described. In this study, Kv4.2 mRNAs were shown to be localized in the dendrites near postsynaptic regions. The dendritic transport of Kv4.2 mRNAs were mediated by microtubule- based movement. The 500 nucleotides of specific regions within the 3`-untranslated region of Kv4.2 mRNA were found to be necessary and sufficient for its dendritic localization. Collectively, these results suggest that the dendritic localization of Kv4.2 mRNAs might regulate the dendritic surface level of Kv4.2 channels and synaptic plasticity.         Background: Korean ginseng (Panax ginseng C. A. Meyer) is recognized for its medicinal properties. Because ginseng is cultivated for 4 – 6 years in one place, the growing environment can affect ginseng growth. Specifically, root growth can be altered by plant hormones, which in turn, affects crop yields. Among plant hormones, cytokinin is known to increase secondary growth by promoting cambium cell division. In addition, ethylene and propiconazole can regulate root development. In this study, the effects and interactions of cytokinin, ethylene, and propiconazole on four-year-old ginseng plants were analyzed. Methods and Results: Ginseng plants were treated with each hormone every two weeks. Cytokinin (6-benzylaminopurine, 500 μM) was applied to the soil, and 100 nM of propiconazole or 100 μM of ethylene was applied in combination with cytokinin. Root width and weight, and ginsenoside content of each plant were the highest when cytokinin and propiconazole were applied together. The highest number of buds was observed under cytokinin treatment. However, no significant differences were observed in shoot growth, except for stem width. Conclusions: The greatest effect on root development was observed when cytokinin and propiconazole were applied together, and ginsenoside content increased following hormone treatment.

        Changes and distributional pattern of microorganism in compost and fruiting body for button mushroom cultivation

        Agaricus bisporus grows on a substrate known as compost, which is a product of aerobic fermentation by various microorganisms. These organisms convert and degrade the straw and form lignin humus complex which is utilized later on by the population of organisms. Theses microflora play a key role in the process of composting and can be regarded as the active agents in the preparation of nutrient medium as many of them may ultimately contribute themselves to the nutrition of A. bisporus. The diversity of microflora according to growing farmhouse and fruiting body of Agaricus bisporus were investigated. The aerobic bacteria and Bacillus as longer of turning stage of compost pile were increased. And, thermophilic actinomycetes and fluorescent Pseudomonas sp. showed high density after the pasteurization stage. But Tricoderma sp. was decreased toward the end of turning stage of compost pile. Ten mushroom farms was selected to research of microflora of fruiting body of button mushroom. The microflora showed significant difference according to mushroom farms. The bacteria density was 0.4~41.6×105 cfu/ml and the fungus was 1.3~3.9×103 cfu/ml. But The microorganism density was not significant change for the storage periods. These isolates were classified into Chryseobacterium indologenes(6 strains), Pseudomonas agarici(5 strains), Sphingobacterium multivorum(2 strains), Flavobacterium anhuiense(2 strains), Microbacterium sp.(10 strains), Pseudomonas sp.(13 strains) on the basis of 16 rDNA analysis. The most dominants of these species were Chryseobacterium indologenes and Pseudomonas agarici.
