Background/Aims: Revaprazan (Revanex(R)) is a novel proton pump inhibitor (PPI) that has a somewhat different effect on proton pump compared with the other PPI`s, also (called as `acid pump antagonist`). We aimed to examine the false negative rate of (13)C-urea breath test (UBT) in the patients with Helicobacter pylori (H. pylori) associated peptic ulcer disease who were treated with revaprazan and evaluate the anti-urease activity of revaprazan. Methods: Total 55 patients were enrolled in this study. They received EGD examination between January 2009 and December 2009 and diagnosed histologically as H. pylori associated peptic ulcer disease. All patients took revaprazan only. Three patients were excluded because of underlying chronic disease and inappropriate breath sampling. The remaining 52 patients had UBT at 0, 2 and 4 weeks of revaprazan use. After 2 weeks of the cessation of revaprazan, they had the fourth UBT. Results: At 2 and 4 weeks, the false negative rates of UBT were 5.8% and 23.1%, respectively (p=0.05). After 2 weeks of the cessation, the cases of the false negative result were five. Four out of five patients had prolonged negative results on two or three successive tests, and baseline (13)C difference value did not predict the false negative results. Conclusions: False negative results of UBT were common and increased with prolonged use of acid pump antagonist. As PPI, it had also anti-urease activity and most patients (47/52, 90.4%) reverted to positive results by 2 weeks after the cessation of taking the medication. (Korean J Gastroenterol 2011;57:8-13)
항산화와 미백효과를 증진시킨 새로운 Sulforaphane 함유 혼합 조성의 개발

본 연구는 새로운 항산화 및 미백 기능성 화장품 성분을 개발하기 위해 기존의 항산화와 미백 기능성 성분으로 알려진 두 가지 성분을 혼합한 새로운 조성을 개발하고, 이를 이용해 항산화 및 미백효과를 알아보고자 하였다. 두 가지 성분은 항산화와 멜라닌 생성억제 기능이 알려진 sulforaphane과 멜라닌 생성 억제 효과가 알려진 PF-3758309이다. 각 성분 및 혼합 조성에 대한 세포독성을 조사하여 세포독성이 나타나지 않는 농도를 결정하였다. 단독 처리 시 sulforaphane는 10 μ M까지, PF-3758309는 100 nM까지 세포독성이 없다는 것을 확인하였다. 두 화합물 10 μ M sulforaphane과 1 nM PF-3758309를 병합 처리 시 세포독성은 없었다. LPS 자극에 의해 생성된 일산화질소는 sulforaphane과 PF-3758309의 병합 처리시 현저히 감소되었다. α-MSH에 의한 멜라닌 생성 역시 병합 처리에 의해 억제되었다. 화장품 소재로 사용 가능성을 확인하기 위해 혼합 조성 시료에 대한 인체적용시험을 실시하였다. 혼합조성 시료는 피부첩포에 의한 안전성 평가는 무자극으로 판정되었고, 피부밝기 증가효과와 피부 멜라닌 침착을 억제함을 확인하였다. 이러한 결과들로 혼합 조성물이 항산화와 미백 기능이 있는 새로운 기능성 화장품 조성으로 개발될 가능성을 확인할 수 있었다. In this study, we analyzed two components, sulforaphane and PF-3758309 to prove that the mixed composition of them has more effective antioxidant and whitening functions compared to each component. We analyzed the cellular toxicity of each component and also the mixed composition to find the safe concentration level for cell viability. From the single component treatment, we discovered that sulforaphane was safe up to 10 μM, and PF-3758309 up to 100 nM. Combination treatment of 10 μM sulforaphane and 1 nM PF-3758309 did not affect the cell viability. The LPS-stimulated NO generation was significantly reduced by the mixed composition of sulforaphane and PF-3758309. Melanogenesis by α-melanocyte stimulating hormone (α-MSH) was also inhibited by the mixed composition. In order to confirm the possibility as the cosmetic material, we carried out clinical studies for the mixed composition samples. Skin safety evaluation using patch test was judged to be unstimulated, skin whitening effect was increased, and melanin deposition was suppressed by treatment of mixed composition samples. These results provide us with the opportunity for applying it into the development of new functional cosmetics.

N/A From early spring 1986 to 1987, serum specimens from 1141 individuals with acute febrile episodes were collected from 9 local collaborators and tested for antibodies to R. tsutsugamushi and R. typhi by indirect immunofluorescent antibody test. 1) Of these, 353 sera (31%) showed positive reaction to R. tsutsugamushi and 115 sera (10%) showed positive reaction to R. typhi. 2) R. tsutsugamushi and R. typhi infections are seemed to occur throughout the country in Korea. The highest positive rate was shown in Chollanamdo. 3) Tsutsugamushi disease and Typhus group rickettsial disease were the most prevalent in October, and November. 4) 132 sera (12%) and 129 sera (11%) among 1141 individuals gave positive reaction to Leptospira and Hantaan virus respectively.
Aspergillus niger 이용 미생물 발효차의 발효 중 미생물학적, 물리적 및 관능적 품질특성

미생물 발효차 제조 시 녹차를 121oC에서 15분간 고압 멸균하여 A. niger를 접종하여 제조한 미생물 발효차와 단일균주를 처리하지 않고 자연 발효시킨 발효차의 색도변화량을 비교하였을 때 서로 비슷한 경향을 나타내었다. 관능검사에서 또한 단일 균주를 이용한 미생물 발효차의 관능적 특성이 단일 균주 처리하지 않은 미생물 발효차와 유의적인 차이를 나타내지 않았다. 이를 통해 본 연구에서는 단일균주를 이용한 미생물 발효차의 제조 가능성을 확인 할 수 있었다. 그러나 A. niger를 이용한 미생물 발효차는 제품화가 가능하나 독성 및 영양적 가치 등에 대한 연구가 더 필요할 것이라고 생각된다. The microbiological, physical, and sensory characteristics of the microbial-fermented tea with A. niger (MFTAN) were investigated during fermentation at 30℃ and 80% RH for 35 days. During the fermentation period, the L* value of the MFTAN was decreased, but a* and b* values increased. Microorganisms from naturally fermented tea were screened and isolated. An isolated microorganism was identified as A. niger by rRNA sequence of analysis. In the sensory evaluation, the result suggested that the sensory properties were only of minor differentiation between MFTAN and control. In conclusion, MFTAN manufactured with Korean green tea would be a good candidate of a microbial-fermented tea and the physicochemical changes of the teas showed supportive evidence, to this fact, during fermentation. The other microorganisms should be considered as starters to make microbial-fermented tea in future studies.
