        Crystallization and preliminary diffraction analysis of human TEAD1, a transcriptional enhancer factor that controls the Hippo signaling pathway

        Transcriptional enhancer activation domain (TEAD) proteins are transcription factors that promote the expression of genes involved in organogenesis, embryonic development, and tumorigenesis. TEAD functions downstream of the Hippo signaling pathway as one of the pivotal regulators that controls cell viability and proliferation, tissue growth, and organ size, by interacting with yes-associated protein (YAP) via its C-terminal YAP-binding domain (YBD). As YAP is a well-known oncoprotein and its interaction with TEAD has been shown to be critical for the function of YAP, TEAD proteins are emerging as a potential therapeutic target for cancer. In this study, the YBD of the TEAD1 protein was produced from an Escherichia coli expression system, purified using a Ni-NTA affinity chromatography, HiTrap Q anion exchange chromatography, and size exclusion chromatography, and then successfully crystallized. X-ray diffraction data were collected to the resolution of 1.70 Å. Preliminary diffraction analysis revealed that the TEAD1 YBD crystals belong to the space group P2 1 2 1 2 1 with unit cell parameters of a = 36.5 Å, b = 89.4 Å, c = 135.6 Å, and that two TEAD1 YBD molecules are contained in the asymmetric unit with a solvent content of 45.2%.

      • Impairment of Driving Ability and Neuropsychological Function in Patients with MHE Disease

        <P>Minimal hepatitis encephalopathy (MHE) is a liver cirrhosis complication that does not show symptoms of overt hepatitis encephalopathy (OHE) but can be detected using specific complementary neuropsychological and neurophysiological evaluations. Patients with MHE show abnormalities in cognitive functions, which can increase the risk of automobile accidents. The aim of this study was to compare the cognitive functioning of patients with MHE to patients with other liver disease and to compare their driving abilities, using a driving simulator and the Stim(2) software. Thirty-eight participants, who were not diagnosed with OHE but were diagnosed with chronic hepatitis and liver cirrhosis, took part in the study. The results indicate that patients with MHE show reduced level of cognitive functioning, which may affect their ability to drive safely. We cautiously predict that any future guidelines based on this knowledge may help reduce the incidence of traffic accidents caused by drivers with decreased with driving abilities.</P>

        Korean Version of the Stroke Rehabilitation Motivation Scale: Reliability and Validity Evaluation

        Objective To translate the Stroke Rehabilitation Motivation Scale (SRMS), developed to evaluate the motivation level of stroke patients during rehabilitation, into the Korean language and to verify the reliability and validity of the Korean version of SRMS (K-SRMS). Methods The K-SRMS was developed following a structured process that included translation, verification, compromise assessment, reverse translation, feedback, and final correction. K-SRMS reliability was evaluated by performing internal consistency and test–retest analyses. The reliability test was conducted in 50 stroke patients. Its validity was assessed by comparing the K-SRMS with the scale and performing exploratory factor analysis. The validity test was conducted in 102 stroke patients. Results The test–retest analysis showed good reliability, and the internal consistency of the K-SRMS was similar to that of the original version for all, except 4, items. Thus, these 4 items were excluded, and then the validity test was conducted. Pearson correlation analysis demonstrated that the K-SRMS score was significantly correlated with the BAS total score (Pearson r=0.207, p<0.05). In the exploratory factor analysis, K-SRMS items were categorized into 7 groups (factors), and factors 1 and 4 showed mutual concordance with K-SRMS subscales, including intrinsic motivation factors and amotivation, respectively. Conclusion The newly developed K-SRMS showed good reliability and validity. It could also be used as a tool to objectify the degree of motivation for rehabilitation among stroke patients in clinical care and research.

      • Coordination between aminofluorescein and long emission wavelength fluorescent dyes for fabrication of ratiometric fluorescent pH sensors

        Aminofluorescein (AF) is a fluorescent dye with high quantum yield and pH sensitivity so that AF is commonly used for pH detection. However, in the area of fluorescence-based sensors, measurements of fluorescence intensity at a single band edge are known to be problematic in practical applications. The use of ratiometric fluorescence method is a useful solution for correction of a variety of analyteindependent factors in fluorescent sensors. Therefore, AF and other referent fluorescent dyes (e.g. ruthenium (Ru), sulforhodamine 101 (S101)) with long emission wavelengths are doped into melamine formaldehyde resin particles to prevent leaking dyes. The dyes are incorporated with the mixture of sol-gel (GA) and polyurethane hydrogel (PU) for fabrication of pH-sensing membranes. These membranes show high sensitivity, selectivity, reversibility and stability in a wide pH detection range (pH5-pH11).

        누에번데기 및 누에애벌레 밀리타리스동충하초(Cordyceps militaris) 열수추출물의 투여가 고형암이 유발된 마우스의 종양성장 억제 및 면역기능에 미치는 영향

        본 연구에서는 누에번데기 또는 누에애벌레를 기주로 하여 인공 재배된 밀리타리스동충하초의 고형암 성장억제 및 면역조절 기능을 평가하였다. 이를 위해 sarcoma-180 세포주를 겨드랑이에 피하 이식받은 ICR 마우스를 대상으로 밀리타리스동충하초 또는 유효성분으로 알려진 cordycepin을 10일간 복강투여한 후 고형암 성장저지능, 면역관련 장기의 무게변화, 비장 내 면역세포수의 변화, 그리고 비장세포에 의한 interleukin-2(IL-2) 생성능을 측정하여 다음과 같은 결론을 얻었다. 겨드랑이에 고형암을 이식받은 마우스에게 50㎎/㎏(CMP50) 또는 100 ㎎/㎏(CMP100)의 누에번데기 밀리타리스동충하초 추출물을 10일간 투여한 결과 생리식염수를 투여한 대조군에 비해 3주 후 고형암 성장이 47.3% 및 57.6% 유의하게 억제되었다(p<0.05). 50 ㎎/㎏(CML50) 및 100 ㎎/㎏(CML100)의 누에애벌레 밀리타리스동충하초를 투여받은 경우 35.5% 및 37.1%, 그리고 1 ㎎/㎏ 및 2㎎/㎏의 cordycepin을 투여받은 군의 경우 26.1%와 29.8%의 고형암 성장 억제효과가 관찰되었다(p<0.05). 고형암을 이식받은 후 누에번데기 또는 누에애벌레 밀리타리스동충 하초를 투여받은 마우스는 대조군에 비해 흉선무게가 38~44% 유의하게 증가하였고(p<0.05), 비장내 CD4+ 및 CD8+T-세포수, NK-세포수(63~110% 증가), 그리고 비장세포에 의한 IL-2 생성능(33~51% 증가)이 모두 유의하게 증가하였다(p<0.05). 이상의 결과를 종합해 볼 때, 밀리타리스동충 하초 열수추출물은 고형암의 성장을 억제하는 효과가 있으며, 이와 같은 밀리타리스동충하초 추출물의 항암활성은 면역기능 증강효과와 연관이 있을 것으로 생각된다. 아울러 밀리타리스동충하초의 고형암 성장억제 및 면역조절 기능은 cordycepin 단일물질에 비해 더 우수하였다. Hot water-extracts prepared from Cordyceps militaris of silkworm pupa (CMP) or Cordyceps militaris of silkworm larva (CML) were tested for tumor growth inhibitory and immunomodulatory activities in ICR mice bearing sarcoma-180 cells solid tumor, and compared with those of the known compound, cordycepin, found in Cordyceps militaris. Mice were subcutaneously injected with sarcoma-180 cells, and i.p. injected with either saline (Control), 50 ㎎/㎏ or 100 ㎎/㎏ of CMP (CMP50 or CMP100, respectively), or CML (CML50 or CML100, respectively), or 1 or 2 ㎎/㎏ of cordycepin (C1 or C2, respectively) for 10 days. Mice injected with CMP50 or CMP100 showed a 47.3% or 57.6% inhibition in the solid tumor growth (p<0.05), while those injected with CML50 or CML100 exhibited a 35.5% or 37.1% reduction (p<0.05) in solid tumor size compared to the value for control mice treated with saline. Animals injected with corcycepin showed a 26~30% inhibition in the solid tumor growth (p<0.05). Mice bearing solid tumor and injected with CMP or CML showed a significantly increased thymus weight (38~44% increase), lymphocyte percentages of CD4+ T-cell, CD8+ T-cell, and NK-cell (63~110% increase) in the spleen, and interleukin-2 excretion (33~51% increase) by the isolated splenocytes compared to those in control mice (p<0.05). These results indicate that the anti-tumor activity of hot water extracts of Cordyceps militaris, raised on both silkworm pupa and silkworm larva, appears to be associated with their immunomodulatory activity, and these activities found in Cordyceps militaris are superior to those for the single compound, cordycepin.

      • Electron transfer and electron carrier of enzymes significantly improved the reduction of indigo to leuco indigo

        In this study, the reduction of indigo to leuco indigo by glucose was enhanced by using enzyme glucose oxidase (GOD) and laccase (LA). Generally, indigo accepted electrons from the glucose oxidation reaction or from intermediates of glucose degradation and after that converted to leuco-form. In case of GOD or LA involved the indigo reduction, the redox centers of GOD and LA acted as an electron carrier from the reducing glucose or direct electron transfer to indigo molecules. Therefore, the turnover rate of electron flow between glucose and indigo through enzymes was increased and led to increase of leuco-indigo production. That was about 5 times higher in cases of using enzymes as compared without using enzymes
