Background This study was conducted to investigate the impact of genetic variants in immune checkpoint genes on the treatment outcome of patients with small cell lung cancer (SCLC). Methods Two hundred sixty-one patients who treated with platinum doublet were enrolled. Ninety-six polymorphisms in 33 immune checkpoint related genes were selected and their association with chemotherapy response and overall survival were analyzed. Results Among polymorphisms studied, CD155 rs1058402G>A and CD226 rs763361C>T were significantly associated with the treatment outcome of SCLC. The rs1058402G>A had worse chemotherapy response and worse overall survival (under a dominant model, odds ratio [OR] = 0.52, 95% confidence interval [CI] = 0.27 - 0.99, P = 0.05; hazard ratio [HR] = 1.55, 95% CI = 1.12 - 2.14, P = 0.01, respectively). The rs763361C>T had better chemotherapy response and better overall survival (under a dominant model, OR = 2.03, 95% CI = 1.10 - 3.75, P = 0.02; HR = 0.69, 95% CI = 0.51 - 0.94, P = 0.02, respectively). When the rs1058402GA/AA and rs763361CC genotype were combined, the chemotherapy response and overall survival were significantly decreased as the number of bad genotypes increased (OR = 0.52, 95% CI = 0.33 - 0.81, Ptrend = 0.004; HR = 1.48, 95% CI = 1.19 - 1.84, Ptrend = 4 × 10-4, respectively). CD155 rs1058402G>A and CD226 rs763361C>T are both non-synonymous mutations and change the protein sequence CD155 A67T and CD226 G307S, respectively. 3D structural model showed that CD155 A67T created new hydrogen bond and structural change on CD155. These changes resulted in extending the distance and losing the hydrogen bonds between CD155 and CD226, thus would weaken the binding activity of CD155/ CD226. Conclusions CD155 rs1058402G>A (A67T) and CD226 rs763361C>T (G307S) may be useful for predicting the clinical outcome of SCLC after chemotherapy.
Amperometric thiol sensor based on Prussian blue-modified glassy carbon electrode

This paper describes the performance of an amperometric sensor for thiol detection. The sensor was designed based on a Prussian blue(PB) glassy carbon (GC) electrode. Prussian blue was chemically deposited onto the glassy carbon electrode by a dropletting method.Thiol compounds were detected at the PB-modied GC electrode by electrooxidation. A PB-modied glassy carbon electrode wasapplied to detect thiol at an applied potential of +0.25 V versus the Ag/AgCl electrode. This sensor showed an excellent electrochemicalresponse for thiol compounds belowl mol level with high sensitivity and selectivity and short response time. In the case of aminoetha-nethiol, the sensor showed a wide linearity range with RSDs <4% for the whole analyses, which reected the highly reproducible sensorperformance. The optimal conditions were investigated. By using the optimized conditions, the detection limit was found to 0.4l M foraminoethanethiol (based onS/N = 3).
COMP-angiopoietin-1 enhances skeletal muscle blood flow and insulin sensitivity in mice.

<P>To test whether chronic enhanced blood flow alters insulin-stimulated glucose uptake, we measured skeletal muscle glucose uptake in chow-fed and high-fat-fed mice injected with adenovirus containing modified angiopoietin-1, COMP-Ang1, via euglycemic-hyperinsulinemic clamp. Blood flow rates and platelet endothelial cell adhesion molecule-1 positive endothelial cells in the hindlimb skeletal muscle were elevated in COMP-Ang1 compared with control LacZ. Whole body glucose uptake and whole body glycogen/lipid synthesis were elevated in COMP-Ang1 compared with LacZ in chow diet. High-fat diet significantly reduced whole body glucose uptake and whole body glycolysis in LacZ mice, whereas high-fat-fed COMP-Ang1 showed a level of whole body glucose uptake that was comparable with chow-fed LacZ and showed increased glucose uptake compared with high-fat-fed LacZ. Glucose uptake and glycolysis in gastrocnemius muscle of chow-fed COMP-Ang1 were increased compared with chow-fed LacZ. High-fat diet-induced whole body insulin resistance in the LacZ was mostly due to approximately 40% decrease in insulin-stimulated glucose uptake in skeletal muscle. In contrast, COMP-Ang1 prevented diet-induced skeletal muscle insulin resistance compared with high-fat-fed LacZ. Akt phosphorylation in skeletal muscle was increased in COMP-Ang1 compared with LacZ in both chow-fed and high-fat-fed groups. These results suggest that increased blood flow by COMP-Ang1 increases insulin-stimulated glucose uptake and prevents high-fat diet-induced insulin resistance in skeletal muscle.</P>

Objectives:This study was conducted to investigate the prevalence of victims of bullying and the demographic characteristics of victims, and their related psychopathology, in a Korean nationwide sample of youths in middle and high school over a one month period. Methods:During the autumn of 2009, students in the 7th to 12th grades at 23 secondary schools participated in a nationwide, cross-sectional study. The study subjects completed the Adolescent Mental Health and Problem Behavior Screening Questionnaire-II (AMPQ-II) and Symptom Checklist-90-Revision (SCL-90-R). Based on the data acquired, descriptive statistics, correlation coefficients and multiple logistic regression analysis were performed. Results:Among the 3364 participants, 2272 (67.54%) completed the questionnaire. The prevalence of victimization was 28.9%. Male gender was positively associated with victimization, and grade level was negatively related to victimization. The AMPQ-II bullying score (Factor 4) was significantly (p<.001) and positively correlated to the AMPQ-II student total score (r=0.50), Worry and thought (Factor 1 ; r=0.38), Mood and suicide (Factor 2 ; r=0.31), Academic and Internet-related problems (Factor 3 ; r=0.24), Rule violations (Factor 5 ; r=0.23), and AMPQ-II teacher total score (r=0.11). Somatization (r=0.23), Obsessive-compulsive behavior (r=0.24), Interpersonal sensitivity (r=0.30), Depression (r=0.33), Anxiety (r=0.26), Hostility (r=0.30), Phobic anxiety (r=0.22), Paranoid ideation (r=0.36), and Psychoticism (r=0.31) results from the SCL-90-R were also found to be positively related to the AMPQ-II bullying score, and remained significant after adjusting for age and gender. A total of 26% of the victims reported suicidal ideations as compared to 9% of non-victims over the month prior to the evaluation (χ2=119.595, df=1, p<.001). The multiple logistic regression analysis indicated that the AMPQ-II bullying score significantly increased the risk of suicidal ideation [Exp(b)=1.55, df=1, p<.001] after adjusting for age and gender. Conclusion:School bullying was highly prevalent among Korean middle and high school students. This study provided strong evidence that suicidal ideation and psychopathology were serious problems among the victims of bullying.

Background Bromodomain and extraterminal domain (BET) proteins are epigenetic readers that regulate gene expression. We investigated whether regulatory polymorphisms in BET genes are associated with clinical outcomes of non-small cell lung cancer (NSCLC) Methods The associations between 77 regulatory polymorphisms in BET family genes and clinical outcomes were analyzed in the 773 surgically resected NSCLC (349 patients in the discovery and 424 patients in the validation cohorts). Results Of the 77 rSNPs investigated, 11 rSNPs are associated with OS or DFS with P value less than 0.05 in the discovery cohort. Among 11 rSNPs, only one rSNP (rs2506711C>T) is replicated with same direction as the discovery cohort, in the validation cohort. BRD3 rs2506711C>T is significantly associated with OS in the discovery, validation and combined analysis (under a recessive model, HR = 1.64, 95% CI = 1.06 - 2.54, P = 0.03; HR = 1.71, 95% CI = 1.01 - 2.92, P = 0.05; HR = 1.71, 95% CI = 1.22 - 2.40, P = 0.002, respectively). In the functional study, knockdown of BRD3 with BRD3 specific siRNA showed significant G2/M arrest in HCC1833 cells compare to control. However, the ability of HCC1833 cell migration or invasion, and apoptosis rate did not differ between the knockdown of BRD3 and the control group. Conclusions We found that BRD3 rs2506711C>T was associated with poor OS survival in NSCLC
