        Bacillus licheniformis KFB-C14가 생산하는 내열성 Chitinase의 정체 및 특성

        Bacillus licheniformis KFB-C14가 생산하는 내열성 chitinase를 30~70% ammonium sulfate fractionation, DEAE-Toyopearl 650M, Butyl-Toyopearl 650M, TSK-Gel Toyopearl HW-55F에 의해 정제도 66배, 수율 21%로 전기영동적으로 균일하게 정제하였다. 정제 단백질은 gel permeation chromatography에 의해 86,000±2,000의 분자량을 나타내었으며, SDS 전기영동에 의해 밝혀진 본 효소의 subunit 구조는 monomer였다. 효소 단백질의 안정성을 검토한 결과 80℃에서 30분 열처리에 의해 56%, 37℃에서 20분간 40% ethanol과 ethyl acetate, 단백질 변성제 등의 처리시에도 50% 이상의 잔존활성을 나타냄으로써 공업적으로 유용성이 높은 안정한 단백질로 판명되었다. 효소반응의 최적 pH와 온도는 pH 6.0과 60℃이었고 Mn^2+ 이온에 의해 효소 활성이 저해되었으나 EDTA, N-ethylmaleimide, p-chloromercuribenzoate 등에 의한 활성감소는 관찰되지 않음으로써 금속효소 또는 thiol계 효소에 속하지 않음을 알 수 있었다. 본 효소는 colloidal chitin, 시판용 chitin에는 반응성이 높았으나 exo형 chitinase의 대표적인 기질인 p-nitrophenyl-2-aectamido-2deoxy-β-glucopyranoside, NN'-diacetylchitobiose에는 전혀 반응성을 보이지 않는 전형적인 endo형의 chitinase였다. 본 효소는 colloidal chitin으로부터 주로 (GlcNAc)_2를, 반응시간 경과에 따라 (GlcNAc)_1과 (GlcNAc)_3을 생성하는 반응성을 보였다. Chitinase (EC 3.2.1.14) from culture fluid of Bacillus licheniformis KFB-C14 was purified 66-folds to homogenity in overall yield of 21% by ammonium sulfate fractionation, DEAE-Toyopearl, Butyl-Toyopearl and TSK-Gel HW-55F column chromatography. The enzyme protein had a molecular weight of about 86,000 and was composed of one subunit. The enzyme was significantly stable not only at high temperature but also on treatment with organic solvents and protein denaturants such as SDS, urea and guanidine·HCl. The optimum temperature and pH for reaction was 60℃ and 6.0,respectively. The enzyme activity was inhibited by only Mn^2+ ion, but not inhibited by EDTA, N-ethylmaleimide and pCMB. The enzyme had high activity with colloidal chitin (V_max: 421) and commercial chitin (V_max :480), but not with typical substrates of exo type chitinase. The thermostable chitinase had an useful reactivity for producing functional chitooligosaccharide, showing the production of (GlcNAc)_1, (GlcNAc)_3, and (GlcNAc)_2 as major product.

        내열성 Chitinase 생산균주의 분리 및 효소생산 특성

        자연계 고온환경으로부터 내열성 chitinase 우수하고 반응산물로 N-acetyl-D-glucosamine 이량체(GlcNAc)_2를 생산하는 균주를 분리 선별하고 Bacillus licheniformis KFB-C14로 동정하였다. 선별균주의 효소생산 특성은 탄소원으로서 효소기질인 colloidal chitin이 첨가될 때만이 생합성이 유도되었으며 유도제의 첨가시기에 의해 효소생산이 크게 영향을 받았다. 각종 무기, 유기태 질소원 중 yeast extract가 활성과 비활성을 각각 약 2배 증가시켰으며 높은 친화도를 나타내었다. 균의 최대생육과 효소의 최대생산온도는 55℃이었다. 본 균주의 내열성 chitinase 생산에 미치는 최적배양조건은 1.2% colloidal chitin, 0.15% K_2HPO_4, 0.05% KH_2PO_4 0.01% MgSO_4·7H_2O, 0.1% yeast extract, pH 6.5의 배지를 55℃, 150rpm에서 40시간 회전진탕배양 하였을 때로 3.89 units/ml의 효소활성과 7.4 units/mg의 비활성을 나타내었다. A strain capable of producing thermostable chitinase suitable for chitooligosaccharide production was isolated from high temperature environment and identified as Bacillus licheniformis. The chitinase from Bacillus licheniformis KFB-C14 was only induced by addition of colloidal chitin into the basal medium as carbon source, showing the decrease of the chitinase production by supplemental addition of other carbon sources into the medium containing 1.0% colloidal chitin. Among organic and inorganic nitrogen sources, yeast extract was the most effective for the increase of total activity and specific activity, and had high affinity for the enzyme production. The optimum temperature of cell growth and thermostable chitinase production was 55℃. The optimum culture medium was composed of 1.2% colloidal chitin, 0.15% K_2HPO_4, 0.05% KH_2PO_4 0.01% MgSO_4·7H_2O, 0.1% yeast extract (pH 6.5). Bacillus licheniformis KFB-C14 produced the thermostable chitinase of 3.89 units per culture fluid and 7.4 units per mg protein under rotary shaking at 150 rpm for 40 hr.

      • 韓·英 俗談 대한 比較 分析 : 健康에 關聯된 俗談을 中心으로 With the Proverbs concerned with Health

        Proverbs are a living lesson, and exercise a great influence over our living by expressing the times and the society ironically and humorously. Proverbs contain one's view of life, and they often reflect the national characteristics, history, customs, belief, institution, living philosophy etc. And proverbs are possession of people in general, they are also songs and poems, tradition and thought expressed by folk wisdom. As we renew our appreciation of proverbs which are a rudder of people in general, make efficient use of such proverbs in our daily life, we can make our life more prosperous and vigorous. In this study I have tried to make a sampling inspection on many Korean proverbs and English proverbs which are often used in our daily life concerning health within the limits of them, and have compared, analyzed the Korean proverbs with English proverbs each other. Through this study, I have tried to awake the porsons to the universal thought and living philosophy, and especially have tried to make a contribution to their health care in a roundabout way. According to the contents directed in l~3, “the Scope of This Study”, the result of comparing and analizing them is as follows : (1) In the Health and Meal ; that good meal is the first essential in human life accounts for 53 percent in Korean proverbs, and 29 percent in English proverbs. It shows that Korean proverbs are given much more weight than English proverbs in good meal. Apart from the current situation of Korea, it brings about that the Korean people have lived their lives in poorer support of food than the Western people. (2) In Health and Disease ; the condition of health or disease and transfer from an original site of disease to another part of the body accounts for 57 perccent in Korean proverbs, and only 15 percent in English proverbs, the former is given much more weight than the latter. It brings about that the Korean people have neglected their health care or have hardly taken a measure to catch a disease. (3) In Health and Old Age ; that the old age act as a negative side in their health accounts for 83 percent in Korean proverbs, and 50 percent in English proverbs, the one is given much more weight than the other. It brings about that the Korean people have not had confidence in their health to reach an advanced age. (4) In Health and Death; that the death is the close showing a negative opinion in one's life accounts for 83 percent in Korean proverbs while nothing in English proverbs. It brings about that the Korean people have a negative view on death strictly. (5) In Health and Disabled person; what takes up a negative opinion toward the disabled persons accounts for 71 percent in Korean proverbs while nothing in English proverbs. It brings about that the Korean people have had much negative view toward the disabled persons strictly. (6) In Health and Drinking; what takes up a negative opinion on drinking accounts for 85 percent in English proverbs while nothing in Korean proverbs. It brings about that the Western people have much negative view to drink. (7) In Health and prevention of a Disease ; in order to control of one's health, what first of all to prevent a disease is most important accounts for 60 percent in English proverbs while few in Korean proverbs. (8) In Health as a General Rule ; to keep one's health, what we have health in mind and body usually a precondition accounts for 82 percent in English proverbs while few in Korean proverbs. (9) In Others ; in this item they show the condition of health or a disease, and a remedy. There are 67 percent concerning them in English proverbs while few in Korean proverbs. In conclusion through this study, to keep one's health we must pay attention to follows ; ① We should have better meal habits. ② We should take good care of our health normally, and must take a measure to meet in the contraction of a disease. ③ In attaining an advanced age they usually have not had confidence in their health in Korea. It is a conventional notion, and we must make away with such an existing opinion, the older we are, the more we should take good care of our health. ④ Death means the close of life, which has been our traditional living philosophy, and death has been transient and desperate in Korea. We must make away with such a living philosophy and should try to have the view of life which should bring forth happiness and hope in the next world. ⑤ They have had a negative opinion toward disabled persons in Korea. We should make away with a. conventional view, and we should not only have moral support but have warm heart and act kindly toward them. ⑥ We should abstrain from alcoholic beverages and must be temperate in drinking. ⑦ Prevention is better than cure. ⑧ We should always keep sound mind, and live a well-regulated life. ⑨ We need to take good care of our health ordinarily.

      • 최근 5년간 제주도에 도래한 월동 수조류 조사

        A study on the wintering birds on Cheju Island was conducted from October 1993 to February 1998. The total number of birds observed in this survey was 37,302 of 73 species, -23,066 birds of 66 species at Hado-ri fishfarm and 14,236 birds of 48 species at sungsan-po fishfarm. The most dorminant species was Anas penelope 26.59%(9,920 individuals), followed by Anas strepera 21.36%(7,996 individuals), and then Anas poecilorhyncha 19.56%(7,287 individuals). The number of species and individuals of wintering birds in each year were recorded: 5.068 birds of 32 species in 1993, 5,068 birds of 36 species in 1994, 8,349 birds of 34 species in 1995, 9,013 birds of 44 species, 9,236 birds of 49 species in 1997. The number of species and individuals of birds became increasingly at Hado-ri fishfarm, but decreasingly at Sungsan-po fishfarm. Species diversity was founded within the range of 0.639∼1.796, and among two areas quotient of similarity was 0.702. The rare birds of 11 species were recorded in this survey.

      • 중학교 과학과 수업 개선을 위한 학급수업과정 분석

        The purpose of this study is to find ways for the improvement of science instruction by describing and analyzing the schooling process in the class-room of the middle school through the participant observation. Four differ-ent middle schools and four different science teachers were selected. And intensive participant observations were made about total 18 hours' school-ing process concerned to the instruction of ' Photosynthesis' and 'Digestion' which are two units in the science textbook of the middle school. The documentary records of the results of field research on schooling the classroom were made. On the basis of these data analytical diagrams of each hour's schooling process were made and each teacher's teaching type, instrucion process type and teaching method about the same teaching contents were comparatively analyzed. Through this analysis, the following conclusions could be obtained. 1. Teaching type was divided largely into two different kinds, such as laboratory lessons in the laboratory and lecture lessons in the classroom. Laboratory lessons were conspicuously grouping lessons, and the lessons in the classroom were lecture lessons practiced by the lead of the teacher. 2. Instrucion process model stood out as an 'Inquiring Model' in the the-oritical model in case of laboratory grouping lessons, that is, in which 'inquiry learning process' was dominant, and stood out as 'Concept Model' explained by the teacher or 'Discovery Model' by suggesting teaching materials in case of lecture lessons. 3. Teaching method, even in case of the same teaching contents, showed various styles according to the different teachers. such as structur-izing lesson, providing instructional cues, questioning and reacting to stu-dents 'response, explanation and writing on the blackboard, student con-trol in class. But in case of the' same teacher the same styles were found in suggesting teaching aims, teaching arrangements, turning over in teach-ing. explanation and writing on the blackboard, of every class hour. 4. In order to make a qualitative improvement for the science instruc-tion, the inquiring instruction process, that is the greatest aim of science education, should necessarily be practiced in each class, and through this teaching process the present limits of science instruction should be over come. Therefore, the exploring attitude of each teachers own to renew the instruction stratege, Comparing the existing instruction process model, and teaching methods of individual teacher's, is thought to be nessessary. For this, through the teacher's training program or the teacher's retrain-ing course, the various ways to improve the science instruction quality should be provided by using ethnographic data on schooling as a tool for the indirect experience about other's schooling process.

      • 제주도 초지대 조류의 군집에 관한 연구

        An investigation into a bird community on grassland in Cheju Island was performed in 4 location : north, south, east and west which are selected as ecotone within a radius of 2km of Mt. Halla. The survey was done twice a month from July 1988 to June 1989 at 4 areas. 1. The total number and species birds observed during survey period were 2,758 and 55 respectively. And the dominant species were Hypsipetes amauroatis Parus major, Passer montarus in order. 2. 7 species were observed continuously during the all period of the survey, 22 species were observed for only a month. There was no species which was observed during the 4 month and 11 month periods of investigation 3. The regression line between the number of individuals and species showed 5% significance and its the correlativity was very high. 4. From the result of log. + ax. = b equation of MOTOMURA, the community structure of January was most simple and the order month was complex. 5. The diversity was high throught the three months of July, March, June and was low in August and January.

      • 제주도의 주요 습지에 도래하는 도요·물떼새류의 현황

        본 연구는 제주도 주요 습지에 도래하는 도요ㆍ물떼새류의 지소적인 연구의 일환으로, 1998년 3월부터 2000년 2월까지 2년간 매월 2회씩, 1) 동부지역(하도리 양어장, 종달리 해안 및 성산포 양어장)과, 2)서부지역(용수리 저수지, 금등리 해안 및 대정읍 일과리 해안)으로 나누어 조사하였다. 조사기간 중 2개 지역에서 관찰된 도요ㆍ물떼새류는 총 45종 5,117개체였으며, 지역별로는 동부지역에서 40종 4,910개체, 서부지역에서 38종 927개체로 나타났다. 계절별로는 봄철 2,821개체, 가을철 1,229개체로 봄철에 도래하는 개체수가 가을철보다 많았다. 우점종은 제1차 조사기간인 봄철에는 민물도요 Calidris alpina 2,024개체(88.5%), 흰물떼새 Charadrius alexandrinus 84개체(3.7%), 흰목물떼새 Charadrius placidus 11개체(2.4%)의 순으로 나타났으며, 가을철에도 민물도요 384개체(82.9%), 흰물떼새 20개체(4.3%), 흰목물떼새 11개체(2.4%)의 순으로 나타났다. 제2차 조사기간에는 봄철에 좀도요 Calidria ruficollis 116개체(21.8%), 민물도요 67개체(12.6%), 청다리도요 Tringa nebularia 58개체(10.9%)의 순으로, 가을철에는 민물도요 191개체(24.9%), 노랑발도요 T. brevipes 156개체(20.4%), 삑삑도요 T. ochropus 104개체(13.6%)의 순으로 나타났다. 동부지역은 서부지역에 비하여 우점도는 높았으나, 종다양도와 균등도는 낮게 나타났다. 동부지역과 서부지역간의 유사도는 0.85였다. 본 연구를 통해 물꿩 Hydrophasianus chirurgus, 검은머리물떼새 Haematopus ostralegus, 바늘꼬리도요 Gallinago stenura, 흰꼬리좀도요 Calidrius temminckii 등 4종의 제주도 미기록종과, 검은머리물떼새, 흰목물떼새, 넓적부리도요 Eurynorhynchus pygmeus, 알락꼬리마도요 Numenius madagascariensis 등 3종의 환경부 지정 보호조류가 관찰되었다. This study was part of a continuous research project on the status of migrating shorebirds on major wetlands in Cheju Island. This study was conducted twice every month for two years, from March 1998 to February 2000. The investigated location are mainly two areas. The first census areas were Hado-ri fishfarm and the coast of Jongdal-ri in Kujaw-up and Sungsan-po fishfarm in Sungsan-up, all located in the eastern part of Cheju Island. The second census areas were Yongsu-ri reservoir and the coast of Kumdung-ri in Hangyoung-myon and the coast of Ilgwa-ri in Dejong-up, all located in the western part of Cheju Island. In total, 45 species consisting 5,117 individuals were observed during the census period. There were 40 species and 4,910 individuals in the eastern area, 38 species and 927 individuals in the western area. The number of individuals of shorebirds in the spring migratory season was greater than in the autumn. The dominant species researched during the first census period were 2,024 individuals of Calidrius alpina (88.5%), 84 individuals of Charadrius alexandrinus (3.7%), 59 individuals of Charadrius placidus (2.6%) in spring and 384 individuals of Calidrius alpina (82.9%), 20 individuals of Charadrius alexandrinus (4.3%), 11 individuals of Charadrius placidus (2.4%) in autumn. The dominant species researched during the second census period were 116 individuals of Calidrius ruficolis (21.8%), 67 individuals of Calidrius alpina (12.6%), 58 individuals of Tringa nebularia (10.9%) in spring and 191 individuals of Calidrius alpina (24.9%), 156 individuals of Tringa brebipes (20.4%), 104 individuals of Tringa ochropus (13.6%) in autumn. Dominance index were recorded better in the eastern area than the western area, but species diversity and evenness index were recorded lower in the eastern area than the western area. Similarity index were 0.85 between the eastern area and the western area. Hydrophasianus chirurgus, Haematopus ostralegus, Gallinago stenura and Calidrius temminuckii, which are observed during the census period are previously unrecorded species on Cheju Island. Rare and endangered species were observed such as Haematopus ostralegus, Charadrius placidus, Numenius madagascariensis, and Eurynorhynchus pygmeus.

        KAL기 추락에 의한 대량재해의 개인식별

        Individual identification is an important part in medicolegal field especially in mass disaster. At July, 27, 1989, KAL KE-803 was crashed on landing at Tripoli International Airport, Liba. The plane was caught in fire and sixty eight Koreans were sacrified. The majority of victims were severely charred and injured. The authors examed all dead bodies and successfully identified all the cases through visual, anthropological, odontological, radiological and pathological methods including fingerprint and blood typing.

      • 제주도의 주요 습지에 도래하는 도요ㆍ물떼새류의 현황

        This study was part of a continuous research project on the status of waterbirds on major wetlands on Cheju Island. This study was researched twice each month for two years, from March 1997 to February 1999. In total, 29 species consisting of 4,887 individual birds were observed. The number of individuals of shorebirds was greater in the spring migratory season than in the autumn - in 1997, 131 birds in the spring, 104 birds in the autumn and in 1998, 2,453 birds in the spring, 635 birds in the autumn Pheasant-tailed Jacana, Eurasian Oystercatcher, Pintail Snipe and Temminck's Stint observed at the Hado-ri or Sungsan-po fishfarm are the first recordes on Cheju Island, Korea. Rare and endangered species were observed such as Eurasian Oystercatcher, Long-billed Plover, Far Eastern Curlew, and Spoon-billed Sandpiper. This survey area used for a suitable roosting and feeding place by many shorebirds. Thus, there is a need of critical plans for the conservation and management of shorebirds migrating in this area.
