      • 수종 식호르몬이 양란의 Somatic embryo분화에 미치는 영향(Ⅰ)

        The effect of indoleacetic acid, kinetin and gibberellin on the development of somatic embryo from cultured cell of Cymbidium janette were observed. The results obtained from the present work are as follows; As compaired with control, homonal treatment brought about promoted level of a number of pro-tocorm and embryo development. Development of somatic embryo, when treatment with various combinations of homones were tested, the maximal promotion was observed with 10^(-6)M IAA+10^(-5)M kinetin+10^(-5)M GA_(3) in suspension culture. With 10^(-6)M IAA+10^(-5)M kinetin promoting a large number of protocorm formation, but these are completely undifferentiated protocorm. The results demonstrated that gibberellin can effectively control somatic embryo development, kinetin and mdoleacetic acid effect on cell division.

        Pretreatment of excised etiolated of cucumber (Cucumis sativus L. cv. Cheongchangmadi)with indoleacetic acid and water-control were continuously irradiated under white light with 2200 lux. The δ-aminolevulinic acid synthetase activity and the rate of δ-aminolevulinic acid convert into prophobilinogen and protochlorophyll were higher than those of the control in which case chlorophyll formation was stimulated by IAA and Light.

      • 수종 식물호르몬이 양란의 Somatic embryo 분화에 미치는 영향(II)

        양란(Cymbidium janette)을 재료로하여 식물생장 촉진 호르몬인 IAA,kinetin,hibberellin을 조합처리 한 후 진탕배양하여 개체분화에 미치는 영향을 조사혀여 몇가지 결론을 얻었다. 대조구에 비하여 호르몬 처리구의 경우protocorm수의 증가와 embryo의 분화가 높았다. ?? kinet처리구에 있어서는 생장분열조직이 embryo의 표면을 이루고 있으며 이것은 전혀 분화되지 않은 것이다. Somatic embryo의 분화는 ?? 처리구에서 root와 shoot 및 물관부의 형성이 현저하다. 이러한 결과로 보아 양란의 배양에 있어서 GA₃는 기관분화에 영향을 주며 kinetin과 IAA는 세포의 증식에 영향을 주고 있다고 사료된다. The effect of indoleacetic acid, kinetin and gibberellin on the development and differentiaiton of somatic embryo from cultured cell of Cymbidium janette were observed. The results obtained from the present work are as follows; As compared with control, hormonal treatment brought about promoted level of a number of protocorm and embryo differentiaion. With ?? kinetin promoting the formation of merstematic tissue on the all surface of embryo,but these are completely undifferentiated protocorm. Differentiation of somatic embryo,when treatment with various combinations of hormones were tested, the max mal differentiation was observed with ?? in suspension culture. The results demonstrated that gibberellin can effectively control differentiation of plant organ, kinetin and IAA effect on cell division.

      • 엽록소-단백질 복합체 형성에 미치는 IAA와 zeatin의 영향

        In greening cucumber(Cucumis sativus L. cv. Cheongchangmadi) cotyledons during the 12 hoursubsequent continuous illumination with 2,200 lux of white light, effect of IAA and zeatin on formation of chlorophyll-protein complex were observed. Cotyledons excised from dark-grown cucumber seedling were pretreated in dark with 10??M IAA, 10??M IAA, 10??M zeatin, 10??M zeatin or incubated with water throughout the dark preincubation period. The results obtained from the present work are as follows: The chlorophyll-protein complex were separated by sodium dodecyl sulfate-polyacrylamde gel electrophoresis. Rapid synthesis of a 30,000 dalton light harvesting chlorophyll protein was observed with 10??M IAA during the 9 hour culture period and 10??M zeatin during the 6 hour incubation time. The CP1a+CP1/C-P complex ratio was gradually increased from 2 to 8 hour but decreased from 8 hour by increasing light harvesting chlorophyll protein. The more rapid decreased was observed with 10??M zeatin, the minimal value of 35.2 elution volume/elution volume in the 12 hours. The data suggest that IAA and zeatin have an influence on the formation of chlorophyll-protein complex.

      • 쥐 심장의 미토콘드리아에서 분리한 Carnitine Acetyltransferase에 대한 DTP의 반응 억제와 Acetyl-CoA 와 Propionyl-CoA의 방어적인 효과에 관한 연구

        Carnitine acetyltransferase was purified from rat liver peroxisomes and mitochondria, as well as rat heart mitochondria. Formation fo acetylcarnitine was assyed using the spectrophotometric forward assay with DTP as the SH complexing agent. When velocity vs. substrate concentration profiles were performed, using acetyl-CoA and propionyl-CoA at low acyl CoA concentrations less than 0.5 μM, an apparant lag in acetylcarnitine, but not propionylcarnitine, production observed. Such data suggest that carnitine acetyltransferase exhibits different kinetic parameter with acetyl-CoA as compared to propionyl-CoA. These experiments were repeated using carnitine acetyltransferase purified from beef heart mitochondria and an isotope forward assay. With th eisotope forward assay, a significant inhibition of carnitine acetyltransferase by DTP was observed at low acetyl-CoA concentrations(i.e., less than 0.5μm); much less inhibition by DTP was observed with propionyl-CoA. These data show that at acyl-CoA concentrations below the K?? for acyl-CoA, DTP is quite inhibitory to CAT. Better substrate protection is provided by propionyl-CoA compared to acetyl-CoA. The cause for the difference is not known.

      • Citrus callus의P-fraction 조성에 미치는 수종 식물 호르몬의 영향(Ⅱ)

        감귤(Citrus unshiu Marc.)을 재료로 진탕배양하여 식물 생장촉진 호르몬인 IAA, Kinetin, Gibberellin을 조합처리를 하여 생장율, 산용성 인의 변화, 지용성인의 변화, 핵산태 인의 변화, 단백태 인의 변화, 전인의 변화를 조사하여 몇가지 결론을 얻었다. 세포생장은 대조구 및 2가지 호르몬 처리구 보다 IAA10??M+Kinetin 10??M+GA₃10??M 처리구가 배양 15일 후 99mg/flask로 가장 높았다. 신용성 인은 배양 3일∼6일 사이에 건물중에 대해 급격히 감소하는 결과를 보였으며 IAA10??M+Kinetin 10??M+GA₃10??M 처리구에서 감소가 가장 빠르며 6일 이후 증가하는 반면 대조구는 배양 12일이후 증가 하였다. 지용성 안의 건물중에 대한 함유율은 배양 3일부터 6일사이에 증가하고 그 이후 감소하는 결과를 보였으며 대조구에 비해 모든 호르몬 처리구의 지용성 인의 함유율은 높았고 대조구는 배양 6일후 0.215mg/g dry weight이 되었으며 IAA 10??M+Kinetin 10??M+GA₃10??M 처리구의 경우 배양 12일에 0.375mg/g dry weight이 되어 가장 높았다. 핵산태 인은 잠복기에서 증가하며 배양 3일 후 건물중에 대한 함유율이 감소되는 경향을 나타내었고 2종의 호르몬 처리구 사이에는 큰 차이가 없으나 IAA 10??M+Kinerin 10??M+GA₃10??M 처리구의 경우 전배양기간을 통하여 2종의 호르몬 처리구 및 대조구에 비해 높았다. 단백태 인은 배양 6일내에 급격한 증가를 보이며 2종의 호르몬을 처리한 시험구의 건물중에 대한 단백태 인의 양은 배양 6일에 2.05mg/g dry weight으로 대조구의 6일째 1.5mg/g dry wieight 보다 높았으나 IAA10??M+Kinerin 10??M+GA₃10??M처리구의 2.34㎎/g dry weight 보다는 낮았다. 전 인은 대조구 및 2 가지 호르몬 처리구보다 IAA 10??M+Kinetin 10??M+GA₃10??M 처리구가 배양 9일에 4.8mg/g dry weight으로 가장 높았다. In suspension culture of Citrus unshiu Marc. cells, effect of various combinations of plant hormones such as 10??M IAA,10??M kinetin, 10??M GA₃ on the cell growth and changes in the amount of various fractions of phosphorus were observed. The result obtained from the present work are as follows: Growth of the cell, when treatments with various combinations of hormones were tested, the maximal promotion was observed with 10??MIAA+10??M kinetin+10??M GA₃. The amount of acid-soluble phosphorus decreased during the 3rd and 6th days of culture, but increased from the 6th day on. The most rapid decrease during this early period of culture was observed with 10??MIAA+10??M kinetin+10??M GA₃. On the contrary, the amount of fat-soluble phosphorus increased during the 3rd and 6th days of culture, and decreased after the 6th day. As compared with the control, hormonal treatments brought about increased levels of this particular phosphorus fraction, the maximal value of 0.375mg/g dry weight being obtained with 10??M IAA+10??M kinetine+10?? GA₃ on the 12th days of culture. The amount of nucleic phosphorus fraction was increasing by the 3rd day, but tended to decrease thereafter. No significant hormonal effect were noticed on this fraction, when combinations of two hormones were tested. However, with 10? M IAA+10??M kinetin+10??M GA₃, the value was higher than that of control or other hormonal treatments. The protein-P fraction increased rapidly until the 6th day of culture followed by aplateau thereatfter, On the 6th day it was 2.34mg/g dry weight with 10?M IAA+10??M kinetin+10??M GA₃, which was higher than that of the control or other treatments. The total phosphorus contents tended to increase throughout the whole culture period. Again, the hightest value was obtained with 10??M IAA+10??M kinetin+10??M GA₃.

        감귤(Citrus unshiu Marc.)을 재료로 진탕배양하여 식물 생장 촉진 호르몬인 IAA, kinetin, gibberellin을 농도별 처리를 하여 생장율, 전 인의 변화, 배지내 인의 흡수를 조사하여 몇가지 결론을 얻었다. 세포생장은 농도별 처리구에 있어 IAA10?M, kinetin 10?M, GA₃10-⁴M∼10?M 처리구가 대조구 및 다른 농도별 처리구보다 높았으며 IAA10-⁴M, IAA10?M 처리구의 경우 생장억제 효과를 보였다. 전 인의 변화에 있어서 배양 9일후 IAA10?M처리구가 4.77mg/g dry weight, kinetin 10-⁴M 처리구는 4.42mg/g dry weight, GA₃10-⁴M 처리구는 4.68mg/g dry weight으로 대조구 및 다른 농도별 처리구에 비해 높았다. 배지내 인의 흡수는 배양 9일에 IAA10?M처리구가 33%흡수되어 가장 흡수가 빠르며 kinetin 10-⁴M처리구는 27.1%, GA₃10-⁴M 처리구는 29.9%의 흡수가 이루어졌으며 같은 시기 대조구는 14.8%의 흡수가 되었다. In suspension culture of Citrus unshiu Marc. cells, effect of plant hormones such as IAA, kinetin and GA₃on the cell growth, uptake of phosphorus by the cells from the culture medium and changes in the amount of total phosphorus were observed. The results obtained from the present work are as follows : Growth of the cells was promoted by 10?M IAA, 10?M kinetin and 10-⁴M to 10?M GA₃respectively. The total phosphorus contents of cells on the 9th day of culure were 4.77㎎/g dry weight with 10? IAA, 4.42mg/g dry weight with 10-⁴M kinetin, and 4.68mg/g dry weight with 10-⁴M GA₃respectively, all of which are higher than those of the control. By the 9th day of culture, 14.8% of the phosphorus in the medium was taken up by the cells for the control, 33% for 10?M IAA, 27.1% for 10-⁴M kinetin, and 29.9% for 10-⁴M GA₃, respectivrly.

        Carnitine acetyltransferase is a short-chain acyltransferase in the cell organelle and was purified from pheasant breast muscle mitochondria. The molecular weight of mitochodrial carnitine acetyltransferase determined by SDS-PAGE is 66,000. Formation of acylcarnitine was assayed using the spectrophotometer forward assay with DTP as the SH complexing agent. When velocity vs. substrate concentration profiles were performed, using acetyl-CoA and propionyl-CoA at low acyl-CoA concentration less than 0.8μM, an apparant lag in acetylcarnitine, but not propionylcarnitine, production observed. Such data suggest that carnitine acetyltransferase exhibits different kinetic parameter with acetyl-CoA as compared to propionyl-CoA. The acylgroup of octanoyl-CoA and decanoyl-CoA was not transferred by camitine acetyltransferase.

        고등학교 생물과 실험실습의 현안과 개선방안에 관한 연구 : 제주도 지역 인문계 고등학교를 중심으로

        The purpose of this study is to examine and analyze the contents of experiment in Biology and the conditions of management which have been Bona on the basis of the 5th national curriculum, and to use them as the fundamental data for the high school biology experiment teaching. The results of the study are as follows : 1. Only five out of college volunteer classes in Cheju-do high schools had laboratories fitting for the criteria of the Ministry of Education, but seven high schools didn't satisfy the criteria. One of them has only one laboratory which covers all experiment and practice for Physics, Chemistry, Biology and Earth Science. hence, it is urgent to secure laboratories to give an efficient experiment lesson for the inquiry learning. 2. Most of high schools have a goon variety of experimental devices except for dissect microscope. They frequently use the experimental devices whose function is limited only in laboratories, nut they don't use the field experimental devices more often. In Cheju-do high schools where learning environments of taxonomy and ecology are favorable, teachers of science should let students interest in Biology through contact with nature. 3. The results of observation on class hours of Science I (High) and Biology per a Large unit show that there is double the difference of the whole class hour-assignment among high schools. The subjects are taught at different semesters and different grades from school to school. It is necessary for teachers to discuss and adjust the class hoax assignment of the 5 major subject in the textbooks of Science I (High) and Biology 4. The results of how high schools did experimental practices in the Science I (High) and Biology Classes are that the average experimental rate is 46.7% for Science I (High), and Biology it is 60% in the seven high schools with experiment. However, 5 high schools don't experiment in Biology. They tend to spend their time doing observation activity in the laboratory in the middle of the experimental practices and inquiry activities, It is desirable to encourage various inquiry learnings in order to cultivate more scientific attitudes toward experiments for students. 5. The number of students per one classroom and experimental pairs show that the number of each group is six to eight. This may hinder the efficient experiment and students' active involvement in that experiment. Therefore, it is important to have a class size with 40 students, to set up some laboratories, and to assign 4 students to each group for the better Inquiry learning. 6. One of the most difficult problems that teachers of science have during the experimental practices is lack of time because of burdening with teaching students for college entrance exam. They want to evade their responsibility to do the experiment and practice, for they are afraid they can not finish them within one class hour, Most of them would like to have two classes consecutively for an efficient experiment. It is advisable to have two consecutive classes, at least more than once a week, to effectively experiment in Physics, Chemistry, Biology and Earth Science.
