      • Down-Regulation of Non-Coding circHIPK3 RNA as Human Hepatocellular Carcinoma Biomarker

        Aims: Hepatocellular carcinoma (HCC) is one of the most common cancers worldwide, with poor prognosis and high risk of recurrence. Previous studies have shown that various microRNAs (miRNAs) are frequently dysregulated in HCC, which contributes to cancer development and progression. Nonetheless, it is still unclear the regulation mechanism of miRNAs. Circular non-coding RNA (circRNA) is highly conserved and stable covalently closed RNA circles with gene-regulatory potential. Recently, one abundant circRNA from HIPK3 gene, circHIPK3, has been identified, and its function has been elucidated as multiple miRNAs sponge. Thus, we aim to investigate the clinical relevance of circHIPK3 in HCC. Methods: We analyzed clinical specimens from 152 pairs of HCC and corresponding normal liver (NL) tissues. Total RNAs were isolated from clinical tissues using qiagen kit. CircHIPK3 expression levels were determined by quantitative real-time PCR (qRT-PCR), and its expression was normalized with GAPDH. In addition, we analyzed the correlation between circHIPK3 expression and various clinicopathological features of HCC patients. Results: CircHIPK3 expression was significantly down-regulated in HCC tissues compared to corresponding NL tissues (P=0.036). In HCC patients, circHIPK3 expression was strongly suppressed in more advanced tumors (P<0.001). Further correlation analysis showed that circHIPK3 expression was significantly associated with T-stage (P<0.001), TNM stage (P<0.001), BCLC stage (P<0.001), and alpha-fetoprotein (AFP) expression (P=0.033). Conclusions: We have determined clinical significances of circHIPK3 expression in HCC, which may provide clinical evidence for the potential of circHIPK3 as novel markers for diagnosis and predicting prognosis in HCC patients.

        COVID-19 Outbreak in a Military Unit in Korea

        OBJECTIVES: This study presents the response of a military unit to an outbreak of coronavirus disease 2019 (COVID-19) in Gyeonggi Province. As soon as 2 soldiers were identified as index cases, the infectious disease investigators of the Gyeonggi Provincial Government, Korea Disease Control and Prevention Agency, and the Armed Forces Epidemiologic Investigation Center discussed the investigation and response plan for an imminent massive outbreak. METHODS: The joint immediate response team (IRT) conducted interviews with confirmed COVID-19 patients, reviewed their medical records, performed contact tracing using global positioning system data, and undertook a field investigation. For risk assessment, the joint IRT visited all 8 sites of the military units and the army chaplain’s church to evaluate the transmission risk at each site. The evaluation items included the size of the site, the use of air conditioning, whether windows were opened, and whether masks were worn. Pooled testing was used for the low-risk population to quickly detect the spread of COVID-19 in the military base. RESULTS: One day before the symptom onset of the index case, the lecturer and >50% of the attendees were infected with COVID-19 while attending a lecture that lasted 2 hours and 30 minutes. Attendees were not wearing masks and were in a poorly ventilated room. CONCLUSIONS: Since COVID-19 can be spread before symptom onset, contact tracing must be performed to investigate potential exposures prior to symptom onset and to manage any exposed persons.

        패션브랜드 매장에 적용된 영상미디어 중심 VMD 특성에 관한 연구

        (연구배경 및 목적)빠르게 변화하는 경제 성장에 따라 최근 패션브랜드 산업에 대한 소비자들의 의식과 소비 패턴 또한 변화하고 있다. 한정된 공간 안에서 단시간에 수많은 정보들을 공유 하고자 하는 소비자들에게 디지털 매체를 활용한 다양한 영상물과 시각적인 표현은 소비활동을 극대화 시키고, 쌍방향 커뮤니케이션이 결합하여 차별화된 경쟁력을 제시할 수 있다. 이에 본 연구에서는 패션브랜드 매장에 적용된 영상미디어를 중심으로 VMD 특성을 분석하는데 목적을 두고 있다. (연구방법)분석의 범위는 첫째, 패션브랜드 매장에 적용된 영상미디어 중심 VMD의 영역별 역할 표현 요소를 분석한다. 둘째, 영상미디어의 특성과 선행 연구를 기반으로 도출된 패션브랜드 매장에 적용된 영상미디어의 4가지 유형인 메커니컬 유형, 코드 유형, 형태적 유형, 인터랙션 유형을 분석의 범주로 설정하여, 패션브랜드 매장에 적용된 영상미디어 중심 VMD특성을 분석한다. 셋째, 패션브랜드 매장에 적용된 VMD의 영역별 역할 표현요소와 영상미디어 분류의 상관성을 분석 하여 결과를 도출한다. (결과)사례분석의 결과 첫째, 패션브랜드 VMD의 영역별 역할 표현요소 분석 결과 인지성> 서비스성> 이미지성> 체험성 순으로 많이 사용되어지고, 이미지성의 표현요소 중에는 쇼윈도가 인지성 표현요소에는 조명이 체험성에는 집기를 이용한 영상미디어가 많은 부분을 차지하고있다. 서비스성에는 미디어 매체, 소셜 네트워크를 이용한 정보공유와 체험적 콘텐츠를 이용한 고객 유도 성격이 강하게 나타나고 있다. 둘째, 패션브랜드 매장에 적용된 영상미디어의 분류를 보면 , 소프트웨어적인 코드유형이 가장 큰 비율을 차지하며 사례조사 결과 영상미디어의 분류 중 가장 많이 적용된 유형이라 할 수 있다. 다시 말해 , 패션브랜드 매장에 적용된 영상미디어 중심 VMD 특성은 디지털 기술의 형태인 동적 동영상을 프로젝션 스크린을 이용한 터치스크린 형식으로 체험적 서비스를 고객들이 경험하여 상품에 대한 흥미를 유발시키고 판매를 유도하는 역할을 하고 있다. (결론)본 논문의 연구 결과, 패션브랜드 매장에 적용된 영상미디어 중심 VMD특성을 보면, 인지성을 강조하는 요소 중 하나인 빛에 의한 시각적 효과와 소셜 네트워크와 미디어적 요소를 통한 체험으로 고객과 브랜드 매장과의 상호 작용 성을 중요하게 보여 주고 있다. 쇼윈도를 활용한 이미지성 브랜드광고와 매장 내 사용되는 집기, 소도구를 이용한 영상미디어의 인터랙션 유형의 미디어는 고객에게 브랜드에 대한 호감도와 구매 유도를 하는 탁월한 방법이라 할 수 있겠다. 체험과 경험의 형태를 통해 매체와사용자, 사용자와 사용자, 매체와 매체간의 쌍방향 커뮤니케이션을 형성해 준다. 영상 미디어를 적용한 공간은 사람들에게 다양한 체험과 즐거움을 줄 수 있고 영상 미디어를 이용한 다양한 판매 전략이 생겨나도록 지속적 연구가 필요하다고 사료되어진다. (Background and Purpose)Due to rapid economic growth, consumers’ awareness of the fashion brand industry and their consumption patterns are changing. For customers who wish to share a lot of information in a limited space and a short time, various videos and other forms of visual expression using digital media can maximize consumption and provide means of differentiation and competitiveness by combining two-way communication. Thus, the purpose of this study is to analyze VMD characteristics based on visual media applied to fashion brand shops. (Methods) The study process is as follows: To complete the analysis, first, the area-specific role expression elements of the visual media-centered VMD applied to fashion brand shops are considered. Second, the visual media-centered VMD characteristics applied to fashion brand shops are addressed by establishing mechanical types, code types, morphological types, and interaction types, which are the four aspects of visual media applied to fashion brand shops derived from the characteristics of, and previous research on, visual media as the scope of the analysis. Third, the results are derived by analyzing the correlation between the area-specific role elements of VMD applied to fashion brand shops and the visual media classification. (Results) The results of the case analysis are as follows: First, with respect to the area-specific role elements of fashion brand VMD, recognition> service> image > experience are used to a great extent, and show window among expression elements of image, lighting among recognition expression elements and the visual media using an appliance among experience elements account for a large portion. In terms of service elements, information-sharing using media outlets, social networks, and other customer-facing aspects using experiential content are used to a large extent. Second, with respect to classification of the visual media applied to fashion brand shops, software code types account for the largest percentage; from the results of the case study, these types can be said to be applied more than other categories of visual media. In other words, the visual media-centered VMD characteristics applied to fashion brand shops play a role in attracting customers’ interest in products and inducing sales by providing services through dynamic videos in the form of digital technology via touch screens or projection screens. (Conclusions) According to the results of this study, the visual media-centered VMD characteristics applied to fashion brand shops are used to increase interactivity between customers and shops by significantly enhancing their experience through visual effects such as light, one of elements emphasizing recognition and social networks and media elements. Image brand advertising using show window and interaction-type visual media using appliance and props within the shop can be said to be an excellent way of inducing customers’ preferences for the brand and inciting them to purchase. Through the form of experience, interactive communication between the media and the user, the user and the user, the media and media has been created. Spaces that use visual media can give people a variety of experiences and enhance their enjoyment, so continuous research is considered necessary to create various sales strategies using visual media.

      • Circulating Exosomal Non-Coding RNAs (microRNA-21, lncRNA-ATB) as Prognostic Biomarkers of Human Hepatocellular Carcinoma

        Aims: It is important to investigate the better prognostic markers of hepatocellular carcinoma (HCC) to improve the clinical management of HCC patients. Tumor-derived exosomes contain tumor-specific non-coding RNAs and having an emerging role in tumor progression and metastasis. Therefore, we determined the prognostic relevancies of two noninvasive circulating exosomal noncoding RNAs (miRNA-21, lncRNA-ATB) for HCC. Methods: HCC patients between October 2014 and September 2015 at Kyungpook National University Hospital, Daegu, South Korea were enrolled for this prospective study. Exosome was isolated by using ExoQuick (System Biosciences). In isolated exosome, exosomal RNA was extracted using miRNeasy serum/plasma kit (Qiagen). Cox regression analysis was used to identify independent predictors of HCC prognostic factors. Results: A total of 79 patients were enrolled in this study. During the study period, disease progression and mortality were found in 44 and 34 patients, respectively. Both miRNA-21 and lncRNA-ATB were successfully detected in the HCC serum sample. Both miRNA-21 and lncRNA-ATB were related to TNM stage and other multiple prognostic factors including T stage and portal vein thrombosis. However, they were not associated with age, gender, presence of cirrhosis, and etiology. In multivariate analysis, both higher miRNA-21 and higher lncRNA-ATB were identified as independent predictors of mortality and disease progression, along with larger tumor size and higher CRP (all P<0.05). The overall survival of HCC patients with a low miRNA-21 expression group (miRNA-21 < 0.09) was significantly higher than in patients with a high miRNA-21 expression group (miRNA-21 >0.09) (log-rank test: P=0.014). The overall survival was also significantly higher in patients with a low lncRNA-ATB expression group (lncRNA-ATB < 0.0016) than in patients with a high lncRNA-ATB expression group (lncRNA-ATB >0.0016) (log-rank test : P=0.005). Conclusions: This study provides strong evidence that circulating exo-somal non-coding RNAs (miRNA-21, lncRNA-ATB) may be novel prognostic markers and therapeutic targets for HCC.

        Diagnostic Efficacy of Serum Mac-2 Binding Protein Glycosylation Isomer and Other Markers for Liver Fibrosis in Non-Alcoholic Fatty Liver Diseases

        Background: Mac-2 binding protein glycosylation isomer (M2BPGi) has been established as a non-invasive biomarker for liver fibrosis. We evaluated the diagnostic efficacy of M2BPGi compared with those of other liver fibrosis markers in liver fibrosis in non-alcoholic fatty liver disease (NAFLD). Methods: We analyzed serum M2BPGi levels in 113 NAFLD patients. A pathologist graded liver fibrosis histopathologically. The diagnostic efficacies of serum M2BPGi and other liver fibrosis markers (aspartate aminotransferase to platelet ratio index, fibrosis index based on four factors, and NAFLD fibrosis score [NFS]) were evaluated using correlation, area under the ROC curve (AUC), logistic regression, and C-statistics. Results: Serum M2BPGi level and other liver fibrosis markers showed a moderate correlation with fibrosis grade. The AUC values of M2BPGi were 0.761, 0.819, 0.866, and 0.900 for diagnosing fibrosis (F)>0, F>1, F>2, and F>3, respectively. Logistic regression analysis showed M2BPGi as the only independent factor associated with F>2 and F>3. Although C-statistics showed that NFS was the best diagnostic factor for F>2 and F>3, M2BPGi with NFS had an increased C-statistics value, indicating that it is a better diagnostic model. Conclusions: The serum M2BPGi level increased with liver fibrosis severity and could be a good biomarker for diagnosing advanced fibrosis and cirrhosis in NAFLD patients. A well-controlled, prospective study with a larger sample size is needed to validate the diagnostic power of M2BPGi and other fibrosis markers in NAFLD.
