EGFR Gene Amplification and Protein Expression in Invasive Ductal Carcinoma of the Breast

Background: The epidermal growth factor receptor (EGFR) is a surrogate marker for basal-like breast cancer. A recent study suggested that EGFR may be used as a target for breast cancertreatment. Methods: A total of 706 invasive ductal carcinomas (IDC) of the breast were immunophenotyped, and 82 cases with EGFR protein expression were studied for EGFR gene amplification. Results: EGFR protein was expressed in 121 of 706 IDCs (17.1%); 5.9% were of luminal type, 25.3% of epidermal growth factor receptor 2 (HER-2) type, and 79.3% of basal-like tumors. EGFR gene amplification and high polysomy (fluorescent in situ hybridization [FISH]-positive) were found in 18 of 82 cases (22.0%); 41.2% of the HER-2+, EGFR+, cytokeratin 5/6– (CK5/6–) group, 11.2% of the HER-2–, EGFR+, CK5/6– group, and 19.1% of the HER-2–, EGFR+, CK5/6+ group. FISH-positive cases were detected in 8.3% of the EGFR protein 1+ expression cases, 15.9% of 2+ expression cases, and 38.5% of 3+ expression cases. In group 2, the tumors had a high Ki-67 labeling (>60%), but the patients showed better disease-free survival than those with tumors that co-expressed HER-2 or CK5/6. Conclusions: EGFR-directed therapy can be considered in breast cancer patients with EGFR protein overexpression and gene amplification, and its therapeutic implication should be determined in HER-2 type breast cancer patients.

A wrinkling is the phenomenon of instability influenced by material properties, part geometry, forming conditions, stress state, etc. In this study, the region in biaxial plane stress state is modeled with a rectangular plate introducing the effective dimension, and critical stress values for the wrinkling are calculated using energy method. Prediction index for the wrinkling is then evaluated by normalizing the actual stress with respect to critical stress.
Impingement wastage experiment with SUS 316 in a printed circuit steam generator

The sodium cooled fast reactor (SFR) is one of the Gen-IV reactors with the most operating experience accumulated. Although the technology level is the most mature among the Gen-IV reactors, there is still a safety problem that has not been solved, which is the sodium-water reaction. Since sodium and water are separated only by a heat transfer tube with a thickness of only a few mm, there is inherently a risk of a sodium-water reaction (SWR) accident in the SFR. In this study, it is attempted to quantitatively evaluate the resistance of SWR accidents by replacing the shell and tube steam generator with printed circuit steam generator (PCSG) as a method to mitigate the SWR accident. To do this, a CATS-S (Compact Accident Tolerance Steam Generator-SWR) facility was designed and built. And for the quantitative evaluation of accident resistance, a methodology for measuring the impingement wastage rate was established. As a result of this research, the impingement wastage rate caused by SWR generated in a PCSG was measured first time. It was confirmed that the impingement wastage phenomenon was suppressed in the PCSG, and the accident resistance was higher than that of the SWR through comparison with the experimental results performed in the existing shell and tube steam generator. In conclusion, a PCSG is more resistant to impingement wastage as a result of the SWR accident than existing shell and tube steam generators, and it is estimated that a PCSG can mitigate SWR accidents, an inherent problem of SFR.
