        체세포복제를 통한 멸종위기동물의 종 보전 평가

        The number of wild animal species is gradually decreasing due to poaching, hunting and habitat loss. While several endangered animal species have been successfully preserved at the zoo, assisted reproductive technology (ART) must be applied to restore wild animals. In the case of critically endangered animals, somatic cell cloning is considered the most appropriate method of ART. Somatic cell cloning can be beneficial for the reproduction of endangered species with limited female populations. However, gene and cell banks, and understanding of reproductive physiology and optimization of ART for wild animals are urgently required for further activation of artificial reproduction of endangered species, which enlarges its application and maintains biodiversity. Care should also be taken to consider ethical and legal issues associated with somatic cell cloning for conservation of endangered animals.

        Identification of a Novel Gene by EST Clustering and its Expression in Mouse Ovary and Testis

        목 적: 본 연구에서는 EST Clustering 방법을 이용하여 이전의 연구에서 발굴한 B357 EST의 염기서열을 포함하는 유전자를 동정하고, 이 유전자의 발현을 생쥐의 난소와 정소를 포함한 여러 가지 조직들에서 살펴보고자 하였다. 연구방법: EST Clustering으로 얻어진 전체 염기서열을 5-day-ovary-specific gene-1 (5DOS1)이라고 명명하여 GenBank에 등록하였으며 (AY751521), northern blotting, real-time RT-PCR, in situ hybridization, western blotting, immunohistochemistry 등의 방법을 이용하여 생쥐 난소와 고환의 발달단계에 따라 그 발현양상을 관찰하였다. 결 과: 5DOS1의 전사체는 성장한 정소, 뇌, 근육에서 높게 발현하였으며, 난소의 경우에서는 원시난포시기부터 모든 난자에서 발현하였으며 특히 생후 5일째 높게 발현하였고 그 이후로는 점차 감소하였다. 반면 정소의 경우는 발달과 함께 계속 증가하였으며, 정모세포를 제외한 모든 발달단계별 정자에서 발현함을 관찰하였다. 결 론: 본 연구결과는 5DOS1에 대한 발견과 동정에 대한 첫 보고로써, 유전자 및 단백질이 생쥐의 난소 및 정소의 생식세포에서 발현하는 것을 관찰하였다. 앞으로 생식세포 발생 및 분화에 관련된 5DOS1의 기능에 대한 심층 연구가 더 필요하다고 사료된다. Objective: Identification of the regulatory mechanism for arrest and initiation of primordial follicular growth is crucial for female fertility. Previously, we found 15 expressed sequence tags (ESTs) that were specifically abundant in the day-S-subtracted cDNA library and that the B357 clone was novel. The present study was conducted to obtain the whole sequence of the novel gene including B357 and to characterize its mRNA and protein expression in mouse ovary and testis. Methods: The extended sequence of the 2,965-bp cDNA fragment for the clone B357 was named ${\underline{5}}-{\underline{d}}ay-{\underline{o}}vary-{\underline{s}}pecific\;gene-{\underline{1}}$ (5DOS1) and submitted to GenBank (accession number ${\underline{AY751521}}$). Expression of 5DOS1 was characterized in both female and male gonads at various developmental stages by Northern blotting, real-time RT-PCR, in situ hybridization, Western blotting, and immunohistochemistry. Results: The 5DOS1 transcript was highly expressed in the adult testis, brain, and muscle as compared to the other tissues. In the ovary, the 5DOS1 transcript was detected in all oocytes from primordial to antral follicles, and highly expressed at day 5 after birth and decreased thereafter. In contrast, expression of 5DOS1 showed a gradual increase during testicular development and its expression was limited to various stages of male germ cells except spermatogonia. Conclusions: This is the first report on the expression and characterization of the 5DOS1 gene in the mouse gonads. Further functional analysis of the 5DOS1 protein will be required to predict its role in gametogenesis.

      • 沈菜類 熟成에 따르는 Microflora, 糖, pH 및 Vitamin B_(12)의 變化와 Pseudomonas sp.의 Vitamin B_(12) 生産性에 관하여

        This study was to investigate the changes of microflora, sugar, pH, and vitamin B_(12) during the aging of fermented vegetables such as Tongkimchi and Dongchimi and the productivity of vitamin B_(12) by Pseudomonas denitrificans D-2-1 selected as vitamin B_12 producing bacteria from Dongchimi. The main bacteria responsible for Tongkimchi fermertation were Lactobacillus plantarum, Leuconostoc mesenteroides and Streptococcus faecalis, but the main bacteria for Dongchimi fermentation were Leuconostoc mesenteroides and the genus Pseudomonas. In TongKimchi and Dongchimi sugar continuously decreased from ca 3% and pH reduced from 5.8 to 4.0 and less, but increased as the over-ripening progresses. Vitamin B_(12) content of TonKimchi was higher than those of Dongchimi at the early stage, but the opposite result at a later period Vitamin B_(12) content of TongKimchi and Dongchimi reached its maximum at the middle period. Pseudomonas denitrificans D-2- 1 cultured for 15 days on the media of radish juice produced vitamin B_12 up to 274 ng/㎖ and the strain cultured on the media of radish juice containing 1.5% NaCl produced vitamin B_12 of 261 ng/㎖. The strain cultured on the media of radish juice containing 3% NaCl and 0.1% L-glutamic acid as a nitrogen source produced vitamin B_(12) of 272ng/㎖. In contrast to the amount of vitamin B_(12)(245ng/㎖) obtained from the media in which L-glutamic acid was not added, the media containing L-glutamic acid increased the productivity of vitamin B_(12) up to 11%

      • 原發性 肝癌患者의 腹水 r-glutamyl transpeptidase 活性値에 關한 硏究

        Gamma-glutamyl transpeptidase (γ-GTP) is a enzyme, cencentrated mainly in kidney, pancreas, liver, spleen, small intestine, brain and heart muscle as a very sensitive indicator of liver cell dysfunction, especially in primary hepatoma. For the purpose of differential diagnosis of ascites with primary hepatoma, the levels of ascitic γ-GTP in various gastrointestinal diseases have been estimated, the correlation between ascitic γ-GTP and serurn alkaline phosphatase, SGOT, SGPT activity, ascitic total protein amount established. Also by peritoneoscopic examination the relationship between γ-GTP levels and extent of involvement of primary hepatoma has been analyzed. The material included 19 cases of primary hepatoma, 18 cases of liver cirrhosis, 12 cases of tuberculous peritonitis, 18 cases of stomach cancer and 6 cases of benign intraabdominal tumor. The results are as follows. 1. The peak age incidence of primary hepatoma was in 6th decade and ratio between male and female was 2.8:1. 2. Mean values of ascitic γ-GTP levels was highest In primary hepatoma (60.2±30.4mU/ml) and they were 28.3±14.2mU/ml in stomach cancer, 14.8±11.6mU/ml in liver cirrhosis, 14.0±6.6mU/ml in benign intraabdominal tumor and 10.2±5.2mU/m1 in tuberculous peritonitis in decreasing order (P<0.05). 3. The correlation between ascitic γ-GTP and serum alkaline phosphatase, SGOT, SGPT activity, ascitic total protein amount were absent in primary hepatoma (P>0.05). 4. Mean values of aecitic γ-GTP levels in primary hepatoma with and without 1iver cirrhosis were 61.2±28.2mU/ml and 59.8± 32.1mU/ml, respectively. The difference of both cases was not statistically significant (P:>0.1). 5. Primary hepatoma was divided into three groups by extent of involvement undel peritoneos-copy. Mean values of γ-GTP levels in group A was 98.6±38.7mU/ml, in group B 49.8±4. OmU/ml and in group C 38.0±3.6mU/ml, respectively (P<0.05). It is concluded that significantly increased ascitic γ-GTP levels in primary hepatoma would be useful for the differential diagnosis of ascites and ascitic γ-GTP levels are related to exten of involvement of primary hepatoma.
