The Interplay between Autophagy and Aging

Numerous studies have established a link between autophagy and aging; however, the relationship has not been clearly defined. Aging is a very complex process caused by the accumulation of various factors due to the gradual failure of cellular maintenance. Recent studies have shown that autophagy reduces the stress responses induced by starvation, reactive oxygen species, and the accumulation of intracellular proteins and organelles through cytoprotection, clearance of damaged mitochondria, and lysosomal degradation. Here, we summarize our current understanding of the relationship between autophagy and the aging process.
Molecules and their functions in autophagy

Autophagy is a self-degradation system of cellular components through an autophagosomal-lysosomal pathway. Over the last 15 yr, yeast genetic screens led to the identification of a number of genes involved in the autophagic pathway. Most of these autophagy genes are present in higher eukaryotes and regulate autophagy process for cell survival and homeostasis. Significant progress has recently been made to better understand the molecular mechanisms of the autophagy machinery. Especially, autophagy process, including the regulation of autophagy induction through mTOR and the nucleation and elongation in autophagosome formation through class III phosphatidylinositol 3-kinase complex and ubiquitin-like conjugation systems, became evident. While many unanswered questions remain to be answered, here, we summarize the recent process of autophagy with emphasis on molecules and their protein complexes along with advanced molecular mechanisms that regulate the autophagy machinery.
매운맛 선호도가 사람의 말초혈액에서 분리한 면역세포 활성에 미치는 영향

고추의 매운맛 성분 capsaicin의 생체에 미치는 영향에 관한 기초 정보을 확보하기 위해, 사람을 대상으로 생체방어기구의 활성화 측면에서 검토하였다. 매운맛을 좋아하는 사람과 싫어하는 사람을 대상으로, 매운맛에 대한 선호도의 차이가 면역세포의 활성 정도에 어떤 영향을 미칠수 있는지를 조사하였다. 먼저, 각 조사대상자의 혈액으로부터 혈청을 분리하여 IgG, IgM, IgA 함량을 ELISA로 측정하였다. 각 조사대상자의 말초혈액으로부터 B 임파구 및 T 임파구를 분리하여, 각 임파구 특이적 유사분열 촉진 물질(mitogen)인 phytohemagglutinin, Concanavalin A, Staphylococcus typhimurium 자극에 따른 임파구의 증식능을 측정하였다. NK세포 활성은 (15)^Cr으로 표지한 종양세포에 대한 세포독성으로 나타내었다. 혈청 IgM 수준은 매운맛 선호 정도에 따른 차이를 보이지 않았으나. IgG 및 IgA 수준은 매운맛 선호도가 높은 그룹에서 약간 높은 경향을 보였다. 임파구의 증식정도는 mitogen 처리를 하지 않았을 때의 증식능과 mitogen 자극 후의 증식능을 비교한 경우, 매운맛 선호도가 높은 군의 mitogen-induced 증식능은 B 임파구 및 T 임파구 모두 비선호군의 증식능보다 약 2~3배 정도 높은 것으로 나타났다. 한편, NK세포 활성은 매운맛 비선호군이 선호군보다 높았다. 이들 결과들은 매운맛을 선호하는 식습관을 지닌 사람과 그렇지 않은 사람에게 있어서 생체내로 침입하는 항원성 물질에 대한 방어작용을 위한 생체내 면역반응에 차이가 있을 수 있음을 시사하는 것으로 사료된다. The effect of hot taste preference on selected immune responses was investigated in human peripheral immunocompetent cells. Human lymphocytes and natural killer(NK) cells were prepared at a concentration of 2×10^6 cells/ml in RPMI-1640 containing 10% fetal bovine serum. Lymphocytes proliferation was determined with the [³H]-thymidine pulse for 18hrs after concanavalin A, phytohemagglutinin, Salmonella typhimurium mitogen, or media alone. NK cell activity was measured by cytolysis of (51)^Cr-labeled target cells K562. Serum antibodies levels such as IgM, IgG, IgA were also measured by ELISA method. There was no difference of serum IgM level among the groups, but IgG and IgA levels were greater in the group with hot taste preference than those of the group without hot taste preference. In lymphocytes of the group with hot taste preference there was a greater mitogen-induced lymphocyte proliferative responses compared to the group without hot taste preference. In addition, NK cell activity in group with hot taste preference was lower than that of the group without hot taste preference. These results suggest that the eating habit of spicy food containing hot components may affect immune status by modulating selective immunocompetent cells function.
