        인삼 비당부와 땅빈대의 뇌암세포 독성작용

        The effects of acid hydrolysis product of Panax ginseng and MeOH extract of Euphorbia humifusa on the growth of human brain tumor cells were evaluated using U-373 MG human astrocytoma and SK-N-MC human neuroblastoma cells as model cellular systems. These plant extracts induced cytotoxicity in both cells in a dose-dependent manner. These cytotoxic effects were significantly inhibited by GSH, an antioxidant, in both cells. BAPTA/AM, an intracellular $Ca^{2+}$ chelator, significantly blocked the cytotoxic effects of these extracts in U-373 cells, but enhanced these effects in SK-N-MC cells. These results suggest that the plant extracts may be a valuable choice for the studies on the treatment of human brain tumors.

        Sesamol 유도체의 합성 및 생물활성

        Sesamol, one of constituents isolated from sesame seed (Sesamum indicum L.) has been known to have natural antioxidant property. Nicotinic acid and clofibric acid have been used for treatment of hyperlipidemia. Sesamol derivatives prepared by conjugation with nicotinic acid (SJ-301) and clofibric acid (SJ-302) were expected to have various biological activity caused by synergistic effect of sesamol. Thus, this study were carried out to investigate the synthesis of sesamol derivatives from seasamol and antihyperlipidemia drugs and their biological activities. As a result, SJ-301 showed a stronger antihyperlipidemia activity in vivo and inhibitory effect on $TNF-{\alpha}$ release in vitro compared to the original agents such as sesamol, nicotinic acid and clofibric acid.

        열처리 및 발효과정이 인삼 및 산양삼의 ginsenoside 함량에 미치는 영향

        Objectives: The aim of this experiment is to provide an objective differentiation of cultivated ginseng, mountain ginseng through component analysis, and to know the change of gin senoside components in the process of heating and fermentation Methods: Comparative analyses of ginsenoside $Rb_1$, $Rb_2$, Rc, Rd, Re, Rf, $Rg_1$, $Rg_3$, $Rh_1$, and $Rh_2$, from the cultivated ginseng 4 and 6 years, and mountain cultivated ginseng were conducted using HPLC (High Performance Liquid Chromatography, hereafter HPLC). And the same analyses were conducted in the process of heating and fermentation using mixed Lactobacillus rhamnosus, Lactobacillus plantarum, Bifidobacterium lactis for 7 days. Results: The change of ginsenosides to the process of red ginseng and fermentation, cultivated ginseng and mountain cultivated ginseng were showed another results. Mountain ginseng showed a lot of change compared with cultivated ginsengs. In the 7 days of fermentation, mountain ginseng showed that ginsenoside $Rg_1$, $Rb_1$, $Rb_2$, Rc, and Rd were decreased and increased ginsenoside Re, Rf, $Rg_3$ and $Rh_1$ were increased compared with cultivated ginseng Conclusions: It seemed that ginsenosides of mountain cultivated ginseng was better resolved than cultivated ginseng because the difference of structure or distribution of ginsenosides in the condition of fermentation.

        지구자의 Glutathione S-transferase 활성과 알코올 분해효과

        For the determination of glutathione S-transferase activity, a new method was established by using HPLC system. Moreover, amount of enzyme for a optimum reaction was determined by a comparative study with a variety concentration of enzyme. Using a established method, activity of glutathione S-transferase that is alcohol metabolizing enzyme was investigated on the fruit of Hovenia dulcis Thunb. As the result of experiment, EtOH and $H_2O$ extracts of the fruit of Hovenia dulcis Thunb showed visible a synergistic effect of glutathione S-transferase activity. On a continuous experiment, EtOH and $H_2O$ extracts of the fruit of Hovenia dulcis Thunb showed alcohol decomposition activity on the in vivo test using rat. These results suggest that the fruit of Hovenia dulcis Thunb may be useful in the prevention of hangover.

        인삼잎의 Dammarane계 사포닌으로부터 Ginsenoside-Rh2의 제조

        The genuine aglycone, 20(S)-protopanaxadiol, obtained from the leaves of Panax ginseng as a result of direct alkaline treatment was isolated and characterized by spectroscopic evidences. The study on the yield of genuine aglycone which is produced from the treatment of some kinds of alkali was carried out. Ginsenoside-Rh2 was synthesized by conjugation of 2,3,4,6-tetra-0-acetyl-alpha-D-glucopyranosyl bromide to 20(S)-protopanaxadiol in the presence of silver carbonate and cadmium cabonate. The preparation of ginsenoside-Rh2 by this method is a new one which the yield of this saponin can be improved in the mild condition.

        대두와 된장의 지용성 성분 비교

        Soybean and beanpaste have been used as one of the important food in Korea. Beanpaste is made from soybean(Seed of Glycine max MERIL, Leguminosae) by traditional fermentation. It was expected that the secondary metabolites might be formed by the action of some microorganisms or enzymes on the constituents primarily contained in soybean during manufacturing or storage of beanpaste. A systematic investigation on the chemical constituents was carried out. As a result, sterol glycoside named daucosterin, lipo-daucosterin have been isolated from the lipid fraction of soybean and their structures elucidated on the basis of chemical and physicochemical evidences. A ratio of composition of the lipid constituents in soybean and beanpaste were estimated.

        Melibiose로부터 M-5 중간체 galactosylglycerol의 합성

        The galactolipid M-5, which showed anti-inflammatory activity is glycoglycerolipid isolated from the Okinawa marine sponge Phyllospongia foliascens. Glycolipids have been synthesized by various methods, especially it were generally known that synthetic method of M-5 analogue and synthetic method of various glycolipids by glycosidation after synthesis of glycerolipid part. The others, it was not suggested that synthetic method via glycosylglycerol obtained by degradation from diglycoside. This study was carried out to investigate the synthesis of galactosylglycerol melibiose as M-5 intermediate. Synthesis of galactosylglycerol was accomplished by selective protection of hydroxy group of sugar and diol cleavage by Pb(OAC)4. As a result, galactosylglycerol was synthesized by 8 steps pathway and their structures were elucidated by analysis instrument.

        괴화, 어성초, 익모초에서 효소 분해에 의한 항산화 활성 변화 연구

        Objectives: The enzymatic hydrolysis is one of the processing methods that improve its effectiveness on medicinal herbs. In this research, changes in ingredients and activity by enzymatic hydrolysis were studied. Methods: For this study, a carbohydrate hydrolase such as viscozyme, which converts glycosides to aglycone, was applied to induce constituent changes in Sophora japonica Linne, Houttuynia cordata Thunberg and Leonurus japonicus Houttuyn. Changes in antioxidant activity were measured using the 1,1-diphenyl-2-picrylhydrazl (DPPH) method, and changes in ingredients were analyzed by high performance liquid chromatography. Results: As a result of enzymatic hydrolysis, the content of quercetin was increased from 1.26 mg/g to 29.66 mg/g in Sophora japonica Linne, from 0 mg/g to 0.66 mg/g in Houttuynia cordata Thunberg and from 0.43 mg/g to 0.71 mg/g in Leonurus japonicus Houttuyn. As a result of the antioxidant experimentation, the IC50 of Sophora japonica Linne decreased from 5 ug/ml (MeOH extract) and 9.1 ug/ml (EtOAc fraction) to 3.0 ug/ml, Houttuynia cordata Thunberg decreased from 15.6 ug/ml (MeOH extract) and 13.6 ug/ml (EtOAc fraction) to 11.2 ug/ml, and Leonurus japonicus Houttuyn decreased from 14.4 ug/ml (MeOH extract) and 12.6 ug/ml (EtOAc fraction) to 10.2 ug/ml. Conclusion: In conclusion, it was confirmed that glycoside rutin contained in the three medicinal herbs was changed to quercetin which is the aglycone, by the enzymatic hydrolysis using viscozyme. In terms of antioxidant activity, Sophora japonica Linne showed a significant antioxidant activity value that closes to the control group butylated hydroxyanisole. Houttuynia cordata Thunberg and Leonurus japonicus Houttuyn showed a minor increase in antioxidant activity.

        비타민 C와 산화 게르마늄 및 셀레늄이 새싹에 미치는 영향

        In this study, for development of functional sprouts, we have investigated the change of antioxidant activity and quantity of vitamin C, germanium and selenium according to the addition of vitamin C, germanium oxide and selenium oxide on the during cultivation of various sprouts. We were cultivated the sprouts using water solution added vitamin C, germanium oxide and selenium oxide on the system of cultivation instrument. Analysis of quantity of vitamin C on control group and treatment group were conducted by HPLC. Quantity of germanium and selenium were analyzed by ICP-MS(Inductively Coupled Plasma-Mass Spectrometer), and antioxidant activities were checked by DPPH method. As a result, quantity of vitamin C, germanium and selenium on the treatment groups have increased tendency compared to the control group. And antioxidant activity of treatment groups showed increasing tendency compared to the control group.

        Nut류의 항산화 및 항균효과

        It has been known that diseases related with aging or cancer result, at least in part, from free radicals, and antioxidants may reduce or prevent the abnormalities associated with free radical formation via its scavenging action. Thus, this study was carried out to investigate the antioxidative and antimicrobial activities of nut species for the purpose of developing antioxidant from natural products. Antioxidant activities of four kinds of nuts such as chestnut, groundnut, walnut and acorn were examined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The extracts from chestnut hull and acorn barnyard millet showed strong antioxidative activities among 10 samples tested. Because of their highest antioxidative activity among 10 samples, radical scavenging effects of 4 different extract compartments (Hexane, EtOAc, BuOH and $H_2O$ extracts) from chestnut hull and acorn barnyard millet, were further examined by DPPH method. EtOAc and BuOH extracts exhibited antioxidative activities similar to those of natural, tocopherol or synthetic antioxidants, BHA. These findings demonstrates that major fraction of the antioxidative activity of chestnut hull or acorn barnyard millet was the EtOAc and BuOH extract compartments. However, antimicrobial activities against food-related bacteria and yeasts was relatively weak.
