        연근 열수추출물 투여가 마우스의 비장세포와 사이토카인의 분비량에 미치는 영향

        Nelumbo nucifera Gaertn has been usedas a traditional remedy and food source in South Korea. It promotes gastrointestinal function and controls blood pressures. Nelumbo nucifera Gaertn water extracts supplement at 5, 10, 50, 100, 250, 500, $1,000{\mu}g/mL$ after a 48 h pre-treatment with the mitogen (ConA or LPS) increased the mouse splenocytes proliferation. Water extract supplement also increased the cytokine production ($IL-1{\beta}$, $TNF-{\alpha}$ and $IFN-{\gamma}$), measured by a cytokine ELISA kit. For the result of in vitro study, the proliferation of splenocytes and cytokine production activated by peritoneal macrophages increased when water extracts were supplemented in the range of $50{\sim}500{\mu}L/mL$ concentration. Specifically, the levels of the splenocytes proliferation, $IL-1{\beta}$, $TNF-{\alpha}$ and $IFN-{\gamma}$ were the highest at $250{\mu}L/mL$ concentration. This in vitro study suggestedthat supplementation with Nelumbo nucifera Gaertn water extracts may enhance the immune function by regulating the splenocyte proliferation and enhancing the cytokine production activating macrophage in vitro.

        가시오가피 물 추출물에 의한 마우스 비장세포 및 대식세포 활성의 항진효과

        In vitro 실험을 통한 가시오가피 물 추출물 첨가가 마우스의 면역세포 증식에 미치는 영향에 대한 연구 결과, 음의 대조군에 비해 가시오가피 물 추출물을 첨가한 모든 농도에서 비장세포 증식능이 증가하였으며, 특히 고농도인 $500{\sim}1,000{\mu}g/mL$ 농도에서 유의적으로 증가하였다. 반면, 복강 대식세포로부터 유도된 사이토카인 생성의 경우, IL-2, IFN-${\gamma}$, TNF-${\alpha}$ 사이토카인 생성량을 측정한 결과, TNF-${\alpha}$ 사이토카인은 가시오가피 물 추출물 $50{\mu}g/mL$ 농도와 $250{\sim}1,000{\mu}g/mL$ 농도에서 대조군보다 유의적으로 높은 분비량을 보인 반면, IFN-${\gamma}$에서는 변화를 보이지 않았다. IL-2의 경우, $100{\sim}500{\mu}g/mL$에서 유의적으로 증가하는 경향을 보여주었다. 이상의 결과에 의하면 가시오가피 물 추출물은 마우스 비장 세포를 증식시키고, 사이토카인 분비량에도 영향을 줄 것으로 보이며, 면역 기관의 주요 기능을 증진시킬 가능성이 있을 것으로 사료된다. 따라서 가시오가피 물 추출물이 면역 증진 기능성식품 개발의 소재로 활용될 가능성이 있을 것으로 기대된다. Acanthopanax senticosus is an herb that has been used as a traditional remedy and medicine source. Its anti-inflammatory and, anti-oxidative effects have been reported in previous studies. This study aimed to investigate the effect of Acanthopanax senticosus water extracts on mouse macrophage cell in vitro. Mouse splenocyte proliferation increased after application of Acanthopanax senticosus water extract supplement of 5, 10, 50, 100, 250, 500, $1,000{\mu}g/mL$ after 48 h pre-treatment with a mitogen (ConA or LPS). The production of cytokines secreted by LPS and non LPS stimulated macrophages was detected by ELISA assay using a cytokine kit. Cytokine production (IL-2, IFN-${\gamma}$, and TNF-${\alpha}$) increased after water extract supplementation. The result of this in vitro study, showed that splenocyte proliferation and cytokine production by activated peritoneal macrophages were increased after Acanthopanax senticosus water extract in the range of $500{\sim}1,000{\mu}L/mL$. Thus, it is suggested that supplementation with Acanthopanax senticosus water extracts may enhance immune function by regulating splenocyte proliferation and enhancing cytokine production by activated macrophage.

        호박고구마 물 추출물 투여가 마우스의 비장세포와 사이토카인의 분비량에 미치는 영향

        Pumpkin sweet potato (Ipomoea batas L.) has been known as a traditional remedy and food source, not only in South Korea but worldwide. It is rich in fiber, potassium, vitamin C, and other minerals and vitamins, making it a nutritional food loved by many. showed that pumpkin sweet potato had antioxidant biological effects. The in vitro study showed that both splenocytes and cytokine production byactivated peritoneal macrophages increased when water extracts were supplemented at 100 and 250 μL/mL. Notably, the production of IL-1β, TNF-α, and IFN-γ by splenocytes was significantly increased at 100 μL/mL. The results suggest that supplementation with pumpkin sweet potato (Ipomoea batas L.) water extract may enhance immune function by stimulating splenocyte proliferation and improving cytokine production, activating macrophages in vitro.

        느타리버섯 물 추출물 투여에 의한 마우스 비장세포 및 사이토카인 활성효과

        In vitro 실험을 통한 느타리버섯 물 추출물 첨가가 마우스의 면역세포 증식에 미치는 영향에 대한 연구 결과, 음의 대조군에 비해 느타리 물 추출물을 첨가한 모든 농도에서 비장세포 증식능이 증가하였으며, 특히 $100{\sim}1,000{\mu}g/mL$ 농도에서 유의적으로 증가하였다. 반면, 사이토카인 생성의 경우, IL-2, IFN-${\gamma}$, TNF-${\alpha}$ 사이토카인 생성량을 측정한 결과, 느타리버섯 물 추출물 $50{\sim}500{\mu}g/mL$ 농도에서 생성된 IL-2, IFN-${\gamma}$, TNF-${\alpha}$ 사이토카인은 대조군보다 높은 생성량을 보였다. IFN-${\gamma}$, TNF-${\alpha}$ 사이토카인 모두 50, 100, 250, $500{\mu}g/mL$의 농도에서 높은 생성량을 나타내었다. IL-2의 경우, 생성량이 유의적인 차이는 보이지 않았지만, $50{\mu}g/mL$ 이상에서 증가하는 경향을 보여주었다. 이상의 결과에 의하면 느타리버섯 물 추출물은 마우스 비장 세포를 증식시키고, 사이토카인 생성량에도 영향을 주어, 면역 기관의 주요 기능을 증진시킬 가능성이 있을 것으로 사료되며, 느타리버섯이 면역 증진 기능성 식품 개발의 소재로 활용될 가능성이 있을 것으로 기대된다.         새송이버섯, 팽이버섯 열수추출물의 항산화 및 항암 활성

        Various studies on the effect of mushroom, a member of phytonutrients, on inflammatory diseases have been reporter over the years, Among various species mushrooms, King oyster mushroom (Pleurotus eryngii) and winter mushroom (Flammulina velutipes) are popular dietary ingredients for Asian cuisine. The extracts from all the three mushrooms were used to determine the antioxidative effect and the cytotoxicity. Result analysis were repeated more than three times to get an average${\pm}$standard deviation, and statistical significance were confirmed by SPSS. As a result, total phenol content of the king oyster mushroom and the winter mushroom were $19.66{\pm}0.10mg/g$ and $22.08{\pm}1.10mg/g$, respectively. Also, the total flavonoid content was $15.21{\pm}1.31mg/g$ for the king oyster mushroom and $20.50{\pm}4.52mg/g$ for winter mushroom. The results in winter mushroom showed higher values in total phenol and flavonoid content than in the king oyster mushroom. All samples of extracts showed free radical DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activity with their 10 mg/mL concentration. These results indicate that the extract of the king oyster mushroom (Pleurotus eryngii) and the winter mushroom (Flammulina velutipes) may be possible phytonutrients with the anti-inflammatory and the anti-cancer effect.

        도라지 물 추출물의 경구 투여가 마우스 면역 세포 활성에 미치는 효과

        Platycodon grandiflorum have been used as a traditional remedy and food source. This study was performed to investigate the immunomodulating effects of Platycodon grandiflorum in mouse, using ex vivo experiments. Six to seven-week old mice were fed ad libitum on a chow diet, and water extract of Platycodon grandiflorum was orally administrated at two different concentractions (50 and 500 mg/kg B.W./day) every other day for four weeks. In ex vivo experiments, the highest proliferation of splenocytes and levels of cytokine (IL-$1{\beta}$, IL-6, TNF-${\alpha}$) production were observed in 500 mg/kg BW/day supplementation group for all three cytokines stimulated by LPS. In conclusion, this study suggests that Platycodon grandiflorum extracts may enhance the immune function by regulating the splenocytes proliferation and cytokine production capacity by activating macrophages in mice.

        아로니아 부산물 분말 첨가 쌀 영양바의 품질 특성

        The quality characteristics, total phenolic content, anthocyanin content, and DPPH radical scavenging activity of rice nutritional bar with added powder of aronia byproducts were investigated in this study. The rice nutritional bar was prepared by adding aronia byproducts powder at concentrations of 0%, 2%, 4%, and 6%. The pH of the rice nutritional bar batter was lowered for the control and 2% aronia byproducts powder was added. The moisture content, baking loss rate, leavening rate and specific volume of the rice nutritional bar decreased with as the aronia byproducts powder content increased. In relation to measurements of the colors of the rice nutritional bar crust and crumbs, as the content of aronia byproducts powder increased, the L and b values decreased while the a value increased. From texture analysis, the hardness and chewiness increased according to the level of added aronia byproducts powder, but the springiness and cohesiveness decreased. The total phenolic content, anthocyanin content, and DPPH radical scavenging activity showed the highest values in the rice bar with 6% aronia byproducts powder added. According to a sensory evaluation, overall preference was highest for the rice bar with 4% added aronia byproducts powder, whereas the color and taste was rated higher for the rice bar added with 4% and 6% aronia byproducts powder.

        옥수수 추출물의 경구 투여가 사이토카인 IL-2 생성과 IFN-γ와 IL-10 Ratio에 미치는 영향

        면역 증진능을 갖는 천연식품소재 발굴 연구의 일환으로서 옥수수의 면역증강 효과를 검색하고자 하였다. 선행 연구의 옥수수의 시험관 내 실험으로 비장세포 증식능과 사이토 카인 생성능을 검색한 결과를 바탕으로 생체 외(Ex vivo)를 통해 옥수수 열수 추출물의 면역작용을 사이토카인 IL-2의 분비량과 1L-10과 IFN-γ의 ratio를 통해 알아보았다. 옥수수 열수 추출물을 4주간 격일로 체중 ㎏당 50 ㎎과 500 ㎎의 두 농도로 각각 마우스에 경구 투여한 후 활성 복강 대식세포에서 분비하는 사이토카인 IL-2, 1L-10과 IFN-γ의 분비능을 측정하였다. IL-2 의 경우 미토젠을 처리한 군과 처리하지 않은 군 모두 500 ㎎/㎏ 농도에서 유의적으로 높은 수준을 나타내었다. IFN-γ의 경우도 500 ㎎/㎏ 농도에서 유의적으로 높은 수준의 생성능을 보여 주었다. 또한 이 두 사이토카인간의 비율이 일정한 범위 내에 있어, 이는 옥수수 열수추출물 투여가 1L-10과 IFN-γ의 분비능을 급격하게 변화시키기보다 서로 일정한 수준으로 균형을 이룰 수 있도록 조절하는 작용이 있음을 시사하고 있다. 이상의 결과를 종합해 볼때 옥수수 추출물 경구 투여를 통하여 면역세포 분비량을 유도, 조절함으로써 체내 면역기능을 증강시킬 수 있는 가능성이 있을 것으로 사료되며, 이를 규명하기 위한 후속연구가 활발하게 진행되어야 할 것으로 생각된다. Corn has been used for a long time as a traditional remedy, as well as a food source. We previously reported that in vitro supplementation of corn water extracts enhanced the proliferation of splenocytes, compared to the control group. In this study, we examined the immunomodulative effect of a water extract of corn. Seven to eight weeks old mice(Balb/c) were fed an ad libitum chow diet, and were orally administrated a water extract of corn every other day, for four weeks, at two different concentrations(50 and 500 ㎎/㎏ B.W). Cytokine production (IL-2, IL-10 and IFN-γ) by macrophages stimulated with LPS or not stimulated with LPS was detected by ELISA assay using the cytokine kit. In an ex vivo study, the cytokines IL-2, IL-10 and IFN-γ were detected at 500 ㎎/㎏ b.w, supplementation group with LPS stimulation in all cases. Also, the ratio of IFN-γ to IL-10 was in the range of 0~3 with mitogen stimulation, such as con A and LPS. In conclusion, this study suggests that in mice, corn extracts may enhance immune function by regulating the cytokine production (IL-2, IL-10 and IFN-γ) of the activated macrophages.

        더덕 추출물의 경구 투여가 마우스의 사이토카인 생성과 IFN-γ, IL-10 Ratio에 미치는 영향

        Codonopsis lanceolatae have been used as one of the traditional remedies as well as food source. We previously reported that in vitro supplementation of Codonopsis lanceolatae water extracts enhanced the splenocytes proliferation compared to the control group. This study, the combined immunomodulative effect of water extract Codonopsis lanceolatae was Seven to eight weeks old mice(balb/c) was fed ad libitum on chow diet and water extract of Codonopsis lanceolatae was orally administrated every other day for four weeks at two different concentrations(50 and 500 mg/kg B.W.). The production of cytokine(IL-2, IL-l0 and IFN-γ), secreted by macrophages stimulated with LPS or not, were detected by ELISA assay using the cytokine kit. The result of ex vivo study showed that the IL-2, IL-10 and IFN-γ was detected at 500 mg/kg B.W. supplementation group with LPS stimulation in all cases. Also, ratio of IFN-γ, IL-10 was the range of 3~7 with mitogen stimulation such as Con A and LPS. In conclusion, this study suggests that Codonopsis lanceolatae extracts may enhance the immune function by regulating the cytokine(IL-2, IL-10 and IFN-γ) prodution capacity by activated macrophages in mice.
