Defensins, a family of antimicrobial peptides, are one of the first lines of host defense. Human beta-defensins (hBD) such as hBD-2 and -3 have anti-HIV activity. Previ-ous studies have shown that HIV-1 virion can induce the expression of hBD, although the exact components of HIV-1 virion that are responsible for hBD expression have not yet been elucidated. In this study, we examined the effect of HIV-1 Tat on the expression of hBD in B cells. Stimulation of B cells with HIV-1 Tat protein significantly increased the mRNA and protein levels of hBD-2. HIV-1 Tat also induced the activation of a reporter gene for hBD-2 in a dose-dependent manner in B cells. Pretreatment of B cells with a JNK inhibitor suppressed HIV-1 Tat-induced hBD-2 expression. Pretreatment of B cells with AP-1 inhibitors or NF-B inhibitors led to a decrease in HIV-1 Tat-induced protein and mRNA expression of hBD-2. Taken together, our results indicate that HIV-1 Tat can up-regulate the expression of hBD-2 via JNK-NF-B/AP-1-dependent pathways in human B cells.

The infiltration of monocytes into the CNS represents one of the early steps to inflammatory events in AIDS-related encephalitis and dementia. Increased activity of selected matrix metalloproteinases (MMPs) such as MMP-9 impairs the integrity of blood-brain barrier leading to enhanced monocyte infiltration into the CNS. In this study, we examined the effect of HIV-1 Tat on the expression of MMP-9 in CRT-MG human astroglioma cells. Treatment of CRT-MG cells with HIV-1 Tat protein significantly increased protein levels of MMP-9, as measured by Western blot analysis, zymography and an ELISA. Treatment of CRT-MG cells with HIV-1 Tat protein markedly increased mRNA levels of MMP-9, as analyzed by RT-PCR. Pretreatment of CRT-MG cells with NF-κB inhibitors led to decrease in Tat-induced protein and mRNA expression of MMP-9. Pretreatment of CRT-MG cells with MAPK inhibitors suppressed Tat-induced MMP-9 expression. Furthermore, HIV-1 Tat-induced expression of MMP-9 was significantly inhibited by neutralization of TNF-α, but not IL-1β and IL-6. Taken together, our results indicate that HIV-1 Tat can up-regulate expression of MMP-9 via MAPK-NF-κB-dependent mechanisms as well as Tat-induced TNF-α production in astrocytes.

HIV-1 Tat is considered to be one of key players to facili-istic feature of AIDS-related encephalitis and de-mentia. This study was performed to determine the reg-ulatory function of superoxide dismutase (SOD) on the HIV-1 Tat-induced signaling pathways leading to NF-κB activation, expression of adhesion molecules, and monocyte adhesion in CRT-MG human as-troglioma cells by using cell-permeable SOD. When cell-permeable SOD was added to the culture medium time-dependent manners. Treatment of astrocytes with cell-permeable SOD led to decrease in Tat-in-duced ROS generation as well as NF-κB activation. Cell-permeable SOD inhibited the activation of MAP kinases including ERK, JNK and p38 by HIV-1 Tat. Treatment of CRT-MG cells with cell-permeable SOD significantly inhibited protein and mRNA levels of ICAM-1 and VCAM-1 up-regulated by HIV-1 Tat, as measured by Western blot analysis and RT-PCR. astrocyte by HIV-1 Tat was significantly abrogated by pretreatment with cell-permeable SOD fusion pro-teins. These data indicate that SOD has a regulatory function for HIV-1 Tat-induced NF-κB activation in as-trocytes and suggest that cell-permeable SOD can be used as a feasible therapeutic agent for regulation of ROS-related neurological diseases.

다양한 생리활성 기능을 가진 파래 분말을 첨가하여 제조한 만두피의 품질특성을 측정한 결과는 다음과 같다. 호화개시온도는 파래 분말의 첨가량이 증가될수록 점진적으로 증가하는 경향을 보였다. 95℃에서 점도, 95℃에서 15분간 유지한 후의 점도 및 최고점도는 파래 분말의 첨가량이 증가함에 따라 감소하는 것으로 나타났다. 만두피의 색도는 파래 분말 첨가량이 증가할수록 명도(L값), 적색도(a값)및 황색도(b값)는 대조군이 가장 높게 나타났으며, 각 시료간에 유의하게 감소하였다. 조리특성에서 파래 분말을 첨가한 만두피는 대조군에 비해서 중량의 증가나 부피의 증가, 수분흡수율 등이 모두 유의적으로 높게 나타났다. 만두피의 조직감은 파래 분말 첨가량이 증가될수록 경도, 탄력성 및 씹힘성은 높아졌으며, 부착성과 응집성은 감소하였다. 만두피의 관능검사 결과 전체적인 기호도는 파래 분말 5% 첨가군이 가장 좋은 것으로 나타났다. 이상의 결과를 종합하여 보면 기능성 만두피 제품 개발을 위해 파래 분말을 첨가할 경우 밀가루 대비 5% 정도 첨가할 때 기호 특성에서 적당한 것으로 사료되며, 기능적인 측면과 영양적인 측면에서 도움을 줄 수 있는 만두피로 사용될 수 있을 것으로 여겨진다.
나트륨 섭취량 추정을 위한 소변 중 Cl<sup>-</sup>의 간이적 측정방법의 유용성 검증

The purpose of this study was to measure concentrations of $K^+$, $Na^+$, $Cl^-$ by ionometer with check salt strip, simple salimeter and Ion-selective electrode (ISE) and compare the results of each mensuration; furthermore, the possibility of inferring the $Na^+$ concentration from $Cl^-$ concentration of urine and the impact of $K^+$ on the concentration of each ion was examined. The results showed that ISE determined $Na^+$ and $Cl^-$ concentrations in the urine are highly interrelated (R=0.9039); in addition, concentrations of $Cl^-$, measured with strip and ISE from urine are highly interrelated (R=0.9338). The concentration of $Na^+$ in urine, inferred by measuring $Cl^-$ concentration with strip, has a high relationship (R=0.8580) with the concentration of $Na^+$ in urine, measured by ISE. The results of our study will increase awareness of $Na^+$ intake and the utility of check salt strip, as well as the possibility of inferred $Na^+$ concentration from measures of $Cl^-$ concentration as a screening test for reducing sodium intake.

One of characteristic features of AIDS-related ence-phalitis and deme ntia is the infiltration of monocytes into the CNS. HIV-1 Tat was demonstrated to facilitate monocyte entry into the examined the effect of HIV-1 Tat on the expression of adhesion molecules, genera tion of reactive oxygen species (ROS) and NF-κB activation in CRT-MG human astroglioma cells. Treatment of CRT-MG cells with HIV-1 Tat protein significantly increased protein and mRNA levels of ICAM-1 and VCAM-1, as mea-sured by Western blot analysis and RT-PCR, in-dicating that Tat increases these protein levels at an mRNA level. In addition, Tat induced the activation of NF-κB in astrocytes. Treatment of CRT-MG with NF-κB inhibitors led to decrease in Tat-induced protein and mRNA expression of ICAM-1 and VCAM- 1. Furthermore, HIV-1 Tat protein increased ROS generation. Inhibition of Tat-induced ROS genera-N-acetyl cysteine, vitamin C and diphenyl iodonium suppressed Tat-induced NF-κB activation, ICAM-1 and VCAM-1 expression, and monocyte adhesion in CRT-MG. These data indicate that HIV-1 Tat can modulate monocyte adhesiveness by in-creasing expression of adhesion molecules such as ICAM-1 and VCAM-1 via ROS- and NF-κB-dependent mechanisms in astrocytes.

Oxidative stress plays a pivotal role in uncontrolled neuro-inflammation leading to many neurological diseases in-cluding Alzheimer’s. One of the major antioxidant enzymes known to prevent deleterious effects due to oxidative stress is Cu,Zn-superoxide dismutase (SOD). In this study, we examined the regulatory function of SOD on the LPS-induced signaling pathways leading to NF-kappaB activa-tion, expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), in BV-2 cells using cell-permeable SOD. Treatment of BV-2 cells with cell-permeable SOD led to a decrease in LPS-induced reactive oxygen species (ROS) generation and significantly inhib-ited protein and mRNA levels of iNOS and COX-2 up-regulated by LPS. Production of NO and PGE2 in LPS stimulated BV-2 cells was significantly abrogated by pre-treatment with a cell-permeable SOD fusion protein. Fur-thermore, cell-permeable SOD inhibited LPS-induced NF-kappaB DNA-binding activity and activation of MAP kinases including ERK, JNK, and p38 in BV-2 cells. These data indicate that SOD has a regulatory function for LPS-induced NF-kappaB activation leading to expression of iNOS and COX-2 in BV-2 cells and suggest that cell-permeable SOD is a feasible therapeutic agent for regula-tion of ROS-related neurological diseases.

Inducible nitric oxide synthase (iNOS) and cyclooxygenase 2 (COX-2) have been known to be involved in various pathophysiological processes such as inflammation. This study was performed to determine the regulatory function of superoxide dismutase (SOD) on the LPS-induced expression of iNOS, and COX-2 in RAW 264.7 cells. When a cell-permeable SOD, Tat- SOD, was added to the culture medium of RAW 264.7 cells, it rapidly entered the cells in a dose-dependent manner. Treatment of RAW 264.7 cells with Tat-SOD led to decrease in LPS-induced ROS generation. Pretreatment with Tat-SOD significantly inhibited LPS-induced expression of iNOS and NO production but had no effect on the expression of COX-2 and PGE2 production in RAW 264.7 cells. Tat-SOD inhibited LPS-induced NF-κB DNA binding activity, IκBα degradation and activation of MAP kinases. These data suggest that SOD differentially regulate expression of iNOS and COX-2 in LPS-stimulated RAW 264.7 cells.
